Description
本発明の記載において、「ナノ粒子」、「ナノ粒子集合体」および「粒子」なる語は、区別することなく使用される。
fnは、各元素と結び付く電子の総数の分数であり、
Znは各元素の原子番号を意味する。
天然ポリマーゲルは、主に、i)温度およびpH変化の結果として、およびii)金属イオンが存在する結果として、分子間結合が形成されることで得られる、かくして、ゲルが形成される間は、可逆的な溶液−ゲルの転移が生じる。
本発明に係るナノ粒子および/またはナノ粒子集合体を含む生体適合性ゲルにおいて、各ナノ粒子および/またはナノ粒子集合体は、無機材料、典型的には原子番号が少なくとも25の金属元素を少なくとも1つ含む無機材料を含むか、あるいはそれのみからなり、各ナノ粒子および/またはナノ粒子集合体は生体適合性コーティング剤で覆われるのが有利である。
本発明の生体適合性ゲルは、多くの分野にて、特にヒトまたは獣医学の分野にて使用され得る。本願明細書に記載される本発明に係る生体適合性ゲルは、哺乳動物にて、より好ましくはヒトにおいてさえ、標的組織を描写するのに、典型的には描写して可視化するのに、とりわけ腫瘍床を描写し、可視化するのに、好ましくはCTスキャナーなどのX線画像化装置を用いながら使用することが望ましい。
実施例1:コーティング剤としてヘキサメタリン酸ナトリウムを用いる生体適合性酸化ハフニウム(HfO2)ナノ粒子またはナノ粒子集合体
水酸化テトラメチルアンモニウム(TMAOH)溶液をHfCl4溶液に添加する。TMAOH溶液の添加は、最終懸濁液のpHが7と13の間からなるpHに達するまで行われる。白色沈殿物が得られる。
塩化金をクエン酸ナトリウム水溶液で還元することにより金ナノ粒子を得る。G.Frens、Nature Physical Science 241 (1973) 21より由来のプロトコルを適応させた。
一容量の実施例1の生体適合性HfO2ナノ粒子の水性懸濁液を、一容量のゲル、典型的には濃度が3.5%w/wと4.5%w/wの間にあるポリマー(メチルセルロース)のゲルに添加する。HfO2ナノ粒子の懸濁液とゲルとの間の容量割合を調整し、ゲル内のHfO2ナノ粒子の最終濃度が0.4%(w/w)に達するようにする。そうして得られる調製物を、典型的には、磁気攪拌器で、またはスパテラでゆっくりと混合する。

Claims (6)
- ナノ粒子および/またはナノ粒子集合体を含み、X線画像装置を用いて腫瘍床を可視化するのに使用するためのセルロース、またはメチルセルロース、カルボキシメチルセルロース、ヒドロキシエチルセルロース、およびヒドロキシプロピルセルロースから選択されるその誘導体の生体適合性ゲルであって、腫瘍床の少なくとも40%を描写するために使用される生体適合性ゲルであって、i)そのナノ粒子および/またはナノ粒子集合体が原子番号Zが少なくとも25である少なくとも1つの金属元素を含む無機材料からなり、そのナノ粒子およびナノ粒子集合体の各々がそのナノ粒子およびナノ粒子集合体の表面に陰性電荷を形成するホスフェート試剤からなる生体適合性コーティング剤で覆われており;ii)そのナノ粒子および/またはナノ粒子集合体の濃度が0.15%〜0.5%(w/w)であり;およびiii)ナノ粒子および/またはナノ粒子集合体を含むゲルの2秒−1での見かけ粘度が、20℃〜37℃で測定した場合に、5パスカル秒〜750パスカル秒である、生体適合性ゲル。
- 無機材料が、金属、酸化物、硫化物またはそれらのいずれかの混合物である、請求項1に記載の生体適合性ゲル。
- ナノ粒子またはナノ粒子集合体が、少なくとも1つの標的化剤をさらに含む、請求項1または2に記載の生体適合性ゲル。
- X線画像装置がCTスキャナーである、請求項1〜3のいずれか一項に記載の生体適合性ゲル。
- 腫瘍床が腫瘍を摘出した後にもたらされるキャビティを覆う組織である、請求項1〜4のいずれか一項に記載の生体適合性ゲル。
- 請求項1〜5のいずれか一項に記載のナノ粒子および/またはナノ粒子集合体を含む生体適合性ゲルを製造するためのキットであって、X線画像装置を用いて腫瘍床を可視化するのに使用され、かつ腫瘍床の少なくとも40%を描写するために使用されるキットであって、生体適合性ゲルと、ナノ粒子および/またはナノ粒子集合体とが別個の容器に入れられている、キット。
