NME1(NM23-H1)は、幹細胞成長を促進し、二量体のみである場合、分化を阻害する。NME6は、略NME1と同じ分子量で、海綿(Suberites domuncula)では、二量体としてそれが存在することが報告される(Perina et al, 2011, "Characterization of Nme6-like gene/protein from marine sponge Suberites domuncula" Naunyn-Schmiedeberg's Arch Pharmacol, 384:451-460)。我々は、NME6を発現し、ELISA分析で、NME6は、MUC1*細胞外ドメインに由来したPSMGFRペプチドに結合することを示した (図20)。この結合実験では、ヒトNME6野生体タンパク質は、あるバリアント(S139G)(NME1をダイマー形成指向にするS120G変異体に類似の変異体(チャン、1986年ら))及び別のバリアント(NME1をダイマー形成指向にする同じ領域が、報告によれば海綿中のNME6は二量体として存在するので、海綿中の該配列をマッチさせるように変異された(S139A、V142D、V143A))と共にテストされた。
そのような修正は、本願発明請求項の範囲内にあることが意図される。以下の実施例は、制限的記載ではなく、本発明の実例を示すものである
ヒト幹細胞が、NME1(H1)およびNME2(H2)に加えてNME6またはNME7を発現するかどうか判断するために、我々は、様々なヒト幹細胞ラインからの溶解物および上澄みについてのウェスタンブロット分析を行った。ヒト胚性幹細胞ラインBGO1v細胞が、a)抗MUC1*単クローン抗体MN-C3で覆われた細胞培養プレート上のみで、ダイマー型のNM23-S120Gで、あるいはb)マウス・フィーダー細胞(MEF)上の4ナノグラム/mLのbFGFで、培養された。培養3日後に、幹細胞は回収され溶解され、次に、NME1、NME6およびNME7の存在を検査のために抗体を使用し、ウェスタンブロットによって分析した。比較のために、同じ分析を、T47D MUC1*陽性の乳癌細胞について、平行に行った。コントロールとして、組み換えNM23-H1野生体(NM23-wt)タンパク質が、ゲルに充填され、3つの異なるNMEを認識する抗体で検査した。NM23-S120G二量体および野生体ヘクサマーの明白な分子量が両方ともモノマーの重量であるかに見えるように、ゲルが変性ゲルであることに注意!
ゲルを検査するために使用される抗体は次のものに対するものである:
NME1:NM23-H1(C-20);
NME6: NM23-H6(L-17)および
NME7: nm23-H7(B9)
(すべてサンタクルーズ・バイオテクノロジー社から購入された)。
各パネルで、レーン1は、抗MUC1*単クローン抗体MN-C3で覆われた細胞培養プレート上でNM23-S120G(ダイマー型)中で培養されたBGO1v幹細胞に相当する;レーン2はMEFでのbFGF中で培養されたBGO1v幹細胞に相当する;レーン3はT47D乳癌細胞に相当する;レーン4は精製された組み換えNM23-H1野生体(NM23-wt)に相当する。
ヒト幹細胞ラインBGO1vおよびHES-3細胞のウェスタンブロット分析は、NME7に特異的な(噂による)NME抗体は、NME7および22-25kDaの明白な分子量をもつ別の種を認識することをしめす(図2)。
本発明者は、ダイマー型、あるいは細胞上でMUC1*受容体を二量化させた型である、NM23が、多分化能幹細胞成長を促進し、分化を阻害し、そして、体細胞を含む、より成熟した細胞の、多分化能を誘導したことを以前に示した。結合実験は、ダイマー型NM23は、細胞上のMUC1*受容体、およびフリーMUC1*細胞外ドメインペプチドに結合することを示した。しかしながら、野生体タンパク質のヘクサマー型あるいはS120G突然変異体のヘクサマー型を使用する実験は、幹細胞を分化させた。細胞の局在性実験は、ヘクサマーが細胞へ結合でき、それが分化を誘導する要素に推測上結合する、核に転位置することを示した。したがって、ヘクサマー及びより高いオーダーのマルチマーを形成する他のNMEファミリーと同様に、低濃度でヘクサマーを形成するNME1(NM23-H1)の発現が、体細胞のようなより成熟した細胞の多分化能を誘導しているインビトロでの幹細胞培養の過程に有害であり、幹細胞集合体を再構成するかあるいは内生の幹細胞集合体をこえる拡張が所望される、幹または先祖細胞を患者に植え付ける過程には有害であることは明らかである。
bFGF中で培養された細胞と比較する、NM23二量体中で培養されたヒト幹細胞のためのプライムドマーカーに対してのナイーブのRT-PCR。
ヒト胚のH9幹細胞は、抗MUC1*単クローン抗体MN-C3で覆われたプラスチックウェアー上に植えつけられ、そして、37回継代で、よりダイマー集合体のみに精製されたNM23-S120G中で培養された。成長している細胞のサンプルは、継代8、12、14、21、26および37で回収された。比較のために、同じスタート貯蔵からの細胞が、マウス・フィーダー細胞(MEF)の層上でbFGFにおいて培養された。細胞は標準の量的PCR技術を使用して分析した。OCT4およびNANOG(多分化能遺伝子)、KLF4、およびKLF2(ナイーブ遺伝子)およびLHX2、OTX2、XISTおよびFOXa2(プライムド遺伝子) の発現レベルは、GAPDHに比較して測定され、MEF上のbFGF中で培養された細胞においてそれらの発現を標準化した。図6で見ることができるように、NM23二量体(NME1-S120G二量体)で培養された幹細胞は、ナイーブ遺伝子のより高いレベル及びプライムド遺伝子のより低いレベルを発現し、それは、該細胞が、bFGF中での幹細胞培養より原始的で、ナイーブ状態であることを示している。図1、パートI.Cに示される実施例1の結果とともに、NME7はプライムド状態ではなくナイーブ状態の特性であり、また、多分化能幹細胞成長をよりサポートできる。これらのデータは、細胞の多分化能のメンテナンスのためのその発現についての、分化の抑制あるいは細胞の多分化能の誘導を導く核酸、及び外生のNME7を加える論理的根拠を提供する。NME7のそのような導入には、分化誘導NME1の抑制を伴う場合がある。
NMEファミリーの抑制
実施例5の実験が、96時間まで継続した。細胞形態学の評価は、付加的警告をもつが、48時間観察をおこなった。96時間後、NMEバリアントノックダウン実験を、他のNMEアイソフォームにおける、1つのNMEアイソフォームを抑制する効果を分析のためと同様に、多分化能遺伝子の発現を分析するためにおこなった。各実験条件からの細胞は、別々にペレット化され、RT-PCRによって分析した。分化遺伝子Foxa2に加えて、多分化能遺伝子Nanog、OCT4およびKlf4及びNMEアイソフォームNME1、NME6およびNME7の発現レベルが測定された。発現レベルは、siRNAのコントロール模擬トランスフェクションに対して標準化した。siRNAは、典型的には、50-75%までターゲット遺伝子を抑制する。ポジティブコントロールとして、Oct4に特異的なsiRNAを使用した。正常な幹細胞培地および成長因子中で培養されたとしても、OCT4siRNAでトランスフェクトされた幹細胞は、予想通り、分化をした。
図14を参照すると、多分化能遺伝子OCT4がノックダウンされる分化コントロールでは、分化遺伝子Foxa2が上昇すること、また3つ全てのNME遺伝子の発現が抑制されることを見られる。同じ分化遺伝子Foxa2も、NME1、NME6あるいはNME7が、ノックダウンされる場合、上昇する。しかしながら、1つのNME遺伝子がノックダウンされる場合、他のNME遺伝子の1つ又は両方における増加があることが観察される。NME1が、ノックダウンされる場合、NME6が増加される。NME6が、ノックダウンされる場合、NME 1およびNME7が増加される。NME7が、ノックダウンされる場合、NME1が増加される。
上述されるように行なわれた別のNMEノックダウン実験では、ヒト胚性幹細胞(BGO1V)が、抗MUC1*抗体(MN-C3)の表面に植えつけられた。特定のNMEタンパク質が、siRNAを使いノックダウンされ、RT-PCRと同様のイメージが、siRNAの96時間ポスト・トランスフェクションでの結果を定量的に示した。成長因子も血清も、siRNAトランスフェクション後に細胞へは加えられなかった。最小の幹細胞培地は、48時間ごとに変更した。図22A、Bは、NME1が抑制され、そして、添加された何らの成長因子も血清もない状態における、96時間後の幹細胞の写真を示す。細胞数および細胞形態は、幹様にみえ、そして、図22C、Dに示される、トランスフェクション試薬が加えられたがナンセンスsiRNAが加えられた、負のコントロール実験からは判別不能に見えることは注目される。
これらの結果は、NME1を抑制することが、幹様の成長に有害な影響がないことを示す。対照的に、図23A、Bは、NME7の抑制が、幹様の成長を阻害し、及び、一般に幹細胞の成長を阻害したことを示す。1個の残存する細胞集合体は、図23Bの中心に細胞のより厚く、より白いクラスタリングに見ることができるように、分化している。多分化能遺伝子OCT4を抑制する(図23C、D)、ポジティブコントロールとの比較によって、NME7の抑制は、重要な多分化能遺伝子OCT4の抑制に匹敵することが確認される。図24A、Bで示されるNME6の抑制は、図24Bで示される図でみることができる分化のエリアのように、適度の程度に、幹様の成長を阻害した。しかしながら、NME6抑制は、掻き混ぜられたsiRNAが加えられたネガティブコントロール細胞に似ているように見える (図24C、D)。
組み換えヒトNME7は、多分化能幹細胞成長を促進し維持する。
400mlのDME/F12/GlutaMAX I(Invitrogen# 10565-018)、100mlのノックアウト血清置換(KO-SR,Invitrogen# 10828-028)、5mlの100xMEMの非本質的アミノ酸溶液(Invitrogen# 11140-050)および0.9ml(0.1mM)のβ- メルカプトエタノール (55mMストック,Invitrogen# 21985-023)。
基礎培地は任意の培地になりえる。好ましい具体化例では、基礎培地は他の成長因子およびサイトカインが存在しない。基礎培地には、NME7-ABかNM23-H1の8nMが加えられ、折りたたまれそして安定した二量体として精製された。培地は48時間ごとに変更され、加速的成長により、回収され、植えつけ後3日目で、継代した。図26は、NM23-H1二量体、あるいはNME7モノマーでの、ヒト幹細胞の培養が、多分化能幹細胞成長に帰着することを示した。NME7およびNM23-H1(NME1)二量体両方は、多分化能に成長し、100%コンフルエントなときでさえ、分化がなかった。写真で見ることができるように、NME7細胞は、NM23-H1二量体中で育てられた細胞より速く成長をした。最初の採取の細胞数は、NME7中での培養がNM23-H1二量体中での培養より、1.4倍多い細胞を生産することを確認した。
ELISA分析は、NME7-ABが、2つのMUC1*細胞外ドメインペプチドに同時に結合することを示す。
ヘクサマー型のNME1は、癌細胞成長とミグレーションを阻害する。
ヘクサマー型NME1は癌細胞ミグレーションを阻害するが、NME1二量体はしない。
ピペット先端あるいは他の道具が、細胞のセクションをこすり取るために使用される。細胞がない各ウェルにおいて、「スラッシュ」又は「クロス」型の領域をもつ成長癌細胞のバンクグラウンドを残して、除去された細胞は、洗い流された。ヘクサマーか二量体のいずれかの純粋な集合体として精製されたNME1が加えられ、さらに16時間あるいは24時間成長させた。

Claims (2)
- 胚性幹細胞、誘導多分化能幹細胞の分化を阻害するインビトロでの方法であって、
生物学的に不活性状態であるNME1(NM23-H1)ヘクサマーを減少させることを含み、及び、MUC1*受容体に結合できる2つの結合部位をもつモノマーであり生物学的にアクティブ状態であるNME7(NM23-H7)が、それを発現する核酸若しくは小分子の導入により、増加されることを含み、並びに、
(i)生物学的に不活性な状態であるNME1(NM23-H1)ヘクサマーが、その発現をダウンレギュレートする核酸若しくは小さな分子の導入により減少する、又は(ii) 生物学的に不活性な状態であるNME1ヘクサマーをダウンレギュレートする第一の核酸と生物学的にアクティブ状態であるNME7をアップレギュレートする第二の核酸を同時に導入することにより、NME1(NM23-H1)ヘクサマーが減少し、かつ、NME7が増加する、方法。 - ダウンレギュレートする核酸が、アンチーセンスDNA、阻害性RNA、siRNA、或いはshRNAであり、及びアップレギュレートする核酸が、コード化DNA、RNA、mRNAあるいはプラスミドである、又は細胞へのエントリーを促進するために修飾されたこれらの核酸の一である請求項1に記載の方法。
