試料(20μL)にエタノールを加え50〜100倍希釈した。これらをフィルター濾過し、分析試料とした。p−クマル酸(4.8mg)、バッカリン(5.57mg)、ドルパニン(3.98mg)、アルテピリンC(4.85mg)をメスフラスコ(25mL容)に秤量し、エタノールでメスアップした。これを順次5倍希釈し、検量線作成用の標準溶液とした。得られた結果を用いて、絶対検量法より、プロポリス抽出物中の上記有機酸及び桂皮酸誘導体の合計含有量を測定した。 Ethanol was added to the sample (20 μL) and diluted 50 to 100 times. (試験例2:プロポリス抽出物の冷却処理後の澄明性の評価)
(A)試験例1において得られたプロポリス抽出物について、4℃の環境下で一晩(16時間)放置した後、ブフナーロートに濾紙(アドバンテック社製のNo.2)をセットして、吸引濾過を行った。試験試料として、実施例1,3,4、参考例2及び比較例1〜8のプロポリス抽出物を使用した。試験例1と同様の評価基準で、濾過性及び澄明性について評価した。尚、濾過装置は、保存環境温度に調整後に使用した。評価結果を表2に示す。

また、各例のプロポリス抽出物は、−20℃の環境下で一晩(16時間)放置した後は、いずれの試料も白濁が生じていた。表2に示されるように、実施例1の試験試料は、零下保存後に簡易な濾過処理によって澄明性の高い抽出液が得られることが確認された。一方、各比較例においては、濾過処理しても澄明性の高い抽出液を得ることができなかった。実施例1のスクワレンを用いたプロポリス抽出物は、零下保存の環境下においても優れた外観特性が得られることが期待される。また、実施例1において、濾過処理後においても、有機酸及び桂皮酸誘導体の含有量が、表1に示される常温保存後のプロポリス抽出物と比較して低下することはなかった。 Moreover, after leaving the propolis extract of each example in the environment of -20 degreeC overnight (16 hours), all samples had white turbidity. 上記のように得られた実施例1,3,4及び参考例2のプロポリス抽出物について、抗酸化活性について測定した。実施例1は−20℃保存及び濾過処理後のプロポリス抽出物を使用し、実施例3,4及び参考例2は、4℃保存及び濾過処理後のプロポリス抽出物を使用した。比較対照として、抽出処理前のスクワレン、しそ油、ドコサヘキサエン酸、及びエイコサペンタエン酸の各油性成分を用いた。ブランクとして蒸留水を使用した。
