Description
本出願は、参照によって本明細書に取り込まれる2012年10月12日に提出された米国仮特許出願61/713,441号に基づく優先権を、米国特許法第119条(e)の定めにより主張する。
a. scFvファージライブラリのネガティブパニングであって、当該ネガティブパニングが望まない抗原と結合するファージを取り除き、当該ネガティブパニングは、ファージを、
(i)一般的なタンパク質、および
(ii)タウの単量体形態、と連続的に接触させ、
原子間力顕微鏡(AFM)画像を使用して、前記ファージと一般的タンパク質およびタウの単量体形態との結合をモニターし、当該AFMによって抗原と結合するファージを観察しなくなるまで、ステップ(i)および(ii)を繰り返し、一定分量のファージを産生し、
b. 一定分量のファージをタウオリゴマーと接触させ、前記オリゴマーへのファージの結合を可能にするのに十分な時間インキュベートし、並びに
c. 結合したファージ粒子をステップ(b)から溶出する。
(a) ファージを、

本発明は、例えば、以下の項目を提供する。

オリゴマータウを特異的に認識するが、単量体タウ、原繊維タウまたは非疾患関連形態のタウと結合しない抗体または抗体断片。

(項目5)
項目1〜4のいずれか1項に記載の抗体または抗体断片であって、前記抗体断片は、以下のステップ:
a. scFvファージライブラリのネガティブパニングであって、当該ネガティブパニングが望まない抗原と結合するファージを取り除き、当該ネガティブパニングは、ファージを、、
(i)一般的なタンパク質、および

アルツハイマー病(AD)は、蔓延している神経変性疾患であり、進行性ニューロン損失および認知障害が観察される。約500万人のアメリカ人がADを有して生きており、この数字は、2050年までに3倍になると考えられている。残念なことに、治療法はまだ見つかっていない。主な薬理学的アプローチは、アセチルコリン抑制物質のような症状の治療である。ADは、2つの神経病理学的特徴、アミロイドベータ原線維の細胞外沈着または拡散および細胞内のタウの神経原線維変化(NFT)を有する。ますます増える証拠は、タンパク質の誤った折りたたみ、凝集、および原繊維形成の特徴がともにADの病因に密接に関連することを示唆する。正常なタウは、ニューロンの微小管をアセンブルし、かつその構造および生理学的機能を安定化することにおいて重要な役割をはたしているが、異常に過剰リン酸化したタウおよびそのオリゴマー並びに凝集形態は、シナプス損失と関連付けられると考えられる。正常なタウ機能を乱すことなく凝集プロセスにより潜在的に妨げる、単量体または凝集されたタウまたはあらゆる非特異的タンパク質を上回ってオリゴマータウを標的とする試薬が必要とされる。その上、そのような試薬により、凝集されたタウではなく、タウオリゴマーを検出することは、AD症例の早期ステージにおける有望な診断上のアプローチである。
I.AFMバイオパニングプロトコルを用いて、精製されたタウ二量体、三量体、混合されたオリゴマー、および単量体サンプルを使用する異なるタウオリゴマー形態に対するscFvを選択する。
A.SH‐SY5Y神経芽細胞腫株およびそのコリン作動性分化形態でLDH試験によるタウの最も毒性のある形態を選択する。
我々の抗体断片ライブラリーにより標的とするための最も有望なオリゴマータウ種を識別するために、我々は、まず、どのタウ種が神経細胞系に対して毒性があるのかを試験した。我々は、単量体、二量体、または三量体タウで処理された未分化かつコリン作動性様のSH‐SY5Yヒト神経芽細胞腫細胞を用いて細胞生存率分析を行った。我々は、タウの異なる2つのアイソフォーム、4N1Rと4N2Rを試験し、記載されているようにLDH分析用いて毒性を測定した。図2(A)および(B)に示されているように、三量体タウ4N1Rと4N2Rは、未分化のSHSY‐5Y細胞に対する毒性があったが、単量体および二量体タウではなかった。本質的に同じ結果が、SHSY‐5Y細胞が単量体、二量体、または三量体タウにより処理する前にコリン作動性様表現型(データは示さない)へと最初に分化される場合に得られた。三量体タウにより引き起こされる毒性は、濃度依存性を示した。これらの結果は、三量体タウがADの進行において決定的に重要な種であるかもしれないことを示唆する。

BSAおよび単量体タウに対するネガティブパニング後に、我々は三量体タウ4N1Rに対するポジティブセレクションから約100のクローンを回収した。オリゴメトリクス(Oligomerix)により我々に提供されるタウサンプルの量は制限されていたため、我々は、その100のクローンが単量体タウを上回る三量体についての高い特異性を有することを測定するために、従来のELISAによるそれぞれのファージをスクリーニングすることはできなかった。代わりに、我々は、異なるタウ種を結合するためにAFMにより、それぞれの個々のファージクローンをスクリーニングした。我々は、マイカ基板にこれまで固定されていた単量体、二量体、および三量体タウサンプルにより各ファージサンプルをコインキュベートした。非結合ファージが過剰なストリンジェントなリンスにより取り除かれ、残りの結合ファージはAFMにより結像された。このような方法で、すべての100のクローンをスクリーニングした後に、二量体または三量体タウのいずれかには選択的に結合するが、単量体タウには結合しないクローンを我々は識別した。分析はかなりの時間がかかるが、AFMパニングプロトコルは、我々が、たった数ナノグラムの抗原を用いて、結合特異性についてすべての100のクローンをスクリーニングすることを可能にする。すべての100のファージクローンをスクリーニングした後に、我々は、三量体タウについての最も高い特異性に基づくさらなる研究のために6つのクローンを選択した。
我々は、全長scFvがコードされたことを確実にするために、6つのクローンのそれぞれのDNA配列を検証した(図4)。6つのケースのそれぞれにおいて、1つの塩基対がコード配列の始めにて欠失していた。さらなる特徴づけのため可溶性scFvを産生するために、我々は、scFvの効率的な発現を可能にするためにフレームシフトを修正することを必要とした。我々が、ポリメラーゼ連鎖反応(PCR)によりそれぞれのcFv配列を変更し、フレームシフトを修正することを可能にする、フォワードおよびリバースプライマーを我々は設計した(例えば、図3参照)。我々は、その後、識別のためのc‐mycタグおよび精製のためのポリヒスチジンタグをも含んだ修正されたscFv配列を発現プラスミドでクローニングした。修正されたF9クローン、F9Tは、非常に高いレベルで発現し、容易に精製され、かつAFMによりみられるファージ形態において、単量体タウおよび原線維タウを上回るオリゴマータウについての高い特異性を維持し、そのため、このクローンは、さらなる研究のために選択された。
A.年齢のアルツハイマー病および認知障害のない脳切片での選択されたナノボディの予備的特異性および親和性試験
単量体タウは、微小管アッセンブリおよび安定性において重要な役割を果たすが、オリゴマータウは細胞に対して毒性がある。オリゴマータウは、タウの過剰リン酸化および他の翻訳後修飾(posttraslational modification)をもたらかもしれない。オリゴマータウは微小管から外れて(dettach)、その後、アルツハイマー病の顕著な特徴である、神経原線維変化(neurofibillary tangle)を形成するためにさらに凝集するかもしれない。タウの誤った折り畳みおよび凝集が、アミロイド‐ベータ(Aβ)の凝集および誤った折り畳みで親密に結合され、そのため、これらのタンパク質の異なるオリゴマー形態検出は、アルツハイマー病の診断および治療における見込みを有するであろう。我々は、神経外のプラーク度数(plaque frequency)により定義された、異なるBraak病期アルツハイマー病の脳の中側頭回から得られた、ホモジナイズされた死後の脳組織を用いて、三量体タウに対するF9Tナノボディの反応性を分析した。すべての脳のサンプルは同齢のものであり、バナー・サン・ヘルス研究所(Banner Sun Health Research Institute)からThomas G. Beachにより寛大に提供された。我々は、認知障害のないソースから得られた6つのサンプルを分析した(ND1からND6)。NDサンプル、痴呆の明白な症状(sympton)がないことを論証された患者は、Aβプラークの存在:ND1、ND2、およびND3がAβプラークを有しない(Braak病期IからII)が、ND4、ND5、およびND6のすべてがわずかなプラーク度数を有する(Braak病期IからII)に基づく2つのカテゴリーに入れられた。6つのアルツハイマー病の脳組織試料(AD1からAD6)がアルツハイマー病を有すると診断された患者由来であった。サンプルはプラーク度数により分けられ、AD1、AD2、およびAD3脳サンプルが中程度の度数のプラーク(Braak病期IIIからIV)を示し、一方、AD4、AD5、およびAD6脳サンプルが最も深刻なプラーク度数(Braak病期IIIからIV)を示す。両方のF9T調製物は類似の反応性を示し、最も強いシグナルはAD2およびAD3から得られるが、強いシグナルが6つのADサンプルのうち5つにより得られる。興味深いことに、いずれのAβプラークも含まない認識的に正常な組織試料のいずれによっても、ほとんど反応性はない。プラークを含む3つの認識的に正常なサンプルは、上記のように、Aβ凝集とタウ凝集間の相互作用を示唆する反応性を示した。別の興味深い傾向は、AD1〜3サンプルがAD4〜6サンプルよりも平均的により高い反応性を有することである。AD4〜6サンプルの高いプラーク度数が、より多い神経原線維のタウおよびより少ないオリゴマータウの存在を示すかもしれない。
CSFにおける総タウ(T‐タウ)およびリン酸化されたタウ(P‐タウ)レベルは、いくつかの理由のためにアルツハイマー病についての重要なバイオマーカーである。ADのCSFにおけるT‐タウレベルの増加は、高い感受性を有する認知障害のない同齢の対照から、散発性のADを判別し得る。T‐タウレベルは、また、クロイツフェルト・ヤコブ病(CJD)およびレビー小体病(LBD)のような、AD以外の他の痴呆へのより広い使用を可能にする、ニューロンのおよび軸索変性を反映する。他の一般的な痴呆および通常の加齢における正常なレベルのP‐タウと比較して、P‐タウレベルがADにおいてきわだって増加する。この特徴をCSFにおける減少したAβ42と組み合わせ、有望な診断アプローチを得ることができる。アミロイドの他の形態を上回ってタウを認識するF9Tの性能が、そのようなアプローチの重要な部分となり得る。ヒトCSFタンパク質とのF9Tナノボディディスプレイファージの相互作用が原子間力顕微鏡法により結像される。予備的な結果は、ADのCSFが増加したレベルの総タウを含むという事実に従っている、NDにおいてファージがないのに対してADにおいて結合するファージの存在を論証する。パラメータが調製されるかもしれないが、このテストが必要とするのは、CSFタウレベルを検出する潜在的により感受性のある方法並びにADおよび他のタウオパチーについての有望な診断技術である。

神経毒性のオリゴマータウ種について選択的な単鎖可変フラグメントの単離および特徴づけ

scFvファージディスプレイライブラリー‐シートファージディスプレイscFvライブラリー(Sheets, M.D., et al., Efficient construction of a large nonimmune phage antibody library: the production of high−affinity human single−chain antibodies to protein antigens. Proc Natl Acad Sci U S A, 1998. 95(11): p. 6157−62)は、6.7×109の見込まれた多様性を有し、Dr. Yu Zhouにより寛大に提供され(Department of Anesthesia, University of San Francisco)、バイオパニングのために使用した。ファージは、これまでに記載されているように産生されかつ精製された(Marks, J.D., et al., By−passing immunization. オリゴマータウに選択的に結合するscFvの単離

それぞれの修正されたscFv配列についての精製された可溶性のscFvタンパク質は、見込みで29kDa、scFvについての全長サイズを有した。scFvは、合成のオリゴマータウ変異体に対して単離されたので、我々は、その後、精製されたF9T、D11C、およびH2AのscFvが、ADマウスモデルの脳組織における天然由来のタウ凝集体を認識し得るかどうか、およびそれらが脳でランダムなタンパク質と交差反応するかどうかを試験した。オリゴマータウ凝集体と結合したすべての3つのscFvクローンが、野生型マウスと比較して、生後9か月の3×TGADマウスモデル由来の海馬組織ホモジネートにおいて優先的に存在する(図8)。結合された凝集体は、scFvがタウ凝集体に選択的に結合していることを検証するために抗タウ抗体AT8を用いて検出された。AT8による検出は、F9T、D11C、およびH2Aにより標的とされた脳組織試料におけるタウ凝集体は、また、最初の抗原標的がリン酸化されていない場合であっても、リン酸化され得ることも示す。
我々は、どのようにオリゴマータウ濃度が、オリゴマータウに対する3つのscFvを用いて、3×TG‐ADマウスにおいて時間とともに変化するのかを分析した。3×TGマウスモデルにおいて、不溶性タウ変化は、典型的に、生後約12〜15カ月で検出され、しかしながら、我々は、オリゴマータウレベルは、生後5カ月ですでに高く、9カ月でピークとなり、13カ月までに減少することを発見する(図10)。予期されるように、各scFvが同じオリゴマータウ種の異なるエピトープを認識するため、我々はすべての3つのscFvにより類似の傾向をみる。同齢の野生型マウス由来のサンプルは、これらのscFvと反応するあらゆるオリゴマータウの存在を示さなかった。このマウスモデルから得られた結果は、オリゴマータウ種の濃度が、不溶性タウ変化が形成を開始する前に、早期のポイント(5〜9カ月)で増加し、その後、神経原線維変化が形成を開始した後(12〜15カ月)に減少することを示し、オリゴマータウ種がNFTに組み込まれるかもしれないことを示唆する。オリゴマータウ凝集体は、NFTの存在する十分に前の5カ月にすでに存在しているので、それらはADについての早期の診断としての見込みを有する。
scFvは、ADマウスモデル由来の脳組織に存在するオリゴマータウ種を有効に検出したため、我々は、次に、F9TscFvを用いて、オリゴマータウの存在についての異なるBraak病期から死後のヒト中側頭回(MTG)抽出物を探索した(図11)。オリゴマータウは、F9Tを用いて、ヒト脳サンプルにおいて、容易に検出された。興味深いことに、サンプルにおけるオリゴマータウの濃度は、NDのBraak病期I‐IIサンプルにおいて最小のオリゴマータウのみ、軽度のプラークを有するNDのBraak病期I‐IIサンプルにおいてより高い値、中程度のプラークを有するADサンプルにおいて再びより高い値(Braak病期III‐IV)、および重度のプラークを有するADサンプルにおいて最も高いシグナル(Braak病期V‐VI)があるので、Braak病期の増進とともに増加する。プラークを有さないNDサンプルおよび軽度のプラークを有するNDサンプルの両方と比較して、ADサンプルにおけるオリゴマータウのレベルとの間で有意な差がある。これらの結果は、F9Tのような形態特異的試薬は、ヒトサンプルにおけるオリゴマータウ種の存在を検出するために使用され得、ADについての早期診断としてのみならず、疾病のステージの進行を補助するための見込みを有することを示す。
タウおよびAβの凝集体は、顕著な老化プラークおよびADの神経原線維変化の主なタンパク質成分である。多くの研究が、Aβ凝集に続く、下流の事象であると考えられるタウ機能障害によるADの病因およびニューロンの死における、開始因子としてのAβの蓄積および凝集に注目し、一方、他の研究は、タウがADの進行を促進するためにAβと相互作用すること、および低減された凝集タウレベルは、また、ADの症状を寛解させるために重要であることを示唆する。Aβおよびタウ凝集は、共通する上流の原因により駆動されるメカニズムを分離することによりリンクされるかもしれない。数多くの研究は、ADにおける毒性を媒介する可溶性オリゴマーAβ種の役割に関し、証拠は、ここで、オリゴマータウが、また、ADにおける毒性の役割を果たすかもしれないことを示唆している。近年の研究は、オリゴマー、プレフィブリル、および未熟なプレフィラメントの形態を含む可溶性タウ種が、むしろ適応性および保護の役割を代わりに果たすかもしれない、顕著なNFTよりもADにおいてより重大な役割を果たすことを示す。オリゴマータウは、プリオン様の自己繁殖の特徴を有することが示され、ニューロン内に形質膜陥入され得、それらは、インビボで、内在性タウ病理を誘発し得る。よって、AD進行におけるオリゴマーおよび原線維タウ種の役割は、高まる注目を得ており、オリゴマータウはADについての有望な治療の標的である。起こるかもしれない代替的な転写後のスプライシングおよび翻訳後修飾に起因して、ヒト脳に存在するかもしれないタウ種の多様性のために、疾病の進行における種々の形態の役割を探索するために、および診断および治療の標的としてそれらの値を評価するために、個々のタウ凝集体変異体を選択的に識別し得る試薬を開発するための重大な必要性がある。
三量体タウは、低ナノモル濃度でヒトニューロン細胞に対して毒性がある
アルツハイマー病(AD)は、診断後4から8年で一般的に生じる死とともに、進行性の認識の障害、大脳の萎縮、およびニューロンの喪失により特徴づけられる、最も一般的な痴呆の形態である。ADの2つの病理的に顕著な特徴、主にアミロイドベータ(Aβ)からなる細胞外の神経突起のプラークおよび主にタウタンパク質からなる細胞内の神経原線維変化(NFT)は、Dr. Alzheimerにより1907年に最初に識別された。顕著な特徴のプラークおよび変化へとタウおよびAβの凝集を促進するメカニズムを理解することにおいて大きな進歩がなされ、一方、比較的わずかな前進が疾病を停止および治療することにおいて達成された。家族性AD症例の分析は、Aβの誤った折り畳みおよび凝集がAD病因を開始し、タウのリン酸化、凝集、および変化形成のような他の影響の引き金を引くことを述べるアミロイドカスケード仮説の浮上を導く、ADの進行における主要因としてAβの産生に関係する。アミロイド仮説は、10年間以上そのフィールドを支配し、Aβを標的とするいくつかの免疫化研究を含む治療の介入についての数多くの臨床研究を動かした。しかしながら、Aβを標的とするいくつかの臨床試験の失敗は、治療の標的としてのその妥当性に疑いをかけた。ますます増える証拠は、タウもまたADの進行に重要な役割を果たすことを示す。タウの誤った折り畳みおよび凝集が、独立したアミロイド形成を生じさせ得、多くの症例で、タウ病変の存在が、Aβ凝集体の存在なしにADと関連される。可溶性タウレベルの低減なしのAβプラークのクリアランスが、AβおよびタウP301Lを過剰発現する二重のトランスジェニックマウスにおける認識の減退を寛解させるのに不十分である。とりわけこれらの結果は、オリゴマータウがADについての重要な治療の標的であるかもしれないことを示す。
2.1.組換え型ヒトタウ(rhTau)調製および精製。rhTauは、pET21BおよびpET29aベクターにおけるタウ構造によるバクテリア(BL21 DE3)クローン由来の単量体として精製された。標準的な方法が、1mMのIPTGによりタンパク質を増殖させかつ誘導するために使用された。ペレットにされた細胞は、製造者のプロトコルに従って、CelLyticB溶解バッファ、リゾチーム、ベンゾナーゼ(benzonase)、およびプロテアーゼ抑制物質により溶解された(Sigma Aldrich, St. Louis, MO)。陽イオン交換(GEHealthcare Life Sciences)が、両方のタウ構造についてのSPセファロース樹脂による精製の第1のステップについて使用され、タウタンパク質を溶出するために300mMのNaCl入りの25mMのトリス‐HCl pH7.4が使用された。Amiconウルトラ遠心力装置(Millipore)がタンパク質調製物を50mMのトリス‐HCl pH7.4にバッファ交換するために使用された。タンパク質濃度が、BSA分析を用いて測定された(Thermo Fisher Scientific)。タウオリゴマーが、37℃で一晩、100mMのNaCl入りの50mMのトリスバッファ pH7.4中に5μMの濃度で、タウ単量体をインキュベートすることにより生成された。単量体およびオリゴマー種が、6%PAGEにより分離され、溶出され、50mMのトリス‐HClにバッファ交換された。分画は、オリゴマータンパク質の劣化を最小限にするための非還元SDS−PAGE並びにシグナルを高めるためにおよびタウ変異体の純度を検証するために銀染色法により分析された。タンパク質濃度がBSA分析を用いて測定された。
3.1.rhTau凝集体分析。我々は、タウあらゆる翻訳後のリン酸化を除去し、よって、リン酸化が機能の損失またはタウの凝集を有するかもしれないあらゆる潜在的な影響を取り除くための、細菌ホストシステムにおける組換え型ヒトタウを表現した。得られた非リン酸化されたヒト組換え型タウ(NPrhTau)単量体は、遊離チオール類を有する反応性システイン基を含み、タウオリゴマーおよびより高い程度の凝集体を産生するための分子間ジスルフィド結合の形成、および安定した非反応性単量体を作成するための分子内ジスルフィド結合の形成を促進する(図15)。重合反応が、インキュベーション時間およびタンパク質濃度により制御される。2N4Rおよび1N4Rスプライス変異体の両方の、非反応性単量体、二量体、および三量体形態は、SDS‐PAGEおよびAFM高さ分布分析により証明されるように、スプライス変異体の長さおよびオリゴマー形成の程度に応じて定義されたサイズのプロファイルを有する安定した凝集体形態を生成する(図16)。各追加の単量体タウユニットについてのオリゴマー高さの増分は、1N4R変異体について0.5nmおよび2N4R変異体について1.0nmである、所定のアイソフォーム内に固定される(図16)。また、各2N4R種のサイズは、それぞれ、タウ2N4Rが、1N4R変異体と比較して、余分なN末端インサート含むとすると、予想されるように、対応する1N4R種(図16)よりも大きい。
アミロイドカスケード仮説は、少なくとも10年間またはそれ以上にわたって、ADの病因への研究を主導し、ADの発症および進行におけるタウの重要性は、着実により明らかになってきている。タウの病理は、子供および若年成人の症例におけるAβ沈着がないことが観察され、嗅内の海馬領域におけるタウ凝集体は、Aβ病理の発症に先行する。数多くの研究は、Aβの種々のオリゴマー形態がニューロンに対して毒性があり、かつ認識の性能を損ない得ることを示し、よって、ADを診断するための有益なバイオマーカーとしてのそれらの潜在的な役割と関係する。ADにおける種々の可溶性オリゴマーAβ種の重要な役割と同様に、タウの異なる可溶性オリゴマー形態は、また、ニューロンの喪失および認知障害をも生じさせ、ADにおいて重大な役割を果たすかもしれない。よって、ADについての診断および治療の療法を促進するために、疾病の発症および進行を伴うキーとなるタウ種を識別することが重要である。タウが複数のスプライス変異体および翻訳後の変更部位を有するので、我々は、2つの非リン酸化ヒト組換え型タウアイソフォーム、1N4Rおよび2N4Rに注目することにより、タウ形態の複合体の多様性を単純化しようとした。これらの2つのタウの4リピート(4R)アイソフォームは、ともに、微小管関連ドメインのすべての4つの繰り返しを有し、凝集体を安定させるためにジスルフィド結合を形成する追加のシステインおよびヘキサペプチドのモチーフを増強する、微小管‐親和性を有するリピート2の存在に起因して、3つのみのリピート(3R)によるものよりも脳タンパク質キナーゼにより容易にリン酸化された凝集体を形成する傾向がよりある。
Claims (16)
- 配列番号1、配列番号9、配列番号11、または配列番号17のアミノ酸配列を含む抗体断片。
- 抗体断片は、配列番号1、配列番号9または配列番号11のアミノ酸配列を含む請求項1に記載の抗体断片。
- オリゴマータウに結合し、単量体のタウ、原繊維タウまたは非疾患関連形態のタウと結合しない結合分子であって、配列番号1、配列番号9、配列番号11、または配列番号17の配列を含む、前記結合分子。
- 請求項1〜3のいずれか1項に記載の抗体、抗体断片または結合分子を含む、タウの凝集を阻害するための組成物。
- 前記タウの凝集は細胞内にある請求項4に記載の組成物。
- 前記タウの凝集は脳組織内にある請求項4に記載の組成物。
- 抗体、抗体断片または結合分子は、組成物または結合分子の非存在下での速度に比べ、タウ凝集体の形成速度を減少させる、請求項4〜6のいずれか1項に記載の組成物。
- 生体試料中のタウの存在を検出する方法であって、試料を、請求項1〜3のいずれか1項に記載の抗体、抗体断片または結合分子と接触させ、前記組成物と前記組織試料との結合を測定し、前記組成物と前記組織試料との結合は、前記組織試料中のタウオリゴマーの存在を示し、前記タウオリゴマーの前記存在が、初期AD、前頭側頭型認知症、他のタウオパチーまたは外傷性脳損傷後の神経変性を示す、方法。
- 生体試料は、脳組織、血清、脳脊髄液(CSF)、血液、尿または唾液である、請求項8に記載の方法。
- タウオリゴマーは二量体または三量体タウである、請求項8〜9のいずれか1項に記載の方法。
- タウオリゴマーは三量体タウである、請求項10に記載の方法。
