Description
リウマチ以外にも,肺,肝臓などにおいてシノビオリンがコラーゲンの過剰な分泌を促し,結果として繊維化を引き起こすことも明らかとなっている(非特許文献3,4)。また,シノビオリンが抗がん遺伝子p53の分解に関わることから,シノビオリンが発がんを引き起こす可能性も示唆されている(非特許文献5)。
まず,脂肪由来の間葉系幹細胞を,バイオミメティクスシンパシ−ズ社製の間葉系幹細胞用無血清培地(sf−DOT培地)で3日間培養し,培養上清を得た。
本実施例では,脂肪由来,臍帯由来及び羊膜由来の間葉系幹細胞培養上清が,リウマチ患者に由来する滑膜細胞のシノビオリン発現を抑制する活性を測定した。まず,脂肪由来の間葉系幹細胞を,sf−DOT培地で3日間培養し,培養上清を得た。リウマチ患者より採取した滑膜細胞を,6ウェル培養プレ−トに1x105/wellで播種した。
DBA/1マウス(体重は25グラム程度)を用いたコラーゲン誘発関節炎モデルに対するヒト脂肪由来間葉系幹細胞の影響を調べた。DBA/1マウスの尾根部皮内に,アジュバンドと混合したウシII型コラーゲンを投与し,感作を行った。初回コラーゲン感作2週間後に,再度のコラーゲン感作を行い,関節炎を作製した。間葉系幹細胞は,初回のコラーゲン感作を行う2週間前から,1週間に2回の頻度で継続的に9週間にわたって,1x106細胞を背部の皮内に投与した。観察期間中は,後肢の厚みをノギスで測定した。
本実施例では,脂肪由来,臍帯由来及び羊膜由来の間葉系幹細胞培養上清が,一般的に研究に使用される細胞株のシノビオリン発現を抑制する活性を測定した。まず,脂肪由来の間葉系幹細胞を,sf−DOT培地で3日間培養し,培養上清を得た。A549,HEK293およびHeLa細胞を,6ウェル培養プレ−トに1x105/wellで播種した。
Claims (2)
- 間葉系幹細胞又は間葉系幹細胞の培養上清を有効成分として含み,シノビオリン遺伝子の発現又はシノビオリンタンパク質の発現を阻害するシノビオリン発現阻害剤。
- 間葉系幹細胞を培養し,間葉系幹細胞又は間葉系幹細胞の培養上清を得る工程と,
前記工程で得られた間葉系幹細胞又は間葉系幹細胞の培養上清を用いて,シノビオリン遺伝子の発現又はシノビオリンタンパク質の発現を阻害するシノビオリン発現阻害剤を調整する工程と,
を含む,シノビオリン発現阻害剤の製造方法。
