Description
マーカー及び/又は標的/分子標的に基づいて抗体若しくは抗原結合フラグメント、一本鎖オリゴヌクレオチド若しくは二本鎖オリゴヌクレオチド、核酸、短ペプチド又はその低分子配合剤を開発及び/又はスクリーニングする工程(ここで、抗体若しくは抗原結合フラグメント、一本鎖オリゴヌクレオチド若しくは二本鎖オリゴヌクレオチド、核酸、短ペプチド又はその低分子配合剤が、マーカー若しくは標的/分子標的の遺伝子発現及び/又はタンパク質活性を低減するか、又はマーカー又は標的/分子標的を標的化した後に細胞傷害性反応を引き起こす)、続いて治療的に有効な量の少なくとも1つの抗体若しくは抗原結合フラグメント、一本鎖オリゴヌクレオチド若しくは二本鎖オリゴヌクレオチド、核酸、短ペプチド又はその低分子配合剤を、それを必要とする被験体に投与する工程を含む、方法を更に提供する。
1)腫瘍発生細胞に富んだ再発性腫瘍源に由来する細胞(Hep−12細胞等)を用いて動物(balb/cマウス等)を免疫化するとともに、腫瘍発生細胞のマーカー及び/又は腫瘍治療の標的/分子標的(CACNA2D1等)を探索、識別又は同定する工程、
2)工程1)において探索、識別又は同定したマーカー及び/又は標的/分子標的(CACNA2D1等)に基づいて、抗体若しくは抗原結合フラグメント、一本鎖オリゴヌクレオチド若しくは二本鎖オリゴヌクレオチド、核酸、短ペプチド又はその低分子配合剤を開発及び/又はスクリーニングする工程(ここで、抗体若しくは抗原結合フラグメント、一本鎖オリゴヌクレオチド若しくは二本鎖オリゴヌクレオチド、核酸、短ペプチド又はその低分子配合剤が、マーカー又は標的/分子標的の遺伝子発現及び/又はタンパク質活性を低減するか、又はマーカー又は標的/分子標的を標的化した後に細胞傷害性反応を引き起こす)、
3)治療的に有効な量の少なくとも1つの工程2)において開発又はスクリーニングした抗体若しくは抗原結合フラグメント、一本鎖オリゴヌクレオチド若しくは二本鎖オリゴヌクレオチド、核酸、短ペプチド又はその低分子配合剤を、それを必要とする被験体に投与する工程、
を含む、方法を提供する。
(i)重鎖(以下、H鎖と称する)(重鎖の可変領域は相補性決定領域(CDR)CDRH1(配列番号1)、CDRH2(配列番号2)及びCDRH3(配列番号3)を含む)、又は、
(ii)軽鎖(以下、L鎖と称する)(軽鎖の可変領域は相補性決定領域CDRL1(配列番号4)、CDRL2(配列番号5)及びCDRL3(配列番号6)を含む)、又は、
(iii)(i)及び(ii)の両方、
を含む。
本発明において使用される幾つかの用語を以下に規定する。

The properties of tumor-initiating cells from a hepatocellular carcinoma patient's primary and recurrent tumor, Carcinogenesis, 2010; 31(2):167-74.の細胞対に関する詳細な背景を参照されたい)。肝臓がん細胞株HuH7(日本学術振興会)、HepG2(ATCC)、SMMC−7721;乳がん細胞株ZR−75(ATCC)、MCF−7(ATCC)、MDA−MB−231(ATCC)、BICR−H1(北京がん病院(Beijing Cancer Hospital)のXinfu Huang教授より提供);肺がん細胞株A549(ATCC)、Calu−3(ATCC)、Calu6(ATCC)、PG(北京大学医学部基礎医学院のBingquan Wu教授より提供);食道がん細胞株KYSE150、KYSE510(北京大学医学部基礎医学院のFengmin Lu教授より提供);胃がん細胞株BGC823、MGC803、SGC7901;前立腺がん細胞株PC3M1E7、PC3M2B4は一般的な細胞株であったため、本発明者らの研究室に保存されている。
a)マウスの免疫化及び細胞融合:
Hep−11細胞及びHep−12細胞を用いて、6週齢の雌性Balb/Cマウス(Vital River Laboratory Animal Technology Co., Ltd.(Beijing))を、サブトラクティブ免疫化(Brooks, P. C., Lin, J. M., French, D. L., and Quigley, J. P.. Subtractive immunization yields monoclonal antibodies that specifically inhibit metastasis. J Cell Biol,1993, 122, 1351-1359、Rasmussen, N., and Ditzel, H. J.. Scanning the cell surface proteome of cancer cells and identification of metastasis-associated proteins using a subtractive immunization strategy. J Proteome Res, 2009, 8:5048-5059)によって免疫化した。ほぼ飽和密度のHep−11細胞をPBSで3回洗浄した後、セルスクレーパーによって細胞を回収した。これらの細胞を遠心分離後に滅菌PBSに懸濁した(約5×106細胞/ml)。その後、4週〜6週齢の雌性Balb/cマウスに、調製した懸濁液を動物1匹当たり0.5ml、合計で4匹の動物に腹腔内接種した。Hep−11細胞の接種後2日目及び4日目に、シクロホスファミド(Sigma-Aldrich(St Louis,MO))をマウスに腹腔内注射した(200mg/kg(体重))。Hep−12細胞を、Hep−11細胞について上記に記載したものと同じ手順で18日目に調製した。各々のマウスにHep−12細胞(2.5×106細胞/0.5ml PBS)を腹腔内接種した。免疫化を、3週間毎に同じ量のHep−12細胞を用いて合計で3回強化した。最後の免疫化強化の3日後に脾臓を分離して、細胞懸濁液を調製した。これらの細胞を108個のSP2/0細胞(ATCC)と混合した。無血清RPMI1640培地(Invitrogen)によって2回洗浄した後、混合細胞を50%PEG4000(Sigma-Aldrich)中で従来のプロトコルに従って融合させた。細胞を、HAT(Sigma-Aldrich)及び15%仔ウシ血清を含有する1640培地に再懸濁した。次いで、細胞を96ウェル細胞培養プレートにプレーティングし、CO2インキュベーター内で培養した。5日後、培養培地を、HAT及び15%仔ウシ血清を含有する新たな1640培地と交換した。細胞融合の約2週間後に(ハイブリドーマの成長に応じて異なる)、上清をサンプリングして、特異的抗体を分泌するハイブリドーマクローンを試験し、スクリーニングした。
15%仔ウシ血清を含有する1640培地中のHep11肝臓がん細胞株及びHep12肝臓がん細胞株を、それぞれ96ウェル細胞培養プレート上にプレーティングした。細胞が壁に接着して収束した(converged)後、上清を捨て、0.125%グルタルアルデヒドを含有する予冷したPBSを添加した。プレートを室温で5分間静置した後、液体を捨てた。細胞をPBSで3回洗浄した。次いで、5%脱脂粉乳を含有するPBSをプレートに添加し、プレートを4℃で一晩ブロッキングした。ブロッキング溶液を除去した後、培養したハイブリドーマクローンを含む上清を添加し、プレートを室温で1時間静置した。上清を除去し、プレートをPBSで2回洗浄した。ホースラディッシュペルオキシダーゼ標識ヤギ抗マウス抗体を、5%脱脂粉乳を含有するPBSで希釈し、均一に混合した。次いで、混合物を96ウェル細胞培養プレートに添加し、プレートを室温で1時間インキュベートした。次いで、上清を捨て、プレートをPBSで5回洗浄した。ELISAの基質溶液を添加した後、反応を遮光下、室温で30分間継続してから、元の溶液と同一体積の12.5%硫酸を添加して反応を停止した。プレートをELISAリーダーに入れ、光学密度を492nmで測定した。Hep11に陰性かつHep12に陽性のハイブリドーマクローンを選抜し、限界希釈法を用いてサブクローニングした。3回の連続サブクローニング後に安定して陽性のハイブリドーマクローンをより大幅に培養し、得られた細胞を冷凍状態で保管した。
拡大培養の後、特異的抗体1B50−1を分泌するハイブリドーマクローンを、プリスタン(Sigma-Aldrich)で前処理した雌性Balb/cマウスに腹腔内接種した(2×106細胞/マウス)。マウスを約1週間後に屠殺し、腹水を更なる試験のために採取した。プロテインGアフィニティークロマトグラフィーを常法により行い、1B50−1を精製した。精製した1B50−1の濃度を、以下の方程式に従って算出した:抗体(mg/ml)=OD280×0.6868。SDS−PAGEを用いて抗体の純度を分析し、純度が電気泳動グレードに達した抗体を関連実験に使用した。
1B50−1ハイブリドーマから分泌された抗体のサブタイプを、Mouse Monoclonal Antibody Isotyping Reagents(カタログ番号Iso2−1KT、Sigmal-Aldrich(St Louis,MO,USA))の使用説明書で推奨される手順に従って決定した。サブタイプ特異的抗体をPBSで1000倍に希釈した後、96ウェル試験プレートに1ウェル当たり100μL、2連(two replicates)で添加した。プレートを37℃で1時間インキュベートした後、PBSで3回洗浄した。その後、1B50−1ハイブリドーマの上清を添加し、プレートを1時間インキュベートしてから、PBSで3回洗浄した。ホースラディッシュペルオキシダーゼ標識ヤギ抗マウス抗体を、5%脱脂粉乳を含有するPBSで希釈した後、試験プレートに添加した。プレートを室温で1時間インキュベートした後、PBSで5回洗浄した。ELISAの基質溶液を添加した後、反応を遮光下、室温で30分間継続してから、元の溶液と同一体積の12.5%硫酸を添加して反応を停止した。
全細胞RNAを1B50−1ハイブリドーマ細胞からTrizol法によって抽出し、逆転写酵素Superscript III(Invitrogen)を用いてcDNAに逆転写した。1B50−1の重鎖可変領域及び軽鎖可変領域をそれぞれ、重鎖可変領域用の5’末端縮重プライマー5’−CTTCCGGAATTCSARGTNMAGCTGSAGSAGTC−3’+5’−CTTCCGGAATTCSARGTNMAGCTGSAGSAGTCWGG−3’、重鎖可変領域用の3’末端プライマー5’−GGAGGATCCAGGGACCAAGGGATAGACAGATGG−3’、軽鎖可変領域用のフォワードプライマー5’−GGAGCTCGAYATTGTGMTSACMCARWCTMCA−3’及び軽鎖可変領域用のリバースプライマー5’−TATAGAGCTCAAGCTTGGATGGTGGGAAGATGGATACAGTTGGTC−3’を用いたPCRによって増幅した。増幅産物をPCR−bluntベクター(Invitrogen)にクローニングした。陽性クローンを選択し、双方向にシークエンシングした。Chothiaの標準ドメイン(Morea V, Tramontano A, Rustici M, Chothia C, Lesk AM. Antibody structure, prediction and redesign. Biophys Chem. 1997 Oct;68(1-3):9-16. Al-Lazikani B, Lesk AM, Chothia C. Standard conformations for the canonical structures of immunoglobulins. J Mol Biol. 1997; 273(4):927-48.)を、重鎖及び軽鎖について、得られたアミノ酸配列に基づき、http://www.bioinf.org.uk/abs/chothia.htmlで提供されるツールによって決定した。このようにして、重鎖及び軽鎖のCDR配列を決定した。
クローニングされた可変領域を同定するために、ベクターを構築して、シグナルペプチドMMP−3によって誘導される一本鎖抗体を発現させた。このベクターを用いて、QM−7筋芽細胞にトランスフェクトした。分化を誘導した後、培地をHep−12細胞とともにインキュベートし、続いて、トランスフェクト細胞がHep−12細胞に特異的に結合し得るか否かを観察することができるように、MYCタグ付き抗体を用いた間接免疫蛍光染色を行った。
培地をHep−11細胞及びHep−12細胞の培養物から除去した後、得られた細胞に1B50−1ハイブリドーマ培養物の上清をそれぞれ添加した。37℃で2時間インキュベーター内でインキュベートした後、培養培地を捨て、細胞をPBSで3回洗浄した。セルスクレーパーを用いて細胞を採取し、遠心分離した。次いで、これらの細胞を5mlの脱イオン水に再懸濁し、超音波処理に供した。2×細胞溶解液を添加した後、これらの細胞を超音波で再度処理した。溶液を4℃で10分間、10000rpmで遠心分離した。上清を採取し、予め平衡化したSepharose 4B−protein G(Pharmacia(Uppsala,Sweden))アフィニティークロマトグラフィーカラムに添加した。室温で1時間維持した後、カラムをPBSでパージした。0.1M グリシン−HClバッファー(pH2.5)を用いて、結合した抗原を溶出させた。得られた溶液を、1M Tris−HCl(pH9.0)を添加することによってpH7.0に調整した。次いで、同量の2×SDS−PAGEサンプリングバッファーを添加して、SDS−PAGE分析を行った。クマシーブリリアントブルーR250で染色した後、Hep−12細胞において特異的発現を示し、1B50−1によって免疫沈降したバンドを切り出し、トリプシン消化後にMALDI−TOF−MS分析に使用した。
培養培地を除去した後、ほぼ飽和密度の細胞をPBSで洗浄し、全細胞RNAをTrizol法によって抽出した。1×逆転写バッファー(50mM Tris−HCl(pH8.3)、75mM KCl及び3mM MgCl2)、20UのRNアーゼ阻害剤(Promega(Madison,WI,USA))、10mM DTT、50mM dNTP、0.5μgのoligo−(dT)15(Promega)及び200Uの齧歯類白血病ウイルス逆転写酵素(M−MLV−RT;Invitrogen)を含む20μlの反応系において、3μgの全細胞RNAを逆転写に供し、第1の鎖のcDNAを合成した。1μLのcDNAに対して、CACNA2D1の遺伝子用のフォワードプライマー:5’−ACAGCAAGTGGAGTCAATCA−3’及びリバースプライマー:5’−ACTGCTGCGTGCTGATAAGA−3’を用いて、Taq DNAポリメラーゼによりPCRを行った。PCRは以下のように行った:94℃×5分間(前変性);94℃×45秒間;56℃×45秒間;72℃×1分間、合計で25サイクル;72℃×10分間(伸長)。産物をアガロースゲル電気泳動に供した後、エチジウムブロマイドで染色し、続いて産物を紫外線下で観察し、撮影した。
cDNAを、全細胞RNAから齧歯類白血病ウイルス逆転写酵素(M−MLV−RT;Invitrogen)によって転写した後、ABIの7500 PCRシステムにおいてSYBR Green PCR蛍光色素混合物(東洋紡株式会社(日本、大阪))を用いてリアルタイムPCR増幅を行った。プライマーを表1に挙げた。遺伝子発現の倍率変化は、GAPDHを内部対照(internal control reference)として2−ΔΔCt法によって算出した(Pfaffl, M. W.. A new mathematical model for relative quantification in real-time RT-PCR. Nucleic Acids Res 2001,29:e45)。
CACNA2D1遺伝子(NM_000722.2;電位依存性カルシウムチャネルα2/δサブユニット1)は、Genebankに記載の配列に従って3つの部分に分けられ、3対のプライマーを以下のように設計した:
パート1のフォワードプライマー:5’−CCGgaattcTATGGCTGCTGGCTGCCTGCTGG−3’、
パート1のリバースプライマー:5’−AACCATTAGGATCGATTGCAAAG−3’、
パート2のフォワードプライマー:5’−TGTGTACCTGGATGCATTGGAACTG−3’、
パート2のリバースプライマー:5’−ACCATCATCCAGAATCACACAATC−3’、
パート3のフォワードプライマー:5’−AGAGACATATGAGGACAGCTTC−3’、
パート3のリバースプライマー:5’−GTCGACTACTTGTCATCGTCATCCTTGTAATCCTCGAGTAACAGGCGGTGTGTGCTG−3’。
細胞を播種し、翌日、飽和密度(80%〜90%)まで成長させた。これらの細胞に、構築したベクターCACNA2D1mychis/pcDNA3.0を、推奨プロトコルに従いLipofectamine 2000(Invitrogen)によってトランスフェクトした。トランスフェクションの24時間後及び48時間後に、遺伝子発現を免疫蛍光染色細胞化学によって分析した。
培養細胞をトリプシン:EDTAで消化して、単一細胞懸濁液を調製した。2×106細胞の懸濁液をとり、精製抗体1B50−1を添加した(100倍に希釈、原液は1mg/mlとした)。混合物を37℃で1時間インキュベートし、PBSで3回洗浄した。次いで、FITC標識ヤギ抗マウスIgG二次抗体(Jackson ImmunoResearch Laboratories(USA)、0.5mg/ml;100倍希釈)を添加し、1時間反応させた。PBSで洗浄した後、細胞をLeicaのSP5共焦点レーザー顕微鏡下で観察するか、又はAriaフローサイトメーターによって分析し、分類した。CACNA2D1mychis/pcDNA3.0をトランスフェクトした細胞を、モノクローナル1b50−1及びウサギポリクローナルmyc抗体を用いて二重染色した。ローダミン標識ヤギ抗マウスIgG及びFITC標識ヤギ抗ウサギIgGを、それぞれ二次抗体として使用した。
臨床側がん組織及び肝臓がん組織のクリオスタット切片を、メタノールで30秒間固定した後、5%脱脂粉乳でブロッキングした。切片を、モノクローナル抗体1B50−1(100倍希釈、5%BSAを含有する)とともに4℃で一晩インキュベートした後、PBSで洗浄した。その後、FITC標識ヤギ抗マウスIgGとの反応を室温で2時間行った。切片をPBSで洗浄し、核小体をDAPI(2000倍)で5分間染色した。グリセロール中の2%DABCOを用いて切片を封入し、LeicaのSP5共焦点レーザー顕微鏡下で観察した。
組織又は培養細胞を、1mM PMSF、完全プロテアーゼ阻害剤カクテル及びホスファターゼ阻害剤カクテル(Roche(Mannheim,Germany))の混合物を含有する放射性免疫沈降アッセイバッファー(Beijing Solarbio Science & Technology Co., Ltd)中で溶解した。10%SDS−PAGEでの電気泳動の後、タンパク質をImmobilon−Pメンブレン(Millipore)に転写した。メンブレンを5%脱脂粉乳でブロッキングした後、タンパク質を一次抗体CACNA2D1(Abcam(Cambridge,MA))又は内部標準であるβ−アクチン(Roche Applied Science)に特異的な抗体、次いでHRP標識ヤギ抗マウス二次抗体(Jackson ImmunoResearch Laboratories Inc.(West Grove,PA,USA))と反応させた。陽性シグナルを、Immobilon(商標)Western Chemiluminescent HRP基質(Millipore)により化学発光(chemical luminance)法で検出した。バンドをChemiImagerスキャナ(Alpha Innotech)によってシグナルについてスキャンし、AlphaEaseFCソフトウェアを用いてグレースケールを定量し、分析した。内部標準としてのβ−アクチンのグレースケールを用いることによってCACNA2D1の相対量を算出した。
CACNA2D1遺伝子の干渉のためのRNA配列は、Origeneによって設計され、合成された。レトロウイルスベースのベクターを、プロモーターとしてU6プロモーターを用いて構築した。ここで、配列1は、CACNA2D1のコード配列中のヌクレオチド546〜574に対するACTCAACTGGACAAGTGCCTTAGATGAAGであり、配列2は、コード配列のヌクレオチド116〜144に対するAGATGCAAGAAGACCTTGTCACACTGGCAであった。Origeneによって無作為に合成された同じ長さの核酸を、対照配列として使用した。これらの配列を保有するベクターを、EcoR I及びSal Iによってそれぞれ切断し、U6プロモーター及びこれらのオリゴヌクレオチドを含有するフラグメントをアガロースゲル電気泳動で分離し、精製した。精製したフラグメントを、同じエンドヌクレアーゼによって切断したPlenti6−リンカーベクター(多重クローニング部位を有するリンカーを、Cla I及びAge Iによって切断し、本発明者らの研究室に保管されているInvitrogenのplenti6ベクターに導入することによって得られる)とライゲーションした。決定の後、レンチウイルスプラスミドベクターplenti6U6CACNA2D1ShRNA−1、plenti6U6CACNA2D1ShRNA−2及びplenti6U6−対照が得られた。レンチウイルスのパッケージングを、Invitrogenによって提示される推奨プロトコルに厳密に従って行った。レンチウイルス粒子を有する上清を、直接Hep−12細胞の感染に使用した。48時間後、6μg/mlのブラストサイジン(Invitrogen)を添加して、レンチウイルスが感染した細胞をスクリーニングし、培地を3日毎に更新した。このようにして、上記のレンチウイルスが感染した細胞集団が得られ、6μg/ml含有されるブラストサイジンを実験の間中連続的に使用して、細胞に対するスクリーニング圧を維持した。遺伝子CACNA2D1に対する阻害効果を、感染細胞においてRT−PCR及び免疫蛍光細胞化学染色によって更に観察した。また、腫瘍の形成及び成長に対する阻害効果を、免疫不全動物における腫瘍原性実験によって観察した。
腫瘍発生実験:
腫瘍発生及び腫瘍発生細胞の自己複製能をアッセイする実験において、種々の供給源に由来し、フローサイトメトリーによって分類された異なる量(104個、103個、102個)の1B50−1陽性細胞又は1B50−1陰性細胞を、同体積のMatrigel(BD Biosciences)(1:1)と混合し、混合物を用いて4週〜6週齢のNOD/SCIDマウス(Vital River Laboratories Animal Technology Co., Ltd.(Beijing,SPF))に皮下接種した(抗体陰性細胞をマウスの片側に接種し、抗体陽性細胞を同じマウスのもう片側に接種した)。各々の群は5匹の動物からなるものであった。腫瘍の成長を毎週観察した。次いで、CACNA2D1を阻害した後の動物における腫瘍形成及び腫瘍成長の変化を観察する実験において、2×106個の細胞をNOD/SCIDマウスに皮下接種した。腫瘍を目に見える大きさまで成長させてから、腫瘍の長径及び短径を3日毎に測定した。腫瘍の大きさを、「腫瘍の大きさ=長径×短径2/2」という方程式に基づいて算出することができ、データを用いて成長曲線を作成した。
4週〜6週齢のNOD/SCIDマウスに、2×106個の肝臓がん細胞を皮下接種した。腫瘍を目に見える大きさ(約0.02cm3〜0.03cm3)まで成長させてから、動物を幾つかの群に無作為に分けた(1群当たり6匹の動物)。これらの動物に、PBS、対照IgG(動物1匹当たり800μg、Zhongshan Golden Bridge Biotechnology)及び1B50−1(動物1匹当たりそれぞれ200μg、400μg及び800μg)をそれぞれ1日おきに腹腔内注射した。各注射の前に長径及び短径をノギスによって測定し、腫瘍の大きさを「腫瘍の大きさ=長径×短径2/2」という方程式に基づいて算出した。合計で7回の投与の後、動物を最後の投与の翌日に屠殺した。腫瘍を解剖し、これらの腫瘍について湿重量及び大きさを測定した。
効果実施例1:1B50−1は膜上の抗原を認識し、陽性率は肝臓がん細胞株によって異なっていた
ハイブリドーマをサブトラクティブ免疫化によって作製し、Hep−11細胞及びHep−12細胞によってスクリーニングした。3回の融合の後、Hep−12細胞に特異的な可能性がある37個のモノクローナル抗体が1回目に得られた。これらの抗体を更なるサブクローニング及び予備分析に供した。細胞膜上に位置する抗体を分泌する1つのハイブリドーマを1B50−1と命名した(図1)。1B50−1陽性細胞の比率を、種々の肝臓がん細胞株及び肝臓がんの臨床検体の初代培養物に由来する細胞においてフローサイトメトリーによって分析した。結果を表2に挙げた。腫瘍発生細胞に富んだ再発性肝細胞がんに由来するHep−12細胞は比較的高い1B50−1陽性率を有し、他の細胞の陽性率は低かった。
1B50−1陽性/陰性細胞を、Hep−11、Hep−12、HuH7、HepG2及びSMMC−7721を含む、肝細胞がんを有する5つの細胞株からフローサイトメーターによって分類した。NOD/SCIDマウスに100個又は1000個の選択した細胞を皮下注射した(抗体陰性細胞をマウスの片側に接種し、抗体陽性細胞を同じマウスのもう片側に接種した)。12週間〜18週間後、100個〜1000個の1B50−1陽性細胞が皮下腫瘍を発生させるのに十分であったが、陰性細胞は成長しないか、又は小結節までしか成長せず(表2、図2)、1B50−1陽性細胞が腫瘍発生細胞としての特性を有することが示された。臨床肝臓がん組織の初代培養物から分類された1B50−1陽性/陰性細胞を用いた腫瘍発生実験でも、同様の結果が得られた(表2)。1B50−1陽性細胞を、1B50−1陽性HuH7細胞によって誘導された腫瘍からフローサイトメーターによって分類した後、NOD/SCIDマウスに皮下接種した。これらの1B50−1+細胞の100%(5/5)が腫瘍を形成することが可能であることが見出され、1B50−1+細胞が自己複製能を有することが示唆された。1B50−1陽性細胞及び1B50−1陰性細胞における幹細胞関連遺伝子の発現を、リアルタイム蛍光定量的RT−PCRを用いて分析した。Nanog、Sox−2、AFP及びABCG2等の幹細胞関連遺伝子が、1B50−1+細胞において高レベルに発現されることが見出された(図D)。1B50−1陽性細胞を、10%ウシ胎仔血清を含有する培地中で培養し、1B50−1陽性細胞の割合を分析した。1B50−1+細胞の割合が親細胞において測定されたレベルまで減少することが判明し(図2C)、1B50−1陽性細胞が1B50−1陽性細胞及び1B50−1陰性細胞の両方に分化し得ることが示された。これらの結果から、1B50−1陽性細胞が腫瘍発生細胞としての特性を有することが示唆された。
約150KDの特異的バンドが、Hep−12細胞における1B50−1による免疫沈降から得られた(図3の左パネル、矢印はバンドを示す)。MS分析から、このタンパク質がCACNA2D1であることが明らかになった。対応する遺伝子を、Hep−12細胞のcDNAを鋳型として用いるPCRによって増幅した。従来のDNA組換えによって、増幅遺伝子のC末端にMYCタグペプチドのコード配列を付加した。次いで、遺伝子を真核細胞発現ベクターpcDNA3.0mychisに導入し、これを続いて、かかる遺伝子を発現しない細胞にトランスフェクトした。その後、ポリクローナルMYC抗体及び1B50−1を用いて二重染色を行った。MYCタグ及び1B50−1に特異的な抗体がトランスフェクト細胞の細胞表面上に一貫して位置し、1B50−1がCACNA2D1を認識することが示された(図3の右パネル)。
クリオスタット切片を、臨床肝細胞がん症例(合計で86人の患者)の新鮮検体のがん又は一対の側がん組織から得た。固定後、これらの切片を免疫蛍光組織化学によって1B50−1で染色した。結果を図4及び表2に示した。これらの症例の72.1%(62症例/86症例)で、1B50−1陽性細胞が病変に分布していた(図4)。側がん組織における陽性細胞の検出率は46.5%(40/86)であり、がん組織における検出率よりも低かった。5つの正常肝臓検体(血管腫に関連する外科手術で摘出された検体に由来する)では、1B50−1陽性細胞は存在が検出されなかった。がん病変及び側がん組織において、1B50−1陽性細胞の存在を、患者の臨床指標と組み合わせて統計的に分析した(表3)。がん病変における1B50−1陽性細胞の存在は、年齢、性別及び肝硬変の発症等の指標とは無関係であった。しかしながら、側がん組織中の1B50−1陽性細胞は肝硬変の発症、外科手術後4年未満の生存及び2年以内の再発と正に相関していた。カプランマイヤー曲線及びコックス回帰モデルを用いた多変量分析から、肝臓がん患者の側がん組織における1B50−1陽性細胞の存在によって、患者の外科手術後の無病生存率及び全生存率が、側がん組織に1B50−1陰性細胞を有する患者よりも不良であることが予測されると示された(図4)。側がん組織は実際に除去されるがんの辺縁部に由来するため、側がん組織における1B50−1陽性細胞の存在の有無は、患者における再発の予測及び予後診断に使用することができる。すなわち、側がん組織における1B50−1陽性細胞の存在は、HCC予後指標として使用することができる。
肝臓がん以外の一般的ながん、例えば胃がん、肺がん、乳がん及び前立腺がんの細胞株におけるCACNA2D1遺伝子の発現を、RT−PCRによって検出した。結果を図5Aに示した。胃がん細胞の中でもMGC−803及びSGC7901、肺がん細胞の中でもPG及びA549、乳がん細胞の中でもBICR−H1及びMDA−MB−231で陽性バンドが見られた。この遺伝子は、高転移性のPC3M1E7前立腺がん細胞で高度に発現されたが、低転移性のPC3M2B4細胞ではバンドは検出されなかった。結果から、全ての細胞が陽性であるわけではないが、上記遺伝子が実際に大半のがんについて部分的な細胞で陽性に発現され、肝臓がん症例において観察されたものと同様の結果を示すことが明らかになった。上記の高転移性の陽性細胞によって、上記遺伝子が腫瘍の転移と正に相関し得ることが示唆された。さらに、1B50−1を一部の細胞の免疫蛍光細胞化学染色に使用した。CACNA2D1がSGC7901胃がん細胞、KYSE−510食道がん細胞及びKYSE−150食道がん細胞並びにZR−75乳がん細胞において陽性に発現され、細胞膜上に位置することが見出された。陽性細胞の数は種々の細胞株によって大きく異なっており、陽性細胞がこれら全ての細胞株のうち僅かな割合しか占めなかった(図5B)。上記の結果は肝臓がんにおいて観察された結果と同様であり、肝臓がんにおける発見が他の腫瘍にも適用され得ることを示している。すなわち、CACNA2D1を腫瘍診断及び腫瘍治療の分子標的として使用することが可能であり得る。
肝臓がん組織において得られた結論が他のタイプの腫瘍にも適用可能であるか否かを更に解明するために、免疫組織化学染色を臨床的な結腸直腸がん、腎臓がん、肺がん及び食道がん並びにそれらの一対の側がん組織(各がんタイプについて10対)に対して行った。1B50−1陽性細胞の分布は、肝臓がんにおいて観察されたものと同様であった。結果を表4に示した。がん組織に1B50−1陽性細胞を有する症例は、側がん組織に1B50−1陽性細胞を有する症例よりも多かった。しかしながら、症例は統計分析を行うには少な過ぎた。免疫組織化学において得られた結果を、臨床組織検体に対してCACNA2D1に特異的な市販の抗体を用いたウエスタンブロットを行うことによって更に検討した。図6に示されるように、一部のがん組織におけるCACNA2D1の発現は、側がん組織における発現よりも明らかに高く(CACNA2D1の発現レベルは、種々の腫瘍タイプによって異なっていた)、がんが発生した場合にCACNA2D1がより高レベルに発現されることを示している。CACNA2D1の高発現が肝臓がん及び他のがんにおいて認められた。このため、CACNA2D1に指向性を有する薬物及び分子マーカーは、肝臓がん以外のがんに使用することができる。
Hep−12肝臓がん細胞及びHuH7肝臓がん細胞を、それぞれ免疫不全動物に皮下接種した。腫瘍を0.02cm3〜0.03cm3の大きさまで成長させてから、種々の投与量の1B50−1を動物に腹腔内注射した。表及び図面に示されるように、腫瘍阻害率は、800μg/マウス1B50−1処理群において、Hep−12肝臓がん細胞株及びHuH−7肝臓がん細胞株(重量により測定)のそれぞれについて80.4%及び65.5%にまで達した。かかる阻害率は投与量に依存し、PBS対照群及びIgG処理群とは統計的に有意に異なっていた。
1B50−1によって認識される抗原CACNA2D1が腫瘍治療の標的分子となるか否かを確認するために、CACNA2D1に干渉するRNAを発現するレンチウイルスベクターplenti6U6CACNA2D1shRNA−1及びplenti6U6CACNA2D1shRNA−2を構築し、対照であるplenti6U6をレンチウイルスに被包させた(enveloped)。これらのベクターを用いてHep−12細胞を感染させ、ブラストサイジン(Invitrogen)を用いて感染細胞をスクリーニングした。1B50−1の免疫蛍光細胞化学染色(図9A)から、細胞にCACNA2D1干渉RNAを含有する2つのレンチウイルスが感染した場合に、CACNA2D1の発現が明らかに抑制され、細胞膜中に見られる陽性蛍光ドットが極めてまばらであることが示された。蛍光強度は対照群で極めて高く、大半の細胞でシグナルが認められた。上記に記載のレンチウイルスが感染した細胞を、NOD/SCIDマウスに皮下接種した(2×106細胞/マウス、1群当たり5匹のマウス)。腫瘍の形成を観察した。図9Bに示されるように、CACNA2D1干渉RNAを担持する2つのレンチウイルスを感染させた細胞は、動物における成長が対照ベクターを有する細胞よりも遅かった。腫瘍阻害率はそれぞれ57.5%及び59.6%(重量により測定)であった。t検定のp値は、対照群と比較してそれぞれ0.0164及び0.014であった。これらの結果から、CACNA2D1の発現の阻害が、実際にin vivoでHep−12細胞の成長を抑制することが示唆された。このため、CACNA2D1は腫瘍治療の分子標的とされた。
QM−7細胞によって発現された1B50−1の可変領域の一本鎖抗体を含有する上清を、Hep−12細胞とともにインキュベートした。染色にはMYCタグ付き9E10を一次抗体として使用し、FITCタグ付きヤギ抗マウス抗体を二次抗体として使用した。染色を蛍光顕微鏡下で観察した。Hep−12細胞の細胞表面上のタンパク質は、MYCタグ付き9E10によって認識されることができた(図10)。蛍光染色パターンは1B50−1を用いた場合と一致し、発現された一本鎖抗体が1B50−1と同様の結合活性を有することが示唆された。また、この結果から、1B50−1の軽鎖可変領域及び重鎖可変領域が正確な配列でクローニングされることが更に確認された。抗体−薬物の更なる修飾を、これらの配列に基づき遺伝子工学によって行うことができた。

- 前記腫瘍が肝細胞がんである、請求項19に記載の治療薬。
- 請求項2又は3に記載のハイブリドーマ細胞株に抗体を産生させる工程を含む、抗体の調製方法。
