Description
《多機能T細胞》
多機能T細胞という用語によって、IFN−γ、IL−2及びTNF−αの総てのサイトカイン、あるいはIL−2及び別の2つのサイトカインIFN−γ及びTNF−αのうちの少なくとも1つを同時に発現するT細胞と解釈される。
本発明における用語「ポリペプチド」の意味は通常のものである。それは、完全長のタンパク質、オリゴペプチド、短いペプチド及びその断片を含む任意の長さのアミノ酸鎖であり、アミノ酸残基は共有結合性のペプチド結合により結合している。
用語「配列相同性」は同一の長さの2のアミノ酸配列の間、又は同一の長さの2つのヌクレオチド配列間の相同性の度合の定量的な測定を意味する。比較すべき2の配列は、ギャップの挿入、あるいはタンパク質配列の末端の切断を可能にする推定される最良の適合度まで整列しなければならない。配列相同性は:
として算出することができる。ここでNdifとは整列した場合の2の配列における非同一性の残基の総数であり、Nrefは配列のうちの1つの残基の数である。従って、DNA配列AGTCAGTCは、配列AATCAATCと75%の配列相同性がある(Ndif=2及びNref=8)。ギャップは非同一な1以上の特異的な残基として計数される。即ち、DNA配列AGTGTCはDNA配列AGTCAGTCと75%の配列相同性がある(Ndif=2及びNref=8)。配列相同性は代替的に、BLASTプログラム、例えばBLASTPプログラムによって計算することができる(Pearson,1988、又は「www.ncbi.nlm.nih.gov/cgi−bin/BLAST」)。発明の一態様においては、アラインメントは、Thompson J.ら、1994に記載のような初期設定のパラメータが「http://www2.ebi.ac.uk/clustalw/」で入手可能な配列アラインメント法Clustal Wで実行することができる。
本発明の好適な実施態様においては、ポリペプチドはB細胞又はT細胞に対するエピトープといったポリペプチドの免疫原性部分を含む。ポリペプチドの免疫原性部分はポリペプチドの一部であり、動物又はヒトにおいて、及び/又は本明細書中に記載の生物学的アッセイのいずれかよって定量した生物学的試料において免疫反応を誘発する。ポリペプチドの免疫原性部分はT細胞エピトープ、又はB細胞エピトープであってもよい。免疫原性部分はポリペプチドの1又は少数の比較的少さい部分に関連づけでき、ポリペプチド配列にわたって分散するか、又はポリペプチドの特異的な部分に位置しうる。いくつかのポリペプチドについては、エピトープが完全配列をカバーするポリペプチドにわたって散在することが示されている(Ravnら、1999)。免疫反応中に認識される関連するT細胞エピトープを同定するために、MHCクラスIIエピトープの検出用の、好適には合成性であり、長さが例えば、ポリペプチド由来の20のアミノ酸残基の重複するオリゴペプチドを用いることが可能である。これらのペプチドは生物学的アッセイ(例えば、本明細書中に記載のIFN−γアッセイ)で試験することができ、これらの一部は、ペプチドにおけるT細胞エピトープの存在の証拠として陽性反応を与える(及びこれによって、免疫原性となる)。ESAT−6及びCFP10については、このような研究によって、抗原の総ての部分がT細胞エピトープを含むことを示した(Mustafaら、2000;Arend SMら、2000)。MHCクラスIエピトープの検出については、結合するペプチドを予測することが可能であり(Stryhnら、1996)、その後にこれらのペプチドの合成物を産生し、関連する生物学的アッセイ、例えば本明細書に記載のようなIFN−γアッセイで試験することが可能である。ペプチドの長さは好適には、ポリペプチド由来の8乃至11のアミノ酸残基となる。B細胞エピトープは、例えばHarboeら、1998に記載のように対象のポリペプチドをカバーする重複するペプチドに対するB細胞の認識を分析することによって定量できる。この定義と一致して、本明細書中に記載のポリペプチドの免疫原性部分は、免疫応答を誘発する部分として同定できる。以下の「免疫応答」の定義を参照。
本発明のポリペプチドの共通の特徴は、実施例に示すように免疫学的応答を誘発する能力である。置換、挿入、付加又は削除により産生される本発明のポリペプチド変異体は更に、本明細書中に記載のアッセイのいずれかによって定量される免疫原性となりうることは理解すべきである。
免疫個体はヒト又は動物として定義され、病原性マイコバクテリアでの感染を排除又は制御しているか、あるいはマイコバクテリウム・ボビスのBCGワクチンでのワクチン接種といった、予防的なワクチン接種を受けている。
免疫応答は下記の方法のうちの1つでモニタリングできる。
一般的には、マイコバクテリウム・ツベルクローシスの抗原、及びこのような抗原をコードするDNA配列は様々な手順のうちのいずれか1つを用いて調製する。それらは、上述したような手順によってマイコバクテリウム・ツベルクローシスの細胞又は培養濾液から天然タンパク質として精製できる。免疫原性抗原は更に、発現ベクターに挿入し、好適な宿主に発現した、抗原をコードしたDNA配列を用いて組換え技術によって産生できる。宿主細胞の例は大腸菌である。そのポリペプチド又は免疫原性部分は更に、約100未満のアミノ酸で、一般的には50未満のアミノ酸で合成的に産生でき、アミノ酸が増殖中のアミノ酸鎖に順次添加される商業上利用可能な固相技術といった、当該技術分野における当業者に公知の技術を用いて産生できる。
免疫原性ポリペプチド更には融合タンパクとして産生でき、それの方法によって、本発明のポリペプチドの優れた特性が得られる。例えば、組換えで産生した場合にポリペプチドの輸送を促進する融合パートナー、ポリペプチドの精製を促進する融合パートナー、及び本発明のポリペプチド断片の免疫原性を強化する融合パートナーは、総て可能性のある対象である。従って本発明は更に、上に規定した少なくとも1のポリペプチド又は免疫原性部分と、少なくとも1の融合パートナーとを含む融合ポリペプチドに関する。免疫原性を強化するために、融合パートナーはマイコバクテリウム・ツベルクローシス由来の他のポリペプチド、例えば、ESAT−6、CFP10、EspA、TB10.4、RD1−ORF5、RD1−ORF2、Rv1036、MPB64、MPT64、Ag85A(MPT59)、MPB59、Ag85C、19kDaリポプロテイン、MPT32及びαクリスタリン(alpha−crystallin)、又は上述の抗原のいずれかのT細胞エピトープといった結核菌群に属する細菌に由来するポリペプチド断片にできる(Skjotら、2000;国際公開公報第0179274号;国際公開公報第0104151号;米国特許出願第09/0505,739号;Rosenkrandsら、1998;Nagaiら、1991)。本発明は更に、本発明のポリペプチド(又はその免疫原性部分)のうちの2又はそれ以上の相互融合を含む融合ポリペプチドに関する。他の融合パートナーは、本製品の免疫原性を強化でき、IFN−γ、IL−2及びIL−12といったリンホカインである。発現及び/又は精製を促進するために、融合パートナーは例えば:線毛成分のピリン(pilin)とpapA;タンパク質A;ZZ−ペプチド(ZZ−融合剤はスウェーデンのPharmacia社によって市販されている);マルトース結合タンパク質;グルタチオンS−トランスフェラーゼ;β−ガラクトシダーゼ;又はポリヒスチジン;といった細菌の線毛タンパク質にできる。融合タンパク質は宿主細胞における組換えで産生でき、大腸菌にでき、それは様々な融合パートナーとの間のリンカー領域を誘発する可能性がある。
《ワクチン》
ワクチンは特定の疾病に対する免疫を確立又は改善する生物学的製剤である。ワクチンは、予防的(例えば、任意の天然型又は「野生型」の病原体によって将来の感染の作用を予防又は改善するために)、事後曝露的(例えば、臨床的な兆候がない潜伏感染した個体の再活性化を予防するために)、又は治療的(例えば、治療を短縮するために、単独で、あるいは抗生物質治療との組み合わせのいずれかで能動的な疾患を治療するために用いられるワクチン)にできる。
マイコバクテリウム・ツベルクローシスで低度の潜伏感染を誘発するために、動物は最初に通常の用量のマイコバクテリウム・ツベルクローシス(肺において約150の細菌)を用いてエアロゾル感染を与える。感染の6週間後、動物は次いで、総てではないが殆どの細菌が根絶する6週間の準最適な化学療法的治療を与える。残余の細菌は潜伏感染を確立する。この化学療法的治療に続いて、一部の動物にワクチン接種して、ワクチンが化学療法的治療後5乃至15週間に自然発生する、潜伏感染の再活性化を予防する能力を試験する。図2参照。
本発明の別の部分は、ポリペプチド(又はその少なくとも1の免疫原性部分)あるいは本発明による融合ポリペプチドを含むワクチン組成物に関連する。このようなワクチン組成物の最適な効能を保証するために、免疫学的及び薬学的に許容される担体、賦形剤、又はアジュバントを含むことが好適である。
本発明の核酸断片は、抗原の生体内発現に影響を与えるために用いることができる。すなわち、核酸断片は、Ulmerら、1993により概説されるようにいわゆるDNAワクチンに用いることができ、引用によって含まれている。従って、本発明は更に、本発明の核酸断片を含む曝露後ワクチンにも関連しており、そのワクチンは、ワクチンを投与したヒトを含む動物によって抗原の生体内発現に影響を与え、発現した抗原の量は、ヒトを含む動物において病原性マイコバクテリアによって引き起こされる感染を治療するのに有効である。このようなDNAワクチンの有効性は、DNA断片が免疫応答を調節する能力のあるポリペプチドをコードするのと一緒に、発現産物をコードする遺伝子を投与することによって場合によっては高めることができる。
曝露後ワクチンの細胞性免疫応答を効果的に活性化するための1の可能性は、非病原性の細菌又はウィルスに、ワクチン中の関連する抗原を発現させることによって得ることができる。このような細菌の公知例としては、マイコバクテリウム・ボビスBCG、サルモネラ及びシュードモナスであり、ウイルスの例としては、ワクシニアウイルス、及びアデノウイルスである。従って、本発明の別の重要な態様は、現在利用可能なBCG生ワクチンの改良であり、上に規定のような1又はそれ以上のポリペプチドをコードするDNA配列の1又はそれ以上のコピーは、細菌がポリペプチドを発現及び分泌できる方法で細菌のゲノムに組み込んでいる。本発明のヌクレオチド配列の2以上のコピーの組込みは、免疫反応の強化を予期するものである。別の可能性は、ワクシニアウイルス又はアデノウイルスなどの弱毒ウイルス内に本発明のポリペプチドをコードするDNAを組み込むことである(Rolphら、1997)。組換えワクシニアウイルスは、感染した宿主細胞の細胞質内で増殖でき、ひいては対象のポリペプチドは免疫応答を誘発でき、TBに備えて防御を誘発することが想定される。本出願で引用した総ての特許文献及び非特許文献はその全体が引用によってここに組み込まれている。本発明を、以下の非限定的な実施例で更に詳細に説明する。
《ワクチン接種用のマウス結核モデル》
コーネルモデルは潜伏性TBの研究用マウスモデルとして広く用いられている。このモデルは、再活性化を予防するワクチン候補の能力の試験用に我々の研究室で適用した。マウスは最初にエアロゾルにより病原性のマイコバクテリウム・ツベルクローシスに感染させ、そして感染後6週間で抗生物質治療を始めて細菌負荷を減らす。これは、マウスに自発発生しないヒト感染の潜伏期を模倣するためである。この潜伏期(細菌数が連続して低いステージ)の間、マウスを2回免疫し、ワクチンによる再活性化を予防する能力は、最後の免疫から20週後に脾臓と肺の生きたマイコバクテリウム・ツベルクローシスを培養することにより定量する。実験の長いスパンは、ワクチン効果の読み出しの必要条件である疾患の再活性化に十分な時間を与えるために必要である(図3)。
《ESAT6により防御を誘発するが、Ag85により防御を誘発しない曝露後ワクチン》
ESAT−6及びAg85Bは単一成分として、更にAg85B−ESAT6(H1)の融合タンパクとしての双方で、防御的であることが証明されている。しかし、これらの抗原は曝露後モデル(実施例1で上述したように)で試験すると、ESAT6のみが防御効果があり、再活性期中の細菌増殖を制御する(図4)。更に、図4Bに示すように感染後18週間程度の早さで、再活性の防御自体が顕在化し、この防御は実験期間中(感染後40週まで)維持された。これは対照と比較して細菌負荷の有意な減少がなかったAg85Bを曝露後ワクチンとして用いた場合に観察されるものとは対照的である(図4C及びD)。更に、我々はTB抗原Ag85B及び予防的設定で有望な有効性を示したESAT−6で構成されるH1融合タンパク質を評価した。この分子は、SSI曝露後モデルにおいて曝露後ワクチンとして用いられた場合、有意に細菌数を減少させることができた(図4E)。
[ESAT6ペプチド混合物の曝露後ワクチンにより誘発される防御]
上述の実施例で示したように、ESAT−6分子は曝露後に投与された場合、非常に活性があり、対照群と比較して、かつ更にAg85Bと比較して細菌負荷が減少する。更に、我々は組換えタンパク質の代わりに、重複するペプチドの統合物として与えたESAT−6が更に、対照群やESAT6の強い活性及び曝露後ワクチンとして機能する能力を示すAg85Bの両方と比較して、再活性に対して更に良好な防御となることを示した。

《多機能性T細胞を誘発するESAT−6での曝露後ワクチン接種》
ESAT−6に特異的な細胞のサイトカイン発現プロファイルでのESAT−6による曝露後ワクチンの効果を実験するため、マウスは最初にエアロゾル病原性のマイコバクテリウム・ツベルクローシスに感染させ、感染後6週間での抗生物質治療を開始して細菌負荷を減少させ潜伏感染を確立させた。潜伏期中に3週間の間隔で3回ワクチンを接種し(図3に示すように)、ESAT−6ワクチンが多機能性T細胞の数に影響し、マイコバクテリウム・ツベルクローシスの再活性を予防する能力は最後のワクチン接種から20週間後に定量した。結果は、非接種群では相応のESAT−6に対する反応があるが、サイトカインの発現プロファイルはESAT−6ワクチン接種群と比較して特に多機能性T細胞の点で著しく異なる(IFN−γ+/TNF−α+/IL−2+CD4細胞)ことを示していた(図5)。従って、非接種群と比較して、IFN−γ/TNF−αのCD4T細胞数の減少と、IFN−γ/TNF−α/IL−2を共に発現している3重に陽性の多機能性CD4T細胞数の増加を観察した。ESAT−6ワクチンを接種した動物の肺で、増加した多機能T細胞の存在が減少した細菌数と相関していた(図5D)。
《初期及び後期段階の感染に関連する他の抗原に比較して、更に一貫して再活性化を予防するESAT6での曝露後ワクチンの接種》
どの抗原が一貫して再活性化を防御するかを決定するため、実施した総ての曝露後の実験に基づいて規格化したデータの統合解析を行った。個々の実験のデータセットは、1のグループ内の個々のマウスの細菌数を対照群の細菌数の中央値と比較することで標準化した。すなわち各データポイントは、対照群の中央値CFUと個々の動物のCFUの差(Log10CFU対照の中央値−Log10CFUワクチン群)を表している(図6)。図6Aにおいては、潜伏期に関連する抗原Rv0569、Rv2031cとAg85B、ESAT6、Rv3871、Rv3905及びRv3445であり、後者の2つはESAT6ファミリーである初期抗原とについての統合したデータの比較によって、他の抗原の評価と比較してESAT6ワクチン接種動物が、顕著に良好に防御されていることを示している。更に、防御レベルはRv3871での曝露後ワクチン接種後に到達し、ESX−1タンパクは更に他の抗原と比較して上昇しているように見える(図6A)。更に、特にESAT6の活性を示すために、我々はESAT6により与えられる防御を、どちらもESAT6を含んでいる2つの融合構造物であるH1(Ag85B−ESAT6)とH56(Ag85B−ESAT6−Rv2660)と比較した(図6B)。分析によって、上述の2つの融合構造物が含まれている場合に再活性化に対する防御におけるESAT6の活性が得られることを示した。
《再活性化のプロセスの阻害効果があると思われるESX−1ファミリーの他のメンバーであるRv3871での曝露後ワクチン接種》
我々はESAT6と並行してESX−1ファミリーの他のメンバーを評価し、ESAT6で誘発される免疫応答ほどではないものの(図7A)、Rv3871での曝露後ワクチン接種はRv3871に特異的な免疫応答(図7B)を誘発をすることを見出した。それにもかかわらずESAT6とRv3871で誘発された免疫応答は両方共、生理食塩水対照動物に比べて大きかった。ワクチンに特異的な免疫応答の誘発は、生理食塩水群と比較して、両方のワクチン群における細菌負荷の低下(中央値)と関連していた。このことは、Rv3871がESAT6と比較した再活性化に対して同様の防御効果が、細菌数が対照群においてレベルがある程度上昇したのと比較して、これら2つの群で同程度の細菌数レベルによって示されたことを意味する(図7C)。

Claims (14)
- 潜伏感染した個体に投与されかつ結核の再活性を予防するワクチン又は免疫原性組成物であって、前記ワクチン又は免疫原性組成物がマイコバクテリウム・ツベルクローシスの感染中に恒常的に発現している抗原ポリペプチド又は当該抗原をコードする核酸を含み、前記ワクチン又は組成物が、IC31又はポリI:Cを有する又は有さないDDA/TDBを具えるアジュバントをさらに含み、
前記恒常的に発現している抗原ポリペプチドが、ESX−1分泌系に属し、かつ:
i)配列番号4;及び
ii)(i)における配列と少なくとも90%の配列同一性を有し、かつ同時に免疫原性であるアミノ酸配列の類似体;
からなる群から選択されることを特徴とするワクチン又は組成物。 - 請求項1に記載のワクチン又は組成物が、配列番号2、3、5、6、7、8、9、10、11、12、13、14、15、16、17、18、32、33及び34またはこれらの免疫原性部分からなる群から選択される少なくとも1つの追加の抗原ポリペプチドを含むことを特徴とするワクチン又は組成物。
- 請求項1に記載のワクチン又は組成物において、前記抗原ポリペプチドがマイコバクテリア科の細菌によって発現した抗原と融合した融合パートナーであることを特徴とするワクチン又は組成物。
- 請求項3に記載のワクチン又は組成物において、前記融合パートナーが恒常的に発現している抗原であることを特徴とするワクチン又は組成物。
- 請求項1乃至4のいずれか1項に記載のワクチン又は組成物が:
マイコバクテリアの遺伝子を発現する細菌又はウイルスから選択される遺伝子改変生物である組換え生ワクチンから選択される追加の送達系;あるいは
配列番号2、3、5、6、7、8、9、10、11、12、13、14、15、16、17、18、32、33及び34またはこれらの免疫原性部分からなる群から選択される抗原ポリペプチドをコードする遺伝子又は遺伝子断片を発現するプラスミドであるDNAワクチン、又はタンパク質自体若しくはアジュバント等の送達系で送達される前記タンパク質自体に由来する合成ペプチドであるタンパク質ワクチンから選択される免疫原性送達系;
を更に含むことを特徴とするワクチン又は組成物。 - 請求項1乃至5のいずれか1項に記載のワクチン又は組成物において、前記抗原ポリペプチドがポリペプチドの自己アジュバント効果を可能にするよう脂質化されていることを特徴とするワクチン又は組成物。
- 請求項1乃至5のいずれか1項に記載の、潜伏結核の治療用のワクチン又は組成物。
- マイコバクテリウム・ツベルクローシス、マイコバクテリウム・アフリカヌム、又はマイコバクテリウム・ボビスから選択される病原性マイコバクテリアによって引き起こされる結核感染の再活性に備えた、ヒトを含む動物の治療用の薬剤の製造のための請求項1乃至5のいずれか1項に記載のワクチン又は免疫原性組成物の使用。
- 請求項8に記載の使用において、前記ワクチン又は免疫原性組成物が急性感染期後に、及び/又は、潜伏感染期中に投与されることを特徴とする使用。
- 請求項8に記載の使用において、前記使用が病原性マイコバクテリアに潜伏感染している対象を同定するステップを具えることを特徴とする使用。
- 請求項10に記載の使用において、病原性マイコバクテリアに潜伏感染している前記対象がマントーツベルクリン皮膚試験(TST)、クァンティフェロン試験、HBHAに対する反応の生体外での検出、又は恒常的に発現している抗原による刺激後のIP10の検出から選択される診断使用で同定されることを特徴とする使用。
- マイコバクテリウム・ツベルクローシス、マイコバクテリウム・ボビス、及びマイコバクテリウム・アフリカヌムから選択される結核菌群の種によって引き起こされる潜伏感染の再活性化に備えるワクチン又は免疫原性組成物の製造用の抗原ポリペプチドの使用であって、
前記抗原ポリペプチドが、ESX−1分泌系に属し、かつ:
i)配列番号4;及び
ii)(i)における配列と少なくとも90%の配列同一性を有し、かつ同時に免疫原性であるアミノ酸配列の類似体;
からなる群から選択されることを特徴とする使用。 - 請求項12に記載の使用において、前記ワクチン又は組成物が急性感染期後、及び/又は潜伏感染期中の投与用であることを特徴とする使用。
- 請求項12又は13に記載の使用において、前記ワクチン又は組成物が、配列番号2、3、5、6、7、8、9、10、11、12、13、14、15、16、17、18、32、33及び34またはこれらの免疫原性部分からなる群から選択される少なくとも1つの追加の抗原ポリペプチドを含むことを特徴とする使用。
