Yはミトコンドリアタンパク質上の官能基と共有結合可能な反応性官能基を表す。

蛍光発色団であるペリレンイミドと、ミトコンドリアに特異的に局在化するトリフェニルホスホニウム基を連結するリンカー(L)としては、ペリレンイミド基の蛍光発光特性(発光波長、発光強度、発光強度のpH依存性、励起波長等)、ミトコンドリアへの局在性等に影響を与えない限りにおいて、任意の原子団を制限なく用いることができる。リンカーLの具体例としては、C−C結合間又は側鎖に、酸素原子、窒素原子、硫黄原子、エステル、アミド、ウレタン、尿素、シクロアルキレン基、アリール基、ヘテロアリール基等を有していてもよく、分岐を有していてもよいアルキレン基が挙げられる。
トリフェニルホスホニウム基の対イオンである陰イオンXとしては、ペリレンイミド基の蛍光発光特性(発光波長、発光強度、発光強度のpH依存性、励起波長等)、ミトコンドリアへの局在性、ミトコンドリアタンパク質との反応性等に影響を与えず、細胞毒性等を示さない限りにおいて、薬学的に許容される任意の有機又は無機陰イオンを特に制限なく用いることができる。陰イオンXの具体例としては、塩化物イオン、臭化物イオン等のハロゲン化物イオン、酢酸イオン、プロピオン酸イオン、乳酸イオン、クエン酸イオン、酒石酸イオン等の有機酸イオン、硝酸イオン、硫酸イオン等の無機酸イオン等が挙げられる。
ミトコンドリアタンパク質上の官能基と反応し共有結合を形成する反応性官能基としては、ペリレンイミド基の蛍光発光特性(発光波長、発光強度、発光強度のpH依存性、励起波長等)、ミトコンドリアへの局在性等に影響を与えず、ミトコンドリアに局在化する前に他の細胞内タンパク質等と反応しない程度の適度な反応性を有する原子又は原子団を一旦側に有し、他端側でピペリジン環の窒素原子上に結合した任意の官能基を特に制限なく用いることができる。反応性官能基Yの具体例としては、ω−クロロアルキレン基、ω−ブロモアルキレン基、4−クロロメチルベンジル基等が挙げられる。
実施例1:pH応答性蛍光化合物の合成
下記に示すスキームにしたがって、上述の式9で表されるpH応答性蛍光化合物(以下、「式n(nは1〜9の整数)で表される化合物」を、「化合物n」と略称する場合がある。)の合成を行った。
金属製オートクレーブに、ペリレン−3,4,9,10−テトラカルボン酸二無水物(化合物1)5.5g(14mmol)、2,5−ジ−tert−ブチルアニリン1.57g(7.6mmol)、酢酸亜鉛二和物1.98g(9.0mmol)、イミダゾール14g(205.6mmol)、水12mLを加えスパーテルでよく混合し、200℃で24時間反応させた。反応終了後、内容物を500mLビーカーに移し、全量200mL程度のEtOHで洗いこんだ。濃塩酸1mLを滴下し、室温で1時間撹拌後、EtOHを留去した。残留物にCHCl3を加え、分液ロートに移し、水で3回洗浄した。CHCl3層をNa2SO3で乾燥後濃縮乾固した。精製はシリカゲルカラムを使用し、クロロホルム/酢酸エチル=9/1で行った(T, Dentani; et al., Dyes Pigments., 2007, 72(3), 303-307.)。
5gの化合物2(9mmol)を反応容器に入れ、THF 10mLを加え溶解し、これにt−BuOH 400mLを加えた。フレーク状のKOH 36g(645mmol)を加え、110℃で2時間還流した。400mLの酢酸を滴下し、室温で2時間撹拌した。析出した黒色結晶をろ取し、減圧乾燥した。得られた化合物は有機溶媒への溶解性が悪く、NMR分析ができないことから、そのまま次段階の反応に使用した。
化合物3 1.5g(4.5mmol)、β−アラニンベンジルエステルp−トルエンスルホン酸塩1.86g(5.3mmol)、酢酸亜鉛795mg(3.6mmol)、キノリン150mLを加え、アルゴン雰囲気下、120℃で一晩撹拌した。反応混合物にCHCl3を加え、3N HClで3回洗浄した。CHCl3層をNa2SO3で乾燥後濃縮乾固した。精製はシリカゲルカラムを使用し、クロロホルム/酢酸エチル=9/1で行った。
化合物4 1.0g(2.0mmol)を、THF80mL−EtOH20mLの混合溶液に溶解した。10%Pd/Cをスパーテル大さじ1杯(50mg程度)加え、H2ガス雰囲気下、室温で終夜撹拌した。得られた化合物は有機溶媒への溶解性が悪く、NMR分析ができないことから、そのまま次段階の反応に使用した。
未精製の化合物5をDMF250mLに溶解した。臭化(2−アミノエチル)トリフェニルホスホニウム(Maryanoff, B. E.;et al., J. Am. Chem. Soc. 1985, 107, 217-226に記載の方法に準拠して合成した。)981mg(1.0当量、2.54mmol)、DMT−MM 830mg(1.2当量、3.05mmol)、DIEA 5mLを加え室温で一晩撹拌した。反応の進行を確認後、反応溶液をエバポレーターで留去した。精製はシリカゲルカラムを使用し、クロロホルム/メタノール=9/1で行った。
化合物6 1gに1,2−ジクロロエタン150mLを加え、K2CO3 539mg(3.0当量、3.9mmol)を加え攪拌した。臭素85μL(2.5当量、3.28mmol)を1,2−ジクロロエタン5mLで希釈し、反応溶液に滴下後、100℃で2時間攪拌した。反応の進行はESI−MS分析により確認した。生成するBr体は原料と同じ極性のため、TLCによる反応の進行確認は困難であった。MS解析の結果、原料のピーク(m/z:761 [M+H+])はほぼ検出されず、生成物のピーク(m/z:839 [M+H+])が得られたため溶媒をエバポレーターで留去した。精製はシリカゲルカラムを使用し、クロロホルム/メタノール=9/1で行った。
化合物7 500mgに、2−メトキシエタノール150mL、ピペラジン3.5g(66.6当量、40mmol)を加え、140℃で1晩攪拌した。反応の進行を確認後、溶媒を留去した。精製はシリカゲルカラムを使用し、クロロホルム/メタノール=8/2で行った。
化合物8 300mg、α,α’−ジクロロ−p−キシレン1245mg(20当量、7.1mmol)、K2CO3 50mg(1.0当量、0.35mmol)をアセトニトリル150mLに溶解し、終夜還流した。TLCにより反応の進行を確認した。過剰のα,α’−ジクロロ−p−キシレンを取り除くため、ろ過後反応溶液をエバポレーターで留去した。精製はシリカゲルカラムを使用し、クロロホルム/メタノール=9/1で行った。
化合物9は共役平面が広い影響により極性溶媒中では自己会合が促進し蛍光強度が弱くなる性能を持つことがわかった。つまり、細胞質中に拡散した化合物9は無蛍光であるため、バックグラウンドが低い高感度のイメージングが期待できる。化合物9の蛍光発光のpH依存性を確認するため、pH4.0及び7.4における励起スペクトル(図1中の「Excitation」)及び蛍光スペクトル(図1中の「Fluorescence」)を、50%アセトニトリルを含む緩衝液中で測定した。得られた励起スペクトル及び蛍光スペクトルを図1に示す。図1より明らかなように、化合物9は、ストークスシフトが大きいという特徴を有している。さらに、pH7.4の場合よりも、pH4.0の場合の方が、蛍光強度が高く、極大蛍光波長は30nm短波長側へシフトすることが分かった。
HeLa細胞をμ-slide 8 well(Ibidi)に播種し、37℃、CO2インキュベーターにて一晩培養した。Hanks’ HEPES bufferで希釈した100nmol/Lの化合物9を添加し30分インキュベートした。Hanks’ HEPES bufferで2回洗浄後、1μMのグルカゴンと7.5μMペプスタチンAを含む血清不含Krebs’ Bufferを加え37℃で適切な時間インキュベートし、飢餓誘導を行った後、蛍光顕微鏡で観察した。マイトファジー誘導前と誘導後のHeLa細胞の観察結果を図3に示す。化合物9のリソソームへの取込を確認するため、Hanks’ HEPES bufferで希釈した1μM Lyso Dyeを37℃で30分間インキュベートした。Hanks’ HEPES bufferで2回洗浄後、蛍光顕微鏡で共染色が起こっていることが確認された。
マイトファジーの誘導は、上述の実施例3で用いた飢餓誘導等のバルク的な方法によるものに加え、Parkin遺伝子と酵素PINK1を介した選択的な方法が挙げられる。後者はパーキンソン病に関係していることが分かってきており、不良ミトコンドリアの品質管理との関連が示唆されている(例えば、Derek Narendra, Atsushi Tanaka, Der-Fen Suen and Richard J. Youle, J. Cell Biol., 2008, 183, 795-803.参照)。ミトコンドリア選択的オートファジーの検出を評価するため、Parkin発現HeLa細胞を調製し、化合物9に加え、ミトコンドリア脱分極剤であるカルボニルシアニド−m−クロロフェニルヒドラゾン(CCCP)を添加してインキュベーションすることにより、マイトファジーを誘導した。
μ-slide 8 well (Ibidi)にHeLa細胞を播種し、37℃、CO2インキュベーターにて一晩培養した。HilyMax (同仁化学研究所製) を用いてParkin プラスミドを細胞に導入し、さらに一晩培養した。血清含有培地で洗浄後、Hanks’ HEPES bufferで希釈した100nmol/Lの化合物9の溶液を添加し、30 分インキュベートした。血清含有培地で洗浄後、10μmol/L CCCPを含む培養培地で24時間培養した。蛍光顕微鏡でマイトファジーが起きていることを確認後、1μM Lyso Dyeを37℃で30分間インキュベートした。Hanks’ HEPES bufferで2回洗浄後、蛍光顕微鏡で共染色を確認した。
24ウェルプレートにHeLa細胞を播種し、37℃、CO2インキュベーターにて一晩培養した。HilyMax (同仁化学研究所製) を用いてParkin プラスミドを細胞に導入し、さらに一晩培養した。血清含有培地で洗浄後、Hanks’ HEPES bufferで希釈した100nmol/Lの化合物9の溶液を添加し、30分インキュベートした。血清含有培地で洗浄後、10μmol/L CCCPを含む培養培地で24 時間培養した。PBSで洗浄後、トリプシン及びEDTAで細胞を剥離した。遠心にて細胞を回収後、0.5mLのHBSSで懸濁させFCM(BD FACS cantII)にて解析した。

- 下記の一般式で表されるpH応答性蛍光化合物。
Lはリンカーを表し、
Xは薬学的に許容される陰イオンを表し、
Yはミトコンドリアタンパク質上の官能基と共有結合可能な反応性官能基を表す。 - 前記pH応答性蛍光化合物が下記の式9で表される化合物であることを特徴とする請求項1に記載のpH応答性蛍光化合物。
- 下記の一般式で表されるpH応答性蛍光化合物を含むマイトファジー検出用組成物。
Lはリンカーを表し、
Xは薬学的に許容される陰イオンを表し、
Yはミトコンドリアタンパク質上の官能基と共有結合可能な反応性官能基を表す。 - 前記pH応答性蛍光化合物が下記の式9で表される化合物であることを特徴とする請求項3に記載のマイトファジー検出用組成物。
- 下記の一般式で表されるpH応答性蛍光化合物を細胞内に投与する工程と、
所定時間インキュベート後、細胞からの蛍光発光を測定する工程とを含むことを特徴とする細胞内におけるマイトファジーの検出方法。
