Description
(b)段階(a)のアルブミン−陰イオン性高分子コンジュゲートをエマルジョン化してマイクロサイズのバブルを形成させる段階と、
(c)段階(b)のマイクロサイズのバブルを架橋して、アルブミンが架橋結合されたマイクロビーズを形成させる段階と、
を含む、抗癌剤吸着能力の向上された生分解性マイクロビーズの製造方法を提供する。
(i)本発明は、抗癌剤吸着能力の向上された生分解性マイクロビーズ、その製造方法及びこれを利用した癌治療方法を提供する。
(ii)本発明によると、アルブミンと陰イオン性高分子とのアミド結合及び架橋結合により優れた抗癌剤吸着能力を有する堅固なビーズを製造することができる。
(iii)本発明のマイクロビーズは、生体適合性及び生分解性高分子で製造されて、人体に適用時安全であり、肝腫瘍に栄養を供給する血管を効果的に塞ぐと同時に、ビーズの表面に吸着されている抗癌剤を持続的に放出することにより、腫瘍の成長を有効に抑制させることができる。
(iv)したがって、本発明は、肝癌の化学塞栓術に有用に活用できる。
アルブミンのアミン基(NH2−)とコンドロイチン硫酸のカルボキシ基(COOH−)をアミド結合させるために、SCBH(sodium cyanoborohydride)またはEDC(1−Ethyl−3−(3−dimethylamino−propyl)carbodiimide)/NHS(N−Hydroxy succinimide)を使用した。まず、陰イオン性高分子をSCBHまたはEDC/NHSを利用して活性化させた後、下記表1の組成1〜5のようにアルブミンと反応させて結合させた。以後、これを1〜2日間透析して未反応物を除去し、ビーズ製造用溶液を収得した。
ドキソルビシン吸着実験を下記のように行った。まず、50mgのドキソルビシンを2mlの蒸留水に溶解した。1mlのビーズを正確に取ってドキソルビシン溶液に入れて混合した。20分間常温で放置した後、上澄み液を取って、紫外線分光器で483nmで吸光度を測定した。予め作成しておいた検量線に対比して濃度を計算すると、50mg/2mlのドキソルビシン溶液から抜け出したドキソルビシンの量を計算することができ、この値は、ドキソルビシンがビーズに吸着された量である。実験結果は、表2に示した。
薬物の溶出挙動を確認するために、溶出実験を進行した。実験方法は、以下のようである。ドキソルビシンで4.0mgに当るビーズを50mlコニカルチューブに入れて、溶出液(PBS、pH 7.4)50mlを満たして37℃でインキュベーションした。溶出液は、回収時点で全て回収した後、新しい溶出液に入れ替えて、これによって溶出曲線を累積値で算出した。溶出された薬物の分析は、HPLCで行った。溶出結果は、図3に示した。
Claims (7)
- アルブミンと陰イオン性高分子がアミド結合されたアルブミン−陰イオン性高分子コンジュゲートを含み、
前記陰イオン性高分子が、コンドロイチン硫酸、デルマタン硫酸、ヘパラン硫酸、ヘパリン、ケラタン硫酸、及びヒアルロナンからなる群から選択され、及び
前記アルブミンが、熱架橋により架橋結合されたことを特徴とする抗癌剤吸着能力の向上された生分解性の化学塞栓用マイクロビーズ。 - 前記マイクロビーズが、前記陰イオン性高分子との静電気的引力によってビーズ表面に吸着された抗癌剤を更に含む請求項1に記載のマイクロビーズ。
- 前記抗癌剤が、アントラサイクリン系抗癌剤である請求項2に記載のマイクロビーズ。
- 前記アントラサイクリン系抗癌剤が、ダウノルビシン、ドキソルビシン、エピルビシン、イダルビシン、ゲムシタビン、ミトザントロン、ピラルビシン及びバルビシンからなる群から選択される請求項3に記載のマイクロビーズ。
- 前記抗癌剤が、イリノテカンである請求項2に記載のマイクロビーズ。
- 前記化学塞栓が、肝癌化学塞栓である請求項1に記載のマイクロビーズ。
- (a)アルブミン及び陰イオン性高分子をアミド結合してアルブミン−陰イオン性高分子コンジュゲートを得る工程であって、前記陰イオン性高分子が、コンドロイチン硫酸、デルマタン硫酸、ヘパラン硫酸、ヘパリン、ケラタン硫酸、及びヒアルロナンからなる群から選択される工程と、
(b)工程(a)のアルブミン−陰イオン性高分子コンジュゲートをエマルジョン化してマイクロサイズのバブルを形成させる工程と、
(c)工程(b)のマイクロサイズのバブルを熱架橋して、アルブミンが熱架橋により架橋結合された化学塞栓用マイクロビーズを形成させる工程と、
を含むことを特徴とする抗癌剤吸着能力の向上された生分解性の化学塞栓用マイクロビーズの製造方法。
