Description
本発明を記述する前に、記述された特定の方法および実験条件は変化することがあるため、本発明はこのような方法および条件に限定されないことを理解しなければならない。また、本発明の範囲は、添付の特許請求の範囲によってのみ限定されるため、本明細書に用いる専門用語は、特定の実施態様を記述するためだけにあり、限定を意図しないことを理解しなければならない。
本明細書に用いる「ErbB3」および「ErbB3フラグメント」という発現は、非ヒト種からのもの(たとえば、「マウスErbB3」、「マウスErbB3フラグメント」、「サルErbB3」、「サルErbB3フラグメント」、など)であると明記されていなければ、ヒトErbB3タンパク質またはフラグメントのことをいう。ヒトErbB3の細胞外ドメインは、示されるアミノ酸配列を有する、例えば、配列番号のアミノ酸1−613:497−499。
本発明の抗体は、ErbB3とそのリガンドニューレグリン−1(NRG1)との間の相互作用を遮断する。本明細書に用いるように、「ErbB3とNRG1との間の相互作用を遮断する」という表現は、ErbB3とNRG1との間の物理的相互作用を検出および/または定量化することができるアッセイにおいて、本発明の抗体を添加するとErbB3とNRG1との間の相互作用が少なくとも50%低減することを意味する。抗体がヒトErbB3とNRG1との間の相互作用を遮断するかどうかを決定するために用いることができる非限定的な例となるアッセイを、本明細書において実施例4に説明する。この実施例では、抗体をErbB3タンパク質と混合し、次いで抗体/ErbB3混合物をNRG1タンパク質でコーティングした表面に塗布する。非結合分子を洗浄した後、NRG1コーティングした表面に結合したErbB3の量を測定する。このアッセイフォーマットでは様々な量の抗体を用いて、NRG1に対するErbB3の結合の50%を遮断するのに必要な抗体の量を算出し、IC50値として表すことができる。本発明は、上記のようなErbB3/NRG1結合アッセイ、または実質的に類似したアッセイにおいて試験したときに約600未満pmのIC50を示す抗ErbB3抗体を含む。たとえば、本発明は、上記のようなErbB3/NRG1結合アッセイ、または実質的に類似したアッセイにおいて試験したときに、約600、500、400、300、290、280、270、260、250、240、230、220、210、200、190、180、170、160、150、140、130、120、110、100、90、80、70、60、50、40、30、20、10、18、16、14、12、10、9、8、7、6、5、4、3、2または1pM未満のIC50を示す抗ErbB3抗体を含む
ErbB3タンパク質は、すべてのErbB/HERファミリーメンバーと同様に、「ドメインI」、「ドメインII」、「ドメインIII」および「ドメインIV」と称する4つの細胞外ドメインを含む。ドメインIは、配列番号:498のアミノ酸1〜190によって表されるアミノ酸の配列であり;ドメインIIは、配列番号:498のアミノ酸191〜308によって表されるアミノ酸の配列であり;ドメインIIIは、配列番号:498のアミノ酸309〜499によって表されるアミノ酸の配列であり;そしてドメインIVは、配列番号:498のアミノ酸500〜624によって表されるアミノ酸の配列である。
完全ヒトモノクローナル抗体を含むモノクローナル抗体を生成する方法は、当分野で知られている。本発明の文脈では、任意のこのような知られている方法を用いてヒトErbB3に特異的に結合するヒト抗体を作ることができる。
本発明の抗ErbB3抗体および抗体フラグメントは、記述された抗体のものとは異なるが、ヒトErbB3を結合する能力を保持するアミノ酸配列を有するタンパク質を包含する。このような変異抗体および抗体フラグメントは親配列と比較したときにアミノ酸の1つまたはそれ以上の付加、欠失または置換を含むが、記述された抗体のもの本質的に同等の生物活性を示す。同様に、本発明の抗ErbB3抗体をコードするDNA配列は、開示された配列と比較したときにヌクレオチドの1つまたはそれ以上の付加、欠失または置換を含むが、本発明の抗ErbB3抗体または抗体フラグメントと本質的に生物学的に同等である抗ErbB3抗体または抗体フラグメントをコードする配列を包含する。このような変異アミノ酸およびDNA配列の例は、上で議論されている。
本発明の特定の実施態様によれば、抗ErbB3抗体は、ヒトErbB3に結合するが、他の種からのErbB3には結合しない。また、本発明は、ヒトErbB3に結合し、かつ1つまたはそれ以上の非ヒト種からのErbB3に結合する抗ErbB3抗体を含む。たとえば、本発明の抗ErbB3抗体は、ヒトErbB3に結合してもよく、かつ、場合によっては、マウス、ラット、モルモット、ハムスター、スナネズミ、ブタ、ネコ、イヌ、ウサギ、ヤギ、ヒツジ、ウシ、ウマ、ラクダ、カニクイザル、マーモセット、アカゲザルまたはチンパンジーのErbB3の1つまたはそれ以上に結合してもよいし、または結合しなくてもよい。
本発明は、サイトトキシン、化学療法薬、免疫抑制薬または放射性同位体のような治療部分に結合した抗ErbB3モノクローナル抗体(「免疫複合体」)を包含する。細胞傷害性薬剤には、細胞に有害なあらゆる薬剤が含まれる。免疫複合体を形成するのに適した細胞傷害性薬剤および化学療法剤の例は、当分野で知られている(たとえば、WO05/103081を参照のこと)。
本発明の抗体は、単一特異性、二重特異性、または多重特異性であってもよい。多重特異性抗体は、1つの標的ポリペプチドの異なるエピトープについて特異的であってもよいし、または1つを超える標的ポリペプチドについて特異的な抗原結合性ドメインを含んでもよい。たとえば、Tutt et al., 1991, J. Immunol. 147:60-69; Kufer et al., 2004, Trends Biotechnol. 22:238-244を参照のこと。本発明の抗ErbB3抗体は、別の機能性分子、たとえば、別のペプチドまたはタンパク質に結合するか、またはそれと共に共発現することができる。たとえば、抗体またはそのフラグメントは、1つまたはそれ以上の他の分子単位、たとえば別の抗体または抗体フラグメントに機能的に結合して(たとえば、化学的カップリング、遺伝的融合、非共有結合的会合またはその他によって)、第2の結合特異性を有する二重特異性または多重特異性抗体を産生することができる。たとえば、本発明は、免疫グロブリンの1つのアームが、ヒトErbB3またはそのフラグメントについて特異的であり、かつこの免疫グロブリンの他のアームが第2の治療標的(たとえば、EGFR、EGFRvIII、ErbB2/HER2、ErbB4、VEGFまたはAng2)について特異的であるか、または治療部分に結合している二重特異性抗体を含む。
本発明は、本発明の抗ErbB3抗体またはその抗原結合性フラグメントを含む医薬組成物を提供する。本発明の医薬組成物は、適した担体、賦形剤および改善された伝達、送達、耐性などをもたらす他の薬剤を用いて処方する。多くの適当な製剤は、すべての薬剤師に知られている処方集:Remington's Pharmaceutical Sciences, Mack Publishing Company, Easton, PAに見出すことができる。これらの製剤としては、たとえば、散剤、ペースト剤、軟膏剤、ゼリー、ロウ、油、脂質、ベシクルを含む脂質(カチオン性またはアニオン性)(たとえばLIPOFECTINTM)、DNAコンジュゲート(DNA conjugates)、無水吸収ペースト剤、水中油型および油中水型乳剤、エマルジョンカーボワックス(emulsions carbowax)(さまざまな分子量のポリエチレングリコール)、半固体ゲルおよびカーボワックスを含む半固体混合物が挙げられる。また、Powell et al. “Compendium of excipients for parenteral formulations” PDA (1998) J Pharm Sci Technol 52:238-311を参照のこと。
本発明の抗体は、とりわけ、ErbB3活性に関連するか、もしくはそれによって媒介されるか、またはErbB3とErbB3リガンド(たとえば、NRG1)との間に相互作用を遮断することによって治療できるあらゆる疾患または障害の治療、予防および/または回復に有用である。本発明の抗体および抗原結合性フラグメントは、たとえば、脳および髄膜、中咽頭、肺および気管支樹、消化管、男性および女性生殖器系、筋肉、骨、皮膚および付属器、結合組織、脾臓、免疫系、造血細胞および骨髄、肝臓および尿路、ならびに眼のような特殊な感覚器官に生じる原発性および/または転移性腫瘍の治療に用いてもよい。特定の実施態様では、本発明の抗体および抗原結合性フラグメントを、以下のがん:腎細胞がん、膵がん、乳がん、頭頸部がん、前立腺がん、悪性神経膠腫、骨肉腫、結腸直腸がん、胃がん(たとえば、MET増幅(amplification)による胃がん)、悪性中皮腫、多発性骨髄腫、卵巣がん、小細胞肺がん、非小細胞肺がん(たとえば、EGFR依存性非小細胞肺がん)、滑膜肉腫、甲状腺がんまたは黒色腫の1つまたはそれ以上の治療に用いる。
本発明は、本発明の抗ErbB3抗体を少なくとも1つのさらなる治療活性成分と併用して投与することを含む治療投与レジメンを含む。このようなさらなる治療活性成分の非限定的な例としては、他のErbB3アンタゴニスト(たとえば、第2の抗ErbB3抗体またはErbB3の小分子阻害剤)、ErbB2/HER2のアンタゴニスト(たとえば、抗HER2抗体[たとえば、トラスツズマブ]またはHER2活性の小分子阻害剤)、ErbB4のアンタゴニスト(たとえば、抗ErbB4抗体またはErbB4活性の小分子阻害剤)、上皮増殖因子受容体(EGFR)のアンタゴニスト(たとえば、抗EGFR抗体[たとえば、セツキシマブまたはパニツムマブ]またはEGFR活性の小分子阻害剤[たとえば、エルロチニブまたはゲフィチニブ])、EGFRvIIIのアンタゴニスト(たとえば、EGFRvIIIを特異的に結合する抗体)、VEGFアンタゴニスト(たとえば、VEGF−Trap、たとえば、US7,087,411参照(本明細書においては「VEGF阻害性の融合タンパク質」とも称する)、抗VEGF抗体(たとえば、ビバシズマブ)、VEGF受容体の小分子キナーゼ阻害剤(たとえば、スニチニブ、ソラフェニブまたはパゾパニブ)、または抗DLL4抗体(たとえば、US2009/0142354に開示された抗Dll4抗体、たとえばREGN421)、など)が挙げられる。本発明の抗ErbB3抗体と併用して有益に投与してもよい他の薬剤としては、小分子サイトカイン阻害剤およびサイトカイン、たとえばIL−1、IL−2、IL−3、IL−4、IL−5、IL−6、IL−8、IL−9、IL−11、IL−12、IL−13、IL−17、IL−18またはそれらのそれぞれの受容体に結合する抗体を含む、サイトカイン阻害剤を含む。また、本発明は、本明細書に記載された抗ErbB3抗体のいずれかおよび1つまたはそれ以上のVEGF、DLL4、EGFRの阻害剤、または上記のサイトカインのいずれかを含む治療的併用を含み、ここで、阻害剤は、アプタマー、アンチセンス分子、リボザイム、siRNA、ペプチボディ(peptibody)、ナノ抗体または抗体フラグメント(たとえば、Fabフラグメント;F(ab’)2フラグメント;Fdフラグメント;Fvフラグメント;scFv;dAbフラグメント;または、他の改変分子、たとえば二重特異性抗体、三重特異性抗体、四重特性抗体、ミニ抗体および最小限の認識ユニット)である。また、本発明の抗ErbB3抗体は、抗ウイルス剤、抗生物質、鎮痛剤、コルチコステロイドおよび/またはNSAIDと併用して投与してもよい。本発明の抗ErbB3抗体は、放射線治療および/または従来の化学療法も含む治療レジメンの一部として投与してもよい。さらなる治療活性成分を、本発明の抗ErbB3抗体の投与の前に、同時に、または後に投与してもよい(本開示の目的では、このような投与レジメンは、本発明の治療活性成分「と併用した」抗ErbB3抗体の投与と考えられる)。
本発明の特定の実施態様によれば、抗ErbB3抗体の複数用量(multiple doses)を、所定の時間経過にわたって対象に投与してもよい。本発明のこの態様による方法は、抗ErbB3抗体の複数用量を対象に順に投与することを含む。本明細書に用いるように、「順に投与する」とは、抗ErbB3抗体の各用量を、たとえば、予め定めた間隔(たとえば、時間、日、週あるいは月)をあけた異なる日の異なる時点で対象に投与することを意味する。本発明は、抗ErbB3抗体の1回の初期用量(initial dose)、続いて抗ErbB3抗体の1回またはそれ以上の二次用量(secondary dose)、場合により、続いて抗ErbB3抗体の1回またはそれ以上の三次用量(tertiary dose)を患者に順に投与することを含む方法を含む。
また、本発明の抗ErbB3抗体を、たとえば、診断目的で、試料中のErbB3の検出および/または測定に用いてもよい。たとえば、抗ErbB3抗体またはそのフラグメントを用いてErbB3の異常発現(たとえば、過剰発現、過小発現、発現の欠如、など)を特徴とする状態または疾患を診断してもよい。ErbB3の例となる診断分析は、たとえば、患者から得た試料を、抗ErbB3抗体が検出可能な標識またはレポーター分子で標識化された本発明の抗ErbB3抗体と接触させることを含んでもよい。別法として、非標識抗ErbB3抗体を、それ自体で検出可能に標識された二次抗体と併用して診断用途に用いることができる。検出可能な標識またはレポーター分子は、放射性同位体、たとえば3H、14C、32P、35Sもしくは125I;蛍光もしくは化学発光部分、たとえばフルオレセインイソチオシアネートもしくはローダミン;または、酵素、たとえばアルカリホスファターゼ、ベータ−ガラクトシダーゼ、ホースラディッシュペルオキシダーゼもしくはルシフェラーゼであることができる。試料中のErbB3を検出または測定するのに用いることができる具体例となるアッセイとしては、酵素結合免疫吸着検査法(ELISA)、放射免疫測定法(RIA)および蛍光活性化セルソーター(FACS)が挙げられる。
ヒトErbB3の外部ドメインを含む免疫原を、免疫応答を刺激するアジュバントと共に、ヒト免疫グロブリン重鎖およびカッパ軽鎖可変領域をコードするDNAを含むVELOCIMMUNE(R)マウスに直接投与した。抗体免疫応答を、ErbB3特異的免疫測定法によってモニターした。所望の免疫応答が達成されたら、脾細胞を集め、マウス骨髄腫細胞を融合させてその生存能力を維持し、ハイブリドーマ細胞系を形成する。ハイブリドーマ細胞系をスクリーニングし、選択してErbB3特異的抗体を産生する細胞系を同定した。この技術を用いて、いくつかの抗ErbB3キメラ抗体(すなわち、ヒト可変ドメインおよびマウス定常を備えた抗体)を得た。このように生成した例となる抗体を、以下のように表わした:H1M1819N、H2M1821N、H2M1824N、H2M1827N、H1M1828N、H2M1829N、H2M1930N、H2M1934N、H2M1936N、H2M1937N、H2M1938N、およびH1M1940N。

以下の実験には、比較のために、さまざまな対照構築物(抗ErbB3抗体)が含まれた。対照構築物は、以下のように表わす:
対照I:US7,846,440に記載された「Mab#6」の対応するドメインのアミノ酸配列を有する重鎖および軽鎖可変ドメインを有するヒト抗ErbB3抗体;
対照II:US7,705,130に記載された「U1−59」の対応するドメインのアミノ酸配列を有する重鎖および軽鎖可変ドメインを有するヒト抗ErbB3抗体;および
対照III:US7,705,130に記載された「U1−53」の対応するドメインのアミノ酸配列を有する重鎖および軽鎖可変ドメインを有するヒト抗ErbB3抗体。
ヒトモノクローナル抗ErbB3抗体の抗体結合親和性および速度定数を、25℃および37℃で表面プラスモン共鳴によって決定した(表2〜4)。測定は、Biacore 2000 またはT100機器において実施した。マウスFc(接頭辞H1M;H2M)またはヒトIgG4Fc(接頭辞H4H)を用いて発現させた抗体を、抗マウスまたは抗ヒトFcセンサ表面(Mabキャプチャーフォーマット)上で捕捉し、可溶性単量体(ErbB3.mmh;配列番号:497)または二量体(ErbB3−hFc;配列番号:498またはErbB3−mFc;配列番号:499)タンパク質を表面に注射した。動的結合(kinetic association)(ka)および解離(dissociation)(kd)速度定数(rate constants)は、スクラバー2.0曲線適合ソフトウェアを用いて、データを処理して1:1結合モデルに適合させることによって決定した。結合解離平衡定数(KD)および解離半減期(t1/2)は、以下のように動的速度定数から算出した:KD(M)=kd/ka;およびt1/2(分)=、(ln2/(60*kd)。いくつかのクローンは、単量体(hErbB3.mmh)ErbB3タンパク質に対してナノモル以下の親和性を示した。
抗ErbB3mAbが細胞表面結合したErbB3を効果的に内部移行するかどうかを決定するため、MCF−7細胞を選択抗ErbB3抗体(10μg/ml)と共に氷上で30分間インキュベートし、洗浄し、次いで氷上で30分間、Dylight 488結合抗ヒトFab(10μg/ml)で染色した。次いでチューブを洗浄し、氷上と37℃のインキュ
ベーションとの間は、1時間離した。インキュベーション後、すべてのチューブを氷上に置き、Dylight 488クエンチング抗体(50μg/ml)を加えた。溶液を氷上でさらに1時間インキュベートした。蛍光シグナルは、Accuri流量血球計算器を用いて測定した。
抗ErbB3抗体を、DU145前立腺がん細胞中のAktのリン酸化を阻害するそれらの能力について試験した。ErbB3に対するNRG1の結合は、ErbB3リン酸化を生じ、ホスファチジルイノシトール3−キナーゼ(PI3−K)の漸増および活性化を導く。活性化されたPI3−KはキナーゼAktをリン酸化し、活性化する。したがって、Aktリン酸化は、ErbB3受容体活性化の下流マーカーである。DU145細胞を96ウェルプレートに接種し、1%FBSを含有する培地中で一夜、血清不足にした。次いで、0.5μg/mlのヒトFc対照タンパク質または0.01、0.1もしくは0.5μg/mlの濃度のさまざまな抗ErbB3抗体の存在下で30分間、細胞を0.5nM NRG1(R&D Systems)で刺激した。抗体の各濃度を3回試験した。細胞を溶解し、ホスホAkt細胞ベースのELISAキット(R&D Systems)を用い、製造元の説明書にしたがって、リン酸化されたAktの相対レベルを決定した。各抗ErbB3抗体対Fc対照基についてのAktリン酸化の平均パーセント阻害を表8に示す。
選択抗ErbB3抗体を、EGFR遮断剤と併用して用いたときのA431表皮がん細胞の成長を阻害するそれらの能力について試験した。96ウェルプレート中のA431細胞を、0.5%FBSを含有する培地中でインキュベートし、抗EGFR抗体(1μg/ml)、抗ErbB3抗体(1μg/ml)または抗EGFR(1μg/ml)+抗ErbB3抗体の存在下、0.05、0.25または1.0μg/mlで1nMニューレグリン−1(NRG1)により刺激した。リガンド(NRG1)および遮断抗体(EGFR及びErbB3)を、72時間実験中0、24および48時間で加えた。処置開始から72時間の時点までに達成された生細胞数における相対変化を、標準的な方法(Cell Proliferation Assay Kit; Promega)を用いて決定した。各抗ErbB3抗体対アイソタイプ対照群についての細胞増殖における平均パーセント減少を表9に示す。
選択抗ErbB3抗体を、A431類表皮がんおよびMCF7乳がん細胞におけるErbB3およびAktのNRG1刺激性リン酸化を阻害するその能力について試験した。第1の実験では、A431細胞を6ウェルプレートに播種し、完全培地中で一夜インキュベートした。次いで、細胞を1時間血清不足(0.5%FBS)にし、5.0μg/mlのアイソタイプ対照または0.05、0.25もしくは5.0μg/mlの抗ErbB3抗体の存在下で1nM NRG1(R&D Systems)を用いて30分間刺激した。細胞を溶解し、ErbB3およびAktに対する抗体ならびにそれらのリン酸化形態を用い、標準方法を用いてウェスタンブロットを実施した。全ErbB3またはAktに対するリン酸化されたErbB3またはAktの比率を算出し、これを用いてアイソタイプ対照と比較した抗ErbB3抗体のそれぞれについてのAktまたはErbB3リン酸化のパーセント阻害を決定した。A431細胞におけるErbB3またはAKTリン酸化の阻害についての結果を、それぞれ表10および11に示す。
選択抗ErbB3抗体を、免疫不全マウスにおいてヒト腫瘍異種移植片の増殖を阻害するその能力について試験した。簡潔には、1〜5×106のA431ヒト表皮がん細胞またはA549ヒト非小細胞肺がん細胞(ATCC)を、6〜8週齢SCIDマウスの側腹部に皮下移植した(Taconic, Hudson, NY)。腫瘍が100〜150mm3の平均体積に達した後、マウスを処置のため群(n=1群につき6匹のマウス)に無作為化した。第1の実験では、A431腫瘍を有するマウスに、ヒトFc(配列番号:500、12.5mg/kg)、または抗ErbB3抗体(0.5または12.5mg/kg)を投与した。すべての抗体は、週2回の皮下注射により約3週間投与した。腫瘍体積を、実験経過の間、週2回測定し、実験の終わりに腫瘍を切除して腫瘍重量を決定した。Fc処置群と比較した平均腫瘍寸法の他に最終的な腫瘍重量を各群について算出した。
結果を表14にまとめる。
ヒト腫瘍異種移植片増殖におけるH4H1821N+阻害性抗EGFR抗体(「抗EGFRmAb」)との併用治療の効果を試験した。簡潔には、2×106のFaDuヒト下咽頭がん細胞(ATCC)を、6〜8週齢SCIDマウス(Taconic, Hudson,NY)の側腹部に皮下移植した。腫瘍が150〜200mm3の平均体積に達した後、マウスを、処置のための群(n=1群につき6匹のマウス)に無作為化した。マウスに、ヒトFc対照タンパク質(12.5mg/kg)、H4H1821N(2.5mg/kg)、抗EGFRmAb(10mg/kg)またはH4H1821N+抗EGFRmAb(2.5+10mg/kg)の組合せを投与した。すべての抗体は、週2回皮下注射により投与した。腫瘍体積を、実験経過の間、週2回測定し、実験の終わりに腫瘍を切除して腫瘍重量を決定した。処置開始からの腫瘍増殖の平均(平均値+/−標準偏差)および腫瘍重量を、各処置群について算出した。腫瘍増殖および腫瘍重量のパーセント減少をFc対照基との比較から算出した。結果を表17に示す。
マッピング
H4H1821Nが相互作用するErbB3のアミノ酸残基を決定するために実験を行った。この目的のために、H/D交換エピトープマッピングを実施した。H/D交換方法の一般的な説明は、たとえば、Ehring (1999) Analytical Biochemistry 267(2):252-259;およびEngen and Smith (2001) Anal. Chem. 73:256A-265Aに記載されている。
進行結腸直腸がん(CRC)、非小細胞肺がん(NSCLC)または頭頸部がん(SCCHN)を有する患者において例となる抗ErbB3抗体H4H1821Nを用いて臨床試験を行う。試験を、二相:用量漸増相および安全性拡張相(safety expansion phase)に分ける。用量漸増相では、すべての患者に、最初にH4H1821Nを3、10または20mg/kgの用量で静脈内に(IV)投与する。H4H1821Nの初期用量の後、治療レジメンは、がんタイプに基づいて変更する。NSCLC患者では、1日1回150mgのエルロチニブを、14日毎に1回、3、10または20mg/kgのIVのH4H1821Nと併用したレジメンを開始し;CRCおよびSCCHN患者では、週1回IVセツキシマブ250mg/m2を14日毎に1回、3、10または20mg/kgのIVのH4H1821Nと併用したレジメンを開始する。安全性拡張相では、NSCLC患者は、14日毎に1回のIVのH4H1821N(推奨された第2相の用量)を1日1回150mgのエルロチニブと併用して受け;CRCおよびSCCHN患者は、14日毎に1回、IVのH4H1821N(推奨された第2相の用量で)を週1回IVセツキシマブ250mg/m2と併用して受ける。
実際に、本明細書に記述したものに加えて本発明のさまざまな変更は、前述の説明および添付の図面から当業者に明らかになる。このような変更は添付の特許請求項の範囲内に帰属するものとする。
Claims (8)
- ErbB3を特異的に結合する、単離されたヒトモノクローナル抗体またはその抗原結合性フラグメントであって、
該抗体またはその抗原結合性フラグメントは、配列番号306/314及び322/330から選択される重鎖可変領域/軽鎖可変領域のアミノ酸配列対の相補性決定領域HCDR1−HCDR2−HCDR3−LCDR1−LCDR2−LCDR3を含み、そして、該抗体またはその抗原結合性フラグメントは、水素/重水素交換によって決定したように、配列番号:498のアミノ酸345−367、配列番号:498のアミノ酸423−439;および配列番号:498のアミノ酸451−463と相互作用し、かつ25℃でELISAにおいて30pM未満のIC50でErbB3に対するNRG1の結合を遮断する、
単離された抗体または抗原結合性フラグメント。 - それぞれ、配列番号:308−310−312−316−318−320;および324−326−328−332−334−336からなる群から選択されるHCDR1−HCDR2−HCDR3−LCDR1−LCDR2−LCDR3ドメインを含み、かつ25℃でELISAにおいて30pM未満のIC50でErbB3に対するNRG1の結合を遮断する、
