Description

本発明は、死亡率を予測する方法に関し、特に、テロメアの長さを決定し、テロメアの長さを生存、死亡の危険性、および心疾患、腫瘍性疾患、および感染症のような年齢関連病に対する罹患性と相関させる方法に関する。さらに、本発明は、そのような年齢関連病を発症する危険のある個人または個人の群を同定するための該方法の使用に関する。

本発明は、真核生物の生存または死亡の危険性を予測するための方法を提供する。具体的には、該方法は、テロメアの長さを決定し、テロメアの長さを集団におけるテロメアの長さと関連した死亡の危険性と相関させることに関する。さらに、本発明は、テロメアの長さを決定し、該長さを、心疾患(例えば、高血圧症、心筋梗塞症等)、腫瘍性疾患(例えば、悪性腫瘍、肉腫等)、神経変性障害、および病原菌に対する罹患性の増加のような、年齢と関連した病気の発症と相関させる方法を提供する。さらなる態様において、本発明は、経時的年齢と対照的に、生物学的年齢を決定して、生物学的年齢を、病気罹患性および死亡の危険性と相関させるためのマーカーを提供する方法を提供する。
平均のテロメア長の決定
ゲノムDNAを、標準的な手順によって血液試料から抽出した。テロメア測定に対する定量PCR対サザーンブロットアプローチを比較するのに使用した試料は、ヒト遺伝子連鎖マップを作成するために世界中で使用されるCentre pour les Etudes du Polymorphisme Humaine(CEPH)コレクションの一員であるユタ家族からの21人の関連のない個人(年齢が61ないし94歳の11人の女性および10人の男性)によって提供された(White, R. et al. (1985) Nature 313:101-105)。精製されたDNA試料を、96−ウェルマイクロタイターソースプレートにおいて、10mM Tris-HCl, 0.1mM EDTA, pH 7.5中の約1.75ng/ul(最終容量300ul/ウェル)まで希釈し、熱サイクラーにおいて5分間、95℃まで加熱し、氷水漕への移動によって5分間素速く冷やし、700xgにて簡潔に遠心分離し、粘着性アルミホイルで密閉し、アッセイの時間まで4℃にて保存した。
ヒトにおけるテロメアの長さおよび死亡
143人のリサーチ対象は、60−97歳の無関連のユタ州住民であり、彼らは、ヒト遺伝子連鎖マップ(White R. et al, Nature 313: 101-105 (1985))を作るのに使用される細胞系統のCEPH(Centre d'Etude du Polymorphisme Humain)コレクションに提供するために、1982年ないし1986年に血液を提供し、追跡生存データが入手可能であった人々である。誕生日および死亡日を、Utah Population Database (UPDB)、およびSocial Security Death Indexから得た。2002年半ばまでに、101の既知の死亡があった。残りの42人の対象に関して、彼らが生存していたと最後に分かる日付を、採血後に確立した。(International Classification of Diseases, 第九および第十改訂版にコードされたユタ州死亡診断書からの)死因による生存分析を、UPDB識別番号を有する124の個人に制限した。この研究は、ユタ大学のInstitutional Review Boardによって承認された。
Claims (12)
- 生物の死亡の危険性を予測する方法であって:
a)該生物のテロメアの長さを決定し;
b)該生物と60歳以上の年齢を適合させた集団において、二分特徴処理を行い、テロメアの長さと死亡の危険性とを相関させ;次いで
c)該生物のテロメアの長さが、該生物と年齢を適合させた集団において、より短い場合、該生物は増大した死亡の危険性を有する生物であると予測する方法。 - 生物の死亡の危険性を予測する方法であって:
a)該生物のテロメアの長さを決定し;
b)該生物と60歳以上の年齢を適合させた集団において、テロメアの長さを分布の下位半数対上位半数または下位25%対上位75%の二分特徴として処理し、テロメアの長さと、テロメアの長さ分布の下位からの個人対該分布の上位の個人に対する死亡率の割合である死亡の危険性と相関させ;次いで
c)該生物のテロメアの長さが、該生物と年齢を適合させた集団において、より短い場合、該生物は増大した死亡の危険性を有する生物であると予測する方法。 - 該生物がヒトである請求項1または2記載の方法。
- テロメアの長さが平均的なテロメアの長さである請求項1または2記載の方法。
