卒中は米国において年間600,000名以上が罹患すると推定されている。1999年の報告では、全死亡数278,000名のうち167,000名以上が卒中で死亡した。1998年には、ショートステイ病院から退院したメディケア受給者だけで36億が支払われ、これには卒中により毎日の生活の活動に機能的制限または困難を有すると報告されている1,000,000名以上に対する長期医療は含まれていない(Heart and Stroke Statistical update, American Heart Association, 2002(非特許文献1))。ニューロンの死からの直接保護によって卒中による脳損傷を軽減する治療法は承認されていない。

I. 定義
「キメラペプチド」とは、融合タンパク質として、または化学結合により互いに連結した、天然には互いに結合することのない2つの成分ペプチドを有するペプチドを意味する。
本発明は、少なくとも部分的にはNMDAR興奮毒性によって仲介される卒中および他の神経学的状態の損傷効果を軽減するのに有用な、キメラペプチドおよびそのペプチド模倣体を提供する。キメラペプチドは少なくとも2つの成分を有する。第一の成分は、NMDA受容体2サブユニットのPLモチーフ(例えば、NMDA NR2BのGenBankアクセッション番号4099612)を含む、またはこれに基づくアミノ酸配列を有する活性ペプチド(すなわち、PLペプチド)である。本発明の実施のためにメカニズムの理解は必要ではないが、そのようなペプチドは少なくとも部分的にはNMDARとシナプス後肥厚部95タンパク質との間の相互作用を阻害することにより作用する(すなわち、PSD-95阻害剤)と考えられる。
に連結している場合には、N型カルシウムチャネルに結合し、それらの活性を阻害するという、本出願に記載する結果を前提としている。シナプス前神経終末上にあるN型カルシウムチャネルは、一次求心性侵害受容器の脊髄末端からのものを含む、神経伝達物質の放出を調節する。N型チャネルへの結合の薬理効果は、薬物ジコノチド(またはプリアルト、イモガイペプチドのオメガ-コノトキシンM-VII-A前駆体の合成体)と関連して詳しく特徴づけられている。N型カルシウムチャネルへの結合は多くの活性と関連づけられており、そのうちのいくつか、またはすべては卒中患者において望ましくないものでありうる。これらの活性には、モルヒネによって誘導されるものよりもはるかに強い鎮痛、低血圧、意識レベルの低下、うつ、認知障害、幻覚、クレアチンキナーゼレベルの上昇、および尿閉が含まれる(例えば、Brose et al., Clin J Pain 13: 256-259, (1997); Mathur et al., Semin Anesthesia Perioperative Med Pain 19: 67-75, 2000, Staats et al., JAMA 291: 63-70, 2004, McGuire et al., J Cardiovasc Pharmacol 30: 400-403, 1997参照)。メイヨークリニックはプリアルトによる治療後に観察された以下の副作用をリストにしているが、これらはN型カルシウムチャネルに高度に選択的である。
本発明において有用な活性ペプチドは、シナプス後肥厚部95タンパク質(PSD-95)のPDZドメイン1および2(Stathakism, Genomics 44(1):71-82 (1997)によって提供されるヒトアミノ酸配列)とニューロンN-メチル-D-アスパラギン酸受容体のNR2Bサブユニットを含む1つまたは複数のNMDA受容体2サブユニットのC末端PL配列(Mandich et al., Genomics 22, 216-8 (1994))との間の相互作用を阻害する。NMDAR2BはGenBank ID 4099612、C末端の20アミノ酸
を有する。活性ペプチドは好ましくはPSD-95のヒト型およびヒトNMDAR受容体を阻害する。しかし、阻害はタンパク質の種変異体からも示されうる。用いることができるNMDAおよびグルタミン酸受容体のリストを以下に示す。

本発明の任意の活性ペプチドを、好ましくはそのN末端で、細胞の形質膜を通しての移動を促進する内部移行ペプチドに連結することができる。内部移行ペプチドは標準のtat配列
の変異体を含む。本発明の実施はメカニズムの理解に依存しないが、膜を通過する能力およびN型カルシウムチャネルに結合する能力はいずれも、ペプチド中の正荷電残基Y、RおよびKの異常に高い出現率によって付与される。本発明において用いるための変異体ペプチドは、細胞への取り込みを促進する能力を保持するが、N型カルシウムチャネルに結合する能力は低下しているべきである。いくつかの適当な内部移行ペプチドは、アミノ酸配列
を含む、またはこの配列からなり、ここでXはY以外のアミノ酸(例えば、他の19の天然アミノ酸のいずれか)であるか、または存在しない(その場合、Gは遊離N末端残基)。好ましいtat変異体はFで置換されたN末端Y残基を有する。したがって、
を含む、またはこの配列からなるtat変異体が好ましい。もう一つの好ましい変異体tat内部移行ペプチドは

を連結するために典型的に用いられる種類のスペーサーもしくはリンカーアミノ酸(例えば、Tang et al. (1996), J. Biol. Chem. 271, 15682-15686; Hennecke et al. (1998), Protein Eng. 11, 405-410参照)でありえ、または隣接残基を含まない変異体の取り込みを付与する能力を検出できるほどに低減させず、隣接残基を含まない変異体に比べてN型カルシウムチャネルの阻害を著しく増大させない、任意の他のアミノ酸でありうる。好ましくは、活性ペプチド以外の隣接アミノ酸の数は

)で観察されたN型カルシウムチャネルの阻害を誘導するのに十分であること、およびC末端に付加した異なるペプチドはTatに結合した場合には阻害に対して緩和な作用しか持たないことを示している。したがって、Tat配列のN末端のチロシンの変更または除去は、結合の低減のために重要であると考えられる。このチロシン近辺の塩基性アミノ酸残基の変異も、N型カルシウムチャネルへの結合およびその阻害の低減を引き起こしうる。以下の表の例示的配列は本明細書において、N型カルシウムチャネルを阻害することなく輸送能力を維持し、したがって卒中または神経外傷治療のための治療指数を高めると予想される。
N型カルシウムチャネルは、α1B-(Cav2.2)、β-、およびα2δ-サブユニットならびに時としてγサブユニットからなるヘテロオリゴマー複合体である。α1B-サブユニットは主となるチャネルを形成し、1つの遺伝子でコードされる。4つのα2δ-サブユニット遺伝子がある(α2δ-1〜α2δ-4)(Snutch et al., Molecular properties of voltage-gated calcium channels. New York: Landes Bioscience, 2005)。種間でN型カルシウムチャネルの緊密な保存がある。したがって、tat変異体を、ヒトまたはラットなどの他の種からのN型カルシウムチャネルを用いて結合の欠如についてスクリーニングすることができる。
1. N型カルシウムチャネルへの結合を測定する
N型カルシウムチャネルに結合することが公知の標識ペプチド(例えば、ジコノチド)を用いての競合結合アッセイにより、内部移行ペプチドのN型カルシウムチャネルへの結合についてスクリーニングすることができる。N型カルシウムチャネルは精製した形で提供することができ、または細胞から自然にもしくは組換えにより発現させることができる。精製した形で提供する場合、N型カルシウムチャネルはビーズまたはマイクロタイターウェルに固定することができる。標識ペプチドおよび試験中の内部移行ペプチドとのインキュベーション後にカルシウムチャネルに結合した標識の量は、試験中の内部移行ペプチドのカルシウムチャネルに結合する能力に反比例する。アッセイはマイクロタイタープレートのウェル内で高処理量で実施することができる。陰性および陽性対照も含めることができる。陰性対照は媒体でありうる。陽性対照はN型カルシウムチャネルに結合することが公知のペプチドの非標識型でありうる。好ましくは、内部移行ペプチドはN型カルシウムチャネルに対して10nMよりも大きい、好ましくは100nMよりも大きいKdを有する。
内部移行ペプチドおよび活性ペプチドなどの活性物質に連結された内部移行ペプチドを含むキメラ物質を、N型カルシウムチャネルによって仲介されるイオン電流を阻害する能力についてスクリーニングすることができる。阻害は、N型カルシウムチャネルによって運ばれるイオン電流測定値の統計学的有意な減少を意味する。そのような減少は電流測定値における20%を超える減少、好ましくは30%を超える減少、より好ましくは40%を超える減少であるべきである。阻害は、N型カルシウム電流が生じる背根神経節ニューロンにおけるホールセルパッチクランプ記録を行うことにより評価することができる。背根神経節(DRG)の培養物は妊娠13〜14日のスイスマウスから調製した。簡単に言うと、DRGを摘出し、トリプシン消化を37℃で20分間行い、機械的に分離し、ポリ-D-リジンでコーティングしたカバースリップ上に播種する。これらを無血清MEM(Neurobasal MEM, B-27 - Gibco Invitrogen Corporation, Carlsbad,CA)中で培養する。3〜5日後、10μM FUDR溶液を加えて、膠細胞増殖を阻害する。培養物を加湿5%CO2雰囲気下、37℃で維持し、1週間に2回供給を行う。ホールセル記録を播種の10〜14日後に室温で実施する。電気生理学記録:ホールセル記録をAxopatch-1B増幅器(Axon Instruments, Foster City, CA)により電位固定モードで実施する。抵抗3〜5MΩの記録電極を薄膜ホウケイ酸ガラス(直径1.5mm;World Precision Instruments, Sarasota, FL)から2段階プラー(PP83; Narishige, Tokyo, Japan)を用いて作製する。データをデジタル化し、フィルターにかけ(2kHz)、pClamp 9(Axon Instruments)のプログラムを用いてオンラインで得る。ピペットに下記を含む溶液を充填する:CsCl 110mM、MgCl2 3mM、EGTA 10mM、HEPES 10mM、MgATP 3mM、GTP 0.6mM。pHをCsOHで7.2に調節する。バス溶液は下記を含んでいた:pH(NaOH)7.4でCaCl2 1mM、BaCl2 10mM、HEPES 10mM、TEA-Cl 160mM、グルコース10mM、TTX 0.0002mM。ホールセル電流を、保持電位-60mVから+20mVまでの40msの脱分極パルスを15sごとに適用して誘導する。使用依存性阻害を試験するために、電流を、保持電位-60mVから+20mVまでの10msの脱分極パルスをそれぞれ0.02s(50Hz)、0.05s(20Hz)、0.1s(10Hz)または15s(0.07Hz)ごとに適用して誘導する。
tat内部移行ペプチドの変異体を動物における輸送活性について試験することができる。内部移行ペプチドを単独で、または活性ペプチド、例えば、
などの活性物質に連結した場合に試験することができる。任意にペプチドなどの活性物質に連結した内部移行ペプチドを、好ましくは塩化ダンシルなどの蛍光標識で標識する。次いで、内部移行ペプチドをマウスなどの動物の末梢に注射する。例えば、腹腔内または静脈内注射が適当である。注射の約1時間後、マウスを屠殺し、固定液(3%パラホルムアルヒド、0.25%グルタルアルデヒド、10%スクロース、10U/mLヘパリン食塩水溶液)で灌流する。次いで、脳を摘出し、凍結して切断する。切片を共焦点顕微鏡を用いて蛍光について分析する。内部移行活性を蛍光から、任意に陽性および陰性対照に対して評価する。適当な陽性対照は試験中の内部移行ペプチドと同じ活性ペプチド(存在する場合は)に連結した標準tatペプチドである。適当な陰性対照は内部移行ペプチドに連結していない蛍光標識した活性ペプチドである。非標識媒体も陰性対照として用いることができる。
活性ペプチドに連結した内部移行ペプチドを含むキメラペプチド(またはそのようなキメラペプチドのペプチド模倣体)の活性を、卒中の様々な動物モデルで試験することができる。一つのそのようなモデルにおいて、成体雄Sprague-Dawleyラットにおいて腔内縫合法(36、37)により90分間の一過性中大脳動脈閉塞(MCAO)を起こさせた。動物を終夜絶食させ、硫酸アトロピン(0.5mg/kg、IP)を注射する。10分後、麻酔を導入する。ラットを経口挿管し、人工換気し、臭化パンクロニウム(0.6mg/kg、IV)で麻痺させる。体温を加熱ランプで36.5〜37.5℃に維持する。大腿動脈および静脈内のポリエチレンカテーテルを用いて、血圧を持続的に記録し、ガスおよびpH測定のために採血する。ポリ-L-リジンでコーティングした3-0モノフィラメントナイロン縫合糸(Harvard Apparatus)を内頸動脈からウィリス動脈環へと導入し、中大脳動脈を効果的に閉塞させることにより、一過性MCAOを90分間行う。これにより、大脳皮質および基底核を含む広範な梗塞を生じる。動物を試験中のキメラペプチドまたは陰性もしくは陽性対照のいずれかで治療する。治療は虚血誘導の前または1時間後までのいずれかでありうる。陰性対照は媒体でありうる。陽性対照は、以前に有効であることが示されたTat-NR2B9cペプチド、
でありうる。キメラペプチドを、MCAOの45分前に1回の静脈内ボーラス注射により送達する(3nmol/g)。化合物を動物に投与した後、梗塞量および/または能力障害指数をもとめる。梗塞量は通常は治療の24時間後に評価するが、3、7、14または60日後などの遅い時期にもとめることもできる。能力障害指数は、例えば、治療の2時間後、治療の24時間後、治療の1週間および1ヶ月後に経時的にモニターすることができる。化合物で治療していない対照動物に比べて梗塞量および/または能力障害指数の統計学的有意な低下を示すキメラペプチドを、本発明の方法を実施するために有用な活性を有すると特定する。
任意に、活性ペプチドおよびそのペプチド模倣体をPSD-95とNMDAR 2Bとの間の相互作用を阻害する能力について、米国特許第20050059597号に記載のアッセイを用いてスクリーニングすることもできる。有用なペプチドは典型的には、そのようなアッセイで50uM、25μM、10μM、0.1μMまたは0.01μM未満のIC50値を有する。好ましいペプチドは典型的には0.001〜1μM、より好ましくは0.05〜0.5または0.05〜0.1μMの間のIC50値を有する。
卒中は原因に関係なくCNSにおける血流障害から生じる状態である。可能性のある原因には、塞栓症、出血および血栓症が含まれる。いくつかのニューロン細胞は血流障害の結果、ただちに死滅する。これらの細胞はグルタミン酸を含む成分分子を放出し、これらは次いでNMDA受容体を活性化し、これらは細胞内カルシウムレベルおよび細胞内酵素レベルを高め、さらなるニューロン細胞死を引き起こす(興奮毒性カスケード)。CNS組織の死滅を梗塞と呼ぶ。梗塞量(すなわち、脳内の卒中により死滅したニューロン細胞の量)を卒中による病理学的損傷の程度の指標として用いることができる。症候性の影響は梗塞の量と梗塞が局在する脳内の位置の両方に依存する。ランキン卒中転帰スケール(Rankin, Scott Med J;2:200-15 (1957))およびバーセルインデックスなどの能力障害指数は、症候性損傷の尺度として用いることができる。ランキンスケールは以下の患者の包括的状態の直接評価に基づいている。
前述のキメラペプチドまたはそのペプチド模倣体を用いて、卒中の患者を治療する。治療は通常は卒中開始後できるだけ早く開始する。時として、高リスクであることが公知の患者では治療を卒中の開始と同時またはその前に開始することができる。危険因子には高血圧、糖尿病、家族歴、喫煙、以前の卒中、および進行中の手術が含まれる。通常、治療は卒中開始後1から24時間以内に最初に投与する。本発明のキメラペプチドの1回用量で十分であることが多い。しかし、複数用量を6〜24時間またはそれ以上の間隔で投与することもできる。
本発明のキメラペプチドまたはペプチド模倣体は薬学的組成物の形で投与することができる。薬学的組成物はGMP条件下で製造する。薬学的組成物は前述の用量のいずれかを含む単位用量剤形(すなわち、1回投与のための用量)で提供することができる。薬学的組成物は通常の混合、溶解、造粒、糖衣錠生成、研和、乳化、カプセル化、包括または凍結乾燥法によって製造することができる。特に、凍結乾燥した本発明のキメラペプチドまたはペプチド模倣体は下記の製剤および組成物中で用いることができる。
本発明は、他の内部移行ペプチドがN型カルシウムチャネルに結合する、および/またはこれらを阻害するかどうかを調べるための、そのようなペプチドのスクリーニング法をさらに提供する。試験ペプチドをそのような結合または阻害について、単独で、または活性物質、特にカーゴペプチドとして知られていることもある活性ペプチドに連結してのいずれかで評価することができる。試験しうる他の内部移行ペプチドには、アンテナペディア内部移行ペプチド(Bonfanti, Cancer Res. 57, 1442-6 (1997))(およびその変異体)、Tat変異体、ペネトラチン、SynB1および3、トランスポータン、アンフィペーシック(Amphipathic)、gp41NLS、ポリArg、および以下の文献に記載の他のペプチドが含まれる(Temsamani, Drug Discovery Today, 9(23):1012-1019, 2004; De Coupade, Biochem J., 390:407-418, 2005; Saalik Bioconjugate Chem. 15: 1246-1253, 2004; Zhao, Medicinal Research Reviews 24(1): 1-12, 2004; Deshayes, Cellular and Molecular Life Sciences 62: 1839-49, 2005)(すべて参照により本明細書に組み入れられる)。
前述のtat変異体は任意の他の活性物質と連結して、細胞膜および/または血液脳関門を通っての物質の取り込みを促進することができる。tat変異体および活性物質を含む、またはこれらからなるキメラ物質の、治療法における使用は、活性物質単独の使用に比べて所期の部位でのバイオアベイラビリティを改善し、標準tatペプチドに連結した活性物質の使用に比べてN型カルシウムチャネルへの結合による副作用を軽減する。tat変異体は細胞内標的を有する活性物質および/または活性を発揮するために血液脳関門を通過する必要がある向神経活性薬のために特に有用である。tat変異体の結合を受け入れられる活性物質のいくつかはペプチドであるが、すべてではない。tat変異体の使用は、低いバイオアベイラビリティ、高用量または短い半減期を有する既存の薬剤にとって特に有用である。
Tat-NR2B9cは標準tatペプチドおよび以前に虚血のラットモデルで有効であることが示されたKLSSIESDV (SEQ ID NO:12)のキメラペプチドである。本実施例はペプチドTat-NR2B9cを約70の受容体タンパク質に対する公知のリガンドの結合を阻害する能力についてスクリーニングする。スクリーニングした受容体の例には、グルタミン酸、ヒスタミンH1、カリウムチャネル、ドーパミンD1、カルシウムチャネル(L型、N型)が含まれていた。
公知のN型カルシウムチャネル阻害剤であるジコノチドと同様、Tat-NR2B9cは多くの正電荷を含む。正電荷はおそらく脳血液関門を通過する能力を促進し、かつN型カルシウムチャネル結合にも貢献すると考えられる。直接の配列比較によって、正(R=アルギニン、K=リジン)電荷ならびにペプチド主鎖に沿ってのこれらの電荷の間隔にいくらかの類似性が見られる(下記の配列参照)。これはTat-NR2B9c N型カルシウムチャネル結合エピトープをTat領域(イタリックで示す)およびNMDAR2Bドメインの1つのアミノ酸にほぼ位置づけている。

)、F-Tat-NR2B9cまたは1994 Tat-NR2B9c KからAは、N型カルシウムチャネル活性の有意な阻害は示さなかった。ペプチド1992、1994および1987はTAT単独に比べてチャネル活性の有意な改善を示したが、なおN型カルシウムチャネル活性の量のいくらかの低減を示した。これらのペプチドはすべて、これらのTat変異体配列の1つを含む薬物の治療指数増大を示す標準Tat単独に比べてN型カルシウムチャネルへの結合低減を提供する。
さらなる実験を図4〜7に示すとおりに実施した。それらの目的はTat-NR2B9cによるN型Ca2+チャネル仲介性イオン電流の阻害をさらに特徴づけることであった。加えて、図4はTat-NR2B9c
によるCa2+電流の阻害の程度を特徴づけており、これを他の変異体:1990 TAT

;1994 Tat-NR2B9c KからA
と比較している。
)を含むすべてのペプチドの漸増する濃度は背根神経節ニューロン(主にN型Ca2+チャネルを発現する)におけるCa2+電流を有意に阻害することを示している。これはN型Ca2+チャネル電流を阻害する性質はTat配列にあることを示唆している。
前述し、実施例4でもさらに記載した、永久虚血のラット軟膜閉塞モデルにおいて、F-Tat-NR2B9cをTat-NR2B9cと、体重1gあたり3nmolの単一の用量で比較した。いずれのケースでも、キメラペプチドを虚血開始の1時間後に投与した。図3に示すとおり、F-Tat-NR2B9cおよびTat-NR2B9cは梗塞サイズの減少において等しく有効であった。
目的:
1. 卒中のインビボ軟膜3血管閉塞(P3VO)モデルを用いて、雄ラットおよび雌ラット両方におけるTat-NR2B9cペプチドの神経保護効果を試験すること。
2. 雄ラットにおいてTat-NR2B9cの6つの変異を試験することにより、作用機序を解明すること。
Tat-NR2B9cとして公知のペプチドが開発され、これまでラットにおける卒中のMCAOモデルで試験された。このペプチドは、梗塞サイズの減少によって明らかなとおり、神経保護的であることが判明している。しかし、卒中のMCAOモデルは広範な神経欠損を伴う大きな梗塞と、寿命の短縮を引き起こす。卒中のP3VOモデルは神経欠損が最小限のはるかに小さい、皮質の梗塞を引き起こし、寿命は通常どおりである。
動物
成体Sprague Dawleyラット(10〜12週齡)(雄約300g、雌約250g)(図8)を12〜18時間絶食させた後、ウィスカーバレル皮質上の中大脳動脈の3つの末端枝の永久軟膜血管閉塞を起こさせた(P3VO)。7つのペプチドそれぞれを雄ラットおよび食塩水対照群で試験した(各群n=8)。Tat-NR2B9cペプチドおよび食塩水対照群を雌ラットで試験した(各群n=8)。研究者は手術中から梗塞サイズの分析中を通して治療群について盲検的に行った。
ラットをケタミン(100mg/kg)、アセプロマジン(2mg/kg)、およびキシラジン(5mg/kg)の0.5ml/kg筋肉内注射で麻酔し、必要に応じて初期用量の3分の1で追加麻酔した。肛門体温プローブを挿入し、動物を37℃に維持した加熱パッド上に置いた。色素注入のために、右外頸動脈(ECA)をPE 10ポリエチレンチューブで挿管した。頭蓋を正中切開により露出し、組織をこそげ取り、右側の側頭筋を頭蓋から切断した。解剖顕微鏡および歯科用空気ドリルを用い、頭蓋に長方形の穴を開け、硬膜を無傷に保ちつつ頭蓋片を持ち上げることにより、右体性感覚皮質(ブレグマの2mm尾側、5mm外側)に6×4mmの頭蓋窓を設ける。人工脳脊髄液で洗浄した後、生理食塩水中の生体色素パテントブルーバイオレット(10mmol/L;Sigma)の少量ボーラス(10から20μL)を右外頸動脈に注入して、皮質の表面血管を通っての移行を示した。バレル皮質周囲のMCAの3つの重要な細動脈枝を選択し、硬膜を通して電気的に焼灼した。焼灼後、ボーラス注入および色素移行を繰り返して、焼灼した細動脈を通る移行の断絶を確実にした。頭蓋の長方形を窓上に戻し、頭皮を縫合した。カテーテルをECAから除去し、ECAを結紮し、前頸部を縫合した。限局性の閉塞開始の1時間後、0.3mlの薬物(体重1gあたり3nMol)または食塩水対照を尾静脈から0.06ml/分の速度で注入した。各ラットを完全に回復するまで体温を維持するために加熱ランプで保温した個々のケージに戻した。食餌および水を自由に摂取させた。
手術の24時間後、動物を1mLのペントバルビタールで再度麻酔し、脳を迅速に採取した。1つの冠状切片を梗塞領域から取り、2%塩化トリフェニルテトラゾリウム(TTC)中、37℃で15分間インキュベートした。画像を走査し、脳切片を-80℃で保存した。
梗塞サイズを試験中の各ラットについて半球のパーセントとして測定した。梗塞サイズ測定値を得た後、動物をそれぞれの群に分けた。治療群間で平均±SEとして比較を行った。
試験した6つの新規ペプチドのうち、Tat-NR2B9c、1976および1977は梗塞サイズの有意な減少をもたらし、1975および1978は梗塞サイズのいくらかの減少を示した(図8)。
Claims (5)
- PSD-95のNMDA受容体への結合を阻害する25アミノ酸以内の活性ペプチド、および該活性ペプチドのN末端に結合した、キメラペプチドの細胞への取り込みを促進する内部移行ペプチドを含み、かつtatペプチド:YGRKKRRQRRR(配列番号:1)に比べてN型カルシウムチャネルに結合する能力が低い、単離キメラペプチドであって、前記活性ペプチドが、そのC末端にESDV(配列番号:6)またはETDV(配列番号:8)を含むアミノ酸配列を有し、かつ、前記内部移行ペプチドが、配列番号:135記載の配列のC末端側に10以内の隣接アミノ酸を伴うFGRKKRRQRRR(配列番号:135)を含むアミノ酸配列を有する、単離キメラペプチド。
- FGRKKRRQRRRKLSSIESDV(配列番号:19)またはFGRKKRRQRRRKLSSIETDV(配列番号:16)を含むアミノ酸配列を有する、請求項1記載の単離キメラペプチド。
- キメラペプチドの有効量を含む、卒中または他のCNSへの傷害を有するかまたはそのリスクがある患者における卒中または他のCNSへの傷害の損傷効果の治療に用いるための薬学的組成物であって、キメラペプチドが、C末端にESDV(配列番号:6)またはETDV(配列番号:8)を含むアミノ酸配列を有する25アミノ酸以内の活性ペプチドと、該活性ペプチドのN末端に結合した、C末端側に10以内の隣接アミノ酸を伴うFGRKKRRQRRR(配列番号:135)を含むアミノ酸配列を有する内部移行ペプチドと、を含む、薬学的組成物。
- 活性物質とキメラ物質の細胞への取り込みを促進する内部移行ペプチドとを含む単離キメラ物質であって、該内部移行ペプチドのC末端に該活性物質を結合し、該内部移行ペプチドが、C末端側に10以内の隣接アミノ酸を伴うFGRKKRRQRRR(配列番号:135)を含むアミノ酸配列を有する、物質。
- FGRKKRRQRRR(配列番号:135)からなるアミノ酸配列を有する単離された内部移行ペプチド。
