Description
「増殖活性」は、例えば、正常な細胞分裂の他、癌、腫瘍、異形成、細胞形質転換、転移、および血管新生を促進する、これらに必要である、またはこれらと特異的に関連する活性を包含する。
典型的な治療的モノクローナル抗体は、4つのポリペプチド、すなわち2つの軽鎖(例えば214個のアミノ酸長)および2つの重鎖(例えば446個のアミノ酸長)から成る。各鎖は今度は、可変ドメインおよび定常ドメインから成る。抗IL−23p19hum13B8−bの可変ドメインは、軽鎖および重鎖に対しそれぞれ108個および116個のアミノ酸であり、定常ドメインは106個および330個のアミノ酸である。抗体のその標的に対する特異性は、いわゆる「超可変」領域または「相補性決定」領域(CDR)(3つのCDRが重鎖および軽鎖の各々の可変ドメインで見出されている)内にある配列により大部分が決定される。CDRは、異なる抗体間で長さが異なり得るが、hum13B8−bにおいてCDRは、重鎖に44個のアミノ酸、および軽鎖に27個のアミノ酸を含む。CDR残基は、異なる抗体間で高度に可変的であり、ヒト生殖細胞系配列(完全ヒト抗体の場合)、または非ヒト(例えば齧歯類)生殖細胞系配列に由来し得る。フレームワーク領域も、抗体によって著しく異なり得る。定常領域は、選択された抗体がラムダ(λ)またはカッパ(κ)軽鎖を有するかどうかに応じて、ならびに抗体のクラス(またはアイソタイプ)(IgA、IgD、IgE、IgG、またはIgM)およびサブクラス(例えばIgG1、IgG2、IgG3、IgG4)に応じて異なるであろう。合計は、「超可変」領域中の71個のアミノ酸を含む224個が可変ドメインにある約650個のアミノ酸から成り、クラス、サブクラス、および軽鎖定常ドメインにおいて定常ドメインが異なる、約150,000Daの抗体分子である。
本発明の抗IL−23p19mAb hum13B8−bの溶液製剤は、IL−23シグナル伝達の選択的拮抗作用が有益であると期待される障害の治療において使用を見出すであろう。皮膚、関節、CNSの炎症性障害、および増殖性障害は、類似した免疫応答を誘発し、故にIL−23ブロックは、全身性感染と戦う宿主の能力を含むことなく、これらの免疫媒介炎症性障害の阻害をもたらすはずである。IL−23との拮抗は、炎症性腸疾患、クローン病、潰瘍性大腸炎、関節リウマチ、乾癬性関節炎、乾癬、強直性脊椎炎、移植片対宿主病、アトピー性皮膚炎、ならびに様々な他の自己免疫障害および炎症性障害に関連した炎症を軽減するはずである。IL−23阻害剤の使用は、増殖性障害、例えば癌、ならびに自己免疫障害、例えば多発性硬化症、I型糖尿病およびSLEの阻害も提供するであろう。これらの様々な障害におけるIL−23の記載は、以下の公開されたPCT出願:国際公開第04/081190号、同第04/071517号、同第00/53631号;および同第01/18051号パンフレットにおいて見出すことができる。IL−23阻害剤はまた、細菌、マイコバクテリア、ウイルスおよび真菌感染などの慢性感染を含む感染の治療における使用も見出し得る。米国特許第8,263,080号明細書および国際公開第2008/153610号パンフレット参照。
本発明は、配列番号2の配列と同一の2つの軽鎖、および配列番号1の配列と同一の2つの重鎖を含み、同時係属中の本発明の譲受人に譲渡された米国特許第8,293,883号(その開示はその全体が参照により本明細書に組み込まれている)に開示された、抗IL−23p19抗体hum13B8−bの高濃度溶液製剤を提供する。ヒト化軽鎖13B8配列(カッパ定常領域を有する)は配列番号2で提供され、軽鎖可変ドメインはその配列の残基1〜108を含む。ヒト化重鎖13B8配列(γ1定常領域を有する)は配列番号1で提供され、重鎖可変ドメインはその配列の残基1〜116を含む。
本発明の選択された製剤の長期安定性は、様々な貯蔵条件下で3〜24ヶ月貯蔵後に試験された。試料は、2mlガラスバイアル(図4)、30mlバッグ(バルク貯蔵)(図5および6)、またはプレフィルドシリンジ(単回投与市販包装)(図7)中で、様々な温度および湿度レベルでインキュベートされた。試料は、高性能サイズ排除クロマトグラフィー(HP−SEC)、イオン交換クロマトグラフィー(HP−IEX)、SDSキャピラリー電気泳動(CE−SDS、還元および非還元)、およびペプチドマッピングといった方法により、分解および凝集生成物の存在について分析された。抗体安定性は、示差走査熱量測定(DSC)により測定された。生物学的活性は、IL−23結合ELISAおよび細胞ベースの機能アッセイにより評価された。抗体濃度は、280nmのUV吸収により決定された。乳光は、350nm(OD350)で光学密度を測定して決定された。結果は図4〜7で提供されている。
本発明の高濃度溶液製剤は高用量皮下投与に特に適しているが、このような製剤は、他の方法で投与することもできる。投与の適切な経路には、例えば、経口、直腸、経粘膜、または腸管投与;筋肉内、皮内、髄内注射を含む非経口送達、およびくも膜下、直接脳室内、静脈内、腹腔内、鼻内、または眼内注射が含まれてもよい。
本発明は、例えば中枢神経系、末梢神経系、および胃腸管の炎症性障害および状態、ならびに自己免疫障害および増殖性障害の治療において使用するための抗ヒトIL−23p19mAb hum13B−bの高濃度溶液製剤を提供する。
[実施例]
[実施例1]
溶液製剤
1つの実施形態において、本発明のhum13B8−bの溶液製剤は、プレフィルドシリンジまたは自動注射器において1.0mL容量中100mg/mL抗体溶液として提供される。1.0mL容量は、充填容量ではなく、全1.0mL用量の送達を確保するのに十分な過充填を含み得る抽出可能な容量である。本発明の製剤1.0mlの例示的な処方は、表2で提供されている。1つの実施形態において、製剤バッチは、15〜30Lスケール、または約15,000〜30,000用量で調製される。
示差走査熱量測定(DSC)
Valerian−Plotnikov示差走査熱量測定(DSC)が、本発明の製剤における抗体の熱安定性をモニターするのに使用された。DSCは、温度の制御された増減中にタンパク質で生じる熱変化を直接測定する。溶液中のタンパク質は、天然の(折り畳まれた)コンフォメーションとその変性(アンフォールド)状態の間で平衡状態にある。DSCは、タンパク質の50%が変性される温度(熱転移中間点、Tm)を決定するのに使用される。より高いTmを有するタンパク質は、一般的により安定である。例えば、DSC曲線は図1Eで提供されている。図1Eは、2つの転移、すなわちpHに応じておよそ64〜70℃で生じる第1の(Tm1)、およびおよそ80℃で生じる第2の主な転移(Tm2)を示す。主な転移は、Fc断片のアンフォールディングと見なすことができる。主な転移に先立つ(すなわちより低い温度での)熱事象は、抗体(例えばFab)内の幾つかの他のドメインのアンフォールディングに起因する可能性があり、これは文献に記載されたIgG1抗体の予期された構造要素と一致する。Vermeer(2000)Biophys.J.78:394。
高性能サイズ排除クロマトグラフィー(HP−SEC)
本発明のhum13B8−b製剤における高分子量(HMW)種、低分子量種(LMW)、および単量体のパーセンテージは、HP−SECにより検出された。テスト溶液が希釈され、サイズ排除カラム(YMCパックDiol−200、細孔径200Å、300×8.0mm、5μmまたは等量;YMC Co.Ltd.、京都、日本)を用いてHPLCにより分離された。ピーク面積は、単量体パーセンテージ、HMW種およびLMW種パーセンテージを決定するのに使用される。
動的光散乱(DLS)
動的光散乱(DLS)は、溶液中0.5nmから6μmの範囲のモノクローナル抗体のサイズ分布プロファイルについて洞察を与えることができる手法である。モノクローナル抗体hum13B8−bは、強度約9.5nm(および体積6.5nm)の平均流体力学的直径を示す。これは、他のモノクローナル抗体の値と一致する。より高いイオン強度製剤は、低イオン強度製剤より低い流体力学的直径を有する傾向があり、緩衝液およびpHなどの他の溶液パラメーターに影響され得る。図1Iおよび1J参照。本発明の製剤における抗体は、本質的に単分散性である。本発明の抗体の流体力学的直径は、抗体がより高いまたはより低いイオン強度の緩衝液に希釈される実験により証明されているように、可逆性である(データ不図示)。これらの結果は、被験者に投与される場合、流体力学的直径が注射部位の局所環境と平衡化されることを示唆している。
高性能陽イオン交換クロマトグラフィー(HP−IEX)
hum13B8−bの電荷バリアントは、試料中のタンパク質と樹脂に固定化された電荷の間の静電相互作用に依存するHP−IEXにより検出された。正電荷hum13B8−bバリアントは、弱陽イオン交換カラム(Dionex ProPac WCX−10、4×250mmまたは等量;Thermo Scientific、バノックバーン、Ill.、USA)上の負電荷樹脂に結合され、HPLCにより分離される。抗体は、pHおよび塩濃度を高めること、該抗体バリアントの電荷を効果的に減少させること、およびそれを等価電荷のイオンと置換することにより溶出される。溶離液中のhum13B8−bおよびバリアントの存在は、UV検出により決定される。ピーク面積は、酸性バリアント、主要ピーク、主要ピーク後ピークおよび塩基性バリアントを決定するのに使用される。
ペプチドマッピング
ペプチドマッピングは、hum13B8−bの一次構造に関する情報を提供し、酸化が起きているかどうかを評価するのに使用することができる。配列包括度を高めるために、2つの異なる酵素的ペプチドマップを用いてhum13B8−bを特徴付けした。最初は、エンドプロテイナーゼLys−C消化後のペプチドマッピングであった。ペプチドマップは、タンパク質を変性させる塩酸グアニジン、およびジスルフィド結合を還元する1,4−ジチオスレイトール(DTT)でhum13B8−bの試料を処理して得た。試料を次いでヨードアセトアミド(IAM)で処理して、DTT処理から生じた遊離チオールをアルキル化した。試料を次いでエンドプロテイナーゼLys−Cで消化した。得られたペプチドの分析は、エレクトロスプレー質量分析に連結された逆相液体クロマトグラフィー(Xbridge C18、5μm、細孔径130Å、2.1×150mmまたは等量;Waters Corporation、ミルフォード、マサチューセッツ、USA)を用いて行った。軽鎖(L)および重鎖(H)に由来するペプチド配列、ならびに質量分析を用いて検出されたその質量は、表3に示されている。保持時間は、90分にわたるクロマトグラフィー中に214nmの紫外線吸収により決定した。質量は、IAMアルキル化システイン残基当たり57Daの正味質量を添加して、システイン残基のアルキル化について補正した。hum13B8−bの相補性決定領域(CDR)に由来する5個のペプチドは特に興味深い。ピログルタミン酸化N末端重鎖ペプチド、リジン切断C末端重鎖ペプチド、ならびに幾つかのメチオニン酸化および脱アミド化ペプチドなどの一般的な翻訳後修飾ペプチドも、同様に観察された。これらの修飾は、該分子のCDRでは見出されない。日常的なペプチドマッピング実験は、質量分析を用いることなく行うことができ、観察されたピークの保持時間は、安定性(テスト)試料および対照(参照)試料などの試料を比較するのに使用することができる。
UV分光法によるタンパク質濃度
本発明の溶液製剤における抗体hum13B8−bのタンパク質濃度は、UV分光法により決定される。決定は、トリプトファン、チロシンおよびシステイン残基のようなアミノ酸による280nmのUV光の吸収に基づく。280nmの吸収は、320nmの吸収を用いる光散乱について補正される。該方法は、水への試料の重量測定希釈(gravimetric dilution)、ならびに280nmの吸収および320nmの吸収を確立するためのUVスペクトルの記録を含む。これらの吸収値および1.44mL mg−1cm−1の実験的に決定された吸光係数は、hum13B8−b濃度を計算するのに使用される。
生物学的活性−細胞ベースの機能アッセイ
本発明の製剤におけるhum13B8−bの、ヒトIL−23の生物学的活性をブロックする能力を評価するために、機能的細胞ベースのアッセイが開発された。このアッセイは、IL−23応答性細胞系(キット225)においてIL−23誘導STAT3活性化を阻害するhum13B8−bの能力を評価する。hum13B8−b基準材料およびテスト試料の連続希釈は、一定濃度のhumIL−23とインキュベートされ、その後キット225 IL−23応答性細胞とインキュベートされる。hum13B8−bによるSTAT3リン酸化の阻害は、STAT3捕捉抗体および抗p−STAT3検出抗体ペアを用いたELISAと、その後のペルオキシダーゼコンジュゲート抗IgGとのインキュベーションおよび化学発光基質の添加により、細胞溶解物において測定される。阻害応答曲線(「標準曲線」)は、IC50値が最大応答の50%を阻害する抗IL−23の濃度を表す、非線形回帰4パラメーターロジスティックフィットを用いて生成される。テスト試料の相対力価は、標準曲線に対するテスト試料の阻害応答曲線の比較により評価され、基準のパーセントとして計算される。同じ試料の複数の複製物の相対力価値が、単一の報告可能な値−相対力価の幾何平均にまとめられる。
生物学的活性−結合ELISA
本発明の製剤におけるhum13B8−bのヒトIL−23に対する親和性は、基準材料およびテスト試料の連続希釈が、ヒトIL−23サイトカインで被覆されたアッセイプレートに適用される平衡結合ELISAにおいて評価される。このアッセイは例えば、様々な可能性のある治療製剤における生物学的活性の保持を評価するのに使用されてもよい。テスト試料の相対力価は、基準材料の用量応答曲線に対するテスト試料の用量応答曲線の比較により評価され、基準のパーセントとして計算される。
SDSキャピラリー電気泳動(CE−SDS)
ドデシル硫酸ナトリウムを用いるキャピラリー電気泳動(CE−SDS)は、サイズに基づきタンパク質を分離する。CE−SDSは、非還元条件下または還元条件下で行うことができる。還元CE−SDSは、分離された重鎖と軽鎖を互いから分解するのに対し、非還元CE−SDSは、試料中の他の種由来、およびまた試料中の他の可能性のある種由来のインタクトな抗体を分解する。例えば、Rustandiら(2008)Electrophoresis 29:3612頁参照。
沈降速度分析的超遠心分離(SV−AUC)
SV−AUCは、本発明の製剤におけるhum13B8−b抗体の四次構造を調べるのに使用された。SV−AUCは、遠心力に応答して分子が沈降する速度を測定する。この沈降速度は、試料中に存在する分子の分子量に関する情報を提供する。hum13B8−bはほとんどが、7.0Sの沈降係数(S20,W)を有する1つの種として沈降する。この種は、約150kDaの推定分子量を有し、該単量体の予期された分子量と一致する。巨大分子の水和および形状に依存する摩擦比は、ロット間で類似する。
Claims (11)
- a)少なくとも50mg/ml抗IL−23p19抗体hum13B8−bと、
b)pH5.5〜6.0のヒスチジン緩衝液と、
c)界面活性剤と、
d)溶液を等張にするために必要な量の等張化剤であって、スクロース、トレハロース又はマンニトールである等張化剤と
を含み、
前記抗体hum13B8−bが、
i)配列番号2の配列を含む軽鎖ポリペプチドと、
ii)配列番号1の配列を含む重鎖ポリペプチドと
を含む、抗IL−23p19抗体hum13B8−bの安定な溶液製剤。 - 少なくとも80mg/ml抗IL−23p19抗体hum13B8−bを含む、請求項1に記載の安定な溶液製剤。
- 少なくとも100mg/ml抗IL−23p19抗体hum13B8−bを含む、請求項2に記載の安定な溶液製剤。
- 80〜120mg/ml抗IL−23p19抗体hum13B8−bを含む、請求項1に記載の安定な溶液製剤。
- 約100mg/ml抗IL−23p19抗体hum13B8−bを含む、請求項4に記載の安定な溶液製剤。
- 自己免疫疾患、炎症性疾患、または増殖性障害の治療において使用するための、請求項1〜5のいずれかに記載の安定な溶液製剤。
- 年4回の皮下投与を含む乾癬を治療する方法に用いる医薬品の製造における、請求項4に記載の安定な溶液製剤の使用。
- 皮下投与がシリンジ、自動注射器、注射器ペンまたは無針注射装置を用いる注射により行われる、請求項7に記載の使用。
- 界面活性剤がポリソルベート80であり、等張化剤がスクロースである、請求項1に記載の安定な溶液製剤。
- 界面活性剤が0〜0.05%(W/V)のポリソルベート80であり、等張化剤が7%(W/V)スクロースである、請求項9の安定な溶液製剤。
