Claims (19)
- マトリックス支援レーザー脱離によるイオン化装置を備えた質量分析計における栄養培地表面の微生物の質量分析のための方法であって、該方法が、次の工程:
(a)サンプル支持体の接触表面の、栄養培地表面への直接接触により、微生物を移動する工程、
(b)該移動された微生物を前記サンプル支持体の前記接触表面上で細胞破壊し、かつ、該破壊された微生物の分子成分を、MALDIサンプルとして調製する工程、及び
(c)該MALDIサンプルを備えた前記サンプル支持体を、該質量分析計に移して、分析を行う工程、
を含む、方法。 - ピン形状サンプル支持体の端表面が、微生物に接触し、該ピン形状サンプル支持体の該接触表面が非常に小さいため、個々のコロニーの微生物のみが該ピン形状サンプル支持体に移動される、請求項1に記載の方法。
- 前記微生物が移動された後、前記ピン形状サンプル支持体がアダプタープレートに挿入され、該ピン形状サンプル支持体の端表面が本質的に、アダプタープレートの表面と揃って平らになるようにされる、請求項2に記載の方法。
- 次の工程:
栄養培地の表面の画像を取得する工程、
該画像から、コロニーの位置を決定する工程、
決定された該位置の微生物を、それぞれにおいてピン形状サンプル支持体に接触移動させる工程、
該ピン形状サンプル支持体を、アダプタープレートに挿入する工程、
該ピン形状サンプル支持体の上の微生物から、MALDIサンプルを調製する工程、
該アダプタープレートを質量分析計に導入する工程、及び、
該アダプタープレート中の該ピン形状サンプル支持体の該位置で、マトリックス支援レーザー脱離によるイオン化のスペクトルを取得する工程、
を含む、請求項3に記載の方法。 - 前記ピン形状サンプル支持体の前記端表面が、9平方ミリメートル未満の表面を有する、請求項2〜請求項4のいずれか1項に記載の方法。
- プレート形状サンプル支持体が、微生物に接触し、該プレート形状サンプル支持体の該接触表面が非常に大きいため、複数のコロニーの微生物が該プレート形状サンプル支持体に同時に移動される、請求項1に記載の方法。
- 前記微生物が前記プレート形状サンプル支持体に移動された後に、前記接触表面の画像が取得され、該プレート形状サンプル支持体上の該移動された微生物の位置が、該画像から決定される、請求項6に記載の方法。
- 前記撮像の後、MALDIサンプルが前記決定された位置でのみ調整され、かつ、前記質量分析が該調製されたMALDIサンプルでのみ実施される、請求項7に記載の方法。
- 前記撮像の後、マトリックス溶液が前記サンプル支持体の接触表面全体に適用され、かつ、前記質量分析が該決定された位置でのみ実施される、請求項7に記載の方法。
- 前記分析のためのサンプルを、前記栄養培地の前記表面に接種する際に、該接種の軌跡を記録し、かつ、前記質量分析が該記録された軌跡に沿ってのみ実施される、請求項6に記載の方法。
- 前記サンプル支持体上の前記微生物が、好適に攻撃的なマトリックス溶液を加えることによって細胞破壊される、請求項1〜請求項10のいずれか1項に記載の方法。
- 前記微生物が、前記栄養培地の前記表面で8時間未満培養されてから、前記サンプル支持体に移動される、請求項1〜請求項11のいずれか1項に記載の方法。
- 前記栄養培地の前記表面が、培養プロセス中に微生物コロニーの成長について繰り返し検査され、コロニーが検出されたときに接触移動が行われる、請求項1〜請求項12のいずれか1項に記載の方法。
- 前記サンプル支持体の前記接触表面が、タンパク質吸着性又は接着性領域を備えて調製される、請求項1〜請求項13のいずれか1項に記載の方法。
- 質量分析計における栄養培地表面の微生物の質量分析のための方法であって、次の工程:
(a)サンプル支持体の接触表面の、栄養培地上の1又は複数のコロニーへの直接接触により、前記接触表面に微生物を移動する工程、
(b)該移動された微生物を前記サンプル支持体の前記接触表面上で細胞破壊する工程、及び、
(c)該破壊された微生物の分子成分を備えた前記サンプル支持体を、前記質量分析計に導入して分析を行う工程、
を含む、方法。 - 分子成分の質量スペクトルを参照用質量スペクトルと比較することにより、栄養培地表面で成長した微生物を同定するための方法であって、次の工程:
(a)サンプル支持体接触表面を、該栄養培地上の1つ以上のコロニーと直接接触させることにより、微生物を移動させる工程、
(b)該移動された微生物を細胞破壊し、その分子成分を調整して、該サンプル支持体接触表面上で質量スペクトルを取得する工程、及び、
(c)該調製されたサンプルを備えた該サンプル支持体を、質量分析計に導入する工程、及び、
(d)該微生物の該分子成分の質量スペクトルを取得してその同定を行う工程、
を含む、方法。 - 分子成分の質量スペクトルを参照用質量スペクトルと比較することにより、栄養培地表面で成長した微生物を同定するための装置であって、
サンプル支持体(3, 5)の接触表面を、該栄養培地上の1つ以上のコロニーと直接接触させることにより、微生物を移動させる機構(25, 26, 27)と、
該移動された微生物を細胞破壊し、その分子成分を調整して、該サンプル支持体接触表面上で質量スペクトルを取得する手段と、
