Description
[1]PSGL−1に対する阻害物質を有効成分として含有する、代謝性疾患の予防及び/又は治療剤。
[2]代謝性疾患が、メタボリックシンドローム、2型糖尿病、耐糖能異常、肥満症又は脂質異常症である、上記[1]記載の剤。
[3]PSGL−1に対する阻害物質が、PSGL−1に対する特異的中和抗体、或いはPSGL−1の発現を特異的に阻害可能なアンチセンス核酸若しくは小分子量干渉RNA、又はこれらを哺乳動物細胞内において発現可能な発現ベクターである、上記[1]記載の剤。
[4]以下の工程を含む、代謝性疾患を予防又は治療し得る物質のスクリーニング方法:
(I)被験物質がPSGL−1の機能を阻害するか検定すること、及び
(II)PSGL−1の機能を阻害した被験物質を代謝性疾患を予防又は治療し得る物質として選択すること。
[5]代謝性疾患が、メタボリックシンドローム、2型糖尿病、耐糖能異常、肥満症又は脂質異常症である、上記[4]記載の方法。
アンチセンス核酸の長さとしては、PSGL−1の転写産物と特異的にハイブリダイズし得る限り特に制限はないが、合成の容易さや抗原性の観点などから、下限としては通常10塩基以上、好ましくは15塩基以上のものが挙げられ、上限としては通常100塩基以下、好ましくは30塩基以下、より好ましくは24塩基以下のものが挙げられる。従って、アンチセンス核酸としては、例えば10塩基以上100塩基以下、好ましくは15塩基以上30塩基以下、より好ましくは15塩基以上24塩基以下のオリゴヌクレオチドが使用され得る。
また、アンチセンス核酸の標的配列についても、アンチセンス核酸がハイブリダイズすることによりPSGL−1の翻訳が阻害される配列であれば特に制限はなく、mRNAの全配列であっても部分配列(通常10塩基以上、好ましくは15塩基以上;且つ通常100塩基以下、好ましくは30塩基以下、より好ましくは24塩基以下)であってもよいし、あるいは初期転写産物のイントロン部分であってもよいが、アンチセンス核酸としてオリゴヌクレオチドを使用する場合は、標的配列はPSGL−1のmRNAの5’末端からコード領域のC末端までに位置することが望ましい。
アンチセンス核酸の具体例としては、例えばヒトPSGL−1の場合、配列番号:1に記載したmRNAの1−1369位までの塩基配列に含まれる、通常10塩基以上、好ましくは15塩基以上、且つ通常100塩基以下、好ましくは30塩基以下、より好ましくは24塩基以下の長さの塩基配列に相補的な配列を持つオリゴヌクレオチドが挙げられ、或いは例えばマウスPSGL−1の場合、配列番号:3に記載したmRNAの1−1412位までの塩基配列に含まれる、通常10塩基以上、好ましくは15塩基以上、且つ通常100塩基以下、好ましくは30塩基以下、より好ましくは24塩基以下の長さの塩基配列に相補的な配列を持つオリゴヌクレオチドが挙げられる。
上記アンチセンス核酸の合成は自体公知の方法で行えば良く、例えば、所望の塩基配列を持つオリゴヌクレオチドを市販のDNA/RNA自動合成機を用いて合成すれば良い。
siRNAの合成は自体公知の方法で行えば良く、例えば、センス鎖及びアンチセンス鎖をDNA/RNA自動合成機でそれぞれ合成し、それらをアニーリングさせることにより合成することができる。
この報告によれば、メタボリックシンドロームとは、内臓脂肪蓄積(ウエスト周囲径が男性は85cm以上、女性は90cm以上)を必須項目として、それに加えて脂質代謝異常(高トリグリセライド血症(150mg/dl以上)且つ/又は低HDLコレステロール血症(40mg/dl未満))、高血圧(収縮期血圧が130mmHg以上、且つ/又は拡張期血圧が85mmHg以上)及び高血糖(空腹時血糖が110mg/dl以上)からなる3項目のうちの2項目以上が該当する場合をいう。
1)ウエスト(腹囲)が男性で102cm以上、女性で88cm以上;
2)空腹時トリグリセライドが150mg/dl以上;
3)HDLコレステロールが男性で40mg/dl未満、女性で50mg/dl未満;
4)血圧が最大血圧で130mmHg以上又は最小血圧で85mmHg以上;
5)空腹時血糖値が110mg/dl以上。
一方、WHOの診断基準では、高インスリン血症(非糖尿病患者の上位25%)又は空腹時血糖110mg/dl以上に加え、内臓肥満、脂質代謝異常、高血圧、マイクロアルブミン尿症のうち以下の条件に2つ以上該当する場合と定義されている。
1)内臓肥満:ウエスト/ヒップ比>0.9(男性)、>0.85(女性)又はBMIが30を上回る;
2)脂質代謝異常:血漿トリグリセリド150mg/dl以上;
3)高血圧:最大血圧140/最小血圧90mmHg以上又は降圧剤内服中;
4)マイクロアルブミン尿症:尿中アルブミン排泄率20μg/min以上又は尿中アルブミン/クレアチニン比30mg/g以上。
本発明の剤は、上記した診断基準により決定されるメタボリックシンドロームの予防及び/又は治療剤として使用され得る。
この報告によれば、糖尿病とは、空腹時血糖値(静脈血漿におけるグルコース濃度)が126mg/dl以上、75g経口ブドウ糖負荷試験(75gOGTT)2時間値(静脈血漿におけるグルコース濃度)が200mg/dl以上、随時血糖値(静脈血漿におけるグルコース濃度)が200mg/dl以上のいずれかを示す状態である。また、上記糖尿病に該当せず、かつ、「空腹時血糖値(静脈血漿におけるグルコース濃度)が110mg/dl未満または75g経口ブドウ糖負荷試験(75gOGTT)2時間値(静脈血漿におけるグルコース濃度)が140mg/dl未満を示す状態」(正常型)でない状態を、「境界型」と呼ぶ。
これらの報告によれば、糖尿病とは、空腹時血糖値(静脈血漿におけるグルコース濃度)が126mg/dl以上であり、75g経口ブドウ糖負荷試験2時間値(静脈血漿におけるグルコース濃度)が200mg/dl以上を示す状態である。
また、上記報告によれば、耐糖能異常とは、空腹時血糖値(静脈血漿におけるグルコース濃度)が110mg/dl以上、126mg/dl未満であり、かつ、75g経口ブドウ糖負荷試験2時間値(静脈血漿におけるグルコース濃度)が140mg/dl以上200mg/dl未満を示す状態である。さらに、ADAの報告によれば、空腹時血糖値(静脈血漿におけるグルコース濃度)が110mg/dl以上126mg/dl未満の状態をIFG(Impaired Fasting Glucose)と呼ぶ。一方、WHOの報告によれば、75g経口ブドウ糖負荷試験2時間値(静脈血漿におけるグルコース濃度)が140mg/dl以上、200mg/dl未満である状態をIGT(Impaired glucose tolerance)と呼ぶ。
本発明の剤は、上記した新たな判定基準により決定される糖尿病、耐糖能異常(IFG、IGT)の予防及び/又は治療剤としても使用され得る。
(I)被験物質がPSGL−1の機能(即ち、活性又は発現)を阻害するか検定すること、及び
(II)PSGL−1の機能を阻害した被験物質を代謝性疾患を予防又は治療し得る物質として選択すること。
当該スクリーニング方法において、用語「代謝性疾患」は、その予防及び/又は治療剤の説明において記述したものと同義である。
以下、本発明のスクリーニング方法を詳述する。
PSGL−1の活性を阻害する物質(PSGL−1活性阻害物質)としては、PSGL−1とセレクチン(例えば、P−セレクチン)との結合を阻害する物質が挙げられる。以下、PSGL−1とセレクチンとの結合を阻害する物質(以下、PSGL−1結合阻害物質ともいう)のスクリーニング方法について説明する。
PSGL−1結合阻害物質のスクリーニング方法(以下、スクリーニング方法Aともいう)は、以下の工程(a)、(b)及び(c)を含む:
(a)被験物質の存在下、セレクチンをPSGL−1に接触させる工程;
(b)被験物質の存在下におけるセレクチンとPSGL−1との結合能を測定し、該結合能を被験物質の非存在下におけるセレクチンとPSGL−1との結合能と比較する工程;
(c)上記(b)の比較結果に基づいて、セレクチンとPSGL−1との結合能の低下をもたらす被験物質を代謝性疾患を予防又は治療し得る物質として選択する工程。
セレクチン及びPSGL−1は自体公知の方法により調製できる。それらは、例えば、セレクチン又はPSGL−1の発現細胞(それぞれ、例えば、血管内皮細胞及び白血球)から単離・精製できる。しかしながら、迅速、容易かつ大量にそれらのタンパク質を調製し、また、それらのヒトタンパク質を調製するためには、遺伝子組換え技術により組換えタンパク質を調製するのが好ましい。組換えタンパク質は、細胞系、無細胞系のいずれで調製したものでもよい。
セレクチン又はPSGL−1発現細胞は、セレクチン又はPSGL−1を発現するものである限り特に限定されず、上述したセレクチン又はPSGL−1の発現細胞、セレクチン又はPSGL−1発現ベクターで形質転換された細胞などであり得る。該細胞は、当業者であれば容易に同定又は調製でき、初代培養細胞、当該初代培養細胞から誘導された細胞株、市販の細胞株、セルバンクより入手可能な細胞株などを使用できる。
次いで、被験物質の存在下におけるセレクチンとPSGL−1との結合能が、被験物質の非存在下におけるセレクチンとPSGL−1との結合能と比較される。結合能の比較は、好ましくは、有意差の有無に基づいて行われる。被験物質の非存在下におけるセレクチンとPSGL−1との結合能は、被験物質の存在下におけるセレクチンとPSGL−1との結合能の測定に対し、事前に測定した結合能であっても、同時に測定した結合能であってもよいが、実験の精度、再現性の観点から同時に測定した結合能であることが好ましい。
次に、PSGL−1の発現を阻害する物質(PSGL−1発現阻害物質)のスクリーニング方法(以下、スクリーニング方法Bともいう)について説明する。スクリーニング方法Bは、以下の工程(a)、(b)及び(c)を含む:
(a)被験物質とPSGL−1タンパク質又はそれをコードする遺伝子の発現を測定可能な細胞とを接触させる工程;
(b)被験物質を接触させた細胞における発現量を測定し、該発現量を被験物質を接触させない対照細胞における発現量と比較する工程;
(c)上記(b)の比較結果に基づいて、発現量を低下させた被験物質を代謝性疾患を予防又は治療し得る物質として選択する工程。
「PSGL−1タンパク質又はそれをコードする遺伝子の発現を測定可能な細胞」とは、PSGL−1遺伝子の産物、例えば、転写産物、翻訳産物(即ち、タンパク質)の発現レベルを直接的又は間接的に評価可能な細胞をいう。PSGL−1遺伝子産物の発現レベルを直接的に評価可能な細胞は、PSGL−1遺伝子を天然で発現可能な細胞であり得、一方、PSGL−1遺伝子の産物の発現レベルを間接的に評価可能な細胞は、PSGL−1遺伝子転写調節領域についてレポーターアッセイを可能とする細胞であり得る。
PSGL−1遺伝子を天然で発現可能な細胞は、PSGL−1遺伝子を潜在的に発現するものである限り特に限定されず、PSGL−1遺伝子を恒常的に発現している細胞、PSGL−1遺伝子を誘導条件下(例えば、薬物での処理)で発現する細胞などであり得る。該細胞は、当業者であれば容易に同定でき、初代培養細胞、当該初代培養細胞から誘導された細胞株、市販の細胞株、セルバンクより入手可能な細胞株などを使用できる。PSGL−1遺伝子を天然で発現可能な細胞の例としては、白血球(例えば、好中球、単球、マクロファージ、B細胞のサブセット、及び全てのT細胞)が挙げられる。
PSGL−1遺伝子転写調節領域についてレポーターアッセイを可能とする細胞は、PSGL−1遺伝子転写調節領域、当該領域に機能可能に連結されたレポーター遺伝子を含む細胞である。PSGL−1遺伝子転写調節領域、レポーター遺伝子は、発現ベクター中に挿入されている。
PSGL−1遺伝子転写調節領域は、PSGL−1遺伝子の発現を制御し得る領域である限り特に限定されないが、例えば、転写開始点から上流約2kbpまでの領域、あるいは該領域の塩基配列において1以上の塩基が欠失、置換若しくは付加された塩基配列からなり、且つPSGL−1遺伝子の転写を制御する能力を有する領域などを挙げることができる。
レポーター遺伝子は、検出可能なタンパク質又は酵素をコードする遺伝子であればよく、例えばGFP(緑色蛍光タンパク質)遺伝子、GUS(β−グルクロニダーゼ)遺伝子、LUS(ルシフェラーゼ)遺伝子、CAT(クロラムフェニコルアセチルトランスフェラーゼ)遺伝子などが挙げられる。
PSGL−1遺伝子転写調節領域、当該領域に機能可能に連結されたレポーター遺伝子が導入される細胞は、PSGL−1遺伝子転写調節機能を評価できる限り、即ち、該レポーター遺伝子の発現量が定量的に解析可能である限り特に限定されない。しかしながら、PSGL−1遺伝子に対する生理的な転写調節因子を発現し、PSGL−1遺伝子の発現調節の評価により適切であると考えられることから、該導入される細胞としては、PSGL−1遺伝子を天然で発現可能な細胞が好ましい。
被験物質とPSGL−1遺伝子の発現を測定可能な細胞とが接触される培養培地は、用いられる細胞の種類などに応じて適宜選択されるが、例えば、約5〜20%のウシ胎仔血清を含む最少必須培地(MEM)、ダルベッコ改変最少必須培地(DMEM)、RPMI1640培地、199培地などである。培養条件もまた、用いられる細胞の種類などに応じて適宜決定されるが、例えば、培地のpHは約6〜約8であり、培養温度は通常約30〜約40℃であり、培養時間は約12〜約72時間である。
例えば、PSGL−1遺伝子の発現を測定可能な細胞として、PSGL−1遺伝子を天然で発現可能な細胞を用いた場合、発現量は、PSGL−1遺伝子の産物、例えば、転写産物又は翻訳産物を対象として自体公知の方法により測定できる。例えば、転写産物の発現量は、細胞からtotal RNAを調製し、RT−PCR、ノザンブロッティング等により測定され得る。また、翻訳産物の発現量は、細胞から抽出液を調製し、免疫学的手法により測定され得る。免疫学的手法としては、放射性同位元素免疫測定法(RIA法)、ELISA法(Methods in Enzymol. 70: 419−439 (1980))、蛍光抗体法などが使用できる。
一方、PSGL−1遺伝子の発現を測定可能な細胞として、PSGL−1遺伝子転写調節領域についてレポーターアッセイを可能とする細胞を用いた場合、発現量は、レポーターのシグナル強度に基づき測定され得る。
次いで、被験物質を接触させた細胞におけるPSGL−1遺伝子の発現量が、被験物質を接触させない対照細胞におけるPSGL−1遺伝子の発現量と比較される。発現量の比較は、好ましくは、有意差の有無に基づいて行なわれる。被験物質を接触させない対照細胞におけるPSGL−1遺伝子の発現量は、被験物質を接触させた細胞におけるPSGL−1遺伝子の発現量の測定に対し、事前に測定した発現量であっても、同時に測定した発現量であってもよいが、実験の精度、再現性の観点から同時に測定した発現量であることが好ましい。
[実験]
8週齢のdb/dbマウス(c57BL/KsJ−db/db)及びC57/BL6マウスの体重、脂肪重量及び各種代謝データを測定した。内臓脂肪組織からRNAを抽出し、リアルタイムRT−PCRによって、マクロファージのマーカーであるF4/80、炎症のマーカーであるMCP−1、接着分子であるP−セレクチン、E−セレクチン、ICAM−1などの遺伝子発現レベルを検討した。更に、DNAマイクロアレイを用いて、内蔵脂肪組織における遺伝子発現プロファイルを解析した。
[結果]
db/dbマウスでは対照群に比し、体重、内臓脂肪重量、LDL、随時血糖及びIRIの有意な増加を認め、HbA1cには差は見られなかった(図1)。DNAマイクロアレイでは、対照群に比しdb/dbマウスで2倍以上の発現増加を認めた遺伝子は1001あり、更にクラスター解析を行った結果、L−セレクチンやPSGL−1をはじめとする40の細胞接着分子関連遺伝子の発現増加を認めた(表1)。また、リアルタイムRT−PCRにより、db/dbマウスにおいて、F4/80、MCP−1、PSGL−1及びP−セレクチンの有意な発現増加を認めた(図2)。
[実験]
7週齢のC57/BL6マウスに低脂肪食及び高脂肪食を各々12週間負荷し、インスリン負荷試験(ITT)及び腹腔内グルコース負荷試験(IPGTT)を施行した。ITTではインスリン0.7単位/kgを腹腔内投与し、負荷前並びに負荷後30分、60分及び120分の血糖値を測定し、負荷前血糖値との変化率を比較した。IPGTTではブドウ糖1.2g/kgを腹腔内投与し、負荷前並びに負荷後30分、60分及び120分の血糖値及びIRIを測定した。また、試験例1と同様に各種代謝データの測定と内臓脂肪組織における遺伝子発現をリアルタイムRT−PCRとDNAマイクロアレイとを用いて検討した。
[結果]
高脂肪食負荷マウスでは低脂肪食負荷群に比し、体重、内臓脂肪重量、LDL、空腹時血糖、空腹時IRI及びHbA1cの有意な増加を認めた(図3)。またITTとIPGTTにて、高脂肪食群で耐糖能異常及びインスリン抵抗性を認めた(図4)。DNAマイクロアレイの結果では、対照群に比し高脂肪食群で2倍以上の発現増加を認めた遺伝子を566認め、クラスター解析にて、PSGL−1を含む37の細胞接着分子関連遺伝子の発現増加を認めた(表2)。リアルタイムRT−PCRでは、db/dbマウスにおいてCD68、MCP−1、PSGL−1及びP−セレクチンの有意な発現増加を認めた(図5)。
[実験]
7週齢のPSGL−1ノックアウトマウス及びC57/BL6マウスに高脂肪食を10週間負荷し、試験例2と同様にITT及びIPGTTを施行し、各種代謝データを測定した。また内臓脂肪からRNAを抽出し、リアルタイムRT−PCRによって炎症関連遺伝子の遺伝子発現レベルを検討した。更に、内蔵脂肪組織のPAS染色を行い、内蔵脂肪細胞の面積を測定した。更に、肝臓重量および肝臓中性脂肪含量の測定を行い、また、肝臓組織のHE染色を行って肝臓組織の変化を検討した。
[結果]
高脂肪食負荷PSGL−1ノックアウトマウスとC57/BL6マウスとでは、体重、脂肪重量、空腹時血糖及びHbA1cに差は見られなかったが、T−Cho、LDL、TG、FFA、空腹時IRI及び血清レプチン濃度はPSGL−1ノックアウトマウスにおいて有意な低下を認めた(図6)。ITTではPSGL−1ノックアウトマウスで有意に耐糖能の改善を認め、IPGTTでも血糖値に差は見られなかったが、PSGL−1ノックアウトマウスで有意にIRIの低下を認めた(図7)。また、内蔵脂肪組織のPAS染色にて脂肪細胞の面積の測定を行い、C57/BL6マウスに比しPSGL−1ノックアウトマウスにて脂肪細胞面積の減少を認めた(図8)。リアルタイムRT−PCRにて、PSGL−1ノックアウトマウスでF4/80、MCP−1、NOS2及びレプチンの有意なmRNAの発現低下を認めた(図9)。PSGLノックアウトマウスでは肝臓重量の有意な減小が見られ、肝臓中性脂肪含量についても減少する傾向がみられた(図10(A))。また、HE染色像において、肝細胞障害(バルーニング変化)がPSGL−1ノックアウトマウスで明らかに抑制されていた(図10(B))。
PSGL−1は、白血球及び血管内皮細胞上に発現し、P−、E−及びL−セレクチンをリガンドとして機能し、炎症巣への白血球浸潤を誘導する分子である。今回の結果から、PSGL−1を阻害することにより、肥満マウスの内臓脂肪組織へのマクロファージの浸潤が減少し、内臓脂肪における炎症が抑制されてインスリン抵抗性が改善することが示された。PSGL−1を阻害することにより、メタボリックシンドロームの予防、及び肥満者におけるインスリン抵抗性の改善と糖尿病発症抑制が可能になる。
Claims (7)
- 以下の工程を含む、2型糖尿病、並びに2型糖尿病に起因する、メタボリックシンドローム、耐糖能異常、肥満症及び脂質異常症からなる群から選択される代謝性疾患を予防又は治療し得る物質のスクリーニング方法:
(a)被験物質の存在下、セレクチンをPSGL−1に接触させる工程;
(b)被験物質の存在下におけるセレクチンとPSGL−1との結合能を測定し、該結合能を被験物質の非存在下におけるセレクチンとPSGL−1との結合能と比較する工程;及び、
(c)上記(b)の比較結果に基づいて、セレクチンとPSGL−1との結合能の低下をもたらす被験物質を前記の代謝性疾患を予防又は治療し得る物質として選択する工程。 - 前記の工程(a)が、該被験物質の存在下、セレクチン又はその発現細胞を、PSGL−1又はその発現細胞に接触させることにより行われる、請求項1に記載の方法。
- 前記の工程(b)における結合能の測定が、フローサイトメトリー法及び表面プラズモン共鳴法からなる群から選択される方法を用いて行われる、請求項1又は2に記載の方法。
- セレクチンがP−セレクチンである、請求項1〜3のいずれか1項に記載の方法。
- 以下の工程を含む、2型糖尿病、並びに2型糖尿病に起因する、メタボリックシンドローム、耐糖能異常、肥満症及び脂質異常症からなる群から選択される代謝性疾患を予防又は治療し得る物質のスクリーニング方法:
(a)被験物質とPSGL−1タンパク質又はそれをコードする遺伝子の発現を測定可能な細胞とを接触させる工程;
(b)被験物質を接触させた細胞における発現量を測定し、該発現量を被験物質を接触させない対照細胞における発現量と比較する工程;及び、
(c)上記(b)の比較結果に基づいて、発現量を低下させた被験物質を前記の代謝性疾患を予防又は治療し得る物質として選択する工程。 - 前記の工程(a)の細胞が白血球である、請求項5に記載の方法。
- 前記の工程(b)における発現量の測定が、RT−PCR法、ノザンブロッティング法、放射性同位元素免疫測定法、ELISA法、蛍光抗体法およびレポーターアッセイ法からなる群から選択される方法を用いて行われる、請求項5又は6に記載の方法。
