Description
本発明は、多能性幹細胞、特に人工多能性幹細胞から、骨格筋細胞への分化誘導方法、当該方法に用いるための試薬キット、並びに当該方法により得られる骨格筋細胞に関する。本発明はまた、前記骨格筋細胞を用いたミオパチーの治療剤のスクリーニング方法に関する。
筋疾患は非常に多くの病気を含んでいるが、その症状の大半は筋肉の萎縮とそれに伴う筋力の低下である。筋肉の萎縮の原因には、筋肉自体に異常がある場合と筋肉を動かす神経に異常がある場合とがあり、前者を筋原性疾患(ミオパチー)、後者を神経原性疾患という。ミオパチーの代表的なものとして筋ジストロフィーおよび筋萎縮症などがある。筋ジストロフィーのうち最も患者数の多いデュシェンヌ型筋ジストロフィーは、点突然変異により1つのコドンがタンパク質の合成終了を意味するストップコドンに変化し、ジストロフィンタンパク質が合成されない病気である。性染色体劣性遺伝で男子だけに発症する疾患であり、人口10万人あたり3〜5人、出生男児2000〜3000人あたり1人といわれている。筋ジストロフィー症に対する良好な治療手段は未だなく、治療法の開発が望まれている。三好型ミオパチー(MM)は、先天性の遠位型ミオパチーの一種であり(Miyoshi, K. et al. Brain 109(Pt 1), 31-54, 1986)、ジスフェリンの変異に起因する筋肉の膜修復異常(defective)によって引き起こされる(Liu, J. et al. Nat Genet 20, 31-36, 1998, and Bansal, D., et al. Nature 423, 168-172, 2003)。
本発明の目的は、iPS細胞を含む多能性幹細胞から骨格筋細胞への分化を誘導する方法を提供することであり、当該方法において一過性に導入される遺伝子を含む分化誘導試薬キットを提供することである。本発明のさらなる目的は、当該方法により得られる多能性幹細胞由来の骨格筋細胞を用いた、筋ジストロフィー、三好型ミオパチーなどを含むミオパチーなどの筋疾患の治療剤をスクリーニングする手段を提供することである。
[1]多能性幹細胞から骨格筋細胞を製造する方法であって、MyoD、Myf5およびそれらをコードする核酸から選ばれる1以上の外因性因子を、多能性幹細胞において発現させる工程を含む方法。
[2]多能性幹細胞がヒト多能性幹細胞である、[1]に記載の方法。
[3]分化誘導条件での培養開始から3日以内に、外因性因子を多能性幹細胞において発現させる、[1]または[2]に記載の方法。
[4]分化誘導条件での培養開始から1日以内に、外因性因子を多能性幹細胞において発現させる、[3]に記載の方法。
[5]分化誘導条件が、胚様体を形成する工程を伴わない、[3]に記載の方法。
[6]分化誘導条件が、血清または血清代替物を含有し、bFGFを含有しない基本培地で培養することを含む、[3]に記載の方法。
[7]外因性因子を発現させた後、ウマ血清を含む培養液(culture medium)でさらに培養する工程を含む、[1]から[6]のいずれかに記載の方法。
[8]外因性因子の発現が、5日間以上10日間以下維持される、[1]から[7]のいずれかに記載の方法。
[9]多能性幹細胞における外因性因子の発現が、MyoDまたはMyf5をコードする核酸を含む薬剤応答性誘導ベクターを導入した多能性幹細胞を、該薬剤の存在下で培養することにより行われる、[1]から[8]のいずれかに記載の方法。

本発明は、多能性幹細胞から骨格筋細胞を製造する方法を提供する。当該方法は、MyoD、Myf5およびそれらをコードする核酸から選ばれる1以上の外因性因子(以下、「本発明の骨格筋細胞誘導因子」ともいう)を多能性幹細胞において発現させる工程を含む。
多能性幹細胞を分化させることによって骨格筋細胞を含有する細胞集団を用意する場合、使用可能な多能性幹細胞は、生体に存在するすべての細胞に分化可能である多能性を有し、かつ、増殖能をも併せもつ幹細胞である。それには、特に限定されないが、例えば胚性幹(ES)細胞、核移植により得られるクローン胚由来の胚性幹(ntES)細胞、生殖幹細胞(germline stem cell)(「GS細胞」)、胚性生殖細胞(「EG細胞」)、人工多能性幹(iPS)細胞、培養線維芽細胞や骨髄幹細胞由来の多能性細胞(Muse細胞)などが含まれる。好ましい多能性幹細胞は、ES細胞、ntES細胞、およびiPS細胞である。
ES細胞は、ヒトやマウスなどの哺乳動物の初期胚(例えば胚盤胞)の内部細胞塊から樹立された、多能性と自己複製による増殖能を有する幹細胞である。
生殖幹細胞は、精巣由来の多能性幹細胞であり、精子形成のための起源となる細胞である。この細胞は、ES細胞と同様に、種々の系列の細胞に分化誘導可能であり、例えばマウス胚盤胞に移植するとキメラマウスを作出できるなどの性質をもつ(M. Kanatsu-Shinohara et al.(2003)Biol. Reprod., 69:612-616; K. Shinohara et al.(2004), Cell, 119:1001-1012)。神経膠細胞系由来神経栄養因子(glial cell line-derived neurotrophic factor(GDNF))を含む培養液で自己複製可能であるし、またES細胞と同様の培養条件下で継代を繰り返すことによって、生殖幹細胞を得ることができる(竹林正則ら(2008), 実験医学, 26巻, 5号(増刊), 41〜46頁, 羊土社(東京、日本))。
胚性生殖細胞は、胎生期の始原生殖細胞から樹立される、ES細胞と同様な多能性をもつ細胞である。LIF、bFGF、幹細胞因子(stem cell factor)などの物質の存在下で始原生殖細胞を培養することによって樹立しうる(Y. Matsui et al.(1992), Cell, 70:841-847; J.L. Resnick et al.(1992), Nature, 359:550-551)。
人工多能性幹(iPS)細胞は、特定の初期化因子を、DNAまたはタンパク質の形態で体細胞に導入することによって作製することができる、ES細胞とほぼ同等の特性、例えば分化多能性と自己複製による増殖能、を有する体細胞由来の人工の幹細胞である(K. Takahashi and S. Yamanaka(2006)Cell, 126:663-676; K. Takahashi et al.(2007), Cell, 131:861-872; J. Yu et al.(2007), Science, 318:1917-1920; Nakagawa, M. et al., Nat. nt ES細胞は、核移植技術によって作製されたクローン胚由来のES細胞であり、受精卵由来のES細胞とほぼ同じ特性を有している(T. Wakayama et al.(2001), Science, 292:740-743; S. Wakayama et al.(2005), Biol. Reprod., 72:932-936; J. Byrne et al.(2007), Nature, 450:497-502)。すなわち、未受精卵の核を体細胞の核と置換することによって得られたクローン胚由来の胚盤胞の内部細胞塊から樹立されたES細胞がnt ES(nuclear transfer ES)細胞である。nt ES細胞の作製のためには、核移植技術(J.B. Cibelli et al.(1998), Nature Biotechnol., 16:642-646)とES細胞作製技術(上記)との組み合わせが利用される(若山清香ら(2008), 実験医学, 26巻, 5号(増刊), 47〜52頁)。核移植においては、哺乳動物の除核した未受精卵に、体細胞の核を注入し、数時間培養することで初期化することができる。
Muse細胞は、WO 2011/007900に記載された方法にて製造された多能性幹細胞である。詳細には、線維芽細胞または骨髄間質細胞を長時間トリプシン処理、好ましくは8時間または16時間トリプシン処理した後、浮遊培養することで得られる多能性を有した細胞であり、SSEA-3およびCD105が陽性である。
本発明の骨格筋細胞誘導因子を多能性幹細胞において発現させる方法は特に限定されないが、例えば、以下の方法を用いることができる。尚、ここで「発現」とは、骨格筋細胞誘導因子がMyoDもしくはMyf5をコードする核酸である場合においては、細胞内で、該核酸からMyoDもしくはMyf5タンパク質が(転写および)翻訳され生成することを意味し、骨格筋細胞誘導因子がMyoDもしくはMyf5タンパク質である場合においては、該タンパク質が細胞内に導入されることと同義である。
本発明は、多能性幹細胞からの骨格筋細胞の製造用キットを提供する。本キットには、上述した本発明の骨格筋細胞誘導因子、即ちMyoD、Myf5およびそれらをコードする核酸から選ばれる1以上の因子を含む骨格筋細胞誘導因子(例えば、核酸であればアルコール沈殿物、凍結TE溶液、凍結乾燥物など;タンパク質であれば凍結乾燥物、適当な緩衝液に溶解した凍結液剤など)、当該因子を導入する上記ベクター、細胞、試薬および培養液を含み得る。本キットには、さらに分化誘導の手順を記載した書面や説明書を含んでもよい。
本発明は、骨格筋疾患の治療または予防に有用な薬剤である候補薬剤のスクリーニング方法を提供する。
(1)候補物質を、骨格筋疾患に罹患した患者由来のiPS細胞から分化誘導した骨格筋細胞と接触させる工程、
(2)候補物質と接触させなかった場合と比較して、当該骨格筋細胞の病態が緩和された場合、骨格筋疾患の治療剤または予防剤として選別する工程。
細胞の培養
ヒトiPS細胞株(201B7、253G1および253G4)の維持培養は高橋らの方法と同様に行った(K. Takahashi et al., Cell 131, 861, 2007)。すなわち、ヒトiPS細胞はフィーダー細胞を用いて維持した。フィーダー細胞は、SNLを10 cm組織培養ディッシュあたり100万個播種して作製した。維持培地は、DMEM Glutamax F12(Invitrogen)500 mlに2 mM L-グルタミン(ナカライテスク)、1×非必須アミノ酸(Non-essential amino acid)(Invitrogen)、50 mU/L ペニシリン/50 μg/L ストレプトマイシン(Invitrogen)、および100 μM 2-メルカプトエタノール(2-ME;ナカライテスク)を加えたものを基本培地とし、これに20%になるようにKnockout Serum Replacement(KSR; Invitrogen)を加え、4 ng/ml bFGF(Wako)を添加したものを用いた。Tetベクター導入後のヒトiPS細胞では、さらに100 μg/mlのネオマイシン(ナカライテスク)を添加した培地にて維持培養した。
本実施例は、施設内倫理委員会の承認を受け、ヘルシンキ宣言の下に実施した。守秘義務を保護するためにMM患者をにコード化し、書面によるインフォームドコンセントを得た。MM患者は、ジスフェリン遺伝子に2つの変異を有することが分かっていた。患者由来のiPS細胞は、房木らに記載されるように線維芽細胞から作製した(Fusaki, N. et al., Proc Jpn Acad Ser B Phys Biol Sci 85, 348-362, 2009)。
MyoDは、MGCクローンよりcDNAを購入(MGC: 71135, GenBank: BC064493.1)した。これに対して下記のプライマーを用い、PCR反応で増幅しcDNAフラグメントを得た。Myf5は、分化したヒトiPS細胞からのcDNAよりKOD plus Neoを酵素としてPCR反応にて増幅しcDNAフラグメントを得た。これらのcDNAフラグメントをpENTR Directional TOPO Cloning Kits(Invitrogen)を用いて添付文書通りにエントリーベクターに組み込んだ。具体的には、PCR産物4 ng、Salt Solution 1 μl、蒸留水3.5 μl、およびTOPOベクターpENTR/D 1 μlを反応させた。その後、大腸菌にベクターを組み込み、大腸菌を増幅させ、エントリーベクターを獲得した。使用したプライマーの配列は以下のとおりである。

テトラサイクリン応答性遺伝子強制発現piggyBacベクターはWoltjenらが開発したKW111を用いた(K. Woltjen et al., Nature 458, 766, 2009)。このベクターはリバーステトラサイクリントランスアクチベーター(reverse tetracycline transactivator)(rtTA)とテトラサイクリン応答性領域(TRE)を両方組み込んだものであり、またターゲット遺伝子と同調してmCherryが発現するようデザインされている(図1aおよび14a)。またネオマイシン耐性遺伝子により薬剤による選別が可能である。このベクターとpENTR/D-TOPOにMyoDもしくはMyf5のcDNAを組み込んだエントリーベクターとを混合し、LRクロナーゼ(Invitrogen)を用いた組換え反応によりテトラサイクリン応答性MyoD(またはMyf5)強制発現piggyBacベクターTet-MyoD(図1a)またはTet-Myf5(図14a)を作製した。
ヒトiPS細胞株は10 cmディッシュ1枚分の細胞数を準備した。前日に、SNLフィーダー細胞をゼラチンコートした6ウェルプレート(IWAKI)に3.3×105細胞/ウェルの密度で培養した。SNLフィーダー細胞がコンフレントになった状態の6ウェルプレートに、維持培養時と同様の方法で回収したヒトiPS細胞株を播種した。翌日、ベクターのトランスフェクションを行った。Tetベクターと、CAGプロモーター下流にトランスポゼースを組み込んだベクター(CAG-PBase)とをそれぞれ1 μg準備し、100 μlのOpti-MEM(Invitrogen)に溶解した。そこに3〜8 μlのFugeneHD(Roche)を添加し、2秒間ボルテックスにて混和した。15分室温で反応させた後、6ウェルプレートの培養液中に混合物を添加し、37℃で培養した。24時間後に培地交換を行い、さらに24時間後に100 μg/mlのネオマイシン(ナカライテスク)含有培地に交換した。その後は毎日ネオマイシン含有培地での培地交換を行い、ネオマイシン耐性に形質転換した細胞を選別した。
得られたクローンは、培地にて50倍に希釈したマトリゲル(Invitrogen)にてコートした6ウェルプレートに播種した。翌日よりドキシサイクリン(Dox; LKT Laboratories)を1 μg/mlの濃度となるように培地に添加した。Dox添加後48時間で細胞を回収し、LSR Fortessa(BD)にてmCherryの発現強度を解析し、強発現のクローンを選別した(図3a、5a、12aおよび14c)。
ジスフェリンcDNAは、EF1αプロモーターによって対象遺伝子を駆動し、ピューロマイシン耐性遺伝子を共発現するデスティネーションpiggyBac(PB)ベクターに挿入した(PB-ジスフェリンベクター)。Tet-MyoDをトランスフェクトしたMM hiPSC(MyoD-MM hiPSC)を、マイトマイシンC処理SNL-PHフィーダー細胞(ネオマイシン、ピューロマイシンおよびハイグロマイシン耐性である)上に播種した。翌日、1μg PB-ジスフェリンベクターと1μg PBトランスポゼースプラスミド16の両方をFuGENE HD(Roche)により製造者のプロトコールに従って、MyoD-MM hiPSCにトランスフェクトした。トランスフェクションから48時間後、Tet-MyoDベクターとPB-ジスフェリンベクターの両方を保持する細胞を選別するために、100 μg/mLのG418(ナカライテスク)および1 μg/mLのピューロマイシン(ナカライテスク)を添加した。選別後、ウェスタンブロッティングにより、ジスフェリンを適正に発現する適切なMyoD-MM+ジスフェリンhiPSCクローンを選別した。
Dox添加によりMyoDを強制発現させた細胞を、4%パラフォルムアルデヒド(ナカライテスク)/PBSを用いて室温で20分間固定し、PBSで各5分間、3回洗浄した後、1%ヤギ血清(Sigma)、0.1%ウシ血清アルブミン(Sigma)および0.2%トライトンX-100(ナカライテスク)添加PBSにてブロッキングを室温で1時間行った。一次抗体は、上記ブロッキング液中に抗MyoD(ウサギポリクローナル;Santacruz)を添加し、1:400の濃度で希釈した。4℃で16〜18時間抗体を反応させ、0.2%トライトンX-100添加PBS(PBST)で3回洗浄した。二次抗体として抗ラットIgG-Alexa488(Molecular Probes)を1:500でPBSTに希釈し、4℃で16〜18時間反応させた。その後、細胞の核を染色するため、5 μg/mlのDAPI(Sigma)をPBSTで5000倍希釈したものと細胞を室温で5分間反応させ、PBSにて3回洗浄した後、倒立蛍光顕微鏡システム(キーエンス)にて観察した。
細胞からのmRNAをSepasol(R)-RNA I Super G(ナカライテスク)で抽出し、SuperScriptIII逆転写キット(Invitrogen)を用いてプロトコールに従いcDNA合成した。その産物に対して下記のプライマーとEx Taq(タカラバイオ)を用いたコンベンショナルPCR反応を行い、アガロースゲル電気泳動で遺伝子発現のバンドを確認した。PCR反応はサーマルサイクラーVeriti(ABI)にて行い、アニーリング温度60℃、25〜30サイクルで反応させた。定量PCRは、プローブセット、SYBR Green(Applied Biosystems)およびStep Oneサーマルサイクラー(Applied Biosystems)を用いて実施した。β-アクチンをインバリアントコントロールとして使用した。標準化のために、7日目のサンプルの値をコントロール値(=1.0)として使用した。(図7bおよびc)。

はじめにクローニングされていないTet-MyoDヒトiPS細胞で、骨格筋分化誘導を行った(図2a)。Mahmoodら(A. Mahmood, et al., J Bone Miner Res 25, 1216, 2010)の骨格筋前駆細胞誘導法に従い、SB outgrowth細胞まで誘導し、その後SB outgrowth培地にDoxを4日間加えた。SB outgrowth培地は、SF-O3(三光純薬)、0.2%ウシ血清アルブミン(Sigma)、0.1%脂質(lipid)(Invitrogen)、100 μM 2-ME、および1 μM SB431542(Milteny Bio)を加えたものであった。4日経過後に、培地をDMEM(ナカライテスク)に2%ウマ血清(Sigma)、IGF-1(Peprotech)10 ng/mlおよび100 μM 2-MEとなるように加えたもの(2%ウマ血清DMEM)に変更した。培地を変更して4日間経過した後、免疫染色を行い、骨格筋への分化誘導がなされているか確認した。
T、Tbx6およびネガティブコントロールに対するsiRNAを購入した(Sigma, SASI_Hs01_00221962、SASI_Hs01_00166068およびSIC-001-10)。6ウェルのプレート中、2.0〜3.0×105細胞密度で播種したhiPS細胞に50 nMのsiRNAを、それぞれ0日目および3日目にトランスフェクトした。siRNAは、Lipofectamine RNAiMAX(Invitrogen)を用いて、製造者のプロトコールに従ってトランスフェクトした。
Tet-Myf5 iPS細胞クローンは6 cmディッシュ1枚分の細胞数を用意し、培地にて50倍に希釈したマトリゲルにてコートした10 cmディッシュに維持培養と同じ方法で播種した。翌日DoxをヒトiPS細胞用培地に添加し、播種後7日目まで毎日培地交換を行った。播種後7日目に、0.25%トリプシン/1 mM EDTAにて細胞を解離し、I型コラーゲンコート24ウェルプレート(IWAKI)に1ウェルあたり1×105細胞密度で播種した。使用した培地はαMEMに100 μM 2-MEおよび50 mU/L ペニシリン/50 μg/L ストレプトマイシンを添加したものに、さらに5%KSRを加えた培地であった。この培地で48時間培養した後、DMEM/F12(Invitrogen)に2 mM L-グルタミン(ナカライテスク)、1×非必須アミノ酸(Non-essential amino acid)(Invitrogen)、100 μM 2-ME、および50 mU/L ペニシリン/50 μg/L ストレプトマイシンを加えたものに、さらに2%ウマ血清を加えた培地に交換し、さらに48時間培養した。
分化した細胞を、2%パラフォルムアルデヒド(ナカライテスク)/PBSを用いて4℃で10分間固定し、PBSで各5分間、3回洗浄した後、メタノール(ナカライテスク)に1%過酸化水素(Wako)を加えたもので脱色を15分間行った。混合物を再度PBSで4℃、5分間、3回洗浄し、2%スキムミルク(BD)/PBST(PBSMT)にてブロッキングを4℃で15分間行った。一次抗体は、上記PBSMTブロッキング液中に、ラットモノクローナル抗体(mAb)ラミニン(1:15;ALEXIS)、マウスmAb抗ヒトミオシン重鎖(MF20)(1:400;R&D)、マウスmAbミオゲニン(F5D)(1:400;SANTA CRUZ)、ウサギポリクローナル抗体(pAb)MyoD(M-318)(1:400;SANTA CRUZ)、マウスmAbヒトスペクトリン(1:100;Leica)、マウスmAb抗ヒト核(1:200;MILLIPORE)、マウスmAb α骨格筋アクチン(1:200;Acris)、ウサギpAbクレアチンキナーゼM(Y14)(1:100;Bioworld Technology)およびマウスmAb抗ジスフェリン(NCL-Hamlet)(1:25;Leica)をそれぞれ希釈して使用した。室温で1時間細胞を反応させ、PBSMTで3回洗浄した。二次抗体として、Alexa fluor488結合ヤギ抗ウサギまたはマウスIgG、Alexa fluor568結合ヤギ抗ウサギまたはマウスIgG(すべて1:500;すべてInvitrogen)、およびHRP結合ヤギ抗マウスIgG(IHCでは1:200、ウェスタンブロッティングでは1:2000;Vector)をPBSMTで希釈したものを添加し、室温で1時間混合物を反応させた。室温でPBSMTにて10分間2回、PBSTにて10分間1回洗浄した後、DAB発色キット(ナカライテスク)を用いて発色させた。適切な発色後に細胞をPBSTで洗浄し、反応を停止させた。サンプルは、LMS710共焦点顕微鏡(Carl Zeiss)または倒立顕微鏡(オリンパス)で観察した。
非肥満糖尿病/重症複合免疫不全(NOD/SCID)マウスはCharles River Laboratoriesから購入し、ジストロフィンを発現しないDMD-ヌルマウスと交配させた。NOD-DMDマウスを作出し、in vivo移植研究のために使用した。筋肉内細胞移植の前に、マウスをジエチルエーテルで麻酔後、カルジオトキシンで損傷させた。カルジオトキシン損傷後24時間にて、6日目のMyoD-hiPSC(1.0×106〜9.5×106細胞/50 ml 10%マトリゲル(aMEM中))を左TA筋に注入した。本研究で使用したマウスはすべて移植後28日目に人道的に犠死させ、組織サンプルを回収した。
分化したMyoD-hiPSCを、GFPが組み込まれた(integrated)C2C12細胞株と共培養した。はじめにMyoD-hiPSCを7日間分化させた。7日目に、培地をDMEMに5%ウマ血清を添加したものに交換し、1.0×105 C2C12細胞をMyoD-hiPSC上に播種した。Bio Station CT(ニコン)を共培養サンプルのタイムラプス観察に使用した。撮影間隔は1時間であった。
MitoTracker Red CMXRos(Invitrogen)を添付のプロトコールに従って使用した。サンプルは、BZ-9000E(キーエンス)で観察した。
Tet-MyoD hiPSCの作製および評価
上述したTet-MyoDベクターをヒトiPS細胞にトランスフェクションし、ネオマイシン耐性を獲得した細胞群をドキシサイクリン(Doxycyclin)(Dox)添加群とDox非添加群に分け、24時間後にmCherryの発現およびMyoDの発現を観察した。Dox添加群においてのみMyoDタンパク質がmCherryと一致して発現していることが確認された(図1c)。また、同様の実験において、mRNAレベルでの発現解析も行った。その結果、Dox添加群では外因性MyoD(Tg)は強く誘導され、内因性MyoD(Endo)も誘導されていた(図1d)。Dox非添加群においても弱く外因性MyoDの発現が認められることから、Dox非添加状態でも発現がリークする細胞が存在すると考えられ、クローンの選別を行う必要性が示唆された(図1d)。Dox投与後7日目には、多核を有する、ミオシン重鎖(MHC)陽性またはミオゲニン陽性筋線維が検出された(図1e)。
上述したようにクローンの選別を行った。ネガティブコントロールである遺伝子改変していない201B7とmCherryの発現強度を比較した。24クローンのうち比較的高度なmCherryの発現を示すクローンNo. 9、16および20(B7 #9、B7 #16、B7 #20)を選択した(図3a)。また、それぞれのクローンをDox添加群および非添加群に分け、48時間後にmRNAを回収し、PCRにて遺伝子発現を解析した(図3b)。Dox非添加群では外因性MyoD(Tg)のリークした発現は認めず、内因性MyoD(Endo)の発現も認めなかった(図3b)。Dox添加群ではいずれのクローンもMyoD(Tg)、MyoD(Endo)両方の発現を認めた。Doxにより外来遺伝子発現が誘導され、内因性のMyoDも発現することが確認された(図3b)。
まずB7 #16を用いて、骨格筋分化能を定性的に評価した。図4aに示すように分化誘導を行い、14日後にMHCの発現を観察した。Dox添加群では褐色に染色されたMHC陽性骨格筋細胞が多数確認された(図4b、左)。高倍率での観察では、多核になり成熟していると考えられる骨格筋細胞も認めた(図4b、中央)。一方、Dox非添加群ではMHC陽性細胞は全く認められなかった(図4b、右)。このことから、選別をしたTet-MyoDクローンにおいても、Doxにより成熟した骨格筋細胞への分化誘導が可能であることが分かった。
比較的高度なmCherryの発現を示すクローンG4 #31、G1 #17およびG1 #23をさらに選択した(図5a)。外因性MyoD1のリークのない内因性MyoD1の発現は、3種の別個のhiPS細胞株201B7、253G1および254G41由来のMyoD-hiPSCクローン間で確認した(図5b)。
まず、Dox投与の開始時点を、mCherry陽性細胞が高度に出現することから、1日目と決定した(図6a)。さらに分化したMyoD-hiPSCは、Dox存在下であっても4日目で非応答性であった(図6b、矢頭)。次に、Dox投与期間について、MyoD-hiPSCを筋原性系列にコミットさせるのに十分であったことから、5日間超と決定した(図6c)。最後に、MyoD-hiPSCの筋原性分化のためのプロトコールを確立した(図7a)。
このような中胚葉遺伝子の発現が、本プロトコールにおける筋原性分化に必須であるかどうかに取り組むために、中胚葉遺伝子発現を分化初期の間にTまたはTbx6に対するsiRNAで抑制した。TまたはTbx6の発現は、該siRNAにより、3日目および5日目においてそれぞれ強力に抑制された(図8aおよび8b)。さらに、MyoD1の上流遺伝子であるPax3の発現も、TおよびTbx6の両方のsiRNAによって抑制された。中胚葉遺伝子の発現を抑制したにも関わらず、筋原性分化の効率は影響を受けなかった(図8cおよび8d)。このことから、未分化hiPSCにおける外因性のMyoD1の発現は、該細胞において前筋原性(premyogenic)の中胚葉遺伝子の発現を促進し得るが、未分化hiPSC集団の大部分は、成熟筋細胞へと直接分化することが示唆された。
本分化系において、3種の別個のhiPSCクローンはいずれもMHC陽性筋線維へと促進され得て(図9a)、MHC陽性細胞の効率は、オリジナルクローンとは無関係に、70〜90%の範囲であった(図9b)。分化したMyoD-hiPSCは、紡錘状に均一に形を変えた。従って、本分化系は、クローン間の差異を克服し、外因性MyoD1の優勢な発現によって効率的かつ均一な筋原性分化を実現する。
筋原性の特性を評価するために、組織学的分析を実施した。未分化hiPSCはほとんどミトコンドリアを有さなかったが、分化したMyoD-hiPSCは核周囲に多くのミトコンドリアを有した(図9c)。さらに、分化したMyoD-hiPSCは、成熟筋細胞マーカーである骨格筋アクチン及びCK-Mを発現した(図9d)。これらの特性はすべて、MyoD-hiPSC由来の、Doxによって誘導される筋原細胞が成熟筋細胞の特徴を有することを示唆する。分化したMyoD-hiPSCのグローバルな遺伝子プロファイルをmRNAマイクロアレイにより解析し、分化したヒト筋芽細胞株Hu5/E1826および未分化hiPSCと比較した(図10)。分化したMyoD-hiPSCのグローバルな遺伝子プロファイルは、分化Hu5/E18のものと非常に近似しており、未分化hiPSCのものとはかなり異なっていた(図10e)。さらに、筋系統に着目し、筋肉分化と関連する特定の遺伝子を選択し、そのmRNA発現プロファイルを解析した。分化したMyoD-hiPSCは、MyoD1の上流転写因子であるMyf5を除いて、分化したHu5/E18と同様に、選択した遺伝子の高発現を示した(図10f)。まとめると、MyoD-hiPSCから作製される、Doxによって誘導される筋原細胞は、分化したヒト筋芽細胞と類似した成熟筋細胞を示す。しかし、その分化プロセスは、MyoD-hiPSCがヒト筋芽細胞とは異なってMyf5を発現しないこと、および未分化細胞からMyoD1陽性筋原細胞へと飛び越え得ることから、全く異なるものである。
Tet-MyoD1ベクターを導入した、2つのMM患者由来hiPSCクローン(MyoD-MM #5およびMyoD-MM #6)は、他のhiPSCと形態学的に同一であり(図12a)、SeVベクターの発現が残存することなく、内因性の未分化マーカー遺伝子を発現した(図12b)。MyoD-MM hiPSCは、MHCまたはミオゲニン陽性成熟筋細胞に分化し得た(図12c)。
上述したTet-Myf5ベクターをヒトiPS細胞にトランスフェクションし、ネオマイシン耐性を獲得した細胞群から単一コロニーをピックアップし、24個のクローンを得た。それぞれのクローンをDox添加群および非添加群に分け、48時間後にmCherryの発現解析にてクローンの選別を行った(図14aおよび14b)。24クローンのうち比較的mCherryの発現の高度なクローンである、クローンNo. 2、18および21(#2、#18、#21)を選択した(図14c)。また、Dox添加群および非添加群のそれぞれにつき、48時間後にmRNAを回収しPCRにて遺伝子発現を解析した(図14d)。Dox非添加群では外因性Myf5(Tg)のリークした発現は認めず、内因性Myf5(Endo)をはじめ、Myf5で誘導されるMyoDやミオゲニンの発現も認めなかった(図14d)。Dox添加群ではいずれのクローンもMyoD(Tg)およびMyoD(Endo)両方の発現を認め、Doxにより外因性遺伝子発現が誘導され、内因性の骨格筋分化遺伝子も発現することが確認された(図14d)。
図15aに示すように、Tet-Myf5 hiPSCからの分化誘導を行った。Dox添加群では48時間後に非常に均一なmCherryの発現を認め(図15b、上段)、同時に、Dox非添加群と比べ紡錘形の細胞形態に変化していることが分かった(図15b、下段)。分化11日後にミオゲニンおよびMHCの発現を観察した。#2、#18および#21の3クローンともミオゲニン陽性、MHC陽性の骨格筋細胞への分化を認めた。#2におけるミオゲニンおよびMHCの発現を図15cに示す。上段はミオゲニンの発現であり、一本の筋線維に多核のミオゲニン陽性細胞が観察される(図15c、矢印)。また下段はMHCの発現であり、非常に多くの骨格筋細胞が均一に分化している様子が観察される。以上の結果より、テトラサイクリン応答性ベクターによるMyf5の強制発現においても、iPS細胞を骨格筋細胞へと効率よく分化させることができることが分かった。
Claims (9)
- 多能性幹細胞から骨格筋細胞を製造する方法であって、以下の工程:
(1)多能性幹細胞を、胚様体を形成せず、bFGFを含有しない基本培地で3日間以内培養する工程、および
(2)工程(1)で得られた細胞において、MyoD、Myf5およびそれらをコードする核酸から選ばれる1以上の外因性因子を、5日間以上10日間以下発現させる工程
を含む方法。 - 多能性幹細胞がヒト多能性幹細胞である、請求項1に記載の方法。
- 前記工程(1)の培養が1日以内である、請求項1または2に記載の方法。
- 前記工程(2)の後、ウマ血清を含む培養液でさらに培養する工程を含む、請求項1から3のいずれか1項に記載の方法。
- 前記多能性幹細胞が、MyoDまたはMyf5をコードする核酸を含む薬剤応答性誘導ベクターを導入した多能性幹細胞であり、前記工程(2)が、該ベクターと対応する薬剤の存在下で培養することにより行われる、請求項1から4のいずれか1項に記載の方法。
- 前記工程(2)の1以上の外因性因子の発現を維持する期間が6日間である、請求項1から5のいずれか1項に記載の方法。
- (1)請求項1から5のいずれか1項に記載の方法で製造された、スクリーニングを行うのに十分な量の、骨格筋細胞を提供する工程、
(2)該骨格筋細胞に被験物質を接触させる工程、及び
(3)被験物質を接触させなかった場合と比較して、該骨格筋細胞の病態が緩和された場合に、該被験物質をミオパチーの治療又は予防の候補物質として選別する工程
を含む、ミオパチー治療または予防剤のスクリーニング方法。 - 前記ミオパチーが、筋ジストロフィー又は遠位型ミオパチーである、請求項7に記載の方法。
- 前記遠位型ミオパチーが三好型ミオパチーである、請求項8に記載の方法。
