Description
しかしながら、NM23-H1及びH-2が、MUC1*細胞外ドメインのリンガンド誘導による二量化を経る幹及び癌細胞成長を促進するMUC1*成長因子受容体のリガンドであるという本願発明者の開示前には、NM23がどうして、分化に、又は活性であるために、それが二量体であらねばならない又はそれはその標的受容体を二量化するということにより重大に関与しているかは知られていなかった。本発明者は、二量体のNM23が、癌細胞と幹細胞において、MUC1*に結合し二量化させ、そして癌の成長と生存あるいは成長と多能性を促進することを見出した。NM23四量体若しくは六量体は、MUC1*受容体のPSMGFRドメインに結合せず、そして二量体としてのものと逆の機能をもっている。六量体のNM23は、幹細胞の分化を誘導する。
その時まで、一般通念は、MUC1は開裂されるが、縦列繰り返しを含む該開裂部位は、細胞表面へ付着のままの膜貫通断片にやってきて結合し、ヘテロ二量体を形成するというものだった(Ligtenberg et al, 1990, Baruch A et al. (1999)。本発明者は、ガン組織の二重染色法実験として、真実ではないが、開裂型(MUC1*)のみを認識する抗体又は脱落ドメイン(縦列繰り返し若しくはコア)にのみ結合する抗体を使うことで、開裂型を染色する抗体は、縦列繰り返しに結合する抗体とは共存(共局在化)しないということを明らかにした。実際、ガン組織の殆どの膜染色は、完全な縦裂繰り返しドメインをもつMUC1には陰性だったか、あるいは最小陽性であった。しかし、切り取った(クッリプ化)MUC1*型には高度に陽性だった。これらの実験は、MUC1が開裂される場合、大容量の細胞外ドメインが細胞表面から遊離されることを示した(Mahanta,et al, 2008)。
1つの態様では、該阻害のために求められうるNMEファミリータンパク質の機能は次のようなNMEファミリータンパク質の能力でありうる: 幹細胞増殖を促進し及び/又は分化を阻害する; 癌細胞増殖を促進し及び/又は分化を阻害する; MUC1*に結合する; DNAに結合する; 転写調節因子として作用する; 細胞によって分泌される; あるいは、二量体を形成する。特異的に、NMEファミリーメンバーは、好適にはNME7あるいはNME7-ABでありうる。
(i)幹若しくは先祖細胞遺伝子又は遺伝子産物の存在のために患者サンプルを分析すること;
(ii)そして、幹若しくは先祖細胞遺伝子又は遺伝子産物の、同様の発現あるいは発現レベルを共有する患者のグループ化をすること。
(iv) ステップ(iii)で決定されるより活発若しくはより少なく活発な癌をもつ患者の処置に釣り合った治療デザインをすること; そして
(v) ステップ(iv)でのデザインに従う治療法で患者を治療する。このような方法では、患者サンプルは、血液、体液あるいは生検でありうる。また、遺伝子又は遺伝子産物は、NMEファミリータンパク質でありうる。1つの実施態様では、初期の幹細胞状態を示す遺伝子か遺伝子産物は、NME7又はNME6でありうる。
(i) NMEファミリータンパク質あるいはペプチド断片で、抗体ライブラリーか抗体断片ライブラリーかエピトープをスクリーニングすること;
(ii)NMEファミリータンパク質あるいはそれらのペプチド断片への結合性を分析すること;そして
(iii)特異的に結合された抗体あるいは抗体様分子を同定すること。
この方法は、当該技術分野において知られた方法を使い、癌の治療か予防のために、患者への投与のために該同定された抗体か抗体様分子を製造することを含む。NMEファミリータンパク質は、NME7若しくはNME7-AB若しくはNME1でありうる。好適には、ペプチド断片は、SEQ ID NOS:88-140、好ましくは88-133、より好ましくは88-121から選らばれうる。

二量体型のNME1は、幹細胞の成長及び多分化能を促進するだけでなく、ヒト幹細胞を最も初期状態、「ナイーブ」状態と呼ばれる最も分化多能な状態に戻すことを誘導する((Nichols J, Smith A (2009); Hanna et al, 2010; Amit M, et al, 2000; Ludwig TE, et al 2006; Xu C, et al, 2005; Xu RH, et al, 2005; Smagghe et al 2013)。現在まで、それらがまさにその初期胚の内部の集合体に存在するので、これは、ナイーブ状態にヒト幹細胞を維持すると示された唯一の天然因子である。
本発明者は、ここに含まれている実験例と実施例において実証されるように、NME7-ABは、自然状態でプロセスされたNME7と本質的に同じである方法で機能するということを示した。しかしながら、NME7の自然に発生する開裂サイトは、本発明者がNME7-AB N-末端の開始場所とは異なるかもしれない。NME7の阻害剤は、N-末端でDM10を含んでいる天然タンパク質に作用しうるか、又はNME7の分泌型への開裂を阻害するために作用しうる。図6A-Cは、ニッケル・カラムによってつづいて精製されるNME7-ABのFPLCトレース(A)、未精製タンパク質のSDS-PAGEゲル(B)及び最終生産物のFPLCトレース(C)を示す。ナノ粒子分析が行なわれ、モノマーとしてのNME7が、MUC1*細胞外ドメインの2つのPSMGFRペプチド(SEQ ID NO: 6)に同時に結合できることを示した。
ヒスチジン・タグ化PSMGFRペプチドは、NTA-SAM-被覆ナノ粒子上に固定化された。分析は、FPLC及び天然のゲルによってモノマーであることが確認された、DM10のN-末端リーダー配列が欠けて発現される、組み換えNME7-ABが、ナノ粒子に加えられた。NME7の添加は、金のナノ粒子溶液をピンクから青に変わらせ、それはNME7が、同時に、2つの別個のナノ粒子上の2つのペプチドに結合していることを示す。それは、粒子を共に密接に近づかせ、特徴的な色の変異をもたらす(図7)。別のELISA実験は、NME7モノマーが、2つのMUC1*細胞外ドメインペプチドを二量化させることを実証した。最初のPSMGFRペプチドは、BSAにカップル化され、多重ウェルプレート上で固定化された。組み換えNME7-ABが添加された。適切な洗浄ステップの後に、ビオチンで修飾された、第2のPSMGFRペプチドが加えられた。その後、ラベル化ストレプタビジン(streptavidin)が加えられ、それはNME7モノマーが同時に2つのMUC1*細胞外ドメインペプチドを結合することができることを明らかに示した(図8)。これらの結果は、その2つのNDPKドメイン経由のNME7が、幹細胞と癌細胞で、結合しそしてMUC1*を二量化させることを示す。
はっきり見ることができるように、幹細胞は、分化の何らのサインなしで等しく成長するように見える。ナイーブ幹細胞が、コロニーで成長しないが、密度が達成するに従いシートになる単層で寧ろ成長することに留意する。図11は、10継代以上NME7-ABで培養された幹細胞が、標準の多分化能マーカーに陽性に染色するということを確認する免疫細胞化学(ICC)実験の写真を示す。図12は、NME7-ABで培養された幹細胞がナイーブ状態であることを確認するICCの実験の写真を示す。パネル(A)の細胞は、標準的手法であるように、マウス・フィーダー細胞上でFGFにおいて培養された。
染色抗体は、それがヒストン3(H3K27me)上の濃縮トリメチル化リジン27に結合し、赤いドットをもたらした;それは1つのX染色体が不活性(XaXi)で、幹細胞が「プライム化(初回刺激を受けた)」状態に進んだことを示している。パネル(B)の細胞は、それらがNME7-ABで10継代培養されたという点を除いて、(A)において撮影されたのと同じ細胞である。その挿入物で見ることができるように、H3K27me抗体は、クラウド染色パターンを生み、それは両方のX染色体が活性(XaXa)であることを示し、細胞はナイーブ状態に戻ったことを証拠づけた。したがって、本発明者は、NME7が、完全に、幹細胞成長及び多分化能をサポートしさらに、後の、プライム化状態より少ない成熟した状態である、ナイーブか基底状態にそれらを戻すことを実証した。
もっと好ましい実施態様では、治療薬は、NME7のNDPK Bドメインへ結合する抗体である。さらにもっと好ましい実施態様では、治療薬は、NME1には存在しないNME7のAかBドメインの配列に結合する抗体である。さらにもっと好ましい実施態様では、治療薬は、NME7及びMUC1*間の相互作用を阻害する抗体である。最も好ましい実施態様では、治療薬は、当該機能が癌腫瘍の成長あるいは癌様状態への復帰の促進をもたらすNME7の機能を阻害する抗体である。
リフォールデイングプロトコルを変えることによって、本発明者は、本質的に100%ヘクサマーあるいは100%二量体である集合体を安定化させることができた。さらに、本発明者は、二量体、四量体及びヘクサマーの混合物である、NME1-S120Gの集合体を単離した。図16は、天然のゲル上でこれらの様々なマルチマーを示す。図17(A)は、SPR実験において使用されたNME1タンパク質のゲルを開示し(B)、そこで、MUC1*細胞外ドメインのPSMGFRペプチドがチップに固定化され、そして、異なるNME1タンパク質が表面を通過した。結果は、NME1の二量体型が、MUC1*細胞外ドメインに結合する型であることを示す。パネル(C)は、PSMGFRペプチドが、NTA-Ni-SAM被覆ナノ粒子に付着され、そして組み換えNME1二量体若しくはヘクサマーが、ナノ粒子に加えられた実験を示す。
相互作用が起こる場合、金のナノ粒子は青くなり、そうでない場合、ピンクのままである。確認できるように、二量体だけが、ナノ粒子上のMUC1*ペプチドに結合し、そして、2つの異なるナノ粒子の2つのペプチドを二量化させ、、本質的に、該粒子を架橋する。溶液に添加された時、MN-C2抗MUC1*抗体のFabは、NME1二量体とMUC1* PSMGFRペプチド間の相互作用を阻止する。パネル(D)は、競争的に相互作用を阻害するために、フリーのPSMGFRペプチドと二量体、ヘクサマー、又はNME1二量体(NM23)で培養されたヒト幹細胞の写真を示す。
写真で見ることができるように、NME1二量体だけが、多能性幹細胞成長を促進する。自然界において、幹細胞の濃度が、臨界量に達し、そして、NME1のそれらの分泌が、それらがヘクサマーを形成する濃度に達する時、分化は誘導される。図18は、本願実験例に支持されるメカニズムを描く漫画である。それによってNME7とNME1は、NME1がそれ自体を制御し、多分化能を促進するために機能する。
これらの変異は、S139A に加えて、V142DとV143Aである。図19 A、B及びCにおいて示されるELISA分析は、NME6が、MUC1*細胞外ドメインのPSMGFRペプチドに結合することを示す。部分Aでは、NME6-wtは、モノマー、あるいは高分子量マルチマーとして精製される。その表面がPSMGFRMUC1*ペプチドで被覆される、ELISA分析は、MUC1*ペプチドへのNME6モノマーの優先的な結合を示す。部分Bでは、NME6マルチマーは、SDSにおいて、稀釈によって解離される。このELISAは、マルチマーが解離されるに従い、MUC1*ペプチドへの結合が増加することを示す。
この図は、NME6-wtと二量体形成を志向する2つの突然変異体が、MUC1*ペプチドに結合することを示す。図19D-Hのゲルは、以下の発現を示す;NME6-wt(D)、ヒトNME1において二量体形成を増加する変異S120Gに相当するS139G変異を持つNME6 (E)、二量体であることが報告される海綿型にヒト型を模倣させる3つの変異を担持するNME6(F)、及び2つのNME6タンパク質をリンクする単一鎖タンパク質(GH)。パネルIは、MUC1の細胞質尾部に対する抗体を使用するプルダウン分析で、NME6が、癌細胞及び幹細胞で、MUC1に結合することを示す。
したがって、有効な抗癌剤は、MUC1*細胞外ドメインペプチドへのNME6二量体の結合を阻止する抗体、小分子あるいは他の薬剤である。好ましい実施態様では、癌の治療用薬は、NME6のNDPK Aドメインへ結合する抗体である。もっと好ましい実施態様では、治療用抗体は、NME1には存在しないNME6の配列に結合する。
非常に早い段階の幹細胞で、まさにハイ・レベルでNME7が発現されることを実証するために、本発明者は、NME1若しくはNME7(ナイーブ)のいずれかにおいて培養されたか、又はbFGF(プライム化)において培養されたヒト幹細胞についてのウェスタンブロット分析を行ない、その後、NME7の存在を検査した。プライム化状態の胚性幹細胞、それらはナイーブ状態の幹細胞よりもっと分化している、は、NME7のトレース量でのみ発現する。ありのままの比較において、初期の「ナイーブ」状態(「グランド」状態とも呼ぶ)の幹細胞は、NME7の高レベルを発現する(図3B、レーン1“ナイーブ”をレーン2“プライム化”に比較せよ)。 NME7は、初期段階の幹細胞でのその発現に匹敵するレベルに癌細胞中で発現される(図3B、レーン3“癌細胞”をレーン1“ナイーブ幹細胞”に比較せよ)。この理由で、それらの主要な役割が、成年期でではなく初期発生にあるので、NME7とNME6は、治療上著しい副作用なしで無力化することができる。
これらのデータは、遺伝子の転写に影響するために、NME1とNME7が、直接あるいは間接的に機能できることを示す。したがって、本発明の1つの態様では、NME1又はNME7の機能は、DNAへのNME1若しくはNME7の結合を阻害する、小分子でありうる薬剤を添加することにより阻害され、そして、NME1又はNME7の転写機能を阻害する薬剤は、癌患者、あるいは癌になるリスクをもつ患者に投与できる抗癌剤である。
これらの結果は、より小分子量のNME7が、分泌された成長因子型であるということができる。本発明者は、〜33kDaの分子量を持ち、NME7-AB と呼ばれる、NDPK A及びBドメインを含み、だがDM10ドメインもたない、NME7変異体を作った。この組み換えNME7-ABは、他の成長因子あるいはサイトカインを欠く無血清培地で分化多能のヒト幹細胞の成長を完全にサポートすることができる。NME7-ABは、さらに完全に、MUC1*陽性の癌細胞の成長をサポートした。これらの実験は、NME7の分泌型が、成長因子型であること、及びそれがNDPK A及びBドメインを含み、大部分あるいは全てのDM10が欠け、及び〜33kDaの分子量を持つことを示した。
図24は、マウス単クローン抗体(A)あるいは単にN-末端DM10配列を認識する別の単クローン抗体(B)を使い、NME7の存在について検査された、様々の細胞溶解物及び相当する調整済み培地のウェスタンブロットの写真を示す。該細胞の調整済み培地からのサンプルでの、〜33kDa NME7種へのDM10特異的抗体の結合の欠如は、NME7の分泌された型が、N-末端DM10リーダー配列のすべてではないが、大部分が欠けていることを示す。
NME7を発現し分泌する癌細胞は、より分化されておらず及びNME7を分泌しない癌細胞より攻撃的な癌細胞である。したがって、NME7及び分泌されたNME7のレベルの分析は、腫瘍の活動性、計画治療、治療のモニター効果を予測するため、及び臨床試験のための患者集団を階層化するために使用されうる。
それゆえ、NME1、NME6あるいはNME7を検知する抗体は、癌の出現を検知するため若しくは癌の活動性を評価する診断のツールとして使用することができ、ここでハイ・レベルのNME1、NME6あるいはNME7は、腫瘍の活動性及び不十分な結果と関連する。ハイ・レベルのNME7及びNME6は、腫瘍の活動性及びしたがって予後不良に特に関連づけられる。NME1、NME6あるいはNME7に対する抗体で検査することができる、患者サンプルは、血液、組織生検、針生検などを含む体液のサンプルでありえる。
a) 分化を阻害している間、完全にヒトES若しくはiPSの成長及び多分化能をサポートする;
b) より幹様若しくはより癌様状態へ、体細胞を戻す; そして、
c) 腫瘍惹起細胞とも呼ばれる、癌細胞を高度に転移性の癌幹細胞状態へ転換する。
幹細胞及び転移がん細胞が、アンカレッジ−独立的に成長する場合があることを思い出して、本発明者は実験を繰り返した。しかし、今回は、細胞を表面に付着させるために、ロー・キナーゼ阻害剤が細胞の1セットに加えられた。フローテイング細胞が、表面に付着することを強いられた時、RT-PCRは、幹/癌マーカーOCT4の7倍の増加が、実際にあったことを示し、及び幹/癌マーカーNanogの12倍の増加と同じくらい高かった (図30) 。
ヒト若しくはバクテリアNMEにおいて培養された時、繊維芽細胞が先祖返りする様に、実験の写真は、形態的な劇的な変異を示した(図31-38)。より少ない成熟した状態への体細胞を戻す効率の相対的な順序は、NME7>NME1二量体>バクテリアNME1であった。報告によると、転写調節因子BRD4及びコファクターJMJD6は、NME7を抑制し、NME1をアップレギュレーとする(Lui With et al, 2013)。本発明者は、これらの因子は、それらが後期段階プライム化幹細胞にあったよりナイーブ幹細胞中で低いレベルで発現されることを見出した(図39)。
この結果は、二量体としては幹細胞成長を活性化するが、該細胞がもっと分泌し、それがヘクサマーsを形成するとき、該ヘクサマーsはMUC1*を結合せず、また、分化が誘導されるような、NME1がする方法で、自己複製を自身中断若しくは制御できないので、NME7はNME1より早く発現された幹細胞成長因子であるという仮説を立証する。
量的PCR分析は、癌幹細胞マーカーの増加を示す。そのいつくかは、幹細胞マーカーのみとみなされ使われた(Miki J etal 2007, Jeter CR et al 2011, Hong X et al 2012Faber A et al 2013, Mukherjee Det al 2013, Herreros-Villanueva M et al , 2013, Sefah K et al, 2013; Su H-T etal 2013)。図44-47は、NMEタンパク質での培養が、癌細胞を、高度転移性腫瘍惹起細胞に戻らせ、転移受容体CXCR4で200倍より以上までアップレギュレートされ、SOX2で200倍以上までアップレギュレートされ、E-カドヘリン(CDH1)、NANOG及びMUC1で10倍までのアップを示す。
結論として、癌細胞は、MUC1を活性化し、癌と癌幹細胞遺伝子のホストをアップレギュレートするNME7及びNME1を分泌する。本発明者は、より適度であるが傾向性をもって、癌幹細胞のマーカーにおける増加及びMUC1陰性だった細胞でさえ転移を観察した(図47)。したがって、本発明者は、NME7は、おそらくMUC1*以外のルートによってこれらの細胞に入ることができ、そこで、それが転写調節因子としてなお作用でき、これらの遺伝子の発現レベルに影響することができると結論した。ヘクサマーとしては、それは幹あるいは癌成長を活性化しないので、NME1はこの方法で機能するために二量体であらねばならない。しかしながら、多くの癌がNME1を変異させ、それが自己複製制限ヘクサマーの形成に抵抗する。NME1と異なり、NME7は常に活性である。
図48に示される結果は、次のことを示す;癌幹細胞マーカーE-カドヘリン(CDH1)、転移受容体CXCR4が、幹/癌マーカーOCT4、SOX2及びNANOGと同様に、2i、2i+NME7-AB、NME7-AB単独で、非常にアップレギュレートされるということを示す。癌幹細胞マーカーを誘導する相対的な性能は、NME7>NME7+2i>2iであった(図48)。
癌細胞が、2iあるいはNME7-ABのいずれかで処置された時、多分化能阻止クロマチンレギュレーター及び転写調節因子BRD4、JMJD6、MBD3及びCHD4の発現は、同様にダウンレギュレートされた(図49)。
BRD4がNME7を抑制して、その一方でそのコファクターJMJD6が、胚形成中のNME7より遅く発現される自己調整の幹細胞成長因子である(本発明者が決定した)NME1を、アップレギュレートしていることは以前に他で報告された(Thompson et al)。さらに最近の報告は、Mbd3若しくはChd4のsiRNA抑制は、iPS生成への抵抗を大きく減少させた(Rais Y et al 2013 et al.)。本発明者の実験による証拠は、多分化能 Mbd3及びCHD4の阻害剤を抑制している間、NME7がBRD4とJMJD6を抑制する相互のフィードバックループがあることを示した。本発明者は、ナイーブヒト幹細胞において、これらの4つの因子BRD4、JMJD6、Mbd3及びCHD4は、後のステージのプライム化幹細胞でのそれらの発現に比較して、抑制されているということに着目した。さらに、本発明者は、マウスプライム化幹細胞をナイーブ状態へ戻す2i阻害剤(Gsk3βとMEKの阻害剤)が、同じ4つの因子BRD4、JMJD6、Mbd3及びCHD4をダウンレギュレートすることにも着目した。
二量体型のNME1は、体細胞を幹/癌様状態に変換することができること、わずかに劣った程度にだが癌細胞を転移性癌幹細胞への変異できることにおいて、ほぼNME7と同様に機能した。同様に、Halomonas Sp 593のような、ヒトNME1あるいはNME7への高い相同性を持つバクテリアNME二量体は、NME1二量体及びNME7モノマーのように、完全に、ヒト幹細胞成長、多分化能及び生存、癌細胞成長及び生存をサポートすることができ、体細胞を癌/幹細胞状態に戻し及び癌細胞をより転移性の癌幹細胞へ変異させた。
好ましい実施態様では、NMEタンパク質の機能を阻止する薬剤は抗体である。別の好ましい実施態様では、薬剤は、NME1二量体の機能又は二量化を阻止する。さらにもっと好ましい実施態様では、薬剤は、NME7の機能を阻止する。これらのNMEタンパク質のうちの1つの機能を阻止する抗癌剤は、二者択一で核酸でありうる。例えば、sh-若しくはsiRNA、アンチセンス核酸などのようなNMEの発現を阻害する核酸が例示される。あるいは、薬剤は、間接的に、NMEの発現を抑制してもよい。例えば、BRD4の増加した発現は、NME7を抑制し、かくして、抗癌剤として働く。
別の実施態様では、標的化NMEタンパク質の機能を阻害する薬剤は、NMEタンパク質に直接作用する若しくはその発現を阻害する、小分子である合成化学薬品でありうる。別々に若しくはコンビネーションで、これらの薬剤は、癌の治療か予防用の有力な抗癌剤である。
例えば、抗NME抗体の部分は、CAR(キメラ抗原受容体)T細胞技術(PorterD et al, 2011)に記述されるような治療用分子の一部であるように修飾できる。該抗体は、二価、一価、二重特異性のヒト化又は部分的にヒト化でありうる。抗体あるいは抗体様分子は、TillerTら(2013年)によって使用されたものを含む、生体外での結合分析、ファージディスプレー技術及びその他を使い、例えばYlanthia(登録商標)システムのような無作為化されたヒト化抗体エピトープライブラリーを使い、生成されうる。
好ましい実施態様では、標的化NMEタンパク質は、ヒトNME1あるいはNME7ドメインA若しくはBへの30%以上の配列同一性をもつバクテリアNMEである。もっと好ましい実施態様では、標的化NMEタンパク質は、ヒトNME1であり、ここで当該抗体は、それにS120G変異、P69S変異あるいはC-末端切断のような、二量体形成を志向させる変異を持つNME1を特異的に標的としうる。さらにもっと好ましい実施態様では、標的化NMEタンパク質は、NME7-AB(SEQ ID NO: 39)として述べられるような開裂型を実質的に含むNME7(SEQ ID NO: 13)である。
NME7-ABをさらに注入されたグループ(点線)には、加速された割合で成長した、より大きな腫瘍があった。NME7-ABが、注入された時に癌細胞と混合した場合、及びマウスが注入の後24時間あるいは48時間ごとに注入された場合、生着速さ、必要された細胞数の減少及びより速い腫瘍成長率が、もたらされた。NME7に対する癌細胞の比率の範囲あるいはNME7の注入スケジュールは、あるマウス種から別の種及びある腫瘍タイプから別のタイプによって変わると予想される。この方法に関する改良で、ヒトNME7あるいはNME7-ABのための遺伝子組み換え動物は、癌細胞の生着レート非常に増加させ、その結果、動物で腫瘍に成長するのに必要な細胞の数を減少させる。これは、ヒトNME7あるいはNME7-ABを発現する動物での原発性患者癌細胞の成長を可能にする。
別の実例では、患者に投与されているか、患者の治療のために考慮されている、第一選択処置若しくは薬が、より少ない成熟状態に振戻された患者の癌細胞を担持する動物に投与される。第一選択治療は、おそらく、癌細胞が第一選択治療を回避するために適合する変異に影響する。処置後の癌細胞は、その後ホスト動物から取り除かれ、ある治療に応じて生じる変異を同定するために分析し特徴付けをなされうる。あるいは、癌細胞はホスト動物に残すこともでき、どの薬剤が耐性細胞若しくは癌幹細胞を阻害若しくは殺すかを決定するために、該ホスト動物は、他の治療薬で処置される。
例えば、動物の成育の間にNME7発現の潜在的な問題を回避するためには、誘導可能なプロモータの上にヒトNME7を持っていることは有利だろう。あるいは、患者細胞を含む癌細胞は、動物で腫瘍を惹起するのにわずか50-200の細胞を必要する癌幹細胞に細胞が変異されるようなヒトNMEを模倣するバクテリアNME 、NME7、或いはNME1二量体において培養されうる。その後、これらの細胞は、NME7、NME1二量体、バクテリアNMEあるいは単一のチェーンのNME1偽二量体を担持する遺伝子組み換え動物を含む生体内で又は生体外でテストされうる。
毒性は、骨髄細胞へ細胞毒素性の薬剤による処置に応じての骨髄細胞の再生応答時間、循環する血球の数とタイプ及び全体として骨髄数を決定することに加えて、心臓、肝臓及びその他同のような器官の検査により評価される。耐えられる副作用での腫瘍惹起能力、腫瘍生着、又は腫瘍成長割合を優位に減ずる、図62-64、ペプチドNo.1-53にリストされたものに由来した免疫ペプチドは、抗癌治療、予防、若しくはワクチンとして、患者の外において若しくは人において、抗体の生成ための免疫ペプチドとして選択される。
NME7あるいはNME7-ABは、既知のトランスジーンの発現をコントロールする多くの方法のうちの1つで誘導可能になりえる。あるいは、NME7の発現あるいは発現のタイミングを、哺乳動物によって自然に発現する別の遺伝子の発現によってコントロールされうる。例えば、それは、心臓のような組織で発現されるNME7変異体又はNME7には好適でありうる。その後、NME7の遺伝子は、MHCのような心臓で発現されたタンパク質の発現に操作可能にリンクされる。この実例で、NME7の発現は、MHC遺伝子産物が発現される時及び場所で、可能になる。同様に、例えば前立腺特異的タンパク質の発現によって、その発現の場所及びタイミングがコントロールされるヒトNME6若しくはNME7の発現を前立腺でおこしたいと思いうる。同様に、ヒト類以外の哺乳動物で、ヒトNME6若しくはNME7の発現が、乳房組織で発現した遺伝子によってコントロールされることができる。例えば、トランスジェニックマウスでは、ヒトNME6若しくはヒトNME7は、プロラクチン・プロモーター、あるいは同様の遺伝子から発現される。
当該技術分野において周知なように、抗体及び抗体様分子は、全NME1、NME6あるいはNME7タンパク質を使用して生成することができる。あるいは、該タンパク質のペプチドあるいは部分を使用することもできる。さらに、他の方法では、ペプチドは、キャリアー分子若しくは免疫応答を誘発するためのアジュバントと共にホスト動物に注入されう。抗体は、その後、ヒト化することができる動物から得られた、抗体産生細胞によって生産される単クローン抗体を含む標準的方法で動物から得ることができる。
本発明は、さらにNME1、NME6若しくはNME7タンパク質、又はその配列がそれらに由来するペプチドをスクリーニング分析で使うことも可能である。1つのそのような例において、抗体ライブラリーは、NME1、NME6あるいはNME7に結合するそれらの能力についてスクリーニングすることができ、ここで標的化NMEタンパク質に結合する抗体は、癌を治療するか予防するために使用される。さらに、ライブラリーは、抗体それ自身で構成される必要はない。抗体エピトープ若しくは断片のライブラリーは、癌患者若しくは癌になるリスクを持つ人の処置のための治療用抗体を同定するために、NMEタンパク質の部分への結合についてスクリーニングされる。
図62-64にリストされた免疫原性のペプチドの1つ以上は、ホスト動物での抗体の生成にとって、あるいは患者への投与のための抗体様分子へ後で修飾できる抗体エピトープの同定及び選択にとって理想的である。別の実施態様では、その配列が、NME1、NME6あるいは好適にはNME7に由来するペプチドは、ヒトに直接投与される。それは、投与されたヒトが、癌ワクチンとして抗体産生を含む免疫応答を生み出す。図62-64(SEQ ID NO.88-140)にリストされたペプチドの1つ以上は、抗体生成あるいは選択のために好適であり、それはホスト動物での抗体生成用に、ワクチンとしての使用に、Ylanthia登録商標システムのような合成ペプチドか抗体エピトープのライブラリーをスクリーニングするための餌としても使用される。ここで、該抗体は、NME7の機能を阻害することによって又はそれを退化のためにマーキングすることによって癌を阻害するだろう。別の態様において、本発明は、NMEファミリータンパク質のペプチド断片を志向し、これらペプチドは、SEQ ID NOS:88-140、好ましくは88-133、より好ましくは、88-121から選ばれる、NME7に結合し及び阻害する、抗癌抗体若しくは抗体エピトープを生成する若しくは選択するために使用される。
さらに、転移がん細胞である細胞集合体(それは癌幹細胞若しくは腫瘍惹起細胞)は、それらを、NME1二量体、バクテリアNME二量体、あるいはNME7(ここでNME7は多くの癌幹細胞を生産した)と接触させることにより、優先的に増大することを本発明者は示した。したがって、NMEタンパク質を不能にする薬剤は、優れた抗癌治療薬であり、特に癌幹細胞あるいは腫瘍惹起細胞の阻害若しくは予防に役立つ。好ましい実施態様では、治療薬によって標的とされるNMEタンパク質は、ヒト若しくはバクテリアNME1であり、ここで該治療薬は、二量化を阻害するか、MUC1*への結合を阻害するか、CXCR4のような多分化能遺伝子若しくは癌幹細胞遺伝子をアップレギュレートする能力を阻害する。もっと好ましい実施態様では、治療薬によって標的とされるNMEタンパク質はNME7であり、ここで治療薬は、NME7の発現を阻害するか、MUC1*へのNME7の結合を阻害するか、DM10ドメインの開裂を阻害するか、CXCR4のような多分化能遺伝子か癌幹細胞遺伝子をアップレギュレートするその能力を阻害する。
1)MUC1*細胞外ドメインに結合する能力;
2)DNAに結合する能力;
3)幹細胞増殖を促進する能力;
4)分化を阻害する能力;
5)転写調節因子として働く能力;そして
6)細胞によって分泌される能力。
NME7ペプチド35〜46(SEQ ID NOS: 122-133)(図63)は、タンパク質の完全性若しくはMUC1*ペプチドへの結合能のために構造上重要に見えるNME7の領域について、適度にユニーク配列であるので選択された。NME7配列の両方のセットが、NME7に結合する抗体を生じさせると予測される。ところが、NME7ペプチドの第2のセットが、NME7あるいはMUC1*ペプチドへの結合能を、不能にするために機能するかもしれない。これらのペプチドは、NME7を認識するか、又はヒトNME1かバクテリアNMEを認識することができ、その結果、癌の治療か予防に使用することができる抗体を生じると予想される。
図64にリストされたペプチド47〜53(SEQ ID NOS: 134-140)は、NME1からのペプチド配列であり、それらは、配列相同性、NME1 ヘクサマーの公表された結晶構造、及びC-末端切除が二量化を志向し、MUC1*への結合若しくは幹及び癌成長におけるタンパク質の機能を阻害しないという知識に基づき、構造的完全性若しくはMUC1*結合にとって重要であると想定されるものである。
これらの配列を含むペプチド若しくはペプチド模倣体から生成された抗体は、癌の治療か予防のために患者に投与することができる抗体を生み出すだろう。これらの配列を含むペプチド又はペプチド模倣体は、ホストで抗体を生じさせて、したがって、癌の治療か予防のための患者に投与することができる、癌治療ワクチンを構成する。
患者の癌によって若しくは多くの患者を苦しめる癌サブセットによって発現されるNMEタンパク質のタイプのこのような決定に基づいて、患者において若しくは患者グループにおいて、NMEタンパク質の存在を特異的に阻害若しくは不能にする抗NEM抗体若しくは他のNME不能化剤が、選ばれ及び患者に投与される。同様に、診断分析は、患者のNMEタンパク質がタンパク質を二量化志向にする変異を持つかどうか、そうならば、その格別な変異体NMEを不能にする薬剤が、癌の治療か予防のために患者に投与されるかどうかを、決定するために使用される。
例えば、MMP14は、それが乳癌細胞にあるより幹細胞でより高いレベルで発現される(図52)。逆に、MMP14及びADAM17、さらに、MUC1開裂酵素は、ヒト幹細胞にそれらがあるより3倍及び5倍高くDU145前立腺癌細胞で発現される;T47D乳癌細胞では、MMP16とADAM17は、それらが幹細胞にあるより2倍高い(図53及び54)。
実際、DU145前立腺癌細胞が注入されたマウスが、抗MUC1*抗体MN-E6のFabで処置される場合、腫瘍成長は非常に阻害され (図55)、MMP14とADAM17の発現は減少し(図56)、MUC1開裂は減少され、及び、分化をシグナルするmicroRNA-145の発現は増加した(図57 A、B)。かくして、MUC1*は、その遠位、10以上のアミノ酸によるN-末端で変わりうる。
MUC1のC末端は細胞内であり、及び、そのN-末端は細胞外である。本発明者の実験は、PSMGFRペプチドのN-10バージョンにNME1二量体が結合することを示す。すなわち、PSMGFRペプチドの最初の10のアミノ酸を除去すること、それは大多数のMUC1*細胞外ドメインに相当する、は、NME1二量体のMUC1*ペプチドに結合する能力に影響しないことを意味する。N-10ペプチド(SEQ ID NO: 86)に優先的に結合する抗体は、それが癌細胞に存在するので、MUC1*に優先的に結合する。逆に、C-10ペプチド(SEQ ID NO: 87)に優先的に結合する抗体は、癌細胞ではなく幹細胞及び骨髄細胞に優先的に結合する(図58-60)。
したがって、癌の治療か予防のためにMUC1*を標的とする抗体は、PSMGFRペプチド、N-10ペプチドあるいはC-10ペプチド、での免疫化によって生成されうる。あるいは、癌の治療か予防用の治療用抗体若しくは抗体様分子は、それが幹と先祖細胞に現われる様とは対照的に、癌細胞に現われるので、MUC1*に結合するものを選択することによって同定することができる。
好ましい実施態様では、抗体は、N-10ペプチドに結合することを志向する。さらにもっと好ましい実施態様では、治療抗体は、幹若しくは先祖細胞ではなく、癌細胞に結合するその能力によって選ばれる。1例で、抗体は、ELISA 若しくは同様の直接結合分析によって、PSMGFRペプチド、N-10ペプチドあるいはC-10ペプチドに結合する能力によってまず選ばれ、、その後、様々なタイプのMUC1*陽性の癌細胞に結合することができることが確認された。ここで1つの抗体は、乳癌細胞よりよりよく前立腺癌細胞に結合でき、あるいは、異なる組織タイプの異なる開裂サイトの仮説を支持して、その逆も又ありうる。
その後、ハイブリドーマ上澄みは、多ウェルプレート上に被覆され。そして、幹細胞はその上に蒔かれた。ト幹細胞は、非付着性であるので、幹細胞(あるいは先祖細胞)に結合された抗体で覆われたウェルは、幹細胞を付着させた。一方、幹細胞を付着しなかった抗体は、癌の治療か予防に好ましい抗MUC1*抗体として選ばれた。
(i) 幹若しくは先祖細胞遺伝子あるいは遺伝子産物の存在のための患者サンプルを分析すること;そして、
(ii) 幹若しくは先祖細胞遺伝子あるいは遺伝子産物の同様の発現あるいは発現レベルを共有する患者のグループ化。
この方法で、その後、患者は、それらの幹若しくは先祖細胞遺伝子あるいは遺伝子産物を阻害する薬剤で治療することができる。

NME1が添加されたレーン4は、ブランクであり、NME7抗体がNME1と交差反応しないということを示す。
本発明者が、それらがbFGFにおいて培養された細胞よりよりナイーブ状態であることを示す表現マーカーであることを示した、NME7がNME1二量体において培養された幹細胞でよりかなり多く発現されるという事実は、NME7が、プライム化細胞でその発現に比べ、ナイーブ細胞でより高いレベルに発現することを意味する。
タンパク質は、SDS-PAGEによって分離され、次に、抗NME1抗体(図3C)、抗NME6抗体(データ示さず)あるいはNME7抗体(図3D)で検査した。結果は、NME7がMUC1*細胞外ドメインペプチドに結合することを示す。これは、幹細胞と癌細胞で、NME7は、その2つのNDPKドメインのその部分を経て、MUC1*細胞外ドメインを二量化させることにより多分化能パスウェイを活性化するということを示す
第1の構成物は、効率的に可溶の型で発現することができる組み換えNME7を作るために調製された。最初のアプローチは、天然のNME7(-1)又はN-末端欠失がある代替的スプライスバリアントNME7(-2)をコードする構成物作ることであった。ある場合には、構成物が、その精製を助けるためにstrepタグ又はヒスチジン・タグを担持した。NME7-1は、大腸菌で貧弱に発現し、そして、NME7-2は大腸菌で発現しなかった。しかしながら、標的配列が削除され、NME7が、31kDaの計算された分子量をもつNDPK A及びBドメインを本質的に含んだ、新しい構成物が作られた。
この新しいNME7-ABは、大腸菌において非常によく発現され、また可溶のタンパク質として存在した。シングルNDPKドメインが発現された構成物、NME-Aは、大腸菌で発現しなかった。NME7-ABは、NTA-Niカラムで最初に精製され、次に、セファデックス200カラムで分子ふるいクロマトグラフィー(FPLC)によってさらに精製された。その後、精製されたNME7-ABタンパク質は、多分化能を促進し、かつ幹細胞の分化を阻害する能力に関してテストされた。
NME7の可溶性変異体、NME7-ABは、生成され精製された。ヒト幹細胞(iPS cat# SC101a-1及びシステム・バイオサイエンス)は、4継代のためにマウス繊維芽細胞フィーダー細胞の層上で4ナノグラム/mlのbFGF中でメーカー指示書によって成長した。その後、これらのソース幹細胞は、単クローン抗MUC1*抗体(MN-C3)の12.5μg/ウェルで被覆された6ウェル細胞培養プレート(VitaTM、ThermoFisher)へ植えつけられた。細胞は、ウェル当たり300,000個の細胞の密度で植え付けされた。
基礎培地は、次のものから成る最小の幹細胞培地であった:
400ml DME/F12/GlutaMAX I(Invitrogen#10565-018)、100mlノックアウト血清代替物(KO-SR、Invitrogen# 10828-028)、5ml 100xMEMの非本質アミノ酸溶液(Invitrogen#11140-050)及び0.9ml(0.1mM)β- メルカプトエタノール(55mMストック Invitrogen# 21985-023)。該基礎培地は任意の培地になりえる。
好ましい実施態様では、基礎培地は、他の成長因子及びサイトカインがフリーである。基礎培地に、NME7-ABの8nMあるいは8nM NM23-H1のいずれかが加えられ、安定した二量体として折りたたまれ精製された。培地は48時間ごとに変更され、加速的成長により回収されなければならなくり、植え付け3日目で継代した。
図9及び10は、NME7モノマーでの成長に対するNM23-H1二量体での成長の日々比較を示す。NME7及びNM23-H1(NME1)二量体の、両方は分化多能に成長し、100%の集密状態でさえ、分化がなかった。写真で見ることができるように、NME7細胞は、NM23-H1二量体中で成長する細胞より速く成長した。最初の回収での細胞数は、NME7での培養がNM23-H1二量体での培養より1.4倍多い細胞の生産を確認した。典型的な分化多能のマーカーのためのICC染色は、NME7-ABがヒト幹細胞成長、多分化能及び抵抗された分化を完全にサポートすることを確認した(図11)。

細胞懸濁は、37℃30分、回転台(275rpm)でインキュベートされ、5分間氷の上で超音波処理された。不溶性細胞残屑は、遠心分離(4℃、30分、20000rpm)によって除去された。その後、透明な溶解液は、ランニングバッファーで平衡に保たれたNi-NTAカラム(Qiagen)に適用された。カラムは、ランニングバッファーの4CVで、30mMイミダゾールが補われたランニングバッファー(4CV)で洗浄され、溶出は、70mMイミダゾールが補われたランニングバッファー(6CV)で、次いで、490mMイミダゾールが補われたランニングバッファーでカラムからタンパク質を溶出させた。NME7-ABは、分子ふるいクロマトグラフィー(Superdex 200)「FPLC」によってさらに精製された。
その後、薄膜は次のもので検査された:
A) 抗-NM23-H1(NME1)抗体(C-20, Santa Cruz Biotechnology);
B) 抗-NME6(Abnova);あるいは
C) 抗-NM23-H7抗体(B-9, Santa Cruz Biotechnology);
D) NME6の染色は、Supersignal(Pierce)を使用して増強された;そして
E) NME7の染色は、Supersignalを使用して増強された。
HRPに結合したそれぞれの二次抗体と培養の後、タンパク質は化学発光によって検知された。写真は、天然のNME1、NME6及びNME7がMUC1*陽性の乳癌細胞、ヒトES細胞、及びヒトiPS細胞に、及びそれらMUC1*に結合しているものに存在することを示す。HES-3ペレットに存在する細胞の数が、他のサンプルに存在する数より少なかったことに留意を要する。
もし幹細胞がプライム化状態(市販の幹細胞はすべてFGFにおいて培養することによりプライム化状態に誘導された)からナイーブ状態に戻っていれば、ヒストン-3は「クロウド“雲”」と見なされるか、あるいはまさに斑点である若しくは検知できないだろう。
図12は、それらが標準プロトコルによってMEF上でのFGFにおいて育てられたという点を除いて、同じ出所由来の、コントロール細胞が、すべて、それらがすべてナイーブ状態ではなくプライム化状態に100%あることを確認できる、核において縮合されたヒストン-3(H3K27me3)を示す。
逆に、10継代の間、NME7において培養された同じ出所細胞は、ほとんどが、それらが前X不活性化であって真のナイーブ状態であることを示す、縮合ヒストン-3もたない幹細胞を有した。図12に示される挿入物は、100%のナイーブである単離された多くのクローンのうちの1つである。
ベッドは、PBSで2度洗われ、また、免疫沈澱したタンパク質は、12%のSDS-PAGE還元性ゲル上で電気泳動によって分離された。タンパク質は銀染色法(Pierce)によって検知された。T47Dサンプル及びBGO1v細胞から、NME7と共に共免疫沈殿させられた、タンパク質の〜23 kDaバンドが切除され、マススペック(Taplin Mass SpectrometryFacility, Harvard Medical School)で分析された。マススペック分析は、切除されたタンパク質バンドが、すべて、以下に示されるようなNME7 NDPK Aドメインからの配列を含むことを示した。NME7のAドメインの下線が付された配列は、マススペックによって同定された。
NME7結合用のプロトコルが使用され、及び、NME7は11.6μg/mLで使用された。

- 薬剤が、SEQ ID NOS:122-133から選択されるいずれか1つのアミノ酸配列からなるペプチドを認識する抗体である、NME7タンパク質の機能を阻害する薬剤、又は
SEQ ID NO: 39若しくはSEQ IDNO:59に示されるアミノ酸配列からなるNME7-ABを認識する抗体である、NME7-ABタンパク質の機能を阻害する薬剤。 - 薬剤が、Fab、一価、二価、又はIgM、二重特異的ヒト若しくはヒト化された抗体である、請求項1に記載の薬剤。
- 阻害されるNME7または7-ABタンパク質の機能は次のことをする能力であって:
幹細胞の増殖を促進する及び/又は分化を阻害する;
癌細胞の増殖を促進する及び/又は分化を阻害する;
MUC1*に結合する;
DNAに結合する;
転写調節因子として作用する;
細胞によって分泌される; あるいは、
