によって成長されうる。
これらの試験では、混合リンパ球培養(MLC)における、CD4+、CD25+T細胞、またはCD4+、CD25−T細胞サブセット、またはこれらのCD4+T細胞サブセットの組合せに対する、CD4+T細胞の増殖を比較した。細胞は、ナイーブDAラットまたは完全に同種のPVG異所性心臓移植片に対して耐性であるまたはDAラットのいずれかのリンパ節および脾臓から調製した。耐性としては、免疫抑制の維持の非存在下での、75日後の移植片の非拒絶として定義した。自己(DA)、および完全にMHC不適合な特異的ドナー(PVG)および完全にMHC不適合な第三者(ルイス)に対する応答を比較した。DA、PVG、ルイスはMHC抗原を共有せず、完全に互いに対して同種である。この耐性の性質は、以前に記載されており、シクロスポリン、および抗CD4または抗CD3モノクローナル抗体療法を含む移植後最初の2週間における様々な療法によって、PVG異所性心臓同種移植片でDAラットにおいて耐性の性質が誘発されうる(J.Exp Med.1990年、171:141、Transplantation、1993年、55:459頁、Transplantation 1997年、64:1559頁)。特異的ドナー刺激に対する非分画末梢リンパ球のMLC増殖性応答は、一般に第三者のものと同等である(Transplantation、1993年、55、380−385頁)。
動物および方法。DA ラット(RT1a)、PVGラット(RT1c)、ルイスラット(RT1l)、およびスプラーグードーレイ(Sprague Dawley)ラットを、以前に記載されている通り繁殖させて維持した(J.Immunol.1998年、161;5146頁)。
脾臓およびリンパ節細胞(LNC)に由来する単一細胞懸濁液を、記載されている通り(34)調製し、RBCを、0.83%NH4Cl、0.1%KHCO3、および10mM EDTAの緩衝液(pH7.2)によって溶解した。細胞をPBS/2%BSA(MultiGel、バイオサイエンス社(Biosciences)、Castle Hill、NSW、オーストラリア)中に再懸濁した。
刺激細胞は、24時間前に8.5グレイ全身照射が与えられたラットの胸腺由来であった。この刺激細胞の集団は成熟リンパ球が枯渇し、抗原提示細胞が増大している。増大した抗原提示細胞は、好ましい。なぜならば、機能的リンパ球様細胞が刺激され、かつ応答細胞を活性化するサイトカインを産生し、またはバックグラウンド刺激をもたらしうるからである。全身照射ドナーに由来する刺激細胞は、24時間以内にインビボで破壊された末梢および胸腺リンパ球様細胞を有する。樹状細胞を増大するために使用されうる代わりの方法が、モノクローナル抗体選択および磁気ビーズ分離を用いたものである。刺激細胞を照射し、一夜放置して末梢リンパ球を照射の効果で死滅させ、抗原提示細胞が増大した集団を残すこともできる。104のこれらの刺激細胞が、インビトロで照射した2×105の脾臓細胞と同様に有効であった。刺激細胞に対する応答細胞の通常の比は、末梢リンパ球様細胞を刺激因子として使用する場合に1:1〜2:1であるが、TおよびBリンパ球の枯渇によって抗原提示細胞の増大が認められる場合、刺激細胞に対する応答は10−100:1でありうる。

ナイーブ非分画リンパ球の増殖の測定、およびCD4+、CD25+T細胞、CD4+T細胞、またはCD4+、CD25−T細胞の濃縮集団の測定の結果は、図1Aに示されている。図1Aからわかるように、CD4+T細胞の4および5日目の応答は、CD4+、CD25+T細胞の枯渇後に得られた細胞(濃縮CD4+、CD25−T細胞で示される)よりも小さい。CD4+、CD25+T細胞の応答は、非分画CD4+T細胞またはCD4+、CD25−T細胞のいずれかによるよりもはるかに小さい。この動態試験は、4および5日目の増殖が、非分画CD4+T細胞およびCD4+、CD25−T細胞の増殖性応答を明らかに示すために最も有用であることを明らかに示す。ナイーブCD4+、CD25−T細胞の応答は、早期に3および4日目にピークに達し、5または6日目には減少するのに対して、非分画CD4+T細胞およびナイーブCD4+、CD25−T細胞の応答は、4日目までは現れることはなく、減少前の5および6日目にピークを示す。
ナイーブCD4+T細胞、ナイーブCD4+、CD25+T細胞、およびナイーブCD4+、CD25−T細胞の自己刺激株、および2つの同種刺激株に対する増殖の比較の結果が図1Bに示されている。図1Bからわかるように、同系DA刺激因子に対する4日目の増殖性応答はすべての場合に低い。PVGおよびルイスに対する応答は、各々の細胞のサブタイプにおいて同様である。上記の通り、CD4+、CD25+T細胞の応答は他の集団よりも小さく、CD4+、CD25−T細胞が非分画CD4+T細胞よりも大きな応答を有する。
図1Cは、ナイーブCD4+、CD4+、CD25+T細胞またはCD4+、CD25−T細胞の連続希釈の結果を示す。図1Cからわかるように、すべての希釈でCD4+、CD25−による応答におけるT細胞は、非分画CD4+T細胞の同等の数のものよりも大きい。非分画CD4+T細胞は、およそ5%のナイーブCD4+、CD25+T細胞と95%のCD4+、CD25−T細胞の混合集団である。したがって、CD4+、CD25−T細胞の大きな増殖は単純に濃縮の効果による(すなわち、5%のCD4+、CD25+T細胞の喪失による)ものではない。このことは、マイナーな集団であるナイーブCD4+、CD25+T細胞が、主要な集団であるCD4+、CD25−T細胞を阻害する効果を有するということと一致するものである。
分離したナイーブCD4+、CD25+T細胞を、分離したナイーブCD4+、CD25−T細胞と混合する効果を試験し、その結果は図1Dに示されている。図1Dからわかるように、ナイーブCD4+、CD25+T細胞は、活性な抑制効果をCD4+、CD25−T細胞に対して有した。これらの実験において、CD4+、CD25+T細胞のCD4+、CD25−T細胞との1:10の混合は、結果として、CD4+T細胞における混合集団(比は1:10−1:20)と同様の増殖性応答をもたらした。比を1:1に増大させことにより、結果として、増殖性応答のほぼ完全な抑制をもたらした。このことは、インビトロでの免疫応答に対するナイーブCD4+、CD25+T細胞の明らかな非特異的阻害効果と一致する。
これは耐性動物に由来するCD4+T細胞、CD4+、CD25+T細胞、またはCD4+、CD25−T細胞の増殖を示す。CD4+T細胞の4および5日目の応答は、第三者ルイスに対してのように耐性株(PVG)と同様である。CD4+、CD25+T細胞の枯渇後においては(濃縮CD4+、CD25−T細胞と示される)、耐性株に対する増殖性応答は第三者に対してよりも小さい。
これは、PVG心臓同種移植片に対して耐性のDAラットに由来するCD4+、CD4+、CD25+T細胞またはCD4+、CD25−T細胞の連続希釈を示す。すべての希釈でCD4+、CD25−T細胞による特異的ドナー(PVG)に対する応答は、第三者(ルイス)に対する応答よりも小さく、かつ自己(DA)に対する応答と同じくらいの大きさである。対照的に、非分画末梢リンパ球またはCD4+T細胞を用いた場合、特異的ドナー(PVG)に対する応答は、第三者(ルイス)と同様であり、かつ自己(DA)と同じくらいの大きさである。すべての希釈で第三者(ルイス)または自己(DA)に対するCD4+、CD25−T細胞の応答は、同等数の非分画CD4+T細胞よりもはるかに大きい。
混合リンパ球培養(MLC)において、CD4+、CD25+T細胞の増殖性応答については、以下の特徴を有する:
a)MHC不適合同種抗原に対するCD4+、CD25+T細胞の増殖性応答は、バックグラウンドを除去する明確な培養条件で再現的に分析されうる。
b)ナイーブ動物に由来するCD4+、CD25+T細胞の増殖性応答は、MHC不適合刺激因子のみで、自己に対しては応答しない。
c)ナイーブ動物に由来するCD4+、CD25+T細胞は、ナイーブCD4+、CD25−T細胞の増殖を抑制する。
d)耐性動物に由来するCD4+、CD25+T細胞の増殖性応答は、第三者に対してのみで特異的ドナーまたは自己に対しては応答しない。
e)耐性動物に由来するCD4+、CD25+T細胞は、特異的ドナーに対する耐性CD4+、CD25−T細胞の応答を阻害することはできないが、第三者に対する応答を阻害することができる。
f)ナイーブ動物に由来するCD4+、CD25+T細胞は、非同種抗原特異的抑制効果を有する。この効果は、CD4+、CD24+T細胞の活性化を刺激し、またはCD4+、CD25+T細胞の生存を延長させ、または増殖を支持することができるサイトカインの非存在下では、応答抑制の能力を第三者同種抗原には保持するが特異的ドナー同種抗原には保持しない耐性動物に由来するCD4+、CD25+T細胞と異なるものである。
次の実験では、CD4+、CD25+T細胞のMLCにおける増殖性応答に対するさまざまなサイトカインの効果を試験し、および個々のサイトカインに対するナイーブおよび耐性濃縮CD25+、CD4+T細胞の応答を比較した。
サイトカインを記載されている通り生成したが、これはIL−2(32、33)を含み、1単位をIL−2依存CTLL系の最大増殖の50%を誘発するために必要とされる単位として定義した。クローン製造およびこれらのサイトカインの分析は記載されており、CHO−K1細胞へのトランスフェクションのための標準方法が使用され(Transplantation Proc.(1999年)31、1574−5頁、1999年、および31、1572ページ、1999年)、IL−4、IL−5、IFN−γ、IL−10、IL−12(p70)、IL−12(p40)、IL−13が含まれた。ヒトTGF−βをシグマ(Sigma)から購入した。サイトカインを各々の該当するウェルに50μlアリコートのCHO−Kトランスフェクト細胞系上清として添加した。これらの上清はml当り5000−50,000単位を有する。したがって、関連サイトカインは最終濃度がml当り1000−12,000単位であった。対照は培地に添加された非トランスフェクトCHO−k1細胞系からの上清とした。
濃縮CD4+、CD25+T細胞を自己(DA)、特異的ドナー(PVG)、および第三者(ルイス)刺激因子に対してMLCにおいて培養した。培養物を上記の通り3−4日で増殖について分析した。
図2Aは、示されているサイトカインの存在下でDA抗原またはPVG抗原のいずれかに対するナイーブDA細胞の応答を示す。図2Aからわかるように、IL−2およびIL−4のみが結果としてナイーブCD4+、CD25+T細胞の増殖の増強をもたらした。これらのサイトカインは、自己、および両方の同種刺激因子に対する増殖を同様の程度に増強した。これは、IL−2、またはIL−4のいずれかによるポリクローナル活性化と一致する。IL−5、IL−10、IL−13、IFN−γの添加はナイーブCD4+、CD25+T細胞の増殖を増強することはなかった。自己に対するバックグラウンド応答が約100cpmであり、蒸留水で得られたカウントと同等であることに注意されたい。すなわち、使用された方法により、(図1に記載されているように)、培地、または刺激細胞のような背景因子に対する非特異的増殖は認められなかった。バックグラウンドカウントは分析した蒸留水と同等である。
図2Bは、TGF−β(D)、IFN−γ(E)、IL−12p70(F)、IL−5(G)、またはIL−10(H)の存在下で自己抗原(黒色)またはドナー抗原(灰色)に対する活性化CD4+、CD25+T細胞の3日目での増殖を示すグラフである。図2Bは、活性化CD4+、CD25+T細胞の応答がいくつかの態様においてナイーブCD4+、CD25+T細胞の応答と異なることを示す。最初に、図1Eに記載されている通り、活性化CD4+、CD25+T細胞は特異的サイトカインの非存在下で特異的ドナーに対して増殖することはなく、それによってナイーブCD4+、CD25+T細胞と異なる。活性化CD4+、CD25+T細胞は、ナイーブCD4+、CD25+T細胞と同様に第三者ルイスに対して増殖する。活性化およびナイーブCD4+、CD25+T細胞は自己(DA)に対して増殖することはない。
サイトカインは、ナイーブおよび耐性CD4+、CD25+T細胞に対する異なる効果を有する。すなわち、
a)IL−2およびIL−4は、自己、特異的ドナー、および第三者に対する、ナイーブまたは耐性細胞ドナーに由来するCD4+、CD25+T細胞の増殖を顕著に増強する。
b)IL−5およびIFN−γは、特異的ドナーに対してのみ耐性CD4+、CD25+T細胞の増殖を増強し、自己または第三者に対しては増強しない。それらは自己または同種抗原に対するナイーブCD4+、CD25+T細胞の増殖に対する効果を有さない。
これらの試験では、耐性を宿主に移動させるCD4+、CD25+T細胞の能力を試験した。
adoptive transferアッセイ
これらを記載されている通り行った(J.Exp.Med 1978年、148;878−889頁およびTransplantation 1993年、55;374−379頁)。手短に言えば、60Co源から7.5−8.5グレイでDAラットを照射し、次いで、やはり照射されていたPVGドナーに由来する異所性成体心臓を移植した(図3A)。この照射源は、記載されている通り(J.Exp.Med 1978年、148;878−889頁)、15−30分にわたる処置を必要とした。その後の実験では、照射源は線型加速器であり、全身照射は2−3分で送達された。これらの照射宿主には異所性心臓移植片が与えられ、CD4+T細胞、非分画または濃縮されたCD4+、CD25−T細胞またはCD4+、CD25+T細胞により照射の24時間以内に回復させた。心臓移植片拒絶を最初に毎日の触診によって、疑いがある場合は、ECGモニタリングでモニタリングした。拒絶は、心電図活性の全喪失と同等であるすべての明白な収縮の喪失がある場合に記録される。
図3Aおよび3Bは、照射宿主へのadoptive transferの拒絶反応をもたらすナイーブCD4+T細胞の能力を示す。
図3Aは、異なる用量のナイーブCD4+T細胞で回復させたadoptive照射DAラットにおける拒絶時間の比較を示す。データはB.ホール(Hall)教授のすべての実験室およびサイトからまとめられた。A群、n=7、B群、n=17、C群、n=86、D群、n=6。宿主における細胞の均衡を変化させるために、DA宿主自体のリンパ球がほぼ致死性の全身照射(700−850rads)によって破壊されたadoptive transfer実験を行った。本モデルでは、異所性PVG心臓移植片は全身照射宿主によっては拒絶されないが、非照射宿主では、最初に設定された拒絶時間である6−9日で生じる。CD4+T細胞の濃縮集団による照射宿主の回復により、拒絶反応が回復されるが、正常な最初の設定時間には回復しない。これらの細胞と用量依存性ではなく、500万個のCD4+T細胞が2000万または1億個のCD4+T細胞と同等に有効である。平均の拒絶時間は、これらの細胞で12日未満である。
照射したadoptive宿主における拒絶時間に対する回復接種におけるナイーブCD25−、CD4+T細胞に対するCD25+、CD4+T細胞の比を変化させることの効果を試験し、その結果のグラフが図3Bに示されている。
インビボでCD4+、CD25+T細胞のCD4+、CD25−T細胞との均衡を変化させることにより、耐性をもたらすことができ、照射宿主における拒絶を回復するために使用されるナイーブ細胞による実験によって、以下が明らかにされる。
a)ナイーブ非分画CD4+T細胞は用量応答効果を示すことはなく、90%を超えるCD4+、CD25−T細胞に対する1−10%のCD4+、CD25+T細胞の恒常性の均衡があることが示唆される。
b)1−10%のCD4+、CD25+T細胞の除去により、CD4+、CD25−T細胞が迅速な拒絶をもたらすことが可能となり、ナイーブCD4+T細胞におけるその自然な混合集団におけるナイーブCD4+、CD25+T細胞が拒絶を抑えることが確認される。
c)CD4+、CD25+T細胞単独では拒絶をもたらすことがなく、ナイーブCD4+、CD25+T細胞のナイーブCD4+、CD25−T細胞との比が増大すると拒絶を阻害しうるが、その比が1:1の場合にほぼ完全な拒絶の抑制を示す。
d)特異的ドナー抗原およびIL−2を用いてMLCにおいて増大させたナイーブCD4+、CD25+T細胞は、1:1の比で抑制する能力を保持し、拒絶をもたらす細胞に戻ることはない。
本試験では、IL−2またはIL−4の存在下での抗原との接触後のCD4+、CD25+T細胞の増殖を試験した。
図4Aは、刺激細胞なし、すなわち抗原なしの場合(第1の列)、自己刺激細胞、すなわちDAラットに由来する抗原を提示する抗原提示細胞を用いた場合(中央の列)、または同種PVG刺激細胞、すなわちPVGラットに由来する抗原を提示する抗原提示細胞を用い場合の、DAラットに由来するナイーブCD4+、CD25+T細胞の培養の結果を示す。
図4Bは、同種PVG刺激細胞を用いて培養した、DAラットに由来するナイーブCD4+、CD25+T細胞の増殖の時間経過を示す。サイトカインの添加なしに培養された細胞は、図1Aに記載されている通り3日目に増殖のピークを示した。バックグラウンドでのカウント(150ccpm未満の蒸留水カウントと同等)により5日目および6日目で検出可能な増殖はなかった。IL−2は、すべての日にちで顕著な増殖を誘発し、5日目にピークを示した。IL−4は、すべての日にちで増殖を増強し、この増殖は4日目でピークを示した。培養の3−4日後のこれら細胞の表現型のプロフィールは、CD8およびCD4を発現する二重陽性集団の出現があり(細胞の10−35%)、CD25を発現し続けることを示す。最初の細胞は1%未満のCD4+、CD8+細胞、および2%未満のCD8+T細胞であった。
図4Cは、IL−2およびIL−4で培養されたCD4+、CD25+T細胞によるサイトカインおよびサイトカイン受容体のmRNA発現の半定量RT−PCRアッセイの結果を示す。
図4Dは、培養された耐性CD4+、CD25+T細胞におけるサイトカインおよびサイトカイン受容体誘発を示す。これにより、特異的ドナー、第三者刺激因子または自己刺激因子によって刺激された耐性細胞を比較した。培養物はサイトカインが補充されず(CTLの上の3つの系)またはIL−2、IL−4、またはCHO−k上清が添加された。−&+CTLSは、アッセイそのものの対照として使用したConA活性化T細胞に由来するcDNAであった(−&+対照)。mRNAは、右から左へのcDNAの連続希釈による半定量RT−PCRを使用して分析された。
図4Eは、IL−5−Rα鎖がIL−4で培養されたCD4+、CD25+T細胞において誘発されることを示す。本試験では、DAラットに由来するナイーブCD4+、CD25+T細胞を4日間、自己DA刺激因子またはPVG刺激因子で培養した。IL−5Rα mRNAを細胞から収集したcDNAにおいて分析した。細胞をnilサイトカイン、IL−2、またはIL−4で培養した。IL−5Rα mRNAのみがIL−4で培養された細胞において検出され、自己抗原で培養されたものよりも同種抗原で培養されたものでレベルが大きかった。対象は非培養CD4+、CD25+T細胞、ConA活性化リンパ球に由来するcDNA、およびIL−5Rαを有する既知のcDNAであった。
図4Fは、IL−2の存在下で抗原との接触させたCD4+、CD25+T細胞でのIFN−γ受容体(IFNGR)のリアルタイムRT−PCRを示す。
IL−2およびIl−4は、培養においてCD4+、CD25+T細胞に対する異なる効果を有し、その効果は、以下を含む。
a)両方は自己および同種抗原に対する増殖を刺激するが、IL−4に対する応答は、IL−2によるよりも早くピークを示し、かつ漸減する。
b)IL−2による培養は、CD4+、CD8+二重陽性CD25+T細胞の発達をもたらす。二重陽性細胞のこの発達は、おそらく、CD8を再発現するCD4+、CD25+T細胞の脱分化である。
c)IL−2またはIL−4による培養ではいずれもIL−2 mRNAの検出可能発現が誘発されないが、IL−2およびIL−4はIL−4、IL−5、IL−12p40、IFN−γ、iNOSのmRNAを誘発する。IL−10、IL−13、およびTGF−βの検出可能な特異的誘発は存在しないように思われる。
d)新しいサイトカイン受容体の遅延発現があり、IL−2および抗原で培養されるとIFN−γRが出現する。IL−5RはIL−4により出現する。IL−12Rβ2は、両方の刺激因子により出現するが、おそらく、IL−4よりもIL−2により多く出現する。
e)IL−2またはIL−4で培養された耐性細胞によるサイトカインmRNA出現のパターンは、ナイーブCD4+、CD25+T細胞によるものと同様であり、これと一致して、CD4+、CD25+T細胞のポリクローナル活性化であり、同種抗原特異的CD4+、CD25+T細胞の活性化ではない。
f)IL−2はIFN−γ/IL−12p70依存抑制細胞を誘発し、本発明者らは、名称をTs1とすることを提案する。
g)IL−4はIL−5依存抑制細胞を誘発し、本発明者らは名称をTs2とすることを望む。
本試験ではCD4+、CD25−T細胞に対するナイーブCD4+、CD25+T細胞の効果を試験した。
図5Aは、DAラットに由来するCD4+、CD25+T細胞、CD4+、CD25−T細胞、および混合CD4+、CD25−T細胞およびCD4+、CD25−T細胞のDAまたはPVG抗原との接触後のGAPDH、IL−2、IFN−γ、IL−4、IL−5、IL−10、TGF−βおよびiNOSmRNAの発現のRT−PCR分析の結果を示す。これらの培養から抽出されたmRNAはわずかであるので、本アッセイでは、cDNAの最大濃度で2つの試料を試験した。自己(DA)刺激因子で培養されたCD4+、CD25+T細胞のみが最小のサイトカイン誘発を示す。PVG刺激因子により、IL−2 mRNAは認められないが、一部のIFN−γ、IL−4、IL−10およびTGF−β発現が認められる。対照的に、CD4+、CD25−T細胞は、PVGによって刺激されると、試験したすべてのサイトカインmRNAの顕著な誘発が認められ、このプロフィールはDA抗原で刺激し場合に確認されたものと同様である。しかし、自己刺激因子により、IL−5 mRNA誘発は認められない。注目すべきは、CD4+、CD25−T細胞は、自己MLCとして周知である自己に対する顕著な増殖性応答を有し、したがって、自己に応答するCD4+、CD25−T細胞におけるサイトカイン mRNAの顕著な誘発は、顕著な自己増殖性応答と一致する。
図5Bは、MLCにおけるCD4+、CD25−T細胞増殖についてのCD4+、CD25+T細胞の阻害効果に対する、IL−5、TGF−β、IL−10およびiNOSを妨害する効果を示すグラフである。
MLCにおけるナイーブCD4+、CD25−T細胞に対するナイーブCD4+、CD25+T細胞の抑制効果は、以下のものと関係がある。
a)ナイーブCD4+、CD25+T細胞は、ナイーブCD4+、CD25−T細胞と1:1で混合されると、MLC増殖を完全に抑制する。
b)MLCにおけるCD4+、CD25−T細胞のみの誘発と比較した、IL−2、IL−4、IL−5 mRNA発現の誘発における顕著な削減、およびIFN−γおよびTGF−γ mRNAの誘発の小さな削減、ただし、iNOSの削減は見られない。
c)抗−IL−5または抗−TGF−βまたは抗−IL−10モノクローナル抗体による妨害が、抑制を部分的に妨害し、IL−5、IL−10およびTGF−βが、MLCにおけるCD4+、CD25−T細胞の増殖に対するナイーブCD4+、CD25+T細胞による抑制の媒介に効果を有する。
PVG同種移植片に対する耐性を有するDAラットに由来するCD4+T細胞の培養における生存に対するIL−4またはIL−5の効果を試験した。耐性媒介細胞の生存を、PVG心臓同種移植片に特異的耐性をadoptive transferし、第三者ルイス同種移植片の拒絶をもたらす能力を保持する能力によって分析した。
図6Aは、PVG抗原とともに、かつCoA上清なしにMLCにおいてIL−4またはIL−5のみで培養された後のPVG心臓同種移植片に対して耐性のDAラットに由来するCD4+T細胞の増殖を示す。この実験は、IL−5のみが特異的抑制CD4+T細胞をトランスファーする耐性を維持することを明らかに示した。同じ細胞は、第三者移植片拒絶をもたらす能力を保持しており、このことから、IL−5のみでは、耐性媒介細胞を誘発しないことを明らかにした。IL−2(Transplantation 1993年、55、374−379頁)でもIL−4(上記のデータを参照)のみでも、特異抗原を有する培養において、これら抑制CD4+T細胞を維持しないが、すべてのこれらサイトカインを有するConA上清がCD4+T細胞の特異的抑制機能を維持する。
図6Bは、耐性CD4+、CD25+T細胞およびCD4+、CD25−T細胞の、異なる刺激細胞に曝露された場合にインビトロで増殖する能力に対する、IL−4および特異的ドナー抗原を用いたMLCにおける培養の効果の試験結果を示す。
IL−4ではなく、Th2サイトカインIL−5は、培養における特異的CD4+、CD25+T細胞耐性媒介抑制細胞の生存を促進し、それらが耐性をトランスファーする能力を保持することを可能にする。
a)MLCにおけるCD4+T細胞の同種抗原特異的耐性媒介細胞の生存は、IL−5またはCon A supによって支持されうる。IL−4の添加のみでは耐性媒介CD4+T細胞の生存は促進されない。
b)耐性CD4+、CD25+T細胞がMLCにおける特異的ドナーに対して増殖ができないことは、特異的ドナー同種抗原およびIL−4による前培養によって回復されない。
この試験ではMLCにおける活性化CD4+、CD25+T細胞の増殖に対するさまざまなサイトカインの効果を試験した。
図7では、PVG心臓同種移植片に対する耐性を有するDAラットに由来する非分画リンパ球のMLCにおける増殖に対する個々のサイトカインの効果を試験する。
耐性動物に由来する活性化CD4+、CD25+T細胞の生存を促進するサイトカイン(具体的にはIL−5、IFN−γ、およびIL−12)の添加により、MLCにおける特異的ドナー抗原に対する非分画末梢リンパ球の増殖が削減される。
本実験では、活性化CD4+、CD25+T細胞を投与することによってラットにおけるEANを治療する能力を試験した。
方法;EANを10−15週齢のメスラットにおいて、記載されている通り(J.Neurol.Sci.1994年、123:162−172頁)、フロイントの完全アジュバント中のウシ末梢神経ミエリンを免疫付与することによって誘発した。体重を計り、臨床所見、および半定量スコアを使用する麻痺の評価によって、疾患活性について動物を毎日モニタリングした。使用したスコアは、5+死亡または安楽死を必要とする全麻痺、4+全肢の麻痺、3+全後肢の麻痺、および虚弱前腕、2+虚弱後肢、1+虚弱尾、0正常であった。

図8Aおよび8Bにおいて、ルイスラットにおける実験的アレルギー性神経炎(EAN)の臨床経過に対する活性化CD4+、CD25+T細胞またはCD4+、CD25−T細胞のadoptive transferの効果を試験した。
急性自己免疫疾患から回復した動物に由来する特異的活性化CD4+、CD25+T細胞は、自己免疫疾患の重篤度を改善しうる。
細胞サブセットおよび培養は、混合リンパ球培養によるものとした。PNMを有する抗原提示細胞を、PNMおよび自己刺激細胞調製物を用いて37度で1時間、前培養することによって調製した。次いで、これらの細胞を洗浄した。培養条件および増殖の測定はMLCについて記載されている通りであった。
これらの試験は、自己抗原(この場合にはEANモデルにおけるPNM)への曝露前および曝露中に、自己抗原に対するCD4+、CD25+T細胞の応答の明確なパターンを明らかに示す。
図9Aについては、PNMを有するまたは有さない自己抗原提示細胞による培養において刺激した場合のナイーブルイスラットに由来するリンパ球の増殖を試験した。増殖は4日目と5日目で比較した。
図9Bについては、本試験では、T細胞サブセットを、免疫付与後30日にEAMから回復したルイスラットから調製した。さらに自己抗原提示細胞のみで刺激した培養物、またはPNMで感作した自己抗原提示細胞で刺激した培養物をセットアップし、増殖を4日目および5日目に分析した。非分画CD4+T細胞およびCD4+、CD25−T細胞の応答は、自己抗原提示細胞に対してよりもPNMに対して大きく、特異的感作と一致した。CD4+、CD25+T細胞のみは、ナイーブCD4+、CD25+T細胞で通常見られる以上の非特異的増殖を有した。これはCD4+、CD25+T細胞のポリクローナル活性化によるものとみられる。
ナイーブ動物に由来するCD4+、CD25+T細胞は、それらが同種免疫応答で作用するように、自己免疫応答において作用する。
a)ナイーブ動物では、自己刺激因子に対するものと同じであるという点で、自己抗原に対する特異的応答は認められない。
b)ナイーブ動物では、CD4+、CD25+T細胞の除去が自己刺激因子に対する応答を増強する。ナイーブCD4+、CD25−T細胞の増殖が非分画CD4+T細胞の増殖よりも大きい。
c)別のナイーブCD4+、CD25−T細胞の増殖が、CD4+、CD25+T細胞と、特に1:1の比で混合することによって抑制されうる。
d)自己免疫の急性エピソードから回復した動物では、CD4+T細胞およびCD4+、CD25−T細胞について、自己抗原と比べ特異抗原に対する応答の増強が認められるが、自己抗原に対するCD4+、CD25+T細胞による特異的応答は認められない。これらのCD4+、CD25+T細胞は、CD4+、CD25−T細胞の増殖を完全に抑制することはなく、特異的同種抗原に対してよりも自己応答に対してより有効である。すなわち、活性化特異的CD4+、CD25+T細胞はインビトロで抑制することはない。
本実施例は、ドナー抗原およびIL−2またはIL−4の存在下でインビトロで培養されるCD4+、CD25+T細胞のドナー心臓移植片拒絶を抑制する能力を示す。
これらの実験では、PVGラットに由来する抗原に活性化され、かつPVG細胞およびIFN−γの存在下で培養されたDAラットに由来するCD4+、CD25+T細胞が、DAラットにおけるPVG心臓移植片の拒絶を抑制することができるかを試験した。
図11は、ナイーブDAラットに由来する非分画CD4+T細胞、CD4+、CD25−T細胞サブセット、およびCD4+、CD25+T細胞サブセットについて、IL−2またはIL−12p70またはIL−2およびIL−12p70の両方を添加した培地において、自己抗原(DA抗原)または同種抗原(PVG抗原)の存在下で4日間T細胞を培養した後の増殖を比較するために行われた実験を示す。細胞増殖に対する抗原とサイトカインの組合せの効果は図11に示されている。非分画CD4+T細胞の増殖がグラフの上の列に示され、CD4+、CD25−T細胞サブセットの増殖がグラフの中央の列に示され、CD4+、CD25+T細胞サブセットの増殖がグラフの下の列に示されている。グラフの左手の列は自己抗原に対する応答の増殖を示し、グラフの右手の列は同種抗原(PVG)に対する応答の増殖を示す。
本実験では、Ts1細胞(抗原およびIL−2の存在下の培養によって活性化されたCD4+、CD25+T細胞)の成長に対するIL−2およびIL−12p70の効果を試験した。
ナイーブDAラットに由来する非分画CD4+T細胞、CD4+CD25+T細胞サブセット、およびCD4+CD25+T細胞サブセットの増殖を、培地にサイトカインの添加なし、IL−4またはIL−12p70またはIL−12およびIL−12p70の両方が添加された場合において、自己抗原(DA抗原)または同種抗原(PVG抗原)に対する4日間の培養後に比較した。増殖は、対照のnilサイトカインと比較した場合、IL−12p70のみによっては増強されなかった。IL−4はすべてのサブセットの顕著な増殖を誘発し、IL−12p70の添加は亜集団のいずれの増殖も増強することはなかった。これは、IL−12p70がTs2細胞の増殖を増強することがないことを示した。
RT−PCRを使用し、同種抗原(PVGラットに由来する細胞)または自己抗原(DAラットに由来する細胞)および100単位/ml超のIL−2またはIL−4による培養後のナイーブDAラットに由来するCD4+、CD25+T細胞におけるIL−2およびIL−12β2受容体mRNAの発現を分析した。RT−PCR分析の結果は図13に示されている。
フロイントの完全アジュバント中におけるウシ末梢神経ミエリン(PNM)による免疫付与によって10−15週齢のメスルイスラットにおいてEANを、J.Neurol.Sci.1994年、123:162−172頁に記載されている通り、誘発した。動物を3群に分けた。すなわち(a)PNMおよびフロイントのアジュバントのみ(対照)で免疫付与した群、(b)PNMおよびフロイントのアジュバントで免疫付与し、CHO細胞上清を投与した群(対照)、および(c)PNMおよびフロイントのアジュバントで免疫付与し、IL−5(5000単位/日を、10日間、臨床の発症日から毎日腹腔内注射)を投与した群。
実施例16に記載されている通り、PNMで免疫付与し、IL−5で処置した動物におけるサイトカインの発現を判定するために、流入領域リンパ節(LN)および馬尾(CE)に由来するmRNAのリアルタイムPCR分析を、PNMによる免疫付与後14および21日目に行った。リアルタイムPCRの結果が図16に示されている。オープンバーは、IL−5で処置されなかった対照ラットのmRNAレベルを表し、クローズドバーはIL−5で処置したラットのmRNAレベルを表す。測定したサイトカインは、図の上部に示されている。
実施例15に記載されている通り、PNMで免疫付与され、未処置(CTL)またはIL−5で処置されていたラットにおける流入領域リンパ節および馬尾でのIL−5、IL−5受容体、IL−4、およびIL−13の発現を、半定量RT−PCRを使用して試験した。半定量RT−PCRの結果は図17に示されている。
MLCにおけるナイーブCD4+、CD25−T細胞の増殖に対するナイーブおよび活性化CD4+、CD25+T細胞の効果を、CD4+、CD25+T細胞の1:2連続希釈を一定数のナイーブ(105)CD4+、CD25−T細胞と混合した限外希釈アッセイで試験した。限外希釈試験の結果は図18に示されている。
本実験は、IL−2またはIL−4の存在下でナイーブCD4+、CD25+T細胞の活性化後、IL−23およびIL−13の存在下でのCD4+、CD25+T細胞のインキュベーションの影響を判定するために行われた。
本実施例は、対象が特異抗原に対して耐性であるかを判定するCD4+、CD25+T細胞の使用を示す。

Claims (22)
- 対象が特異抗原に活性化されているCD4+、CD25+T細胞を含んでなるかどうかを評価する方法であって、
(a)前記特異抗原と、IL−5、IL−12、IL−23およびIFN−γからなる群から選択される少なくとも1つのサイトカインとの存在下で、前記対象から得たCD4+、CD25+T細胞を含んでなるリンパ球の試料の少なくとも一部分をインキュベートするステップと、
(b)その後に、特異抗原に活性化されているCD4+、CD25+T細胞が前記試料に存在するかどうかを判定するステップと
を含んでなる方法。 - 対象が特異抗原に耐性であるか、または耐性になることができるかを判定する方法であって、
(a)前記対象から得たCD4+、CD25+T細胞を含んでなるリンパ球の試料の少なくとも一部分を前記特異抗原と接触させるステップと、
(b)前記リンパ球の試料の少なくとも一部分を、IL−5、IL−12、IL−23およびIFN−γからなる群から選択される少なくとも1つのサイトカインの存在下でインキュベートするステップと、
(c)その後に、前記特異抗原に活性化されているCD4+、CD25+T細胞が前記試料に存在するかを判定するステップと
を含んでなる方法。 - (i) 前記試料における特異抗原に活性化されているCD4+、CD25+T細胞の存在がCD4+、CD25+T細胞の増殖を検出することによって判定され、または
(ii) 前記試料における前記特異抗原に活性化されているCD4+、CD25+T細胞の存在が活性化されたCD4+、CD25+T細胞の存在を示す分子を検出することによって判定され、または
(iii) 前記試料における特異抗原に活性化されているCD4+、CD25+T細胞の存在がCD4+、CD25−T細胞の増殖の阻害を検出することによって判定される、
請求項1または2に記載の方法。 - 前記リンパ球の試料の少なくとも一部分が前記特異抗原と接触され、かつIL−5、IL−12、IL−23およびIFN−γからなる群から選択される少なくとも1つのサイトカインの存在下で、さらに抗−IL−2および/または抗−IL−4抗体の存在下でインキュベートされ、かつ、前記試料における前記特異抗原に活性化されているCD4+、CD25+T細胞の存在が、CD4+、CD25+T細胞の非増殖または低増殖を検出することによって判定される、請求項1に記載の方法。
- 前記少なくとも1つのサイトカインが、前記リンパ球の試料の少なくとも一部分が外因性起源からインキュベートされる培地に添加される、請求項1から4のいずれか一項に記載の方法。
- 前記少なくとも1つのサイトカインが精製されたタンパク質である、請求項5に記載の方法。
- 対象が特異抗原に耐性であるか、または耐性になることができるかを評価する方法であって、前記対象が前記特異抗原に活性化されているCD4+、CD25+T細胞を含んでなるかどうかを請求項1に記載の方法によって評価するステップを含んでなる方法。
- 特異抗原に活性化されたCD4+、CD25+T細胞をインビトロで増殖させる方法であって、IL−5、IL−12、IL−23およびIFN−γからなる群から選択される少なくとも1つのサイトカインの存在下で前記特異抗原に活性化されたCD4+、CD25+T細胞を培養するステップを含んでなる方法。
- 前記特異抗原に活性化したCD4+、CD25+T細胞が、ナイーブCD4+、CD25+T細胞をインビトロで前記特異抗原と接触させ、かつ、CD4+、CD25+T細胞の活性化を支持することができる1つもしくは複数のサイトカインの存在下でCD4+、CD25+T細胞を培養することによって、インビトロで前記特異抗原に活性化される、請求項8に記載の方法。
- CD4+、CD25+T細胞の活性化を支持することができる前記1つもしくは複数のサイトカインが、IL−2およびIL−4からなる群から選択される、請求項9に記載の方法。
- 前記特異抗原に活性化したCD4+、CD25+T細胞がインビトロで培養する前にインビボで前記特異抗原に活性化されたものである、請求項8に記載の方法。
- ナイーブおよび活性化CD4+、CD25+T細胞の混合物が、ナイーブおよび活性化CD4+、CD25+T細胞をインビトロで前記特異抗原と接触させることによって、かつ、前記ナイーブおよび活性化CD4+、CD25+T細胞を、IL−2および/またはIL−4、ならびに、IL−5、IL−12、IL−23およびIFN−γからなる群から選択される少なくとも1つのサイトカインの存在下で培養することによって、増殖される、請求項8に記載の方法。
- 前記少なくとも1つのサイトカインが、CD4+、CD25+T細胞が培養される培地に外部から添加されている、請求項8に記載の方法。
- 前記特異抗原が、自己抗原、同種抗原、異種抗原、アレルゲン、腫瘍抗原および感染症病原体に由来する抗原からなる群から選択される、請求項1から13のいずれか一項に記載の方法。
- 活性化CD4+、CD25+T細胞および薬剤として許容される担体を含んでなる、特異抗原に対する耐性を増大させるための組成物であって、前記活性化CD4+、CD25+T細胞が、IL−5、IL−12、IL−23およびIFN−γからなる群から選択される少なくとも1つのサイトカインの存在下でインビトロで培養された組成物。
- (i)IL−5、IL−12、IL−23およびIFN−γからなる群から選択される1つもしくは複数のサイトカイン、ならびに(ii)IL−2および/またはIL−4を含んでなる、活性型CD4 + 、CD25 + T細胞を増殖させることによって特異抗原に対する耐性を増大させるためのキット。
- それを必要とする対象中の特異抗原に対する耐性を増大させるための薬剤の製造における、請求項8〜14のいずれか一項に記載の方法によってインビトロで増殖させた特異抗原に活性化したCD4+、CD25+T細胞の使用。
- 特異抗原に対する免疫応答に起因する疾患を治療または予防するための薬剤の製造における、請求項8〜14のいずれか一項に記載の方法によってインビトロで増殖させた、前記特異抗原に活性したCD4+、CD25+T細胞の使用。
- 対象の特異抗原に対する免疫応答に起因する疾患の治療のために前記対象における特異抗原に対する耐性を誘発する薬剤の製造における、IL−2およびIL−4からなる群から選択される1つもしくは複数のサイトカインの使用であって、前記治療が、薬剤の有効量、およびIL−5、IL−12、IL−23およびIFN−γからなる群から選択される少なくとも1つのサイトカインの有効量を投与するステップを含んでなる使用。
- 対象の特異抗原に対する免疫応答に起因する疾患の治療のために前記対象における特異抗原に対する耐性を誘発する薬剤の製造における、IL−5、IL−12、IL−23、IFN−γからなる群から選択される1つもしくは複数のサイトカインの使用であって、前記治療が、薬剤の有効量、およびIL−2およびIL−4からなる群から選択される少なくとも1つのサイトカインの有効量を投与するステップを含んでなる使用。
- 特異抗原に対する免疫応答に起因する疾患を治療または予防するための薬剤の製造における、IL−2およびIL−4からなる群から選択される1つもしくは複数のサイトカインの使用であって、前記治療が、薬剤の有効量、およびIL−5、IL−12、IL−23、IFN−γからなる群から選択される少なくとも1つのサイトカインの有効量を投与するステップを含んでなる使用。
- 特異抗原に対する免疫応答に起因する疾患を治療または予防するための薬剤の製造における、IL−5、IL−12、IL−23およびIFN−γからなる群から選択される1つもしくは複数のサイトカインの使用であって、前記治療が、薬剤の有効量、およびIL−2およびIL−4からなる群から選択される少なくとも1つのサイトカインの有効量を投与するステップを含んでなる使用。
