ヒト単球を、Tat誘導体(Nani−P2)、Toll様受容体(TLR)の免疫活性化配列(ISS)(図1)、又はリポポリサッカライド(LPS)(図2)とともに24−28時間培養し、細胞をその後洗浄し、蛍光ラベルしたCD86で染色した。Tat誘導体は、ISS(TLR)又はLPSに比べて高度にCD86の発現を促進した。
(マテリアル及び方法)
10匹のBALB/cマウスの4グループにおいて、マウスの乳房脂肪体に、1×104の4T1細胞をSCで移植した。10日目、腫瘍の直径が4−5mmに到達したときに治療を開始した。コントロールマウスに3日間隔にPBSをIV注射した。代わりに治療マウスには10日サイクルを繰り返して3日おきに3回投薬した。腫瘍負荷(腫瘍サイズmm3)を、標準公式を用いて算出した。CY:(シクロホスファミドのみ)マウスには、10日目から始めてマウスごとに80mg/kgで週1回IP注射した。Cy/Nani−P2:(最初にシクロホスファミド、次にNani−P2)マウスには、最初に、10日目から始めて3日間隔の3回の投薬でシクロホスファミド(80mg/kg)をIP注射し、その後、順番で3日間隔の3回の投薬でNani−P2(40ng)をIV注射した。CYの3回投薬その後Nani−P2の3回投薬のサイクルは、50日目まで繰り返された。Nani−P2/CY:(最初にNani−P2、次にシクロホスファミド)マウスには、10日目から始めて3日間隔の3回の投薬でNani−P2(40ng)をIV注射し、その後、順番で3日間隔でシクロホスファミドをIP注射した。Nani−P2の3回投薬その後CYの3回投薬のサイクルは、50日目まで繰り返された。

Claims (18)
- 配列番号2又は配列番号3と少なくとも85%同一であるアミノ酸配列を有するヒト免疫不全ウイルス(HIV)転写のトランス活性化因子(Tat)タンパク質を含有する、腫瘍を治療するための医薬組成物。
- 前記Tatタンパク質は、配列番号3のアミノ酸配列を有する、請求項1に記載の医薬組成物。
- 配列番号2又は配列番号3と少なくとも85%同一であるアミノ酸配列を有するTat誘導体ポリペプチドを含有し、
必要に応じて対象に治療上有効量投与され、前記対象において腫瘍の増殖の停止又は腫瘍の退縮を引き起こすように用いられる、
ことを特徴とする腫瘍を治療するための組成物。 - 前記Tat誘導体ポリペプチドは、複数回投与される、
ことを特徴とする請求項3に記載の組成物。 - 前記組成物は、反復投与サイクルにおいて投与され、
各々のサイクルは、後に休薬期間が続く規定された期間において、前記Tat誘導体ポリペプチドを複数回投与することを含み、
前記サイクルは、複数回繰り返される、
ことを特徴とする請求項3に記載の組成物。 - 前記組成物は、反復投与サイクルにおいて投与され、
各々のサイクルは、規定された期間における治療剤の1回又は複数回の投与が後に続く、規定された期間において、前記Tat誘導体ポリペプチドを複数回投与することを含み、

ことを特徴とする請求項3に記載の組成物。 - 前記治療剤は、シクロホスファミドである、

ことを特徴とする請求項3に記載の組成物。 - 前記Tat誘導体ポリペプチドは、配列番号3のアミノ酸配列と少なくとも85%同一であるアミノ酸配列を有する、
ことを特徴とする請求項3に記載の組成物。 - 配列番号2又は配列番号3と少なくとも85%同一であるアミノ酸配列を有するTat誘導体ポリペプチドを含有し、
必要に応じて対象に治療上有効量投与され、前記対象において腫瘍の退縮を引き起こすように用いられる、
ことを特徴とする腫瘍負荷を低減させるための組成物。 - 前記Tat誘導体ポリペプチドは、複数回投与される、
ことを特徴とする請求項11に記載の組成物。 - 前記組成物は、反復投与サイクルにおいて投与され、
