シャルコー・マリー・ツース (CMT) 末梢神経障害は、末梢神経系に影響を及ぼす疾患の遺伝的な混成群を表す。常染色体優性 CMT 1C 型 (CMT1C)。染色体16p13.1−p12.3 に対して遺伝的にマッピングした。染色体16p13.2 に対してマッピングする上皮 膜タンパク質 2 の遺伝子(EMP2)は、CMT1C の候補遺伝子である (Street V. A., 2002, Am J Hum Genet 70(1):244−250)。
本発明のタンパク質を用いて in−situ 発現パターンから取得した本発明に記載のタンパク質が、島細胞 (例えば、DP685; DP160; RA770)、膵臓の間充織 (RA770)、膵臓上皮細胞 (例えば、DP685; DP160)、膵管細胞 (DP160)、同様に、神経管 (DP160; DP444) に沿った神経節、体節 (DP444)、背側菱脳 (DP444)、肝臓 (DP685)、心臓 (DP685)、胃腸 (DP444) および腸内細胞 (DP685; DP444) などの他の細胞のような膵臓細胞において特異的に発現されることが結論できる。以上の点から、本発明の核酸およびタンパク質およびそのエフェクター/修飾因子は、診断目的および治療目的に関与する、例えば、限定するものではないが、上記臓器または組織に関連する糖尿病および肥満症のような代謝障害および機能障害、肝臓疾患および神経疾患、例えば神経変性障害および他の疾患および障害において有用である。従って、本発明のタンパク質は、診断マーカーとして、または小分子スクリーニングの標的として、および糖尿病および/または肥満症および代謝障害および神経変性疾患、心臓、肝臓、胃腸、または腸内障害のような他の疾患の 予防 または治療において有用であり得る。

ニワトリ DPd6 の cDNA ライブラリは、6 日目のニワトリ胚から採取した背側膵芽を使用して構築したものである。凍結組織はホモジナイズかつ溶解した。Brinkmann POLYTRON homogenizer PT−3000 (Brinkman Instruments, Westbury, N.J.)をグラニジウム イソチオシアネート溶液において使用した。溶解物に遠心力 5.7 M CsCl 以上をかける。Beckman SW28 ローター、Beckman L8−70M 超遠心分離機 (Beckman Instruments、Fullerton、Calif.)使用。 18 時間、25,000 rpm、室温。RNA を、4.7 pH 酸フェノールで抽出し、0.3 M 酢酸ナトリウムと容積 2.5 のエタノール沈殿させ、無 RNA 水で再懸濁し、37℃ にてDNAe 処理した。以前の通り RNA を4.7 pH 酸フェノールで繰返し抽出し、酢酸ナトリウムおよびエタノールで沈殿させた。 その後、Micro−FastTrack 2.0 mRNA 分離キット(Invitrogen、Groningen、Netherlands)を使用して mRNA を単離し、cDNA ライブラリを構築するために使用した。mRNAs の取り扱いは、SUPERSCRIPT cDNA synthesis and plasmid cloning system (Gibco/BRL) の推奨プロトコルに従って行なった。DH10B 宿主細胞への形質転換に続いて、単一コロニーを摘出し、PCR 法を用いてクローン化された cDNA インサートを増幅した。単一 cDNA インサートを代表する PCR 法によって増幅された断片を、その後に生体外転写し、Digoxygenin 標識 RNA プローブ(Roche)を生成した。RNA プローブをホールマウント in−situ スクリーニングにおいて使用して初期のニワトリ胚におけるそれぞれの遺伝子産物の発現を確定した。本発明のタンパク質をコードする遺伝子を含有するプラスミドを、膵臓組織におけるその高発現のおかげで同定した。
当業者であれば周知の標準プロトコルおよび以前に説明した(例えば、Pelton, R.W. らの(1990) Development 110, 609−620、Belo, J.A. らの(1997) Mech. Dev. 68, 45−57) に従って in−situ ハイブリッド形成法を実行した。
REAL プレP 96 ウェルのプラスミド分離キット (QIAGEN) を用いて、プラスミドDNA を細胞から開放し精製した。このキットは、多重試薬ディスペンサーを使用した96 ウェル ブロックにおいて、96 個のサンプルの同時精製を可能にした。 下記の変更を除いてメーカーの推奨するプロトコルを使用した。変更(i): 細菌を 1 ml の消毒した Terrific Broth (LIFE TECHNOLOGIESTM, Gaithersburg, Md, USA)でカルベニシリンを 25 mg/L で、グリセロームを 0.4 % で用いて培養した。変更(ii): 接種後、培養を 19 時間インキュベートした後、細胞を 0.3 ml 溶解バッファで溶解した。変更(iii): イソプロパノール沈殿に続いて、プラスミドDNA ペレットを 0.1 ml の蒸留水で再懸濁した。プロトコルの最後のステップの後、サンプルを 4℃ にて保管する 96 ウェル ブロックへ移した。cDNA を GATC Biotech AG (Konstanz、Germany)によって、当業者であれば既知の標準プロトコルに従って配列せしめた。
リーディング・フレームを決定した後に、本発明のヌクレオチド配列から演繹したアミノ酸配列のみならず、本発明のヌクレオチド配列も、GenBank、SwissProt、BLOCKS、および Pima II のようなデータベースに対する問い合わせ配列として使用した。以前に同定され注釈された配列を含むこれらのデータベースで、相同性(類似性) の領域を検索した。使用したのはBLAST(Basic Local Alignment Search Toolの略語、Altschul S.F. (1993) J. Mol. Evol. 36:290−300; Altschul、S.F. らの(1990) J. Mol. Biol. 215:403−10)である。BLAST 検索により、ヌクレオチドおよびアミノ酸配列両方のアライメントを産生し、配列類似性を決定した。ローカル特性をもつアライメントのおかげで、BLAST 検索は、完全なマッチの決定において、または相同体真核(細菌)または原核(動物、カビ、植物) の起源の識別に特に有用である。ここに引用として取り込まれたSmith らの(1992、Protein Engineering 5:35−51)において説明されたような他のアルゴリズムは、一次配列パターンおよび二次構造ギャップのペナルティーを取り扱う場合に使用された。BLAST 検索の方法は、Karlin らの(前出)で詳細に説明したように、ここに引用として取り込まれ、問い合わせ配列とデータベース配列との間のマッチを検索した。BLAST 検索によって見出された任意のマッチの統計的な重要性が評価され、重要性をもつユーザ選択限界を満足するマッチのみが報告された。本出願において、限界値はヌクレオチドに対して 10〜25、およびペプチドに対して 10−14 と設定した。ヌクレオチド配列は、GenBank データベースの霊長類、げっ歯類、および他の哺乳動物の配列に対して検索し、それから同一クローンからの演繹アミノ酸配列を、GenBank の機能的タンパク質データベースの哺乳動物、脊椎、真核生物の相同性に対して検索した。
本発明のタンパク質をコードする完全長核酸配列を用いて、部分的ヌクレオチド配列を完全長に拡張するため、またはゲノム ライブラリから 5' または 3' イントロンまたは他の調節配列を取得するために、オリゴヌクレオチドプライマーを設計する。単一プライマーを合成してアンチセンス方向における伸長を開始し、他を合成してセンス方向における配列を伸長する。プライマーを用い、領域目的に対して新規ヌクレオチド配列、未知ヌクレオチド配列を含む既知の配列「outward」生成 amplicons の伸長を促進する。初期プライマーは、オリゴ 4.06 プライマー分析ソフトウェア(National Biosciences)、または別の適切なプログラムを用いて、ヌクレオチド長が 22〜30 となるように、GC 含有率が 50% 以上を有するように、および温度約 68℃ 〜 72℃ で標的配列にアニールするように cDNA から設計した。ヘアピン二量化をもたらすようなヌクレオチドは避けた。元の選択したcDNA ライブラリ、またはヒトゲノム ライブラリを用い、配列を伸長した。後者(ヒトゲノム ライブラリ)は5' 上流領域を取得するために最も有用である。より多くの伸長または所望の必要な場合は、追加プライマーセットをデザインして既知領域を伸長する。XL−PCR キット(Perkin Elmer) の指示に従い、充分に酵素および反応混合液を攪拌することによって、高い忠実度増幅を得た。40 pmol の各プライマーおよび推奨濃度の他の全ての組成物キットの準備を発端として、Peltier thermal cycler (Peltier thermal cycler (PTC200; M.J. Research、Watertown、Mass.)および下記のパラメータを用いて、PCR 法を実行した。

本発明に記載の核酸に由来するハイブリダイゼーションプローブを用いて、cDNA、ゲノム DNA、または mRNA をスクリーニングした。約 20 塩基対から成るオリゴヌクレオチドの標識について特に記載するが、より大きなcDNA 断片に対しても実質的には同手順を用いる。オリゴヌクレオチドを、OLIGO4.06 プライマー分析ソフトウェア(National Bioscience)のような最新式のソフトウェアを用いてデザインし、50 pmol の各オリゴマーおよび 250 マイクロ Ci の [.gamma.−32 P] アデノシン三リン酸(Amersham) および T4 ポリヌクレオチドキナーゼ (DuPont Nen(r), Boston, Mass.)とを組み合わせることにより標識する。標識したオリゴヌクレオチドは、SEPHADEX G−25 超細繊分子樹脂カラム(Pharmacia & Upjohn)を用いて十分に精製する。毎分 107 カウントの各センス オリゴヌクレオチドおよびアンチセンス オリゴヌクレオチドを含む部分を、(Ase I, Bgl II, EcoRI, Pst I, Xba I, or Pvu II; DuPont NEN(r)) の 1 つの膜で消化されたヒトゲノム DNA の典型的膜ベースのハイブリダイゼーション解析において用いる。各消化物から得た DNA は、0.7% アガロースゲル上で分画してナイロン膜(NYTRAN PLUS membrane, Schleicher & Schuell, Durham, N.H.)に移す。スクリーニングハイブリダイゼーションは40℃にて 16 時間かけて行う。非特異的シグナルを取り除くため、最大 0.1 x クエン酸食塩水(SSC)および 0.5% ドデシル硫酸ナトリウムの一層ストリンジェントな条件の下、ブロットを順次室温にて洗浄する。XOMAI AR オートラジオグラフィーフィルム (Kodak Rochester、N.Y.)をブロットに晒した後、またはブロットを PHOSPHOIMAGER (Molecular Dynamics、Sunnyvale、Calif.)に数時間置いた後にオートラジオグラフィーパターンを肉眼で比較する。
本発明のタンパク質をコードする配列、またはその一部に対するアンチセンス分子を用い、生体内または試験管内の天然の本発明のタンパク質の発現を抑制する。約 20 塩基対を含むアンチセンスオリゴヌクレオチドの使用は、特に記載するが、より大きなcDNA 断片に使用される手順と本質的に同一である。オリゴヌクレオチドを用いて、天然の本発明のタンパク質の発現を抑制する。アンチセンスオリゴヌクレオチドは多数の方法で遺伝子機能を抑制することができる。アンチセンスオリゴヌクレオチドは、転写物の 5'UTR に結合して、翻訳をブロッキングすることができる。あるいは、アンチセンスオリゴヌクレオチドの結合は、RNAseH による転写物の切断を誘発することもできる。また、アンチセンスオリゴが、プレ−mRNA のスプライシングをブロッキングすることも証明されており、その結果、特異的なスプライス形態の形成がブロッキングされるか、またはスプライシングされていない、成熟タンパク質を作ることができない不安定なメッセージの蓄積につながるかのいずれか、またはその両方が起こり得る。特定のアンチセンスオリゴヌクレオチドの作用機構は、オリゴヌクレオチドの化学組成および/または 標的転写物内の結合部位によって決定される。
本発明のタンパク質および相同タンパク質のような本発明のタンパク質の発現は、cDNA の適切なベクターへのサブクローニングおよびベクターの宿主細胞への形質転換によって達成する。この場合、以前に cDNA ライブラリを作成するために使用したクローンニング ベクター、PSPORT 1 を用いて、大腸菌において本発明のタンパク質を発現させる。クローンニング部位の上流では、このベクターにはプロモーター βガラクトシダーゼ、継いで、アミノ末端 Met を含有する配列、およびその結果生じた βガラクトシダーゼの 7 残基が含まれる。これらの 8 残基の直後に続くのは、転写に有用なバクテリオファージ プロモーター、および多数のユニークな制限部位を含むリンカーである。標準方法を使用するIPTG を有する単離形質転換細菌株の誘発は、β−ガラクトシダーゼの最初の8 つの残基から成る融合タンパク質、約 5〜15 のリンカー残基、および全長タンパク質を産生する。シグナル残基は、活性化のため下記のアッセイで直接使用することができる細菌の成長培地中に本発明のタンパク質の分泌物を誘導する。
PAGE 電気泳動法(前出の Sambrook)または他の精製技術を用いて十分に精製した本発明のタンパク質を用いて、標準プロトコルでウサギを免疫化して抗体を産生する。DNASTARアミノ酸配列をDNASTARソフトウェア(DNASTAR 社)を用いて解析して免疫原性の高い領域を決定し、対応するオリゴポリペプチドを合成してこれを用いて当業者に周知の方法で抗体を産生する。C末端に近傍するエピトープ、または親水性領域内のエピトープなどの適切なエピトープの選択については前出のAusubel らによって説明されている。
相同タンパク質および核酸分子コーディングは、それゆえ昆虫または各種の脊椎動物、例えば、哺乳動物または鳥から入手可能である。配列に対して相同性を持つニワトリタンパク質および核酸分子は、公的に利用できる全米バイオテクノロジー情報センター(NCBI) の非重複タンパク質データベースのプログラムBLASTP 2.2.3を用いて同定した(Altschulらの、1997、Nucleic Acids Res. 25:3389−3402を参照)。
Claims (1)
- RA770ポリペプチドと結合標的/薬剤との相互作用を調整する薬剤をスクリーニングする方法において、
(a) 下記
(aa) RA770ポリペプチド;
(ab) 前記RA770ポリペプチドの結合標的/薬剤、その際、該結合標的/薬剤が、膵臓細胞に関連する多成分受容体であり、該多成分受容体がRETチロシンキナーゼ及びグリコシル・ホスファチジルイノシトール・アンカードコレセプターである;および
(ac) 候補薬剤
を含む混合物を、基準親和性にて前記RA770ポリペプチドが特異的に前記結合標的/薬剤に結合する条件下でインキュベートするステップ;
(b) (候補)薬剤偏向親和性を決定するため、前記RA770ポリペプチドの前記結合標的に対する結合親和性を検出するステップ;および
(c) (候補)薬剤偏向親和性と基準親和性間の差異を確定するステップ
を含む、前記方法。
