(1).変性(≒94℃)を2から10秒
(2).アニール(54〜60℃)を5秒
(3).伸長(≒72℃)を2から10秒
(1).注入ウェル17から排出ウェル18に反応液を連続的又は断続的に流しながら、チャネル19内の反応液の全体をPCTサイクルとなるように温度制御する。
(2).反応液をチャネル19内に注入し、チャネル19内に停止させた状態で、反応液の全体をPCTサイクルとなるように温度制御する。
(3).電極対22をいくつかに分割し、それぞれの電極対22にPCRサイクルの各ステップに対応する交流電圧を常に印加しておき、注入ウェル17から排出ウェル18に反応液を連続的又は断続的に流す。
各構成要素の材質は上述のものに限られず、一般的なマイクロチャネルに用いられる材質のものを採用することができる。

Claims (6)
- ポリメラ−ゼ連鎖反応(PCR)を行う容器の内面に、反応液の流れに沿うギャップを挟んで対向して配置される電極対を設け、該電極対に交流電圧を印加して反応液に交流電流を流すことによってジュール熱を発生させて前記反応液の温度を制御することを特徴とするPCR方法。
- ポリメラ−ゼ連鎖反応(PCR)を行う容器と、
該容器の内面に、反応液の流れに沿うギャップを挟んで対向して配置される電極対と、
該電極対に交流電圧を印加して反応液に交流電流を流すことによってジュール熱を発生させて前記反応液の温度を制御する制御手段と
を備えることを特徴とするPCR装置。 - 前記容器は、管状のチャネルであることを特徴とする請求項2記載のPCR装置。
- 前記チャネルは、鉛直方向を向いていることを特徴とする請求項3記載のPCR装置。
- 前記容器は、鉛直方向に立っている平たいチャンバーであり、前記電極対は、該チャンバーの一方の側壁内面に鉛直方向のギャップを挟んで対向して配置されていることを特徴とする請求項2記載のPCR装置。
- 前記容器は、鉛直方向に立っている平たいチャンバーであり、前記電極対は、該チャンバーの一方の側壁内面と他方の側壁内面に、チャンバーをギャップとして挟み、対向して配置されていることを特徴とする請求項2記載のPCR装置。
