PluriX(商標)栓流バイオリアクター(Pluristem社、ハイファ、イスラエル;図1gに示す、米国特許第6,911,201号も参照されたい)に、ポリエステルの不織布マトリックスから作られた1〜100ml充填用の3D多孔性担体(porrosive carrier)(直径4mm)を充填した。これらの担体は、比較的少量で大細胞数の増殖を可能にする。ガラス製品は、Pluristem社が設計し、製造した。このバイオリアクターは、37℃のインキュベーター内で維持され、流速は、バルブ(図1gの6a)、および蠕動ポンプ(図1gの9)によって調節され、監視された。このバイオリアクターは、サンプリングポイントおよび注入ポイント(図1gの4)を含み、細胞の連続播種を可能にする。培地を、貯蔵所(図1gの1)から、pH6.7〜7.4で供給した。この貯蔵所には、バイオリアクター中の細胞密度に応じて、異なる比率で空気/CO2/O2を含む濾過したガス混合物(図1gの2、3)が供給された。O2の割合は、モニター(図1gの6)によって測定される、バイオリアクターの出口における溶存O2のレベルに適していた。このガス混合物は、シリコンチューブまたは拡散器(Degania Bet社、Emek Hayarden、イスラエル)を経て貯蔵所に供給された。培地を、循環している非接着細胞の収集を可能にする分離容器(図1gの7)に通過させた。培地の循環は、蠕動ポンプ(図1gの9)が行った。このバイオリアクターは、さらに、追加のサンプリングポイント(図1gの10)および連続培地交換用の容器を備えていた。
上記のように成長させたコンフルエントでない初代ヒト接着間質細胞の2D培養物を、トリプシン処理し、洗浄し、10%FBS、Pen−Strep−ナイスタチン混合物(100U/ml:100μg/ml:1.25un/ml)および2mMのL−グルタミンを補充したDMEMに再懸濁し、注入ポイントを介して、無菌の栓流バイオリアクター(図1g参照)内の3D担体上に播種した(103〜105細胞/ml)。接種前に、バイオリアクターに、PBS−Ca−Mg(Biological Industries社、Beit Ha’emek、イスラエル)を充填し、これをオートクレーブ処理(120℃、30分間)し、10%熱不活性化ウシ胎児血清およびPen−Strep−ナイスタチン混合物(100U/ml:100μg/ml:1.25un/ml)を含むダルベッコ成長培地で洗浄した。流れを、0.1〜5ml/分の速度で維持した。播種プロセスには、2〜48時間の循環の中断が含まれ、それによって、細胞がこれらの担体上で定着するのを可能にした。バイオリアクターを、必要に応じて、無菌の空気およびCO2が供給されるインキュベーターを用いて、制御された温度(37℃)およびpH条件(pH=6.7〜7.4)下で維持した。成長培地を週に2〜3回交換した。循環培地を、4時間〜7日ごとに新鮮なDMEM培地と交換した。1×106〜1×107細胞/mlの密度(成長の12〜40日後)で、培地の全量をこのバイオリアクターから除去し、担体をPBSで3〜5回洗浄した。その後、3D−ASC細胞を、トリプシン−EDTA(Biological Industries社、Beit Ha’emek、イスラエル;3〜15分穏やかに攪拌しながら、1〜5回)を用いてこれらの担体から脱離させ、その後、DMEMに再懸濁して、凍結保存した。
凍結保存した3D−ASC細胞を解凍し、数を数えた。細胞生存率の評価のために、2×105細胞を150cm2組織培養フラスコに播種し、それらの接着力および再増殖を、播種後の7日以内に評価した。その後、3D−ASCの膜マーカーの表現型を、蛍光モノクローナル抗体のフローサイトメーター(Beckman Coulter社、フラートン、カリフォルニア州)を用いて分析した。
2D培養物および3Dフロー方式培養物の100,000〜200,000個の接着間質細胞を、5mlチューブ中の培地0.1mlに懸濁し、飽和濃度の以下のMAbのそれぞれとインキュベートした(4℃、30分、暗い条件):FITC結合抗ヒトCD90(Chemicon International社、テメキュラ、カリフォルニア州)、PE結合抗ヒトCD73(Bactlab Diagnostic社、Ceasarea、イスラエル)、PE結合抗ヒトCD105(eBioscience社、サンディエゴ、カリフォルニア州)、FITC結合抗ヒトCD29(eBioscience社、サンディエゴ、カリフォルニア州)、Cy7−PE結合抗ヒトCD45(eBiosience社)、PE結合抗ヒトCD19(IQProducts社、グローニンゲン、オランダ)、PE結合抗ヒトCD14MAb(IQProducts社)、FITC結合抗ヒトCD11b(IQProducts社)およびPE結合抗ヒトCD34(IQProducts社)またはFITC結合抗ヒトHLA−DRのMAb(IQProducts社)。インキュベーション後、これらの細胞を、1%熱不活性化FCSを含む氷冷PBSで2回洗浄し、0.5%ホルムアルデヒド500μlに再懸濁し、FC−500フローサイトメーター(Beckman Coulter社、フラートン、カリフォルニア州)を用いて分析した。
2Dおよび3Dの培養手順によるASCを、上述したとおり胎盤から生産した。簡単に説明すると、これらの2D培養物は、60〜80%コンフルエンスに達するまで37℃の加湿5%CO2雰囲気下で4日間、175cm2フラスコ内で0.3〜0.75×106細胞を培養することによって生産した。これらの3D培養物は、2000担体を含むバイオリアクター中で2〜10×106細胞/gを播種し、18日間培養することによって生産した。収集後、細胞を洗浄し(×3)、全ての血清を除去し、ペレット化して、凍結させた。タンパク質を、(Tri試薬キット(Sigma社、セントルイス、米国)を用いて)ペレットから単離し、トリプシンで消化し、メーカーのプロトコールに従ってiTRAQ試薬(Applied Biosciences社、フォスターシティ、カリフォルニア州)で標識した。簡単に説明すると、iTRAQ試薬は、非ポリマー性の等圧タグ付き試薬である。各試料中のペプチドを、それらのN−末端および/またはリジン側鎖を介して4つの等圧の同位体コード化タグのうちの1つで標識した。これらの4つ標識試料を混合し、ペプチドを、質量分析で分析した。ペプチド断片化の際、各タグは異なる質量のレポーターイオンを放出するので、4つのレポーターの比率は、試料中の特定のペプチドの相対存在量を与える(情報元:http://docs.appliedbiosystems.com/pebiodocs/00113379.pdf)。
2Dおよび3Dの培養手順によって胎盤から生産させるASCを、上述したとおり、24日間、3D培養で生産した。その後、馴化培地を収集し、3つの独立した実験で、ELISA(R&D Systems社、ミネアポリス、ミネソタ州)を用いて、Flt−3リガンド、IL−6、トロンボポエチン(TPO)および幹細胞因子(SCF)について分析した。結果を、1×106細胞/mlに対して規準化した。
PluriX(商標)バイオリアクターシステムは、生理的環境のような微小環境を作り出す
接着間質細胞にとって効率的な培養条件にさせるために、生理的環境のような環境(図1aに示す)を、PluriXバイオリアクター(Pluristem社、ハイファ、イスラエル;担体を図1gに示し、図1bに播種前の状態を示す)を用いて人工的に作り出した。図1c〜図1fに示すとおり、骨髄生産3D−ASC細胞を十分に培養し、播種後20日間(図1b〜図1c、それぞれ150倍および250倍に拡大)ならびに40日間(図1c〜図1d、それぞれ350倍および500倍に拡大)、3Dマトリックス上で増殖させた。
異なる生産ロットの胎盤由来の3D−ASC細胞を、PluriXバイオリアクターシステム内で成長させた。播種密度は、13,300細胞/担体(全部で2×106細胞)であった。播種後14日目に、細胞密度は15倍増し、約200,000細胞/担体(図2)、または150担体のバイオリアクター中30×106個に達した。別の実験では、細胞を、1.5×104細胞/mlの密度でバイオリアクターに播種し、播種後30日目に、これらの担体は、50倍を超える高い細胞数、すなわち、約0.5×106細胞/担体、または0.5×107細胞/mlを含んでいた。様々なレベルの成長カラムの担体上の細胞密度は一貫しており、細胞への酸素および栄養素の移送が均質であることが示された。したがって、この3D培養系は、生着および移植の成功を支援する目的のために十分な量まで効率よく成長させることができる、高密度の間葉系細胞培養物の成長および長期維持のための支援条件を提供することが証明された。
3D培養法を模倣する骨環境によって与えられる可溶性分子の分泌プロファイルおよびタンパク質生産の違いを定義するために、FACS分析を行った。図3aに示すとおり、細胞マーカーのFACS分析は、3D−ASCが2D条件で成長させた接着間質細胞とは異なるマーカー発現パターンを示すことを表す。2D培養細胞は、3D培養細胞と比較して、著しく高いレベルの陽性膜マーカーCD90、CD105、CD73およびCD29を発現した。例えば、CD105は、3D培養細胞中では56%の発現を示し、それに対して、2D培養細胞中では87%の発現を示した。2D胎盤培養物と3D胎盤培養物の両方のASCは、造血膜マーカーを発現しなかった(図3b)。
造血のニッチには、豊富なサイトカイン、ケモカインおよび成長因子を生産する支援細胞が含まれる。2D培養ASCと3D培養ASCの違いをさらに定義するために、2D ASC培養物および3D ASC培養物の馴化培地中の4つの主要な造血分泌タンパク質のプロファイルをELISAにより調べた。図4a〜図4cは、3D条件で成長させた細胞が、高レベルのFlt−3リガンド(図4a)、IL−6(図4b)、およびSCF(図4c)を有する馴化培地を生成するが、2D培養物の馴化培地では、低レベルのIL−6、ならびに0に近いレベルのFlt−3リガンドおよびSCFが検出されたことを示す。トロンボポエチン(TPO)の生産は、非常に低く、両方の培養物で等しかった。
2D培養ASCと3D培養ASCの違いをさらに定義するために、これらの細胞のタンパク質プロファイルを、質量分析によって分析した。図4dは、2D培養ASCおよび3D培養ASCが、非常に異なるタンパク質発現プロファイルを示すことを示す。以下の表1に示すとおり、3D培養細胞は、非常に高い発現レベル(それぞれ、9倍および12倍を上回る高さ)のH2AFおよびALDH X、ならびに高レベルのタンパク質EEEF2、RCN2およびCNN1を示す(それぞれ、約3倍、約2.5倍および約2倍)。さらに、3D培養細胞は、タンパク質HnrphlおよびCD44抗原アイソフォーム2前駆体が約半分の発現レベルであり、Papss2およびrpL7aが約3分の1の発現レベルであることを示す。
2D培養ASCおよび3D培養ASCによるリンパ球反応の抑制
接着間質細胞、特に、3D−ASCは、MLRアッセイにおいてヒト臍帯血単核細胞の免疫反応を抑制することが見出された。
混合リンパ球反応(MLR)アッセイ
胎盤から生産される2Dおよび3D培養手順によるASCの免疫抑制ならびに免疫特権の特性は、反応(増殖)細胞および刺激(非増殖)細胞の混合培養における不適合リンパ球の増殖率によって影響されるように、HLA遺伝子座で組織適合性を測定するMLRアッセイによって影響を受けた。ヒト臍帯血(CB)単核細胞(2×105)を反応細胞として使用し、放射線照射(3000Rad)した等量(105)のヒト末梢血由来の単球(PBMC)と共培養するか、胎盤から生産された2Dもしくは3D培養の接着間質細胞と共培養するか、または接着間質細胞とPBMCの組み合わせで共培養することによって刺激した。各アッセイを3回繰り返した。細胞を、96ウェルプレート中の(37℃の加湿5%CO2雰囲気下で、20%FBSを含む)RPMI 1640培地で4日間共培養した。プレートを、培養の最後の18時間中に、1μCの3H−チミジンでパルスした。その後、細胞を、グラスファイバーフィルター上で収集し、チミジン取り込みを、シンチレーションカウンターを用いて定量した。
図7は、PBMCで刺激した時に、CB細胞の増殖が上昇することによって示されるCB細胞の免疫反応を示し、これは、理論によって縛られることなく、おそらく、HLA不適合性に反応するT細胞増殖に関連付けられる。しかし、本発明の接着間質細胞とインキュベートした時に、かなり低いレベルの免疫反応がこれらの細胞によって示された。さらに、PBMCに対するCB免疫反応は、これらの接着細胞と共培養したときに、実質的に低下した。したがって、ASCは、GvHDに特有の、ドナー細胞のT細胞増殖を低下させる潜在能力を有することが見出された。2Dおよび3Dの両方の培養物は、リンパ球の免疫反応を低下させるが、上記の3D−ASCの他の利点と一致して、これらの3D−ASCはより強い免疫抑制を示した。
胎盤由来の3D−ASCのHSC生着を改善する能力の評価
HSC生着の3D−ASC支援を、亜致死線量の放射線照射または化学療法で前処理した免疫欠損NOD−SCIDマウスで検出されるヒト造血細胞(hCD45+)のレベルにより評価した。
CD34+細胞の単離
臍帯血試料を、分娩中無菌条件下で取得し(Bnei Zion医療センター、ハイファ、イスラエル)、単核細胞を、Lymphoprep(Axis−Shield PoC As、オスロ、ノルウェー)密度勾配遠心分離を用いて分画し、凍結保存した。解凍した単核細胞を洗浄し、抗CD34抗体とインキュベートし、midi MACS(Miltenyl Biotech社、Bergish Gladbach、ドイツ)を用いて単離した。2種類以上の試料からの細胞を、所望の量(50,000〜100,000細胞)に達するまでプールした。
7週齢の雄および雌のNOD−SCIDマウス(NOD−CB 17−Prkdcscid/J;Harlan/Weizmann Inst.,Rehovot、イスラエル)を、無菌のオープンシステムケージ内で維持し、無菌の食事およびオートクレーブした酸性水を与えた。これらのマウスに亜致死線量(350cGy)の放射線を照射し、その後(放射線照射後48時間)、胎盤または脂肪組織由来の追加のASC(0.5×106〜1×106)を含む場合と含まない場合とで(各群3〜7匹のマウス)、外側尾静脈への静脈注射によって50,000〜100,000個のhCD34+細胞を移植した。移植後4〜6週間目に、これらのマウスを脱臼により屠殺し、BMを、FACS緩衝液(PBS50ml、FBS5ml、5%アジ化ナトリウム0.5ml)で大腿骨と脛骨の両方を洗い流すことによって収集した。マウスBMにおけるヒト細胞をフローサイトメトリーによって検出し、治療したNOD−SCIDマウスにおけるヒトおよびマウスのCD45造血細胞マーカー発現細胞の割合を、抗ヒトCD45−FITC(IQ Products、Groningen、オランダ)と細胞をインキュベートすることによって測定した。明確なヒト生着についての最低の閾値は0.5%であった。
6.5週齢の雄のNOD−SCIDマウス(NOD.CB17/JhkiHsd−scid;Harlan、Rehovot、イスラエル)を、放射線照射マウスについて上記に記載したとおりに維持し、ブスルファンを腹腔内に注射した(2日連続−25mg/kg)。第2のブスルファン注射後2日目に、マウスに、CD34+細胞を単独で、または胎盤から生産された0.5×106個のASCと共に注射した。移植後3.5週目に、マウスを屠殺し、放射線照射マウスについて記載したとおりに、ヒト造血細胞の存在を決定した。
放射線照射マウスにおける3D−ASCによって改善されたHSCの生着
ヒトCD34+造血細胞および胎盤または脂肪に由来する3D−ASCを、放射線照射NOD−SCIDマウスに共移植した。生着率を共移植後4週目に評価し、HSCを単独で移植したマウスと比較した。表2および図5に示すとおり、3D−ASCおよびUCB CD34+細胞の共移植は、UCB CD34+細胞単独で治療したマウスと比較して、レシピエントマウスのBMにおいてかなり高い生着率およびより高レベルのヒト細胞を与えた。
ヒトCD34+造血細胞を、化学療法で前処理したNOD−SCIDマウスに、胎盤由来の500,000個の2D−ASCまたは3D−ASCと共移植した。共移植後3.5週目に、生着効率を評価し、HSCを単独で移植したマウスと比較した。表3に示すとおり、ASCおよびUCB CD34+細胞の共移植は、UCB CD34+細胞単独と比較して、レシピエントマウスのBMにおいてより高レベルの生着が得られた。さらに、表3に示すとおり、生着の平均レベルは、従来の静的な2D培養条件(フラスコ)で成長させた同じドナー由来の細胞を共移植したマウスよりも、PluriXバイオリアクターシステムで成長させた胎盤由来の接着間質細胞(3D−ASC)と共移植したマウスにおいてより高かった。
放射線照射および化学損傷後にHSC回復を改善する胎盤由来の3D−接着間質細胞の
能力の評価
レシピエントの内因性HSC回復の3D−接着間質細胞による支援を、亜致死線量の放射線照射または化学療法で前処理した免疫欠損NOD−SCIDマウスにおいて検出されるマウスの造血細胞(mCD45+)のレベルによって評価した。
放射線照射マウスにおける回復した細胞の検出
7週齢の雄および雌のNOD−SCIDマウス(NOD−CB17−Prkdcscid/J;Harlan/Weizmann Inst.,Rehovot、イスラエル)を、無菌のオープンシステムケージ内で維持し、無菌の食事およびオートクレーブした酸性水を与えた。これらのマウスに亜致死線量(350cGy)の放射線を照射し、その後(放射線照射後48時間)、2Dまたは3D条件下で成長させた胎盤由来の接着間質細胞(0.5×106〜1×106)を含む場合と含まない場合とで(各群3〜7匹のマウス)、50,000〜100,000個のhCD34+細胞を移植した。細胞を、外側尾静脈への静脈注射によって投与した。移植後4〜6週間目に、これらのマウスを脱臼により屠殺し、BMを、FACS緩衝液(PBS50ml、FBS5ml、5%アジ化ナトリウム0.5ml)で大腿骨と脛骨の両方を洗い流すことによって収集した。マウス造血系の回復を表す、治療したNOD−SCIDマウスにおけるマウスCD45造血細胞マーカー発現細胞の測定を、抗マウスCD45−FITC(IQ Products、Groningen、オランダ)と細胞をインキュベートすることによって行った。
6.5週齢の雄のNOD−SCIDマウス(NOD.CB17/JhkiHsd−scid;Harlan、Rehovot、イスラエル)を、放射線照射マウスについて上記に記載したとおりに維持し、ブスルファンを腹腔内に注射した(2日連続−25mg/kg)。第2のブスルファン注射後2日目に、マウスに、ヒトCD34+細胞を単独で注射するか、または胎盤から生産された0.5×106個の接着間質細胞と共に注射した。移植後3.5週目に、マウスを屠殺し、放射線照射マウスについて上記に記載したとおりに、ヒト造血細胞の回復を測定した。
放射線照射マウスにおける3D−接着間質細胞によって改善されたHSCの生着
ヒトCD34+造血細胞および胎盤または脂肪組織に由来する3D−接着間質細胞を、放射線照射NOD−SCIDマウスに共移植した。マウス造血系の回復効率を共移植後4週目に評価し、胎盤接着間質細胞を含めないで、hHSCを移植したマウスの自己回復と比較した。表4に示すとおり、2D−接着間質細胞とUCB CD34+細胞および3D−接着間質細胞とUCB CD34+細胞の両方の共移植は、UCB CD34+細胞単独で治療したマウスと比較して、mCD45の発現レベルによって示されるように、かなり高い回復率を与えた。改善は、3D増殖細胞においてより高かったことに留意されたい。
ヒトCD34+造血細胞を、化学療法で前処理したNOD−SCIDマウスに、胎盤由来の接着間質細胞と共移植した。レシピエントマウスの造血系の回復効率を、共移植後3.5週目に評価し、HSCを単独で移植したマウスと比較した。表5に示すとおり、接着間質細胞およびUCB CD34+細胞の共移植が、UCB CD34+細胞単独と比較して、レシピエントマウスの造血系のより高い回復率を与えた。さらに、表5に示すとおり、回復の平均レベルは、投与した接着間質細胞の数に対して用量依存的であった。
放射線照射マウスの生存に対する3D−ASC(PLX)細胞の効果を、放射線照射(850cGy)後24時間に、C3Hマウスに3D増殖ASCを静脈内投与した後に検討した。
調製
マウス(C3H雄、20g、約8週齢)を、Harlan社から購入した。動物を、実験前の馴化のためにSPF施設で1週間飼育した。30匹のC3H雄マウスに全身放射線照射(850cGy)した。放射線照射後24時間に、15匹のマウスに、外側尾静脈の1つへのゆっくりとした静脈注射(約1分)によって、マウス1匹あたりplasmaLyte A250μl中の3D−ASC細胞(1×106個)を注射した。凝集を防ぐために、注射ステップの初めから細胞を穏やかに混合した。15匹のマウスの残りの対照群に、等量(250μ1)のplasmaLyte A(ビヒクル)を注射した。
図9は、BALB/cおよびC3Hマウスにおける2種類の放射線量の電離放射線照射後のマウス(3D−ASC治療なし)の生存率の追跡調査を示す。
放射線照射マウスの生存に対する胎盤由来の3D−増殖接着間質細胞(PLX)の効果を、放射線照射(770cGy)後24時間に、C3Hマウスに静脈内投与した後に検討した。
調製
マウス(C3H雄、20g、約6週齢)を、Harlan社から購入した。動物を、実験開始前の馴化のためにSPF施設で2週間飼育した。30匹のC3H雄マウスに全身放射線照射(770cGy)した。放射線照射後24時間に、15匹のマウスに、外側尾静脈の1つへのゆっくりとした静脈注射(約1分)によって、マウス1匹あたりplasmaLyte A250μl中の3D−ASC細胞(1×106個)を注射した。凝集を防ぐために、注射ステップの初めから細胞を穏やかに混合した。15匹のマウスの残りの対照群に、同量(250μ1)のplasmaLyte A(ビヒクル)を注射した。
PLX細胞で治療したマウス(●)およびPLX治療を受けていないマウス(○)についての、770cGyの放射線量での放射線照射後の生存率を図17に示す。図18は、規準化した体重変化(図18A)または平均体重変化(図18B)のいずれかの18日間の体重変化を経時的に示す。
これらの結果は、使用する放射線照射線量が使用するマウス系統に致死的であったことを示す。770cGyにおいて、ビヒクル治療群の12匹のマウスのうち1匹だけが18日目まで生存した(生存率8%)。PLX細胞の静脈注射は、同じ線量の放射線照射後に、生存マウスの割合を75%(9/12)まで増加させる。追跡調査期間中、PLX治療マウスは、ビヒクル治療マウスよりもよく食べた。18日目に、PLXマウスは平均体重まで増えたが、たった1匹生き残ったビヒクル治療マウスは、さらに体重を失っていた。IV PLX注射によって治療した群も、8日目および18日目の両方において、より高い骨髄細胞数を有していた。18日目の血液パラメータ、特に、RBCおよび血小板も、一般に、PLX治療マウスで良好であった。これらの効果は、生存率が低下する時期の開始前である8日目においては明らかでなかった。
放射線照射マウスの血清サイトカインプロファイルに対する胎盤由来の3D−増殖接着間質細胞(PLX)の効果を、放射線照射(770cGy)後24時間に、C3HマウスにPLXを静脈内投与した後1日目および4日目に検討した。
調製
マウス(C3H雄、20g、約6週齢)を、Harlan社から購入した。動物を、実験前の馴化のためにSPF施設で2週間飼育した。4匹のマウスを未治療対照マウスとする一方で、26匹に770cGyの放射線を照射した。放射線照射後24時間に、8匹のマウスにPLXをIV注射し、8匹のマウスにPlasmalyteを注射した。IV注射後の予期せぬ死亡の場合のために、6匹を予備として飼育した。PLX注射後1日目および4日目に、2匹の非放射線照射対照マウス、2匹の放射線照射した対照マウス、4匹のビヒクル注射マウス、および4匹のPLX注射マウスから動脈血を収集した。血清をこの血液から分離し、次いで、650μlの十分な量を得るために、各2匹のマウスからの血清を一緒にプールした。以下のサイトカイン/成長因子IL1A、IL1B、IL2、IL4、IL6、IL10、IL12、IL17A、IFNγ、TNFα、G−CSF、およびGM−CSFについて「マウスの炎症性サイトカイン多検体ELISAアレイキット(Mouse Inflammatory Cytokines Multi−Analyte ELISArray Kit」(SABiosciences社;カタログ番号MEM−004A)を用いた解析まで、収集した血清を−20℃で保管した。
図26A〜図26Bは、PLX細胞またはビヒクルでの注射後1日目(図26A)および4日目(図26B)におけるサイトカインプロファイルを示す。最も顕著な変化は、放射線処理した全マウスにおいてG−CSFレベルが増加したことであった。
放射線照射マウスの生存に対する3D−ASC(PLX)細胞の効果を、放射線照射(770cGy)後24時間に、C3Hマウスに3D増殖ASCを筋肉内投与した後に検討した。
調製
マウス(C3H雄、約24g、7週齢)を、Harlan社から購入した。動物を、実験前の馴化のためにSPF施設で2週間飼育した。C3H雄マウスに全身放射線照射(770cGy)した。放射線照射後約24時間に、12匹のマウス(「放射線照射+PLX」群)に、インスリン注射器および25g針を使用して、それぞれの尾筋肉に、2×106細胞/マウスの総量に対して3D−ASC細胞50μl(バッチPD061210 153B04、plasmaLyte A中20×106細胞/mL)を注射した。第2群(「放射線照射+PLX−2X」)には、最初に同じ量の注射をしたが、さらに、4日の間隔で、第2の2×106個の3D−ASCをそれぞれの尾筋肉に筋肉注射した。全ての注射について、凝集を防ぐために、注射ステップの初めから細胞を穏やかに混合した。放射線照射マウスの対照群は、plasmaLyte A(ビヒクル)を同量(全部で100μl、尾筋肉あたり50μ1)、同じ方法で筋肉注射した。
図27は、770cGy電離放射線を与えたC3Hマウスの生存率(図27A)および体重変化(図27B)を示す。放射線照射後1日目および5日目(丸)に、2×106細胞/用量の2回の筋肉注射を与えたマウスは、放射線照射後1日目に1回の筋肉注射を与えたマウスまたはPLX細胞を一切与えなかった対照の放射線照射マウスと比較して、生存率が改善した。
放射線照射マウスの生存率に対する胎盤由来の3D−増殖接着間質細胞(PLX)の効果を、放射線照射(770cGy)後24時間および/または5日目に、C3Hマウスに2種類の異なる用量(1万細胞または2万細胞/注射)の3D増殖ASCを筋肉内投与した後に検討した。
44匹のC3Hマウスに、ハダサヘブライ大学医療センターの腫瘍学のシャレット研究所(Sharett Institute of Oncology at Hadassah Hebrew University Medical Center)で全身放射線照射(770cGy)した。放射線照射マウスを4群に分け(11匹/群)、以下のように治療した:
1.1×106PLX細胞を2回注射した:放射線照射後24時間および放射線照射後5日目(注射した全細胞数2×106個)。
2.2×106PLX細胞を2回注射した:放射線照射後24時間および放射線照射後5日目(注射した全細胞数4×106個)。
3.放射線照射後5日目に2×106PLX細胞を1回注射した。
4.対照群として、PlasmaLyte A(ビヒクル)のみを注射した。
図28A〜図28Bは、770cGyの放射線量での放射線照射後の生存率(図28A)および平均体重変化(図28B)を示す。ビヒクルで治療した11匹のマウスのうちの3匹(△)が、監視期間生存した。5日目に2×106PLX細胞単独で治療した11匹のマウスのうちの5匹(○)が生存した。1日目および5日目に1×106PLX細胞で治療した11匹のマウスのうちの9匹(●)が生存した。1日目および5日目に2×106PLX細胞を投与した場合(上部の黒丸のセット)、11匹のうち10匹が生存した。
3D−増殖させ、混合した母方/胎児PLX細胞と比較した、放射線照射マウスの生存率に対する3D−増殖させた胎盤由来の母方接着間質細胞(PLX)の効果を、放射線照射(770cGy)後24時間および5日目に、C3Hマウスに2百万細胞/注射の3D増殖ASCを筋肉内投与した後に検討した。
1.2×106細胞/マウスでPLX−1(混合)を2回注射した:放射線照射後24時間および5日目(注射した全細胞数4×106個)。
2.2×106細胞/マウスでPLX−2(母方)を2回注射した:放射線照射後24時間および5日目(注射した全細胞数4×106個)。
3.PlasmaLyte Aを2回注射した:放射線照射後24時間および5日目。
図33A〜図33Bは、770cGyの放射線量での放射線照射後の生存率(図33A)および平均体重変化(図33B)を示す。混合PLX細胞(□;9/9生存)は、母方PLX(△;7/9生存)よりも23日の生存率は良好であったが、両群とも、ビヒクル治療マウスと比較して生存率が改善した(◇;3/9生存)。図33Bに示すとおり、混合PLX細胞で治療したマウスは、高い割合の初期体重も保持していた。
IM投与した母方PLX細胞とPLX細胞の混合バッチの両方は、生存率を改善した。この混合PLXバッチは、生存率をより効果的に改善した。混合PLXも、BM有核細胞および末梢血白血球百分率(differential count)のパラメータによって示されるとおり、BM再増殖を引き起こすのにより効果的であった。
3D−増殖させ、混合した胎盤由来の母方/胎児接着間質細胞と比較した、放射線照射マウスの血清サイトカインに対する3D−増殖させた胎盤由来の母方接着間質細胞(PLX)の効果を、放射線照射(770cGy)後24時間および5日目に、C3Hマウスに2百万細胞/注射の3D増殖ASCを筋肉内投与した後に検討した。
図36は、PLX細胞またはビヒクルの注射後8日目のサイトカインプロファイルを示す。G−CSFレベルは、放射線処理した全マウスにおいて増加した。この増加は、母方PLX細胞で治療したマウスで最大であった。
全身的な炎症の急性発症は、放射線照射後8日目のマウスでは検出されなかった。顆粒球コロニー刺激因子(G−CSF)レベルにのみ顕著な変化があった。G−CSFは骨髄を刺激し、顆粒球および幹細胞を生産し、その後、骨髄を刺激して、それらを血液中に放出する。G−CSFは、放射線照射マウスの血清、特に、母方PLX注射群において上昇した。
3D−混合(PLX)細胞と比較した、放射線照射マウスの生存率、血液学的パラメータ、および血清サイトカインに対する3D−母方(PLX)細胞の効果を、放射線照射(770cGy)後48時間および5日目に、C3Hマウスに2百万細胞/注射の3D増殖ASCを筋肉内投与した後に検討した。
1.2×106個の混合PLX細胞を2回注射した:放射線照射後48時間および5日目(注射した全細胞数4×106個)。
2.2×106個の母方PLX細胞を2回注射した:放射線照射後48時間および5日目(注射した全細胞数4×106個)。
3.PlasmaLyte Aを2回注射した:放射線照射後48時間および5日目。
図40A〜図40Bは、770cGyの放射線量で放射線照射し、放射線照射後48時間および5日目に治療した後の生存率(図40A)および平均体重変化(図40B)を示す。第1の注射を、放射線照射後24時間に行った場合のように、混合PLX細胞(□)は、母方PLX(△)よりも良好な日数の生存を与えたが、本例においても、両群とも、ビヒクル治療マウス(◇)と比較して生存率が改善した。図40Bに示すとおり、混合PLX細胞で治療したマウスは、対照の生存マウスと比較して、より高い割合の初期体重も保持していた。母方の細胞の性質または混合細胞の性質に関わらず、24時間および5日目における治療と比較して、第1の治療を48時間まで遅らせると、全体的な生存率がわずかに減少した。
第1の治療が24時間〜48時間遅れたとしても、I.M.投与した母方PLX細胞とPLX細胞の混合バッチの両方は生存率を改善した。しかし、本例においても、混合PLX細胞が、良好な生存率を与えた。混合PLXは、本例においても、BM有核細胞および末梢血白血球百分率のパラメータによって示されるとおり、BM再増殖を引き起こすのにより効果的でもあった。
Claims (13)
- 接着間質細胞を含む、放射線または化学療法に曝露された対象における腸損傷を軽減するための組成物であって、前記接着間質細胞が、筋肉注射により対象へ投与され、且つ、前記接着間質細胞が、三次元培養条件下で培養されたものである、組成物。
- 前記放射線または化学療法への曝露が進行中である、請求項1に記載の組成物。
- 前記接着間質細胞の入手源が、胎盤、脂肪組織、または骨髄である、請求項1または2に記載の組成物。
- 前記三次元培養条件が、細胞増殖を支援する、請求項1〜3のいずれか1項に記載の組成物。
- 前記接着間質細胞が、分化なしでの、細胞増殖を支援する三次元培養条件下で、バイオリアクター中で培養された胎盤接着間質細胞である、請求項4に記載の組成物。
- 前記接着間質細胞の約60%未満が、アイソタイプ対照と比較して、フローサイトメトリーによって検出される、マーカーCD200が陽性である、請求項1〜5のいずれか1項に記載の組成物。
- 前記接着間質細胞の約60%未満が、アイソタイプ対照と比較して、免疫蛍光法によって検出される、マーカーOCT−4が陽性である、請求項1〜6のいずれか1項に記載の組成物。
- 前記接着間質細胞の少なくとも70%が、胎児細胞である、請求項1〜5のいずれか1項に記載の組成物。
- 前記接着間質細胞が胎児細胞及び母方細胞の混合物である、請求項1〜5のいずれか1項に記載の組成物。
- 外因性造血幹細胞が前記対象に投与されない、請求項1〜9のいずれか1項に記載の組成物。
- 第1の投与後約2日〜約21日目に少なくとも1つの追加的な治療有効量の接着間質細胞を投与する、請求項1〜10のいずれか1項に記載の組成物。
- 少なくとも1つの追加的な治療有効量の投与が、筋肉注射によるものである、請求項11に記載の組成物。
- 外因性造血幹細胞が、前記第1の治療有効量の投与のときに前記対象に投与されず、少なくとも1つの追加的な治療有効量が、マッチした外来性造血幹細胞の同定後に投与される、請求項11または12に記載の組成物。
