Description
実施例1では、低分子リガンドと極性補助基を備え、かつそれらが30Å以下の距離にある化合物を検討した。
比較例1では、低分子リガンドと極性補助基を備え、かつそれらが30Å以上の距離にある化合物を検討した。
比較例2では、低分子リガンドのみ備えた化合物を検討した。
比較例3では、極性補助基のみ備えた化合物を検討した。
比較例4では、スペーサであるε−ポリリジンを検討した(試料5)。
試料1A、2A、3〜5およびプロテインAの抗体に対するアフィニティを、試料をIgGアガロースカラムに通液した際の保持時間から測定した(図3)。それぞれ試料1A、2A、3〜5をPBS(pH7.4、5mM)溶液とし、直径5mmのIgGアガロースカラムに流速0。5mL/minにて通液し、10CV分のPBS(pH7.4、5mM)によって洗浄した(図3)。
試料1〜3、プロテインAを吸着させたIgGアガロースカラムに、PBS(pH7.4、5mM)100%から、PBS(pH7.4、5mM)20%と酢酸緩衝液(pH4.0、0.1mM、0.5mMのNaCl含む)80%の組成にリニアグラジエントにて溶媒を置換しながらカラムを洗浄し、溶出時間を測定した(図4)。
実施例2では、低分子リガンドと極性補助基を備える試料をビーズに固定化し、抗体精製を実施した。
比較例5では、低分子リガンドを備え、極性補助基を備えない試料をビーズに固定化し、抗体精製を実施した。
比較例6では、低分子リガンドと極性補助基を備えるがそれぞれの間の距離が30Å以上となるよう試料をビーズに固定化し、抗体精製を実施した。
実施例3では、種々の低分子リガンドと極性補助基とスペーサの組み合わせに関して、実施例2と同様のカラムを用いた実験にてIgG溶出の容易性を検討した(図5)。

- 低分子リガンドと極性補助基と担体を備え、
前記低分子リガンドは溶液のpH変化に基づきイオン化する官能基を備え、
前記極性補助基は、前記低分子リガンドがイオン化状態で有する電荷と逆の電荷を有し得る状態にあり、
前記低分子リガンドと前記極性補助基の平均距離が10Å以上30Å以下であることを特徴とする吸着材。 - 請求項1に記載の吸着材において、
前記低分子リガンドがpH変化に基づきアニオン化する官能基を備え、
前記極性補助基がカチオン性極性補助基を備えることを特徴とする吸着材。 - 請求項2に記載の吸着材において、
前記pH変化に基づきアニオン化する官能基がフェノール性水酸基、ボロン酸、カルボン酸、ホスホン酸、スルホン酸のいずれかを含み、
前記カチオン性極性補助基が含窒素複素環、アミノ基、アンモニウム基のいずれかを含むことを特徴とする吸着材。 - 請求項1に記載の吸着材において、
前記低分子リガンドがpH変化に基づきカチオン化する官能基を備え、
前記極性補助基がアニオン性極性補助基を備えることを特徴とする吸着材。 - 請求項4に記載の吸着材において、
前記pH変化に基づきカチオン化する官能基が含窒素複素環、アミノ基、アンモニウム基のいずれかを含み、
前記アニオン性極性補助基がフェノール性水酸基、ボロン酸、カルボン酸、ホスホン酸、スルホン酸のいずれかを含むことを特徴とする吸着材。 - 請求項1に記載の吸着材において、
前記低分子リガンドと前記極性補助基がスペーサを介して結合していることを特徴とする吸着材。 - 請求項6に記載の吸着材において、
前記スペーサがポリアルキレングリコール、ポリ(N−イソプロピルアクリルアミド)、ポリ(N,N−ジアルキルアクリルアミド)、ε−ポリリジン、ポリアミノ酸、直鎖ポリエチレンイミン、分岐ポリエチレンイミン、ポリメタクリレート、ポリアクリレート、ポリビニルエーテル、のいずれかを含むことを特徴とする吸着材。 - 請求項1に記載の吸着材において、
前記担体がアガロース、架橋アガロース、疎水化アガロース、セルロース、ポリスチレン、シリカ、酸化鉄、フェライト、のいずれかを含むことを特徴とする吸着材。 - 請求項1に記載の吸着材において、
前記低分子リガンドが抗体を吸着可能であることを特徴とする吸着材。 - 請求項1に記載の吸着材を充填したことを特徴とする精製装置。
- 標的物質を精製する精製方法であって、
吸着材に標的物質を含む第1の溶液を接触させることにより前記標的物質を前記吸着材に吸着させる工程と、
前記標的物質が吸着した前記吸着材に第2の溶液を接触させることにより前記標的物質を前記吸着材から脱着させる工程を含み、
前記吸着材は低分子リガンドと極性補助基と担体を備え、
前記低分子リガンドは溶液のpH変化に基づきイオン化する官能基を備え、
前記極性補助基は、前記低分子リガンドがイオン化状態で有する電荷と逆の電荷を有し得る状態にあり、
前記低分子リガンドと前記極性補助基の平均距離が10Å以上30Å以下であり、
前記第1の溶液のpH値と前記第2の溶液のpH値は異なることを特徴とする精製方法。 - 請求項11に記載の精製方法であって、
前記第1の溶液のpH及び前記第2の溶液のpHは、4以上10以下であることを特徴とする精製方法。 - 請求項11または12に記載の精製方法であって、
前記標的物質が抗体であることを特徴とする精製方法。
