Description
〔発明の概要〕
本発明は、部分的には、本発明の抗体が結合するポリペプチドに加え、RSV、MPV、およびPVM感染を中和する、広範に中和する抗体の発見に基づく。それゆえ、本発明のある局面において、本発明は単離された、RSV、MPVおよびPVMの感染を交差中和する抗体、例えばモノクローナル抗体、ヒト抗体、ヒトモノクローナル抗体、変異抗体、または抗原結合フラグメントを含む。
〔図面の簡単な説明〕
図1は、インビトロでのRSVまたはMPVウイルス感染を中和する能力についての、7人の提供者(Don.1〜7)から得られたEBV不死化記憶B細胞によって生成されたモノクローナル抗体の、スクリーニング結果を示す。
〔発明を実施するための形態〕
本発明は、部分的には、RSVおよびMPVの両方、またはRSVと、MPVとPVMとを共に交差中和する抗体ならびに本発明の抗体が結合するエピトープの発見および単離に基づく。前記抗体は、1種類のみまたは数種類の抗体が、RSVおよびMPVの両方、またはRSVとMPVとPVMとを中和するために必要とされるため、好ましいものである。さらに、交差中和する抗体は、RSVおよび/またはMPV感染の処置のための、抗体を含む医薬の生産コストを低減するため、高い力価で製造される。さらに、当該抗体によって認識されるエピトープは、RSVおよびMPVの両方に対する幅広い防御を誘導可能なワクチンの一部であり得る。
本発明に係る抗体は、当該技術分野において公知のいかなる方法でも作製され得る。例えば、ハイブリドーマ技術を用いてモノクローナル抗体を作製するための一般的な方法論はよく知られている(Kohler, G. and Milstein, C,. 1975; Kozbar et al. 1983)。一実施形態においては、国際公開第WO2004/076677号に記載された代替的なEBV不死化法が使用される。
上述のように、本発明の抗体は、それらの結合するエピトープをマッピングするために用いられ得る。発明者らは、融合前Fタンパク質に見いだされるが、融合後Fタンパク質には見いだされない、エピトープに関する本願発明の中和抗体を発見した。一実施形態において、抗体またはその抗原結合フラグメントは、RSV融合前Fタンパク質に結合し、RSV融合後Fタンパク質には結合しない。他の実施形態において、抗体またはその抗原結合フラグメントは、RSVおよびMPVの融合前Fタンパク質に結合し、RSVおよびMPVの融合後Fタンパク質には結合しない。さらに他の実施形態において、抗体またはその抗原結合フラグメントは、RSV、MPVおよびPVMの融合前Fタンパク質に結合するが、RSV、MPVおよびPVMの融合後Fタンパク質には結合しない。
本発明は、本発明の抗体もしくは抗体フラグメント;当該抗体もしくは当該フラグメントをコードしている核酸;当該核酸をコードしているベクター;または本発明の抗体もしくは抗原結合フラグメントによって認識されるポリペプチドを含む、医薬組成物を提供する。また、医薬組成物は薬学的に許容可能な担体または賦形剤を含み得る。担体または賦形剤は投与を容易にし得るが、それ自身は組成物を受容する個体に有害な抗体の生成を誘導するものではない。有毒性でないだけではなく、好適な担体は大きな、ゆっくりと代謝された巨大分子(タンパク質、ポリペプチド、リポソーム、多糖類、ポリ乳酸、ポリグリコール酸、重合体のアミノ酸、アミノ酸共重合体および不活性なウイルス粒子など)であり得る。
鞘内、心(脳)室内(intraventricular)、経皮、経皮膚、局所、皮下、鼻腔内、経腸、舌下、膣内、または直腸の経路が挙げられるがこれらに限定されない)。ハイポスプレーもまた、本発明の医薬組成物を投与するために用いられ得る。典型的に治療組成物は、液体の溶液または懸濁液のいずれとしても注射可能なように調製され得る。注射の前の液体のビヒクル中の溶液または懸濁液に、好適な固体の形態が調製され得る。
本発明の抗体および抗体フラグメントまたはその派生物および変異体は、RSVもしくはMPVの感染またはRSVおよびMPVの両方の同時感染の処置のため;RSVもしくはMPVの感染、またはRSVおよびMPVの両方の同時感染の予防のため;またはRSVまたはMPVの幹線の診断のために用いられ得る。
本明細書に用いられるとき、“抗原結合フラグメント”“フラグメント”および“抗体フラグメント”という用語は抗体の抗原結合活性を維持している本発明の抗体の任意のフラグメントを指すために相互交換可能に用いられる。抗体の例としては、単鎖抗体、Fab、Fab’、F(ab’)2、FvまたはscFvが挙げられるがこれらに限定されない。さらに、本明細書で用いられる“抗体”という用語は、抗体とその抗原結合フラグメントの両方を含んでいる。
〔実施例〕
本発明の例示的な実施形態は、以下の実施例において供される。以下の実施例は、実例として、かつ、当業者が本発明を使用するに際し助けとなるという目的のためのみに提示されている。この実施例は、いかなる方法によっても本発明の範囲を制限することを意図していない。
125人の血液提供者のコーホートから、RSVおよびMPVに対し高い血清抗体価を示す7人を選択した。凍結保存された末梢血単核細胞(PBMCs)からCD22+IgG+B細胞をソートし、エプスタイン・バールウイルス、および、CpGオリゴデオキシヌクレオチド2006、および、フィーダー細胞として、放射線照射された同種異系のPBMCsを用いて、ウェルあたり3から5細胞の割合で不死化した。培養上清を14日後に採取し、Hep−2細胞へのRSVのA2株の感染、または、LLC−MK2細胞へのMPV A1 I−PV−03/01−6621株の感染に基づくマイクロ中和アッセイを用いて、中和抗体の存在を解析した。それぞれ6日または8日培養したHep−2標的細胞またはLLC−MK2標的細胞の添加に先立ち、上清の原液を、50−100 TCID50のウイルスと共に1時間室温で培養した。培養物に対してWST−1試薬(ロシュ)を添加し3〜4時間おき、分光光度計を用いて生存している細胞を検出した。
HMB2430およびHMB3210の反応性の範囲を評価するため、以下に示すRSV株およびMPV株を用いて得たHep−2 RSV感染細胞またはLLC−MK2 MPV感染細胞に対する結合性に関して、精製されたmAbsをFACSによって試験した。RSV株およびMPV株:RSV A2(A、1961、オーストラリア;A/A2/61)、RSV Long(A、メリーランド、アメリカ、1956;A/Long/56)、RSV Randall(A、シカゴ アメリカ、1958;A/Randall/58)、RSV 9320(B、マサチューセッツ アメリカ、1977;B/9320/77)、WV/14617/85(B、ハンチントン ウエストヴァージニア、1985;B/14617/85)、18537(B、ワシントン・コロンビア特別区 アメリカ、1962;B/18537/62)、MPV I−PV−03/01−6621(A1、パヴィーア イタリア、2001;A1/6621/01)、MPV I−PV−02/06−8938(A2、パヴィーア イタリア、2006;A2/8938/06)、I−PV−03/04−4702(B1、パヴィーア イタリア、2004;B1/4702/04)およびI−PV−02/04−3817(B2、パヴィーア イタリア、2004;B2/3817/04)。並行して、3種類の前述のmAbsを試験した:(i)モタビズマブ(RSV特異的);(ii)mAbs234(MPV特異的);および(iii)FO32(インフルエンザA特異的。ネガティブコントロールとして使用)。全てのmAbsは10μg/mLの濃度で、感染細胞および非感染細胞に対する結合性の観点で試験された。HMB2430およびHMB3210は、既知のRSVおよびMPVのグループおよびサブグループを代表する株である試験された6種類のRSV株全ておよび4種類のMPV株全てと反応した(表5)。対照的に、モタビズマブは試験された6種類のRSV株全てと反応したが、試験された4種類のMPV株のいずれとも反応しなかった。反対に、234mAbsは試験された4種類のMPV株全てと反応したが、試験された6種類のMPV株のいずれとも反応しなかった。
HMB2430およびHMB3210により認識される、RSVおよびMPVウイルスの標的抗原を同定するため、一過性導入された293F細胞から生産されたRSV(A2株)のホモ3量体可溶Fタンパク質との結合能について、モタビズマブおよびmAb234と並行して、上述の2種類のmAbsを解析した。図3に示すように、HMB2430およびHMB3210は、ELISAによってRSVのFタンパク質と特異的に反応し、モタビズマブと比較して明らかに異なる結合プロファイルを示した。加えて、HM2430およびHMB3210は、RSV(A2株)もしくはMPV(NL/1/99 B1株)のFタンパク質の全長をコードしている哺乳類発現ベクターを一過性導入された293F細胞を、その細胞内的に染色し(表6)、HMB2430およびHMB3210が、RSVのFタンパク質およびMPVのFタンパク質に共通して存在するエピトープを認識することを示唆した。この発見は、RSVのFタンパク質およびMPVのFタンパク質に、33〜35%のアミノ酸配列同一性があるのみであることを考慮すると、特に際立ったものである。予想された通り、パリビズマブおよびモタビズマブは、RSVのFタンパク質を発現している細胞と結合したが、MPVのFタンパク質を発現している細胞とは結合しなかった(表6)。反対に、234mAbは、MPVのFタンパク質を発現している細胞と結合したが、RSVのFタンパク質を発現している細胞とは結合しなかった(表6)。
HMB2430およびHMB3210によって認識されるFタンパク質のエピトープに関する識見を深めるため、RSV A2株を感染させたHep−2細胞を用いて結合阻害アッセイを行った。以下に名前を示す、RSVのFタンパク質に特異的なmAbsは購入されたか、または遺伝子合成によって製造した:(i)モタビズマブ(抗原サイトIIに特異的);(ii)101F(抗原サイトIVに特異的);(iii)D25(特異性が不特定);(iv)131−2a(抗原サイトIに特異的)。前記mAbsはビオチンで標識され、最大量の70〜80%の結合を達成するのに必要な、mAbsの至適濃度を決定するため、Hep−2感染細胞に対する結合試験が行われた。その後、ビオチンで標識されたmAbsは、RSVのA2株感染細胞を、50倍量の同種または異種の非標識化mAbsとともに前培養することによって、その結合(蛍光体とコンジュゲートしているストレプトアビジンを用いて計測)が阻害されるかを評価するためのプローブとして使用された。
予想された通り、ビオチンで標識されたHMB2430の結合は、非標識化HMB2430と共に上記細胞を前培養することによって阻害されたが、同様に非標識化HMB3210によっても、その結合は、部分的に阻害された(図4)。対照的に、試験された他の全てのビオチン標識化mAbsは、HMB2430もしくはHMB3210のいずれとの前培養によっても阻害を受けなかった(図4)。これらの結果をまとめると、HMB2430およびHMB3210は、Fタンパク質上にある、RSVとMPVとの間で共通しかつ互いに部分的に重複するエピトープを認識すること、またこれらのエピトープはFタンパク質の抗原サイトII(モタビズマブおよびパリビズマブにより認識)、抗原サイトIV(mAb 101Fにより認識)、および抗原サイトI(mAb 131−2Aにより認識)とは異なることを示す。加えて、RSVのFタンパク質上に存在する、mAbsHMB2430およびHMB3210により認識されるエピトープは、mAb D25により認識される未知のエピトープと異なる。
HMB2430およびHMB3210が、RSVのFタンパク質およびMPVのFタンパク質の両方を認識するという発見をさらに確認するため、これらの2つのmAbsに関して、RSVおよびMPVのFタンパク質をウエスタンブロットにおいて染色する能力を試験した。Hep−2細胞をRSVに感染させ、LLC−MK2細胞をMPVに感染させ、緩和な界面活性剤に溶解し、還元状態および非還元状態でSDS−PAGEゲルに泳動させた。タンパク質はPVDFメンブレンに転写され、HMB2430、HMB3210、モタビズマブ、および234 mAbのいずれかと共に培養した。mAbの結合は抗ヒトHRP結合抗体をECL Western Blotting Detection reagentと併用して検出した。HMB2430およびHMB3210は、非還元状態において、RSVに感染した細胞由来(図5)およびMPVに感染した細胞由来(図6)のFタンパク質に結合した。MPV特異的なmAb 234(RSVのFタンパク質の抗原サイトIIに相当する、MPVのFタンパク質上のエピトープを認識する)は、非還元状態においてMPVのFタンパク質に結合するが、RSVのFタンパク質には結合しなかった。対照的に、RSV特異的なmAbであるモビズマブは、還元状態と非還元状態の両方においてRSVのFタンパク質と結合し、主として線状エピトープを認識することが裏付けられた。これらの結果は、モタビズマブおよびパリビズマブと異なり、HMB2430およびHMB3210は、RSVのFタンパク質およびMPVのFタンパク質内のアミノ酸残基間のジスルフィド結合の存在に依存する、コンフォメーショナルエピトープを認識することを示唆する。
精製されたHMB2430およびHMB3210mAbsの、RSVに感染したHep−2、または、MPVに感染したLLC−MK2細胞を中和する能力を試験した。以下のRSVおよびMPV株を試験した。:RSV A2(A、1961、オーストラリア;A/A2/61)、RSV Long(A、メリーランド、アメリカ、1956;A/Long/56)、RSV Randall(A、シカゴ アメリカ、1958;A/Randall/58)、RSV 9320(B、マサチューセッツ アメリカ、1977;B/9320/77)、WV/14617/85(B、ハンチントン ウエストヴァージニア、1985;B/14617/85)、18537(B、ワシントン・コロンビア特別区 アメリカ、1962;B/18537/62)、RSV 9727/2009(B、パヴィーア イタリア、2009;B/9727/09)、RSV 9736/2009(B、パヴィーア イタリア、2009;B/9736/09)、RSV 9847/2009(B、パヴィーア イタリア、2009;B/9847/09)、MPV I−PV−03/01−6621(A1、パヴィーア イタリア、2001;A1/6621/01)、MPV I−PV−02/06−8938(A2、パヴィーア イタリア、2006;A2/8938/06)、I−PV−02/06−8908(A2、パヴィーア イタリア、2006;A2/8908/06)、I−PV−02/06−8909(A2、パヴィーア イタリア、2006;A2/8909/06)、I−PV−03/04−4702(B1、パヴィーア イタリア、2004;B1/4702/04)およびI−PV−02/04−3817(B2、パヴィーア イタリア、2004;B2/3817/04)。
HMB3210、パリビズマブおよび234 mAbの、RSVもしくはMPVのモノクローナル抗体抵抗性ミュータント(MARMs)を選択する能力をインビトロで試験した。何回かの試みにも関わらず、HMB3210は、32x 10e6 RSV A/Long/58 TCID50および16x 10e6 MPV A1/6621/01 TCID50に対して試験した場合に、RSVおよびMPVのMARMsを選択できなかった。対照的に、パリビズマブは、高い頻度でMARMsを選択した(16x−10e6 RSV A/Long/58 TCID50の1回のインプットから、計85の独立したパリビズマブMARMsを単離し、これは185000TCID50につき1の頻度に相当する)。同一の実験条件において、mAb 234はMPV MARMsを1つも選択しなかった。234 mAbのMARMsを単離することの困難さは、少数のMARMsを単離するために高いレベルのウイルスが必要であることを示したUlbrandt et alによる報告(J General Virol 2008)と一致する。エスケープしたウイルスは、単離NL/1/99 MPV B1を用いてK242N変異にマッピングされた。独立したパリビズマブMARMs(PZ−MARMs)が収集され、そのうちの10のFタンパク質が完全に配列解析された(表8)。PZ−MARM2、PZ−MARM3、PZ−MARM4、PZ−MARM5、PZ−MARM6、PZ−MARM8およびPZ−MARM10は、同じ2つのアミノ酸変異(P101S/K272T)を共有していた;PZ−MARM1も、同じ位置に2つのアミノ酸変異を有していたが異なるアミノ酸変異であった(P101S/K272Q);PZ−MARM7もまた、位置272に、1つのアミノ酸変異(K272N)を有していた;そして、PZ−MARM9は、他のPZ−MARMsと共通のアミノ酸変異、および262の位置におけるユニークな変異(P101S/N262Y)を有していた。この領域(ヌクレオチド位置827および828)における点突然変異は既に記述されており(Zhao et al. Virology 2004)、この変異は位置272における2つの異なるアミノ酸変異(K272QおよびK272M)をもたらすものであった。最初の突然変異(すなわちK272Q)は、本明細書に記載されるPZ−MARM1にも存在した。これらの点突然変異を保有するウイルスは、コットンラットにおいて、パリビズマブの感染予防効果に対し完全な耐性を有し(Zhao et al. Virology 2004)、他の変異を伴っている同じ変異が、予防的にパリビズマブで処置された、RSVに感染した免疫抑制コットンラットにおいて記述された。
HMB3210の軽鎖可変部は、N−グリコシル化モチーフ(NxS/T、xはプロリン以外のいかなるアミノ酸でもあり得る)をLCDR3の113位(IMGT番号)に有する。113位のアスパラギン(N113)が、ジャームライン配列に存在するセリンと置き換えられている。可変領域におけるグリコシル化モチーフの存在は、抗体の活性にポジティブもしくはネガティブな影響を与える可能性があり、また抗体の不均一性の原因と認識されている。HMB3210の軽鎖におけるグリカンの存在は、N−グリコシダーゼであるPNG−アーゼFの存在下および非存在下における培養に続き行った、還元SDS−PAGEゲルにおいて評価した(図8)。この解析により、少数の軽鎖が、実際にグリコシル化されていることが示された。そしてN113残基を取り除いて、対応するジャームラインがコードするセリン残基に戻した。並行して、軽鎖のフレームワーク−1領域(P7T)におけるもう1つの体細胞変異も同様に、ジャームラインがコードする7位のプロリン残基(対応するIGLV1−40*02遺伝子におけるIMGT番号で示す)に戻すため、取り除かれた。HMB3210重鎖VH.2(配列番号17)およびHMB3210軽鎖VL.3によって構成されるHMB3210の新しい変異体(HMB3210v3と命名された)が生産され、HMB3210v2(HMB3210重鎖VH.2(配列番号17)および配列番号14に示す軽鎖VLによって構成される)とともに、RSV A2株およびMPV I−PV 03/01−6621株を中和する能力を試験した。この変異体(HMB3210v3)は、試験したRSV株およびMPV株の両方に対するわずかに改善された中和能力を示し(図9)、すなわち、軽鎖のグリコシル化サイトの除去は、RSVおよびMPV上の標的エピトープに対する結合に影響を与えないことが示される。2種類の抗体は、その後並行してRSVおよびMPV一群に対して試験され、試験された全てのウイルスに対し同等の活性を有することを示した(図10)。結論を述べると、HMB3210v3は可変軽鎖においてグリコシル化されておらず、またジャームラインの重鎖遺伝子および軽鎖遺伝子と比較して全体でほとんど変異が無く、重鎖において8つのアミノ酸のみに体細胞変異を有し、軽鎖において4つのアミノ酸のみに体細胞変異を有していた:S58A(HCDR2)、165S(HCDR2)、Y66D(HFR3)、V71A(HFR3)、N85T(HFR3)、Y88F(HFR3)、V101I(HFR3)、Y103F(HFR3)、G56D(LCDR2)、S65N(LCDR2)、G78A(LFR3)およびS109R(LCDR3)(全ての位置はIMGT番号で示されている)。
RSVおよびMPVに対するHMB3210およびHMB2430の交差反応性における体細胞変異の役割についての見識を得るため、両方のmAbのジャームラインバージョンを合成し、RSV A2株およびMPV I−PV 03/01−6621株を中和する能力を試験した。HMB2430およびHMB3210の両方のジャームラインmAb(それぞれHMB2430−GLおよびHMB3210−GL)は、HMB2430−GLの場合はVH.3およびVL.2から、HMB3210−GLの場合はVH.3およびVL.4から成る。ジャームラインの形態のmAbは両方とも、元の体細胞変異したHMB3210およびHMB2430で観察されたのと同等のレベルで、RSVを効果的に中和した(図11)。しかしながら、HMB3210−GLおよびHMB2430−GLはMPVを中和せず、それゆえMPVの中和には体細胞変異が不可欠であることを示している。重鎖もしくは軽鎖の両方の体細胞変異がMPVの中和に関与しているかをさらに理解するため、これらの重鎖もしくは軽鎖のいずれかを、ジャームラインの構成において有する抗体を生産した:VH.3およびVLにより作られるHMB2430−VHGL−VLSM;VH.2およびVL.2により作られるHMB2430−VHSM−VLGL;VH.3およびVLにより作られるHMB3210−VHGL−VLSM;およびVH.2およびVL.4により作られるHMB3210−VHSM−VLGL。HMB3210の重鎖の体細胞変異の除去は、RSVもしくはMPVウイルスの中和に影響を与えなかった。その一方、HMB2430の重鎖の体細胞変異を除去すると、RSVの中和力は維持されたが、MPVの中和に影響を与えた。HMB2430およびHMB3210の軽鎖の体細胞変異の除去は、MPVの中和力を失わせたが、RSVの中和には影響を与えなかった(図11)。まとめると、これらの発見は、HMB3210およびHMB2430は最初にRSVによって選択され、次いで、体細胞変異の蓄積を通じてMPVに対する交差活性を発達させたことを示唆している。全体でわずか3か所の軽鎖CDRの体細胞変異が、RSV特異的ジャームライン抗体がMPV交差活性を獲得した理由である。注目すべきは、HMB2430およびHMB3210(クローン的関係は無い)は、LCDR3における、109位のSからRへの体細胞変異を共有していることである。
C末端ヒスチジンタグを伴ったRSV Fの26〜136および147〜512の残基(RSV A2株の融合ペプチドのないFエクトドメインに対応する)をコードするDNA構築物がコドン最適化され、合成された。組み換えFは、Sf21細胞内でバキュロウイルス発現ベクターを用いて発現され、上清からニッケルアフィニティークロマトグラフィーおよびサイズ排除クロマトグラフィー(SEC)によって精製された。類似の構築物は、既に融合後Fタンパク質の結晶構造を解明するために、他者に使用されている(Swanson et al. PNAS 2011 and McLellan et al. J Viol 2011)。タンパク質は、非還元状態においてSDS−PAGEによって解析され、約65〜70kDaのバンドを示した。また、SECによってS200カラムで解析されたとき、“融合後”RSV Fタンパク質は、三量体Fタンパク質の分子量に相当する、見かけ上約150kDaの分子量を示す一つの対称ピークとして溶出し、ヒトIgG1抗体の溶出体積と重なり合った。“融合後”Fタンパク質は、HMB3210もしくはパリビズマブのいずれかともに培養され、その2つの混合物がS200カラムにかけられた。 “融合後”Fタンパク質とパリビズマブとの培養物は、Fタンパク質単独の場合と比較して溶出ピークの位置がより低い溶出体積側に移動しており(見かけMWが300kDaに相当する)、既に報告されたとおりパリビズマブが“融合後”Fタンパク質と結合されていることを示している。注目すべきは、HMB3210および“融合後”Fタンパク質の培養物は溶出体積の移動が起こらなかったことである(図12)。HMB3210と“融合後”Fタンパク質とが独立した物質として溶出するという事実は、HMB3210はパリビズマブとは異なり、“融合後”Fタンパク質と結合しないことを示す。
我々は、HMB3210およびRSV特異的抗体D25の、中和活性とは異なる抗RSV抗体の特性として報告されている、細胞間のウイルス拡散を防ぐ能力を測定した。我々は、HEP−2細胞をRSV AもしくはB株に感染させ、20時間後に異なる濃度の抗体を加え、3日目にシンシチアの形成を試験することで、50%抗体ウイルス拡散阻止濃度(IS50と定義した)を決定した。両方の抗体ともより高い濃度であれば、ウイルス拡散を阻止することができた。興味深いことに、このアッセイにおいて、HMB3210はより強力な中和抗体であるD25と同等のIS50を示した(図15)。
RSVマウスモデルにおいて、HMB3210はパリビズマブよりも、肺におけるRSVの力価を低減することに関して平均で5〜10倍強力であり、0.12mg/kgかそれ以下の濃度で有効であった(図16a)。MPVマウスモデルにおいても同等に有効であった(図16b)。HMB3210の治療における可能性を試験するため、我々はSTAT1欠損マウスをRSVに感染させ、HMB3210を感染後の第1、2もしくは3日に投与した。ウイルスの複製が低いことによるこのモデルの限界にもかかわらず、HMB3210は全ての時点で治療上の有効性を示し、肺におけるウイルスの力価および炎症性サイトカインを低減した(図17)。
HMB3210のPVMに対する交差反応性を利用した。PVMは、マウスにおいて非常に低い接種量で致命的な病気を起こし、ヒトの重篤なRSVおよびMPV感染の特徴を再現するウイルスである。予防の設定においては、HMB3210は、0.12mg/kgでマウスを死から保護し、0.6mg/kgで体重減少から保護した(図18a)。加えて治療の設定においては、感染後3日以内に30mg/kg投与したとき、および5mg/kg投与したときの両方においてマウスを死から保護し、第4日もしくは第5日に30mg/kg投与したときには著しい保護を与えた(図18b〜d)。この系において、以前に説明されたように(Bonvill et al. 2004, journal of Virology 78:7984-7989)、ヒトの治療において唯一の認可された標準治療であるリバビリンは無効であった。重要なことに、HNB3210の治療的送達は、肺におけるウイルスRNAのさらなる増加を効果的に阻止した(図19)。インビボでのエフェクター機構の役割に焦点を当てるため、IgG1 HMB3210を、補体およびFcレセプター結合を欠く変異体(HMB3210−LALA)と比較した。2種類の抗体は、インビトロでの中和活性を比較され、同等と示された。予防の設定における制限量を投与されたとき(0.12mg/kg)、HMB3210−LALAの活性は大きな減少を示した(図20a)。対照的に、治療の設定において使用したとき、HMB3210−LALAは試験された全ての用量においてHMB3210と同等の効果があった(図20b)。リバビリンが有効ではない、PVM感染モデルにおけるHMB3210の治療上の有効性は、複数の因子、たとえば強力な中和性および拡散阻止活性、融合前タンパク質の選択的な認識でもって、デコイとして働く大量の融合後タンパク質により抗体が消費されないようにする、およびエスケープミュータントを選択しないこと、の組み合わせによるものである。驚くべきことに、PVMマウスモデルにおけるHMB3210の治療上の有効性にはエフェクターの機能は必要なく、このことはインビボの抗体の活性は主としてウイルスの中和およびウイルス拡散の阻止に依存することを示唆している。
HMB3210によって認識されるエピトープを同定するため、ヒトRSVウイルスのFタンパク質の全体の配列をカバーする7095個の構造ペプチドのライブラリーでスクリーニングを行った。実施例5に示される実験は、HMB3210が、ウエスタンブロットにおいて、非還元状態で、RSVおよびMPVのFタンパク質と反応することを示し、このことはHMB3210の標的が、その構造が陰イオン界面活性剤であるSDSの存在下で安定であるエピトープであることを示唆する(図5および図6)。ライブラリーのスクリーニングの手法により、F2のN末端領域における、残基SAVSKGYLSALRTGWYTSVIT(配列番号63)にわたる推定上のHMB3210エピトープを同定した。この領域の中心の配列であるYLSALRTGW(配列番号64)は、RSV、MPV、BRSVおよびPMV間で高度に保存されており(図21)、変異体YLSVLRTGW(配列番号65)、YFSALRTGW(配列番号66)、YFSVLRTGW(配列番号67)、YKSALRTGW(配列番号68)およびYKSVLRTGW(配列番号69)もまたHMB3210により認識される。この配列は、融合後RSV Fタンパク質においては表面に露出していないが、PIV5 Fタンパク質(Yin, et al., 2006, Nature 439;38-44)の構造に基づき作られたRSV融合前Fタンパク質モデルにおいては、パリビズマブの標的であるループ領域に近接した、露出したベータストランドに位置していると予想される(図22)。このマッピングは、実施例10で示された、HMB3210の融合前Fタンパク質への特異性と一致する。このエピトープはパリビズマブにより認識されるものと近いが、異なるものであり、364HRSV、162HMPV、8BRSVおよび5PVM株(図23)を含む551株のウイルスの間で高度に保存されているYLSVLRTGW配列に集中している。
Claims (20)
- 配列番号1、配列番号2および配列番号3でそれぞれ規定される重鎖CDR1、CDR2およびCDR3配列、ならびに、配列番号4、配列番号5および配列番号35でそれぞれ規定される軽鎖CDR1、CDR2およびCDR3配列を含み、RSV、MPVおよびPVMの感染を中和し、
RSV、MPVおよびPVMの融合前のFタンパク質に特異的に結合し、かつRSV、MPVおよびPVMの融合後のFタンパク質に特異的に結合しない、単離された抗体、またはその抗原結合フラグメント。 - グループAおよびグループBの両方のMPVならびにグループAおよびグループBの両方のRSVの感染を中和する、請求項1に記載の抗体、またはその抗原結合フラグメント。
- MPVのサブグループA1、A2、B1およびB2の感染を中和する、請求項1または2に記載の抗体、またはその抗原結合フラグメント。
- 抗原部位I、抗原部位IIまたは抗原部位IVと重複する部位においてRSVのFタンパク質と結合しない、請求項1〜3のいずれか1項に記載の抗体、またはその抗原結合フラグメント。

- 治療上有効量の請求項1〜12のいずれか1項に記載の抗体またはその抗原結合フラグメントを含み、それを必要とする被験体においてRSVもしくはMPV感染を低減するまたはRSVもしくはMPV感染のリスクを低下させるための、医薬組成物。
