Micromer−M PEG−NH2
市販の単分散Micromer(登録商標)−M粒子(Micromod Partikeltechnologie GmbH)をこれらの検討に用いた。各粒子は、アミノ官能基を有する二官能性ポリエチレングリコール(−NH−(CH2−O−CH2)200−NH2)で修飾された架橋メタクリル酸メチル−メタクリル酸コポリマーマトリックスからなる表面コーティングを施したスチレン−マレイン酸コポリマーに埋め込まれたマグヘマイト(γ−Fe2O3)ナノ粒子のコアからなっている。ビーズは、4.9μmの平均直径及び0.2μmの標準偏差を有していた。それらは、7×108粒子/mlの濃度、50mg/mlの磁気物質含量で水中に分散させた。置換度は、製造業者によれば、1mg当たり5〜6nmolNH2基又は1粒子当たり2.2×108NH2基であった。
Micromer(登録商標)−M PEG−NH2(100μL、7×108粒子/ml)を1000μLのPBS(10mMリン酸塩、150mM NaCl、10mM EDTA、pH7.4)で3回洗浄し、1000μLのPBSに再懸濁した。3−(2−ピリジルジチオ)プロピオン酸N−スクシンイミジル(SPDP)(25μL、DMSO中20mM)をビーズ懸濁液に加え、90分間反応させた。ビーズを1000μLのPBSで5回洗浄し、1000μLのPBSに再懸濁し、4℃に維持した。
Micromer−M PEG−SS−ピリジル(100μL、7×107粒子)を1000μLの2%w/vDTT含有PBS中ジチオトレイトール(DTT)と混合し、室温で20分間インキュベートした。粒子を磁石で収集し、上清をSS−ピリジルの分光光度分析にさらに用い、SS−ピリジル含量(図2)をShimadzu(京都、日本)UV−2101PC分光光度計及び石英キュベットを用いて波長343nm(ξ=8080M−1cm−1)で分光光度法により測定した。約1.0×108SS−ピリジル基が各粒子に結合していた。
電気酸化の直前に、1000μLの懸濁液中のMicromer−M PEG−SS−ピリジル粒子(7×107粒子/ml)を外部磁石で収集し、緩衝液を1000μLの酢酸緩衝液(pH4.5、2%w/vDTT)中ジチオトレイトール(DTT)に変え、室温で20分間インキュベートした。ビーズをPBS(10mMリン酸塩、150mM NaCl、pH7.4)で5回洗浄し、80mlのPBSに8.75×106粒子/mlの最終濃度になるように再懸濁し、室温に維持した。
セファロース6Bをガラスフィルター上で蒸留水で洗浄し、吸引乾燥した。3gの乾燥ゲル粒子を2.4mlの1M水酸化ナトリウム溶液に再懸濁し、室温で撹拌しながら0.45mlのエピクロロヒドリンを1滴ずつ加えた。温度を60℃に上昇させ、2時間維持した。セファロースゲルをガラスフィルター上で中性まで蒸留水で洗浄した。さらにエポキシド活性化ゲル(3g)をガラスフィルター上で50mlの0.5Mリン酸緩衝液pH6.3で洗浄し、吸引乾燥した。ゲルをリン酸緩衝液(6ml)に再懸濁した。3mlの2Mチオ硫酸ナトリウムを加え、混合物を室温で6時間振とうした。過剰のチオ硫酸ナトリウムを蒸留水でチオ硫酸エステルゲルから洗い流した。チオール基含有ゲルを調製するために、チオ硫酸エステルゲルをジチオトレイトール(DTT)で還元した。1gのチオ硫酸エステルゲルをガラスフィルター上で洗浄し、3mlの0.1M重炭酸ナトリウム溶液に再懸濁した。2ml(1mM)EDTA溶液中0.1gのDTTを加え、エステルゲルを30分間還元した。還元ゲルを30mlの0.1M重炭酸ナトリウム溶液(1M塩化ナトリウム及び1mM EDTA)で、最後に100mlの1mM EDTA溶液で洗浄した。チオール基含有セファロース6B粒子を10mMリン酸ナトリウム緩衝液に懸濁した。
粒子の電気酸化に用いた反応セルは、金プレートの上部に取り付けたテフロン(登録商標)円筒からなっていた。金プレートの下に取り付けた永久磁石(金プレートの内表面で350mT)を用いて垂直磁場を印加した。対及び参照電極を反応セルの内側に取り付け、作用電極として機能する金プレートを反応セルの外側に接続した(図2)。
実験の前に、反応セル及び作用電極を7.5mlの30%(w/v)過酸化水素(Merck Inc.、P.A.グレード)と15mlの硫酸(P.A.グレード)の混合物で洗浄した後、水ですすいだ。3mlのPBSに入れたチオール基含有粒子、0.75mg若しくは0.25mg磁気粒子又は2〜6mgアガロース粒子を次に反応セルに加えた。粒子を重力(アガロース粒子)及び磁場(磁気粒子)により5分間にわたり金表面上に分布させた。Ag/AgCl参照電極に対する0.45〜0.90Vの範囲の電位を1秒間印加した。チオール基含有粒子の異なる酸化の間に作用、対及び参照電極をPBSで洗浄した。その後、粒子を再懸濁し、永久磁石により磁気粒子を、遠心分離によりアガロース粒子を収集することにより、容積を200μLに減少させた。測定の前に、金作用電極が適切に機能していたことを確認するために粒子の非存在下でPBS溶液中でサイクリックボルタンモグラム(−0.2〜+1.5V、50mV/s)を記録した。
電気酸化実験からの粒子懸濁液を100μLの生体分子(IgG、β−アラニン、α−ラクトアルブミン、α−ラクトアルブミン(FITC)及びプロテインA)(PBS中2mg/ml)と混合し、40分間インキュベートした。約10mlのPBSで粒子を洗浄することにより、非結合生体分子を除去した。反応は、粒子表面の蛍光の程度に関して、及びアミノ酸分析により評価した。蛍光は、Nikon Coolpixカメラを装備したNikon Eclipse蛍光顕微鏡により評価した。494nmの励起波長及び520nmの発光波長を得るように蛍光フィルターを調整した。アミノ酸分析は、特殊フィルターの使用によるアンモニアの除去がUltrapac 8樹脂を含む4.6×200mm高分解能PEEKカラム(Biachrome)上のすべてのアミノ酸の分離を可能にする、Spackman、Stein及びMooreにより開発された古典的方法の改善された変法により実施した。アミノ酸の量は、Biachrome20及びBiachrome30分析機器を用いることにより2種の波長440nm及び570nmで検出した。検出限界は25〜50pmolであり、定量限界は50〜100pmolであった。2mlの6M HCl(内部標準を含む)を既知重量の一定容積の粒子懸濁液に加え、次にこれを100℃で24時間加水分解した。アミノ酸分析の前にアガロース粒子を凍結乾燥した。
電気酸化時の異なる電位の試験
固定化分子は、FITCマーカー付き分子を用いることにより検討した全ビーズ表面に均一に分布していた。分子を結合する能力は、電気酸化ステップ中に印加された電圧に依存し(表1a参照)、ゼロボルトで分子の非特異的吸着が起こり、0.9Vの電圧で最大であった。表1に示す結果から、印加電圧及び粒子濃度の両方により粒子上の分子の置換の程度を制御することが可能であることがわかる。生体分子の結合容量をアガロース(セファロース6B)において検討したところ、凍結乾燥アガロース1g当たりIgG29.4mgであることがわかった。
α−ラクトアルブミンをアガロースに結合する能力を各種ボルトについて検討した。電気酸化を60秒間実施した。アミノ酸分析を実施して、結合α−ラクトアルブミンの量を計算した。表1bにおける結果を参照のこと。
安定性試験
表面と固定化生体分子との間の結合の安定性を検討するために、IgG及びβ−アラニンを電気酸化磁気粒子に固定化した。粒子を大過剰のpH7.2のリン酸緩衝生理食塩水で数時間にわたり徹底的に洗浄し、徹底的な洗浄処置の前後にアミノ酸分析を実施した。
低pHにおける安定性試験
電気化学的酸化アガロースへの固定化タンパク質の低pHにおける安定性を検討するために、上述のようにプロテインA及びIgGを成功裏に固定化した。その後、アガロース−タンパク質粒子をグリシン緩衝液pH3で20分間処理した。表3に示すように、固定化プロテインA及びIgG間の結合は、pH3で少なくとも20分間安定であった。固定化分子は低pHで安定であると結論付けることができる。

アガロース−SH粒子の化学的酸化と電気酸化との比較
過酸化水素による化学的酸化。2mgのチオール基含有アガロース粒子を2mlの酢酸緩衝液pH5中3.5%H2O2で20時間酸化した。酸化後、粒子をガラスフィルター上で10mlのPBSで洗浄した。生体分子である500μlのα−ラクトアルブミン(FITC)(PBS中2mg/ml)を酸化アガロース−SH粒子に加えた。40分後及び24時間後に反応を粒子表面の蛍光の程度に関して評価した。40分後に蛍光は認められなかった。24時間後に200倍の蛍光顕微鏡で見える弱い蛍光が粒子表面上に出現した。α−ラクトアルブミン(FITC)の蛍光が40分後に肉眼で見える電気酸化アガロース−SH粒子と比較して、これは、アガロース粒子上の天然生体分子の固定化の非効率的な技術である。
8mgのチオール基含有アガロース粒子を10mlの酢酸緩衝液pH5中3.5%H2O2で12時間酸化した。酸化後、粒子をガラスフィルター上で50mlのPBSで洗浄した。次に粒子を10mlの塩化金(III)(PBS中8mM)と2分間混合した。粒子をガラスフィルター上で100mlのPBSで洗浄し、次に生体分子である500μlのα−ラクトアルブミン(FITC)(PBS中2mg/ml)と40分間混合した。粒子を再びガラスフィルター上で50mlのPBSで洗浄した。粒子は、強い均一な蛍光を示し、生体分子がアガロース粒子上に成功裏に固定化されたことがわかる。
8mgのチオール基含有アガロース粒子を10mlの塩化金(III)の溶液(PBS中8mM)と2分間混合した。粒子をガラスフィルター上で100mlのPBSで洗浄し、次に生体分子である500μlのα−ラクトアルブミン(FITC)(PBS中2mg/ml)と40分間混合した。粒子を再びガラスフィルター上で50mlのPBSで洗浄した。粒子は、強い均一な蛍光を示し、生体分子がアガロース粒子上に成功裏に固定化されたことがわかる。
Claims (19)
- 少なくとも1つのアミノ基を含む少なくとも1つの分子の共有結合固定化の方法であって、
a.−SH基を含む表面を準備するステップと、
b.少なくとも1つの貴金属イオンの存在下で酸化還元反応により−SH基を含む表面を酸化するステップと
c.前記表面を少なくとも1つのアミノ基を含む少なくとも1つの分子と接触させて、前記表面への前記少なくとも1つの分子の共有結合を得るステップと
の順次的なステップを含み、前記少なくとも1つのアミノ基が前記共有結合を得るステップに関与する上記方法。 - 前記少なくとも1つの貴金属イオンがAu及びPtからなる群から選択される、請求項1に記載の方法。
- 前記酸化還元反応が前記表面上の少なくとも1つの官能基を生じさせ、前記少なくとも1つの官能基がチオール、ジスルフィド、チオスルフィネート基及びチオスルホネート基からなる群から選択される、請求項1または2に記載の方法。
- 前記表面上の前記官能基が前記貴金属イオンと反応する、請求項1から3までのいずれか一項に記載の方法。
- 酸化還元反応を用いて前記表面を酸化するステップの前に−SH基の少なくとも一部を互いに反応させる、請求項1から4までのいずれか一項に記載の方法。
- 酸化還元反応を用いて前記表面を酸化するステップの前にすべての−SH基を互いに反応させる、請求項1から4までのいずれか一項に記載の方法。
- 前記少なくとも1つのアミノ基を含む少なくとも1つの分子が、炭素原子を含む分子、アミノ酸、ペプチド、タンパク質、抗体及びアプタマーからなる群から選択される、請求項1から6までのいずれか一項に記載の方法。
- 前記少なくとも1つのアミノ基を含む少なくとも1つの分子が、タンパク質及び抗体からなる群から選択される少なくとも1つの分子である、請求項1から7までのいずれか一項に記載の方法。
- 前記酸化還元反応を水溶液中で実施する、請求項1から8までのいずれか一項に記載の方法。
- 前記酸化還元反応を2種以上の溶媒の混合液中で実施する、請求項1から8までのいずれか一項に記載の方法。
- 前記表面を前記少なくとも1つのアミノ基を含む少なくとも1つの分子と接触させるステップを水溶液中で実施する、請求項1から10までのいずれか一項に記載の方法。
- 前記表面を前記少なくとも1つのアミノ基を含む少なくとも1つの分子と接触させるステップを溶媒の混合液中で実施する、請求項1から11までのいずれか一項に記載の方法。
- 15℃〜300℃の温度で実施される、請求項1から12までのいずれか一項に記載の方法。
- 前記酸化還元反応が電気酸化を含み、前記電気酸化が0.5〜3Vの電位を用いて実施される、請求項1から13までのいずれか一項に記載の方法。
- 前記酸化還元反応が電気酸化を含み、前記電気酸化が1秒から10分までの期間中に実施される、請求項1から14までのいずれか一項に記載の方法。
- 前記表面と前記少なくとも1つのアミノ基を含む少なくとも1つの分子との接触が10分から10時間までの期間中に実施される、請求項1から15までのいずれか一項に記載の方法。
- 前記表面上の官能基を得るために前記表面を誘導体化に供し、前記官能基が−SH基及び−SS−基からなる群から選択される、請求項1から16までのいずれか一項に記載の方法。
- 少なくとも1つのアミノ基を含む少なくとも1つの分子と後に共有結合するために、−SH基を含む表面上で貴金属イオンの存在下に酸化還元反応を実施して、アミノ基と反応性で共有結合を形成し得る硫黄-金属イオンコンプレックス又はクラスターを形成する、−SH基を含む表面を修飾する方法。
- 貴金属イオンの存在下での酸化還元反応により生成される表面結合チオスルフィネート及び表面結合チオスルホネートの少なくとも1つと、アミノ基とを共有結合させる、少なくとも1つのアミノ基を含む分子を固定化する方法。
