1H NMRスペクトルをCDCl3中で400.1MHz(Bruker DRX分光計)にて得た。NOE実験を、異なる混合時間(250から2000ms)を用いた2D位相敏感NOESY実験を使用して脱気試料において実施した。典型的に、位相サイクルについて16パルスおよび2ダミースキャンを用いて1s混合時間を使用した。各次元において0.3Hzのラインブロードニングを用いてデータを処理し、ゼロ充填して、512×4096 2D等高線図を得る。負交差ピークの積分強度を、標準的なBruker NMRソフトウェア特性を使用して決定した。
この調製物は、乾燥THF(1ml)中のNaH(0.050g、2.1mmol)の懸濁液、乾燥THF(1ml)中のトリエチルホスホノセネシオエート(phosphonoseneciaote)(0.11g、0.40mmol)の溶液、乾燥THF(1ml)中のHMPA(0.048g、0.22mmol)および(all−E)−4(0.065g、0.31mmol)の溶液を利用して、エステル(all−E)および(13)−5の2:1混合物を得た(0.12g、78%収率)。この混合物を、ヘキサン中の1%Et.sub.2O、0.5%THFを使用してシリカゲルでHPLCによって分離した。
メタノール(最終濃度0.061M)中のエステル5の溶液(1当量)に、2M KOH(10当量)の水溶液を加えた。この溶液を還流にて加熱し、反応進行をTLCによってモニタした。90分後、加熱溶液を氷(40g)のビーカー内に注ぎ、pH2まで10%HClにより酸性化した。次いで混合物をEt2Oで抽出し、それをブラインで洗浄し、乾燥させ(Na2SO4)、減圧下で濃縮して、生成物を得た。NMRにより、加水分解が異性化せずに生じたことが明らかになった。以下の酸をこの方法によって合成した。
この調製物は、KOH(0.258g、4.61mmol)水溶液(2ml)およびエステル(all−E)−5(0.122g、0.378mmol)のメタノール温溶液(10ml)を利用して、黄色固体として酸(all−E)−UAB30(0.104g、93%収率)を得た:mp 192〜197℃(シクロヘキサン)。
この調製物は、KOH(0.15g、2.8mmol)水溶液(1ml)およびエステル(9Z)−5(0.073g、0.23mmol)のメタノール温溶液(3ml)を利用して、酸(9Z)−UAB30(0.065g、98%収率)を黄色固体として得た:mp 182〜185℃(シクロヘキサン)。
この調製物は、KOH(0.085g、1.50mmol)水溶液(1ml)およびエステル(13Z)−5(0.040g、0.12mmol)のメタノール温溶液(4ml)を利用して、酸(13Z)−UAB30(0.034g、93%収率)を黄色固体として得た:mp 180〜185℃(シクロヘキサン)。
a.概要
化合物UAB30、4Me−UAB30、UAB111およびタザロテンを、無毛SKH1−Eマウスにおいて14日の局所適用の後、起こり得る皮膚効果(表皮および真皮肥厚ならびに皮膚炎)について評価した。タザロテンなどの公知のRAR選択的レチノイドは、局所的に適用される場合、ヒトおよび齧歯動物の皮膚において表皮角化細胞の増殖を刺激する。ケラチノサイト増殖の刺激(表皮過形成として組織学的に報告されている)は、上皮細胞増殖因子受容体活性化の増加に起因すると考えられ、レチノイド効果を反映することが知られている(Fisher G.、Vorhees J.、Molecular mechanism of retinoid actions in skin.FASEB J.1996;10:1002−1013)。表皮過形成と併せて、RAR選択的レチノイドは顕著に皮膚炎に関連している。以前に、RXR選択的レチノイドは、表皮過形成を効果的に誘導しないことが報告されている。驚くべきことに、本発明のRXR選択的化合物は、本明細書において、有効なRAR選択レチノイド(タラゾレン)と比較してケラチノサイト増殖効果を示すと識別された。タザロテンと比較して本発明のRXR選択化合物によって示されるさらなる利点は、局所的に適用される場合、最小または全くない炎症を観察した。
1.試験物質および投薬パターン
試験化合物は、UAB30、4Me−UAB30およびUAB111であった。タザロテン(0.1%クリーム)を、Ricerca Taiwan Ltdにより購入した。0.1%および0.3%における3つの化合物、タザロテン(0.1%クリーム)およびビヒクル(70%エタノール/30%プロピレングリコール)を各々、2週間1日に1回、背部(100μL)および右耳(20μL)に経口投与した。

22±2gの体重のメスのSKH1−Eマウスを、Charles River、USAにより与えられた。動物を、実験の全体にわたって無菌領域下で個々に通気したケージラック(IVCラック、36ミニアイソレーター(Mini Isolator)システム)内に収容した。2または3匹の全てのマウスを動物ケージ(26.7長さ×20.7幅×14.0高さ(cm))に維持し、12時間の明/暗サイクルで制御温度(20〜24℃)および湿度(50%〜80%)下で維持した。動物は、滅菌した標準的な実験用食餌[MF−18(Oriental Yeast Co.,Ltd.Japan)]を自由に取ることができ、無菌の水道水を自由に与えた。この作業の全ての態様、すなわち、動物の収容、実験および処分は、一般的な実験動物のケアおよび使用のガイド(Guide for the Care and Use of Laboratory Animals(National Academy Press、Washington,D.C.、2011))に従って実施した。
無水エタノール(Merck、Germany)、マウスMPO ELISAキット(HK210、Hycult(登録商標)biotech、The Netherlands)、プロピレングリコール(Sigma、USA)およびZorac(登録商標)(タザロテン、1%クリーム)(Allergan、USA)。
動物ケージ(Techniplast、イタリア)、Dyerモデルマイクロメートルゲージ(Peacock、日本)、Pipetman(Gilson、フランス)およびTimer(Wisewind、台湾)。
1.皮膚、局所、過形成、炎症アッセイ
22±2gの体重のメスのSKH1−E無毛マウス(Crl:SKH1−Hrhr)を使用した。動物を各々5匹の群に分けた。1日に1回、100μLのビヒクル、タザロテンおよび試験物質を、マイクロピペットを用いて背面に適用し、背面皮膚の約8cm2(2×4cm)に手袋をはめた指で均一に塗りつけ;20μLを右耳に適用した。試験動物を2週間連続で処置した。体重を一週間に2回モニタした。
背面皮膚における表皮過形成をレチノイド活性のマーカーとして使用した。組織学的評価のために、マウスを15日目に屠殺した。背面皮膚を切断し、フィルタ上で元のサイズに伸ばした。パンチ生検(ED8mm)を得て、10%緩衝化ホルマリン中で固定し、パラフィンに包埋した。5マイクロ厚さの断片を切断し、H&Eで染色し、各生検の代表的な切片を組織学的評価のために選択した。
表皮および真皮の厚さを、接眼レンズにおけるマイクロメートルにより顕微鏡を使用して測定した。詳細を組織病理報告に記載する(以下の組織病理のdを参照のこと)。
背面皮膚における紅斑、落屑(剥離皮膚)および皮膚病変/皮膚剥削を、2週間の処置の終わりに個々の動物において視覚的に評価し、0から4+のスケールでスコア付けした。4+が最大である。
0:正常
1+:軽度の紅斑
2+:中度の紅斑
3+:重度の紅斑/落屑
4+:病変/剥削。
組織構造評価のために、背面皮膚を切断し、フィルタ上に元のサイズに伸ばした。パンチ生検(ED8mm)を得て、10%緩衝ホルマリン中に固定し、パラフィンに包埋した。5マイクロ厚の切片を切断し、H&Eで染色し、各々の生検の代表的な切片を組織構造評価のために選択した。マウスMPO ELISAキット、好中球浸潤の測定によるミエロペルオキシダーゼ活性の測定のために右耳を15日目に収集した。
組織病理の所見は以下の通りであった:
1)表皮の過形成が、ビヒクル処置した群を除いて全ての処置群において示された。市販の製剤の0.1%タザロテンと比較して、UAB30、4Me−UBA30およびUAB111処置は、表皮過形成の同等の発生を生じた。
2)過角化および/または血清細胞痂皮が、0.1%タザロテン、UAB111および0.3% 4Me−UBA30で処置したほとんどのマウス(5匹のうちの3〜5匹)に見られたが、この所見は、ビヒクルおよび0.3%UAB30で処置したマウスにおいて報告されず、0.1%UAB30群の1匹の動物および0.1% 4Me−UBA30群の2匹のマウスにおいてのみ報告された。
メスのSKH1−E無毛マウスにおいて、0.1%および0.3%UAB30、4Me−UBA30およびUAB111の14日の局所適用は、0.1%タザロテン処置と同等の表皮過形成を生じた。0.1および0.3%におけるUAB30ならびに0.1%における4Me−UBA30は、タザロテン処置より顕著に低い炎症および過角化を生じた。
Claims (18)
- 表皮過形成によって治療され得る非腫瘍性皮膚病態を治療、予防または改善するための薬学的に許容可能な皮膚用組成物であって、前記組成物は、皮膚用途のための薬学的に許容可能なビヒクルまたは担体と、式III:
R3は、独立して、H、F、ジフルオロ、CF3、C1−C15分枝鎖もしくは直鎖アルキル基、置換もしくは非置換アリール基、または置換もしくは非置換アラルキル基、置換もしくは非置換シクロアルキル基を含む、1つまたは複数の基であり;
R6およびR7は、独立して、H、CF3、C1−C15分枝鎖もしくは直鎖アルキル基、または置換もしくは非置換シクロアルキル基であり;
R9は、独立して、H、CF3、C1−C15分枝鎖もしくは直鎖アルキル基、置換もしくは非置換アリール基、または置換もしくは非置換アラルキル基、または置換もしくは非置換シクロアルキル基を含む、1つまたは複数の基であり;
R11は、H、またはC1−C15分枝鎖もしくは直鎖アルキル基であり;
nは0〜3である)
として示されるレチノイド化合物またはその薬学的に許容可能な塩とを含む、組成物。 - 前記組成物は、局所投与のために製剤化される、請求項1に記載の組成物。
- 1つまたは2つ以上の式IIIで表されるレチノイド化合物を含む、請求項1に記載の組成物。
- さらに、日焼け止め剤を含む、請求項4に記載の組成物。
- 少なくとも2つの式IIIで表されるレチノイド化合物を含む、請求項4に記載の組成物。
- 少なくとも1つの非レチノイド化合物をさらに含む、請求項4に記載の組成物。
- 非レチノイド化合物をさらに含む、請求項6に記載の組成物。
- 前記式IIIで表されるレチノイド化合物は約0.01%から約10%(w/w)の濃度で提供される、請求項1に記載の組成物。
- 前記式IIIで表されるレチノイド化合物の濃度は約0.1%から約3.0%(w/w)である、請求項9に記載の組成物。
- 前記組成物は経口投与用に製剤化される、請求項1に記載の組成物。
- 前記組成物は錠剤の形態である、請求項11に記載の組成物。
- 前記組成物はカプセル剤の形態である、請求項11に記載の組成物。
- 1つまたは2つ以上の式IIIで表されるレチノイド化合物を含む、請求項11に記載の組成物。
- 少なくとも2つの式IIIで表されるレチノイド化合物を含む、請求項14に記載の組成物。
- 少なくとも1つの非レチノイド化合物をさらに含む、請求項14に記載の組成物。
- 前記式IIIで表されるレチノイド化合物は、約0.01mg/kg体重〜約300mg/kg体重の用量で投与される、請求項11に記載の組成物。
- 前記式IIIで表されるレチノイド化合物は、約0.1mg/kg体重〜約30mg/kg体重の用量で投与される、請求項17に記載の組成物。
