Description
前記第1工程によってメチルチオ化したシステインと、前記シスチンとを、誘導体化剤により誘導体化する、第2工程と、
前記第2工程後のシステイン誘導体及びシスチン誘導体を、光学異性体を区別して分析して、前記システイン及び前記シスチンの各々を、光学異性体を区別して定量する、第3工程と、
を有するシステイン及びシスチンの定量方法が提供される。
次に、本実施形態に係るシステイン及びシスチンの定量方法について、詳細に説明する。図1に、本実施形態に係る定量方法の一例のフロー図を示す。
システイン及びシスチンを含む試料にメチルチオ化剤を添加する、第1工程(S100)と、
前記第1工程によってメチルチオ化した前記システインと、前記シスチンとを、誘導体化剤により誘導体化する、第2工程(S110)と、
前記第2工程後のシステイン誘導体及びシスチン誘導体を分析する、第3工程(S120)と、
を有する。
本実施形態の定量方法により、システイン及びシスチンの各々の含有量を正確に定量できることを確認した実施形態について、説明する。
逆相分離:
カラム:monolithic ODS column(0.53mm i.d.×1000mm)、
流速:25μL/min、
温度:45℃、
移動相:アセトニトリル及びトリフルオロ酢酸を含む水溶液、
とし、
光学分割:
カラム:Sumichiral OA−2500S(1.5mm i.d.×250mm)、
流速:200μL/min、
温度:25℃、
移動相:ギ酸を含むメタノール及びアセトニトリルの混合溶液、
とした。
次に、本実施形態に係る定量方法が、蛍光検出以外の検出方法でも実施可能であることを確認した実施形態について、説明する。
Claims (5)
- システイン及びシスチンを含む試料にメチルチオ化剤を添加する、第1工程と、
前記第1工程によってメチルチオ化したシステインと、前記シスチンとを、誘導体化剤により誘導体化する、第2工程と、
前記第2工程後のシステイン誘導体及びシスチン誘導体を、光学異性体を区別して分析して、前記システイン及び前記シスチンの各々を、光学異性体を区別して定量する、第3工程と、
を有するシステイン及びシスチンの定量方法。 - 前記メチルチオ化剤は、メタンチオスルホン酸S−メチルを含む、
請求項1に記載の定量方法。 - 前記誘導体化剤は、4−フルオロ−7−ニトロ−2,1,3−ベンゾオキサジ アゾールを含む、
請求項1又は2に記載の定量方法。 - 前記第3工程は、蛍光検出液体クロマトグラフ装置を使用して、各々の前記誘導体を検出する工程を含む、
