Description
[1]バルプロ酸及び/又はゾニサミドを有効成分として含有する、神経前駆細胞の移植補助剤。
[2]上記神経前駆細胞を移植する2日前以降に、投与されるように用いられる、上記[1]に記載の移植補助剤。
[3]上記神経前駆細胞がiPS細胞由来である、上記[1]又は[2]に記載の移植補助剤。
[4]ドパミン作動性神経の変性疾患治療用である、上記[1]〜[3]のいずれかに記載の移植補助剤。
[5]上記ドパミン作動性神経の変性疾患がパーキンソン病である[4]に記載の移植補助剤。
[6]1日当り、100〜1200mgの上記バルプロ酸、又は1日当り、10〜600mgの上記ゾニサミドが、ヒトに対して投与されるように用いられる、上記[1]〜[5]のいずれかに記載の移植補助剤。
[7]上記神経前駆細胞を移植する2日前以降に、ヒトに対して投与されるように用いられる、上記[1]〜[6]のいずれかに記載の移植補助剤。
[8]バルプロ酸及び/又はゾニサミドの有効量を、神経前駆細胞が移植された哺乳動物に投与することを含む、移植後の上記神経前駆細胞から誘導されるドパミン作動性神経細胞の残存率を向上させるための方法。
[9]上記哺乳動物がヒトである、上記[8]に記載の方法。
[10]上記神経前駆細胞を移植する2日前以降に、バルプロ酸及び/又はゾニサミドの有効量を投与する、上記[8]又は[9]に記載の方法。

ドパミン作動性ニューロンのマウスiPS細胞からの分化
マウスiPS細胞株である440A3細胞は、10−25継代の後に使用された。Oct3/4、Klf4及びSox2の三つの遺伝子を有するプラスミドベクターによって生じた440A3細胞は、Nanogエンハンサー及びプロモーターの制御下にある、緑色蛍光タンパク質(GFP)及びピューロマイシン耐性遺伝子を有していた。上記GFP遺伝子及びピューロマイシン耐性遺伝子は、440A3細胞が分化していないときにのみ活性化する。440A3細胞において、外来性遺伝子の組込みは報告されていない。
SFEB法を開始してから9日目に、440A3細胞をリン酸生理食塩水(PBS(−))で2度洗浄した。その後、37℃でAccumax(Innovate Cell Technologies社製、商品名)を使用した5分間培養によって440A3細胞を単一細胞に分離した。上記細胞は、FACSバッファーで回収された。上記FACSバッファーは、2%のFBS、20mMのD型グルコース及び1%のペニシリン/ストレプトマイシン(P/S、インビトロジェン社製)を含有するPBS(−)で構成されていた。回収された細胞は、緩やかなピペッティング操作で単一細胞浮遊液へと機械的に分離された。次に、上記細胞をマウス抗PSA−NCAM抗体(希釈倍率1:200,Millipore社製)とともに4℃で30分ほどインキュベートした。その後、遠心分離機を用いて洗浄操作を2度行い、更に上記細胞を二次抗体であるAlexaFluor 594が標識されたロバ抗マウスIgG(希釈倍率1:400,インビトロジェン社製)とともに30分間インキュベートした。死細胞及び細胞の破片は、7−アミノアクチノマイシン−D(7−AAD,BD Pharmigen社製)染色剤を用いて除去した。残った生存細胞は再度、最終濃度(細胞密度)1×107細胞/mlで懸濁させた。細胞ソーティングは、488nmアルゴンレーザー、633nmヘリウム−ネオンレーザー、100μmノズル及びFACSDivaソフトウェアプログラムを備えたFACSAriaIIセルソーター(Becton Dickison社製)を用いて行われた。PSA−NCAM陽性率は、一次抗体を用いていないネガティブコントロールを基準として決定された。
細胞ソーティングの後、細胞の再集合を誘発するため、DMEM/F12培地(Wako社製)において、PSA−NCAM+細胞群を96ウェルプレート上に、20000細胞/ウェルの濃度(細胞密度)で播種した。上記DMEM/F12培地は、1%のN−2サプリメント、200nMのアスコルビン酸、2%のB27サプリメント(インビトロジェン社製)、0.5mMのL−グルタミン及び1%のP/Sを含んでいた。アポトーシスを防ぐために、ROCK阻害剤Y−27632(Wako社製)が、細胞ソーティングの過程とその後の一晩の培養とにおいて、30μMの濃度で使用された。SFEB法を開始してから10日目に、バルプロ酸(VPA)(Sigma社製)、ゾニサミド(ZNS)ナトリウム塩(大日本住友製薬株式会社製)、17β エストラジオール(E2)(Sigma社製)、グリア細胞株由来神経栄養因子(GDNF)(R&D Systems社製)、又はPBS(−)のいずれかが、4日間培地に加えられた。VPA、ZNS及びE2はそれぞれ三つの異なる濃度で使用された。すなわち、VPAの濃度は0.01mM、0.1mM及び1mMであり、ZNSの濃度は1μM、10μM及び100μMであり、E2の濃度は1nM、10nM及び100nMであった。GDNFは、20mg/ml加えて、ポジティブコントロールとした。VPA及びE2の効果を中和するため、アデニル酸シクラーゼ阻害剤である2,5−ジデオキシアデノシン(ddA,100μM;Santa Cruz Biotechnology社製)又はエストロゲン受容体拮抗薬であるICI182780(ICI,2μM;Wako社製)が、SFEB法を開始してから10日目に培地に加えられた。
生後10週間のSprague−Dawleyラット(SDラット、清水実験材料株式会社製)は、京都大学動物実験ガイドラインに従って取り扱われた。上記SDラットに麻酔をかけ、両側の線条体中にドナー細胞を定位固定的に注射することによって移植した。SFEB法を開始してから9日目の二つの細胞集合体(平均3.1x105細胞)を1μlのPBS(−)中に集めた状態で、ドナー細胞として各トラクトへの移植に使用した。上記PBS(−)には、終濃度で30μMのY−27632が加えられていた。その後、VPA(150mg/kg/日)、ZNSナトリウム塩、(30mg/kg/日)、E2(80μM/kg/日)、又は生理食塩水の腹腔内注射を、上記ドナー細胞を移植する2日前から施し、剖検される日まで続けた。免疫抑制のために、全てのSDラットに対して、1日用量である10mg/kgのサイクロスポリンA(CsA、Wako社製)を投与した。ドナー細胞の移植後4週間で、上記SDラットは、深麻酔の下、4%のパラホルムアルデヒドを心臓内にかん流することによって脳を洗浄及び固定した。剖検の日、最後の試験化合物又はCsAの注射から1時間後に各SDラットから血液サンプルが採取された。これらのサンプルはSRL,Inc.(東京、日本)に送られ、上述の投与された医薬(試験化合物)の血中濃度が測定された。
生後12週間のSCIDラット(京都大学医学部動物実験施設にて作製)は京都大学動物実験ガイドラインに従って取り扱われた。上記SCIDラットに麻酔をかけ、両側の線条体中にドナー細胞を定位固定的に注射することによって移植した。ヒトiPS細胞株である1039A1細胞から調製したドパミン神経前駆細胞(平均2.7x105細胞)を2μlのPBS(−)中に集めた状態で、ドナー細胞として各トラクトへの移植に使用した。上記PBS(−)には、終濃度で30μMのY−28632が加えられていた。その後、VPA(150mg/kg/日若しくは600mg/kg/日、それぞれ高用量、低用量という場合がある)、ZNSナトリウム塩(30mg/kg/日若しくは60mg/kg/日、それぞれ高用量、低用量という場合がある)、又は生理食塩水の腹腔内注射を、上記ドナー細胞を移植する2日前から施し、剖検される日まで続けた。ドナー細胞の移植後4週間で、上記SCIDラットは、深麻酔の下、4%のパラホルムアルデヒドを心臓内にかん流することによって脳を洗浄及び固定した。剖検の日、最後の試験化合物の注射から1時間後に各SCIDラットから血液サンプルが採取された医薬(試験化合物)の血中濃度が測定された。
トータルRNAはRNeasy Plus Miniキット(Qiagen社製)を使用して抽出された。抽出されたトータルRNAは、Super Script III First−Strand Synthesis System(インビトロジェン社製)を使用して逆転写された。各PCRは、Hot StarTaq DNA ポリメラーゼ(Qiagen社製)を使用して行われた。逆転写酵素を加えないことで、各プライマーに対するコントロール増幅反応が行われた。MEFがその他のネガティブコントロールとして使用された。RT−PCRの検出対象となった遺伝子の配列はいずれも公知であり、その遺伝子の配列に基づいて、プライマーの設計及び増幅産物の分子量の見積もりが行われた。
インビトロ実験では、SFEB法を開始してから14日目に上述の試験化合物で処理された細胞集合体を、4%のパラホルムアルデヒドで固定した。その後、固定した細胞集合体を冷凍し、免疫細胞化学用のミクロトームを使用して10μM厚の薄片に切断した。一方、インビボ実験(移植実験)では、移植実験の後SDラット又はSCIDラットの脳を取り出し、4%のパラホルムアルデヒドで再度2日間固定した。その後、固定したSDラット又はSCIDラットの脳を30%のスクロース中で3日間低温保存し、冷凍し、免疫組織化学のために40μM厚の薄片に切断した。スフィア(球状の細胞塊)及び脳の凍結切片に透過処理を施し、PBS(−)中に室温で一時間ブロッキングすることで、サンプルとした。上記PBS(−)には、0.3%のトライトン−X及び2%のロバ血清が含まれていた。その後、上記切片を一次抗体とともに4℃で一晩インキュベーションを行った。本実施例で使用した一次抗体は、ウサギ抗チロシンヒドロキシラーゼ抗体(希釈倍率1:400,TH;Millipore社製)、マウス抗TH抗体(希釈倍率1:200,Millipore社製)、ヒツジ抗TH抗体(希釈倍率1:400,Millipore社製)、マウス抗Tubβ3抗体(希釈倍率1:1000,Tujl;Covance社製)、ラット抗NURR1抗体(希釈倍率1:1000,カン研究所、神戸、日本)、ウサギ抗Ki67抗体(希釈倍率1:1000,Novocastra社製:NCL−Ki67p)、ウサギ抗カスパーゼ3抗体(希釈倍率1:500,Santa Cruz Biotechnology社製)、ラット抗M2M6抗体(希釈倍率1:50,Developmental Study Hybridoma Bank社製)、マウス抗Nestin抗体(希釈倍率1:50;Millipore社製)、ウサギ抗Pitx3抗体(希釈倍率1:500;Chemicon社製)、ヤギ抗HNF−3β抗体(希釈倍率1:500,Foxa2;Santa Cruz Biotechnology社製)、マウス抗ヒトNuclei抗体(希釈倍率1:1000;Millipore社製)及びマウス抗NeuN抗体(希釈倍率1:500,Chemicon社製)である。PBS(0.05%のTween−20)中で3度洗浄した後、サンプルをAlexa Fluor結合二次抗体とともに室温で1時間インキュベートした。さらに3度洗浄した後、上記サンプルを核染色のためDAPIとともにインキュベートし、パーマフロー(Dako)を使用して標本にした。免疫反応性細胞は、共焦点レーザーマイクロスコープ(Fluoview FV1000D;Olympus社製)で可視化した。各マーカーの陽性細胞の割合を決定するため、標識細胞を、少なくとも3度の独立した実験において手動で数えた。Ki67+/Nestin+細胞の移植片の体積及び数は、BZ−II解析ソフトウェアプログラム(Keyence)を使用して決定した。各移植片における免疫反応性細胞の数を予測するため、6片毎に細胞数を手動で数え、Abercrombie Correctionを適用した(Abercrombie,1946)。
統計分析はグラフパッドプリズムソフトウェアプログラム5.0bバージョン(GraphPad Software)を使用して行った。全ての量的データは平均値±SD(標準偏差)として示され、One−way ANOVA及びNewman−Keuls事後解析テストが使用された。差はP<0.05で統計的に有意と判定した。

SFEB法を用いてマウスiPS細胞株である440A3細胞からドパミン作動性ニューロンを分化誘導した。上記440A3細胞は、SFEB法を開始してから14日目まで継続して増殖していた。分化誘導に伴って、Nanog−GFPの発現が徐々に減少し、SFEB法を開始してから9日目には、ほとんど発現が確認されなかった。各遺伝子マーカーの経時的な発現変化を示したRT−PCRの写真を図2に示す。多能性を示す細胞集団(Oct3/4+、Nanog+)が、SFEB法を開始してから6〜9日目に未熟神経前駆細胞(NPC)(Nestin+)に分化し、その後Tuj1+神経細胞に分化したことが確認された(図2)。このTuj1+神経細胞は、ドパミン作動性ニューロンの特異的マーカーであるLmx1a、Nurr1及びTHも発現していた。
VPA、ZNS及びE2がインビトロにおいて、ドパミン作動性ニューロンへの分化誘導に影響があるか調べた。SFEB法を開始してから10日目〜14日目の間、再集合したPSA−NCAM+細胞を、VPA、ZNS又はE2の存在下で培養した。SFEB法を開始してから14日目に免疫細胞化学法を行ったところ、どの試験化合物においても90%以上の細胞が神経細胞マーカーであるTuj1を発現していた(図3(A))。コントロールの細胞では、5.2±1.1%がTH+であった。これに対し、VPA(0.01mM、0.1mM)又はE2(10nM)の存在下で培養した細胞は、TH+細胞の割合が、コントロールに対して約2倍にまで増加していた(それぞれ、12.1±1.5%、11.7±0.4%及び12.2±2.3%)(図3(B))。VPA及びE2のこのような効果が環状AMP経路又はエストロゲン受容体を介して行われているのかどうかを調べるため、アデニル酸シクラーゼ阻害剤であるddA又はエストロゲン受容体拮抗剤であるICIをそれぞれ用いた。100μMのddA又は2μMのICIをそれぞれ0.1mMのVPA又は10nMのE2と共に細胞に加えて培養したところ、TH+細胞の増加の割合が顕著に減少した(図3(C))。一方、ddA又はICIを単独で細胞に加えてもコントロールと比較して、TH+細胞の割合に変化は見られなかった。
次に、VPA、ZNS又はE2の全身投与が、移植片中のドパミン作動性ニューロンの生存率及び分化に影響を及ぼすかどうか調べた。この移植実験では、SFEB法を開始してから9日目に、FACSによるソートを行っていない細胞集団(2つの細胞集合体中に3.1×105細胞、PBS中)をSDラットの線条体に移植した。移植に使用したSDラットは、移植を行う日の2日前から殺処分される日まで(移植後4週間)毎日、上述の薬剤のうちの1つ及び免疫抑制剤であるCsAを腹腔内投与された。殺処分の日、CsAの血中濃度は平均で3700±898ng/mlであった。VPA、ZNS及びE2の血中濃度は、それぞれ、158.5±3.9μg/ml、2.43±0.13μg/ml及び1141±926pg/mlであった。
移植後4週間目の移植片中におけるTH+細胞の数を、4つのグループ間で比較した。二重標識を用いた免疫組織化学法を行ったところ、VPAを投与したSDラットの移植片では、コントロールのSDラットの移植片と比較して、TH+細胞の数が顕著に多かった(それぞれ、1396±864細胞、393±311細胞、図7(B))。コントロールのSDラットの移植片において、TH+細胞のごく一部が中脳マーカーであるFOXA2を共発現していた(24.7±9.3%)。これに対して、VPA又はZNSを投与したSDラットの移植片では、大部分のTH+細胞が、FOXA2+であった(それぞれ、81.8±33.6%及び80.4±21.1%)。統計学的な分析によって、VPA又はZNSを投与したSDラットの移植片中における中脳ドパミン作動性ニューロン(TH+FOXA2+)の数は、コントロールのSDラットの移植片と比較して、有意に増加していることが示された(それぞれ、984±770細胞、835±540細胞及び97±76細胞、図7(C))。これらの結果から、VPA又はZNSの全身投与によって、移植した神経前駆細胞から分化したドパミン作動性ニューロンの残存率が向上することが示唆された。ZNSを投与した場合、移植したNPCの神経細胞への分化の割合が、コントロールと同程度であること(図6(B))から、移植後の残存率と、NPCの神経細胞への分化の効率とは互いに独立した関係にあることが示唆された。
移植後4週間目の移植片中におけるTH+細胞の数を、5つのグループ間で比較した。二重標識を用いた免疫組織化学法を行ったところ、高用量のZNSを投与したSCIDラットの移植片では、コントロールのSCIDラットの移植片と比較して、TH+細胞の数が顕著に多かった。(それぞれ、6480±2145細胞、3026±1349細胞、図8(A)。図中の「*」は、p<0.05であることを示す。)。コントロールのSCIDラットの移植片においては、TH+細胞のうち中脳マーカーであるFOXA2を共発現していた細胞が76.3±9.7%であったのに対し、高用量のZNSを投与したSCIDラットの移植片では、TH+細胞のうち91.8±6.2%がFOXA2+であった。統計学的な分析によって、高用量のZNSを投与したSCIDラットの移植片中における中脳ドパミン作動性ニューロン(TH+FOXA2+)の数は、コントロールのSCIDラットの移植片と比較して、有意に増加していることが示された(それぞれ、5889±1821細胞、2297±1116細胞、図8(B)。図中の「*」及び「**」は、それぞれ、p<0.05、及び、p<0.01であることを示す。)。これらの結果から、ZNSの全身投与によって、移植した神経前駆細胞から分化したドパミン作動性ニューロンの残存率が向上することが示唆された。
