Description
本発明による血液試料内の標的分子の例証的な検出は、内部全反射(FTIR)に基づく検出を使用したMagnotech(登録商標)バイオセンサシステム上で実行される。当該システムは、ハンドヘルド式分析機器及び使い捨ての自蔵カートリッジを含む。標的分子の検出に必要な全てのアッセイ試薬は、カートリッジ内に含有される。試料の液滴が与えられると、毛管力によって試料は自動的にカートリッジ内に導かれる。そこで、抗体で官能性を持たされた磁気ナノ粒子が、標的分子に結合し、さらに、検出のためのラベルとして役立つ。磁力が、磁気ナノ粒子をセンサ表面まで移送するために使用され、センサ表面も、標的分子が結合することができる抗体で官能性を持たされている。実験準備が、図1において描かれている。磁気粒子の磁気作動が、2つの磁石、すなわち上方磁石(洗浄磁石)及び下方磁石(結合磁石)の交互の作用を介して発生する。光が、臨界角よりもわずかに狭い角度でカートリッジの表面上に放出され、カメラセンサ上で画像処理される反射を引き起こす。ナノ粒子がセンサ表面上で結合する場合、光の一部が反射且つ散乱され、磁気粒子の非存在下での測定と比較して強度を減らす。
実施例1において例証されたのと同じ測定を、メタ重亜硫酸ナトリウムの代わりにアスコルビン酸を使用して実行することができる。
Claims (23)
- 血液試料における少なくとも1つの標的分子を検出するための方法であって、少なくとも以下の、
(a)血液試料を提供するステップ、
(b)抗酸化物質を含むフィルターに前記試料を与えて前記試料から血液細胞を分離するステップ、
(c)前記標的分子を検出するステップ、
を含み、
前記標的分子の検出は、少なくとも以下の、
(c1)第1の結合分子に前記試料を接触させるステップであり、前記第1の結合分子は磁気粒子に付着させられ、さらに、前記第1の結合分子は前記少なくとも1つの標的分子に特異的に結合する能力を持つ、ステップ、
(c2)第2の結合分子に前記試料を接触させるステップであり、前記第2の結合分子はセンサ表面に付着させられ、さらに、前記第2の結合分子は、前記少なくとも1つの標的分子に特異的に結合する能力を持つ、ステップ、及び、
(c3)前記センサ表面上の前記磁気粒子を検出するステップ、
を含む、方法。 - 前記ステップ(c1)及び(c2)は同時に行われる、請求項1に記載の方法。
- 請求項1に記載の血液試料における少なくとも1つの標的分子を検出するための方法における抗酸化物質の使用。
- 血液試料における少なくとも1つの標的分子を検出するためのアッセイ装置内に挿入するためのカートリッジであって、抗酸化物質を含む、血液細胞を除去するためのフィルターを含むカートリッジ。
- (iii)磁気粒子に付着させられる第1の結合分子、及び、
(iv)センサ表面に付着させられる第2の結合分子、
をさらに含み、
前記第1の結合分子及び前記第2の結合分子は、前記少なくとも1つの標的分子に特異的に結合する能力を持つ、請求項4に記載のカートリッジ。 - 前記抗酸化物質は前記フィルター内に組み込まれる、請求項4又は5に記載のカートリッジ。
- 還元剤をさらに含む、請求項4、5又は6に記載のカートリッジ。
- 前記アッセイ装置は、ハンドヘルド式アッセイ装置である、請求項4、5、6又は7のうちいずれか一項に記載のカートリッジ。
- 前記標的分子は副甲状腺ホルモンである、請求項1又は2に記載の方法。
- 前記抗酸化物質はフェノール性抗酸化物質である、請求項1、2又は9に記載の方法。
- 前記抗酸化物質は、アスコルビン酸、メタ重亜硫酸ナトリウム、Trolox(登録商標)、5−ブロモ−5−ニトロ−1,3−ジオキサン、エトキシフェノール、フェニルサリチレート、ビタミンA、ビタミンE、BSA、メラトニン、アルファカロチン、ベータカロチン、Aspirin(登録商標)、Topanol(登録商標)OC、Irganox(登録商標)1076、Ionox(登録商標)220、Ethanox(登録商標)330、Cyanox(登録商標)2246及びIrganox(登録商標)1010を含む群から選択される、請求項1、2、9又は10に記載の方法。
- 前記標的分子は副甲状腺ホルモンである、請求項3に記載の使用。
- 前記標的分子は副甲状腺ホルモンである、請求項4、5、6、7又は8に記載のカートリッジ。
- 前記抗酸化物質はフェノール性抗酸化物質である、請求項3又は12に記載の使用。
- 前記抗酸化物質はフェノール性抗酸化物質である、請求項4、5、6、7、8又は13に記載のカートリッジ。
- 前記抗酸化物質は、アスコルビン酸、メタ重亜硫酸ナトリウム、Trolox(登録商標)、5−ブロモ−5−ニトロ−1,3−ジオキサン、エトキシフェノール、フェニルサリチレート、ビタミンA、ビタミンE、BSA、メラトニン、アルファカロチン、ベータカロチン、Aspirin(登録商標)、Topanol(登録商標)OC、Irganox(登録商標)1076、Ionox(登録商標)220、Ethanox(登録商標)330、Cyanox(登録商標)2246及びIrganox(登録商標)1010を含む群から選択される、請求項3、12又は14に記載の使用。
