Description
なお、該文献では、ゲニスティンのE2との相関係数が0.93であり、ダイゼインのそれは0.74であることが注目される。しかしながら、ゲニスティンはヘム鉄による生体防御因子ヘムオキシダーゼ(HO-1)の発現誘導を抑制する一方、ダイゼインは抑制しない。ヘム鉄とイソフラボンは食品として併用される可能性も高いことから、食品成分としてはHO-1誘導を抑制しにくいダイゼインが好ましいと考えられる。またエストラジオールはHO-1誘導を抑制しなかったことなどから、イソフラボンによるHO-1誘導抑制は、エストロゲン受容体を介する既知の機序とは異なるものによる可能性が示唆されている。(非特許文献12)。
すなわち、本発明は、以下の(1)〜(5)を提供するものである。
(1) ダイゼインの微生物代謝産物であるエコールとほぼ同等のエストゲン類似活性を有する作用物を含有するレッドクローバーと亜麻種子、およびレッドクローバー由来のイソフラボン抽出物と亜麻種子由来のリグナン抽出物のそれぞれの混合物のケカビ目に属するカビ発酵産物。
(2) (1)記載の発酵産物からの抽出物。
(3) (1)記載のカビは、リゾプス(Rhizopus)属では、リゾプスニベウス(R. niveus)、リゾプス オリザ(R. oryzae)、リゾプス オリゴスポラス(R.oligosporus)、 リゾプス ストロニファー(R.stolonifer)、及びムコール(Mucor)属としてはムコール シルシネロイデス(M. circineloides)、ムコール ヒエマリス(M.hiemalis)、ムコール プシラス(M. pusillus)などの内から選ばれる少なくとも1種である。
(4) (2)記載の抽出物で、DNAチップとMCF-7細胞を用いて解析算出した、該抽出物投与の有無による発現変動遺伝子とエストロゲン応答遺伝子の発現型の相関係数(R)が、少なくとも0.7以上であることを特徴とする抽出物及びその基質発酵産物。
(5) (1)及び(2)記載の発酵産物あるいは抽出物を含有する食品、飲料、化粧品、香水、医薬部外品及び医薬品。
さらに、該発酵産物および該抽出物を含有する食品、飲料、化粧品、育毛剤、香水、医薬部外品及び医薬品等を提供することができる。
ケカビ目に属するカビの発酵物からの抽出物のエストロゲン類似活性は、エストロゲン応答細胞であるMCF-7細胞およびエストロゲン応答遺伝子を搭載したDNAチップを用いて、それぞれ該抽出物およびE2のMCF-7細胞における遺伝子発現プロファイルの示す相関を解析し、相関係数を求めることにより測定することができる。
ケカビ目に属するカビとしてはリゾプス(Rhizopus)属およびムコール(Mucor)属のカビを使用することができる。
本発明に係る微生物については、独立行政法人製品評価技術基盤機構等から入手することができる。
固体培養法(仕込み量約50%, w/w)によるカビ発酵プロセスを以下に示す。各種原料に水分含量が約50%になるように製造水を混合して、加圧滅菌(121℃,15分間)した後、40℃以下になるまで放冷した。これに初発胞子数としてグラム当たり約104個になるようにカビ胞子を接種し、好気的な条件を維持するために培養層を薄く平らにして30℃で48時間培養することにより発酵産物を得た。また、原料が粉体の場合には、蒸米や加圧滅菌した亜麻種子を一定割合添加して間隙を形成させて同様に固体培養に、あるいはそのまま滅菌してジャーファーメンターによる液体培養(仕込み量約10%, w/w)に供することができる。
供試する各種抽出物サンプルは、カビとしてムコール プシラス(M. pusillus) を用いて実施例1に従い調製したレッドクローバーおよび亜麻種子のそれぞれ単独、およびレッドクローバーと亜麻種子の混合物の発酵産物から調製したそれぞれの抽出物の少量を70%エタノールに溶解し、メンブランフィルター(0.22μ)でろ過滅菌後、遠心濃縮機により濃縮・乾固して調製した。
これらの結果から、エストロゲン応答遺伝子148種を搭載したDNAチップによるアッセイには、これら細胞増殖促進活性を示す上記濃度で培養したMCF-7細胞から抽出したmRNAを用いた。
1.実験方法
エストロゲン応答遺伝子(148種類)を酵素(enzymes), シグナル(signaling), 細胞増殖(proliferation), 転写(transcription), 細胞輸送(transport), およびその他(others)の機能別グループに分けて、機能別グループごとの発現における各抽出物とE2との相関解析を実施した結果を表2及び図4に示す。
Claims (5)
- (a)レッドクローバーの花部若しく葉花、及びそのイソフラボン抽出物から選ばれる少なくとも1種と(b)亜麻種子および亜麻種子由来のリグナン抽出物から選ばれる少なくとも1種との混合物からの、ムコール属に属するカビによる発酵産物である、エストロゲン類似活性を有する発酵産物。
- 請求項1記載の発酵産物からの抽出物。
- 上記ムコール属に属するカビが、ムコール シルシネロイデス(M. circineloides)、ムコール ヒエマリス(M.hiemalis)及びムコール プシラス(M. pusillus)から選ばれる少なくとも1種である、請求項1記載の発酵産物又は請求項2記載の抽出物。
- 請求項2又は3記載の抽出物で、DNAチップとMCF-7細胞を用いて解析算出した、該抽出物投与の有無による発現変動遺伝子とエストロゲン(E2)応答遺伝子の各発現の相関係数(R)が、少なくとも0.7以上であることを特徴とする請求項2又は3記載の抽出物。
- 上記抽出物が、エタノール抽出物である、請求項2〜4のいずれか1項記載の抽出物。
