Description
ウスバノコギリモク酒精抽出物の製造
本発明において用いたウスバノコギリモクは、釜山市機張郡で採集して使用し、下記のような方法を通してウスバノコギリモクの酒精抽出物を得た。先ず、酒精抽出物の製造のために、ウスバノコギリモクを日陰または日向で自然乾燥後、摩砕して得た粉末2kgと酒精(95%エタノール)をそれぞれ8Lずつ入れ、70℃で3時間の間3回繰り返して還流冷却器が取り付けられた抽出器で抽出した後、限外ろ過装置(MWCO、50kDa)でろ過して異物を除去した後、ろ過された抽出物を真空回転濃縮機で40℃で溶媒を除去してから、抽出された残渣としてウスバノコギリモク粗抽出物(200±25g)を得た。
ウスバノコギリモクの酒精抽出物のリウマチ関節炎の治療効果
<2−1>ウスバノコギリモク抽出物処理による体重変化の測定
本発明者等は、ウスバノコギリモク抽出物でリウマチ関節炎を治療することができるか否かを確認するために、先ず、リウマチ関節炎が誘導された実験動物を作製した。
本発明者等は、ウスバノコギリモク抽出物によるリウマチ関節炎の治療効果を確認するために、関節炎の指標となる足の厚さ(paw thickness)の変化を測定した。
本発明者等は、ウスバノコギリモク抽出物のリウマチ関節炎の治療効果を確認するために目視で関節炎指数を観察し、それぞれに対する関節炎指数は、0〜4点と点数をつけ、非実験者5名により測定された。
−評価基準−
0点:何の症状もない
0.5点:一つの指が赤く、むくみがある
1点:2〜5個の指が赤く、若干のむくみがある
2点:2〜5個の関節が赤く、中度のむくみがある
3点:全体足が赤く、むくみがある
4点:減少したむくみと奇形
本発明者等は、ウスバノコギリモク抽出物がリウマチ関節炎を治療することができるか否かを確認するために脾臓の重さを測定した。
本発明者等は、ウスバノコギリモク抽出物処理による関節炎の治療程度を目視で観察した結果、関節炎が誘発された実験動物群では、後足が激しくむくんでおり、紅斑症状を現した。しかし、本発明のウスバノコギリモク抽出物を0.01%、0.05%と19日間処理した処理群では、むくみが減少し、紅斑症状が緩和されたことを確認することができた(図7参照)。
通常、リウマチ関節炎が発達すると、炎症部位に集まった活性化された免疫細胞によりインターロイキン(IL)とPGE2のような炎症媒介体(inflammatory mediator)が関節と血液に放出されるが、特に、TNF−α、IL−6、IL−1βは、リウマチ関節炎の疾病程度の有無を把握できる指標として用いられ得る。
本発明者等は、炎症部位である後足を摩砕して抽出したタンパク質中の炎症性サイトカインの発現程度を測定するためにウェスタンブロット(Western blot)で測定した。
NOとPGEsは、nitric oxide synthase(NOS)とcyclooxygenase(COX)により、それぞれL−アルギニン及びリン脂質から合成される。病理的な条件下でNOとPGE2の過度な増加は、炎症過程を刺激し、他の炎症媒介体と共に相乗効果を示す。このような状態下で過度なNOとPGE2は、inducible NOS(iNOS)とCOX−2により合成される。また、iNOSとCOX−2は、炎症性サイトカインとバクテリア性内毒素(endotoxin)にさらされたとき、強く発現される。
CIA処理によるiNOSとCOX−2及び前駆炎症性サイトカインであるTNF−α、IL−1β及びIL−6等は、転写因子であるNF−κBによって調節される。NF−κBは、一般的な状態ではinhibitory κB−α(IκB−α)と共に結合して細胞質で不活性化状態で存在する。しかし、コラーゲンのような抗原を動物体内に注入すると、大食細胞、単球細胞等の免疫細胞が活性化されて炎症作用を起こす。このような外部刺激により、細胞内不活性化状態のNF−κBは、リン酸化によって活性化されて核内に移動し、炎症関連タンパク質の転写作用に寄与することとなる。
様々な外部刺激によるNF−κB転写因子の活性化を通してiNOSやCOX−2または前駆炎症性サイトカインの発現を誘導する過程を調節する分子信号伝達機序は、主要MAPKサブファミリーに属する3つの酵素、即ち、extracellular−regulated protein kinase(ERK)、c−Jun NH2−protein kinase(JNK)、p38 MAPKとPI3K/Aktが挙げられる。そこで、関節内タンパク質のリン酸化に関する本発明のウスバノコギリモク酒精抽出物の効果を確認するためにウェスタンブロットを遂行した。
CIAでリウマチ関節炎を誘導すると、関節部位の軟骨組織を分解するMMP−1、MMP−2及びMMP−9の発現の増加、関節滑膜組織内への炎症細胞浸潤を誘導するMCP−1の増加、そして、関節の滑膜組織に炎症細胞の浸潤を増加させるICAM−1とVCAM−1が増加する。そこで、関節炎症部位のICAM−1、VCAM−1、MCP−1、MMP−1、2、9の生成抑制効果に関する本発明のウスバノコギリモク酒精抽出物の効果を確認するためにウェスタンブロットを遂行した。
様々なホンダワラ抽出物での関節炎治療効果の確認
本発明のウスバノコギリモク抽出物と比較して他のホンダワラ種でも関節炎治療効果があるか否かを確認するために、様々な炎症性サイトカインのうちTNF−αの発現程度をELISA kitを用いて測定した。
Claims (4)
- ウスバノコギリモク抽出物(Sargassum serratifolium)を有効成分として含み、
前記ウスバノコギリモク抽出物は少なくともエタノールを利用して得た抽出物(但し、ヘキサンを利用して得た抽出物を除く。)である、関節炎予防または治療用薬学的組成物。 - 前記ウスバノコギリモク抽出物は、炎症性サイトカインIL−1β、IL−6またはTNF−aの活性を減少または抑制することを特徴とする、請求項1に記載の組成物。
- 前記関節炎は、退行性関節炎、リウマチ関節炎またはループス(Lupus)関節炎であることを特徴とする、請求項1に記載の組成物。
- ウスバノコギリモク抽出物(Sargassum serratifolium)を有効成分として含み、
前記ウスバノコギリモク抽出物は少なくともエタノールを利用して得た抽出物(但し、ヘキサンを利用して得た抽出物を除く。)である、関節炎予防または改善用健康機能食品。
