試料内の濃度が非定常のときを考えた場合、試料内の温度分布と温度の時間的変化の間は、試料の密度をp、定圧比熱をCpとすると、以下の式(2)の熱拡散方程式で表される。
熱拡散率αと熱伝導率λとは、次の式(3)に示す関係を有する。
(i)測定すべき試料片方の面で試料温度が交流状に変化する。
(ii)温度波は無限に拡散する。
(iii)測定すべき試料の板厚dが、下記式に示すように、熱的に厚い。

は、長さの次元をもつことより、熱拡散長とよばれ、本測定法において重要なパラメーターの一つである。試料の厚みdと熱拡散長μの関係は、図11(a)および(b)に示すように、

と定義される。熱拡散長は温度変化の波長であるため、それが試料の厚みより大きい、すなわち熱的に薄い場合、試料全体が同じ周期で温度変動を起こしてしまう。この場合、試料表面と裏面における温度変動の位相差は0に近づき、熱拡散率は式(8)からは求められなくなる。
(1)液体中の浮遊細胞等の試料としての微粒子1個を誘電泳動法により確実に固定した状態で観察することができるため、迅速な温度変化の測定が可能になる。また、迅速な赤外線分析、二次元的(ないしは擬似三次元的)な赤外線分析が可能になる。
(2)微粒子1個を観察するため、迅速な赤外線分析が可能になる。
(3)二次元的(ないしは擬似三次元的)な赤外線分析が可能になる。

図12、図13は本発明の前提となる試料温度測定装置のシステム構成を示す前提技術1である。
図16は、本発明に好適に使用可能な赤外線像拡大手段(顕微鏡等)65を備えた赤外線カメラ62の配置の一例を模式的に示した前提技術2の斜視図である。温度コントローラ54により温度制御されるホットステージ53に収容された試料Sを赤外線顕微鏡65で観察し、観察像を赤外線放射温度計である赤外線カメラ62により撮影し、撮影データをデータ処理装置63で処理する。
図13に示す前提技術1の試料温度測定装置50による通電で発生させた温度波の位相遅れの測定例を、図14および図15のグラフに模式的に示す。

(2)測定すべき試料および参照試料を一定速度で昇温または降温させつつ、前記試料および参照試料の少なくとも一部を顕微鏡により拡大し、その際の温度変化を赤外線放射温度計により測定し、測定試料および参照試料の温度変化の差を比較することにより、試料のDTA分析を行う態様において好適に使用可能な条件の例は、以下の通りである。
(3)(2)の熱分析を行いながら、測定すべき試料の一部を交流状に加熱して、距離d離れた位置に到達した温度波の位相差の遅れから熱拡散率を測定する態様。
交流熱源に好適に使用可能な導電性物質は、電流を流すことでジュール熱により発熱するものである限り、特に制限されない。このような導電性物質の例としては、例えば、金、銀、白金、銅、鉄、亜鉛、アンチモン、イリジウム、クロメル、コンスタンタン、ニクロム、アルミニウム、クロム、ニッケル、カーボン等が挙げられる。
図20〜22は本発明において好適に使用可能な画像データ取り込み装置の構成を示す前提技術3の回路ブロック図である。
(1)記憶時間を出来る限り長くできる点からは、PC本体のメモリに取り込むよりも、PCとは別個に設けたメモリに取り込むことが好ましい。
(2)取り込み速度を速くし、且つ上書き機能をも具備させる点からは、HDDを補助記憶として使用するタイプよりも、HDDをメインの記憶として使用することが好ましい。
(3)ハードディスクを用いる場合、ヘッド移動や命令発行時間が必要で、その間の記録ミスを防ぐため、バッファメモリおよび複数のハードディスク搭載が好ましい。
(4)取り込み制御をPC上の専用ソフトウェアで行うことを必須でなくする点からは、PC上のスロットに装着するタイプでない方が好ましい。

温度計測には、CCD素子のアンプ込みの感度むら、ドット抜け、窓材・レンズを通した光量低下、レンズの口径食や周辺光量低下、サンブルの反射率や輻射率等を補正することが好ましい。このため、擬似黒体板の温度を幾つか設定して面全体が均一温度と仮定する方法で構成することが好ましい。同時に赤外線カメラに内蔵する補正用温度板も使用することができる。100度程度の狭い範囲でシュテファン・ボルツマン則が成り立つと仮定して、受光量を温度へ換算することができる。この装置により、高速熱現象や突発的現象の解析が可能となる。例えば、突然の急激な発熱をモニターでき、および/又は周期加熱の時間分解能を増大させることができる。
図36および図37は前提技術4を示すフローチャートで、前提技術1〜3において取り込んだ赤外画像の解析ソフトウェアである。
(1)赤外線カメラからのデジタル信号を受け取り、同期信号部分を除いた部分(画像データの部分)をバッファメモリに格納する。
(2)バッファメモリの内容をHDDの書き込みタイミングにあわせて送出し、HDDに書き込む。
(3)再生時には、外部に接続したPC等に画像データを送出する(この態様においては、取り込みと再生を同時に行わなかった)。
ATAハードディスクドライブ(通常のPC用の接続端子を持ったHDDの意味)。好適な条件としては80Gバイト以上の容量を持つ回転数7200RPM以上のHDDを2台以上使用し、並列に書き込み動作を行うこと。HDDの例としては、IBM社(HITACH1)製 IC35L090AVV が例示できる。
200MHz以上で動作する32BitCPU。実際には、画像取り込みボードにもCPUが搭載されていてもよい(後述するFPGA等)。取り込み装置全体においては、通常のPCに使用するCPUが載っているものを使用可能である(具体名 Intel社 Pentium)。
好適な条件としては、アクセス速度12ns以下の高速スタティツクRAM等が好ましい。具体には、Cypress社製CY7C−1041があげられる。
生体材料の凍結・融解過程の熱解析法として、細胞冷凍保存液中における細胞冷凍の生死判別における高速顕微2次元熱分析の応用例を前提例1として示す。
前提例1に用いた測定試料のサイズは標準で2mm角であるが、1つの細胞サイズとしてはおよそ10×10μm程度を測定対象とした。本前提例1では、熱ショックによる細胞の破壊と物性値の変化に着目し、室温から−20℃程度までの冷却を何サイクルか繰り返した。赤外線カメラ1での計測は、細胞単位での温度計測を冷却固化過程で試みた。
輻射、熱伝導の影響を考慮して設計した専用の冷却容器内のステージに試料を静置し、高速冷却および昇温過程の赤外線画像を2.7ms毎に撮影した。試料温度は、ペルチエ素子を用いて温度制御された冷却容器内の観察用ステージ上に静置した。また、走査温度範囲は室温から−25℃付近である。凍結過程の見かけの冷却速度は約0.1〜1000℃/minで制御可能であるが、典型的には50℃/minとした。温度の絶対値の決定は比較的に困難であるが、黒色塗料を塗布した試料台の温度を一定に保ち、そのときの台に取り付けた白金抵抗温度計の読み値で較正した。

前提例1の測定データとして、ウシ血管内皮細胞培養液中 1回目冷却凝固過程の赤外線画像を図23(a)に、ウシ血管内皮細胞培養液中 2回目冷却擬固過程の赤外線画像を図23(b)に、ウシ血管内皮細胞DMSO10%水溶液中1回目冷却凝固過程の赤外線画像を図24(a)に、ウシ血管内皮細胞DMSO10%水溶液中2回目冷却凝固過程の赤外線画像を図24(b)に示した。
1.培養液、および保存液中の血管内皮細胞について、凍結過程の潜熱発生による湿度上昇を、細胞単位で観測すること、すなわち細胞単位の凍結過程熱分析を可能とした。
2.細胞凍結は、溶液の凍結に対して、いずれの場合も時間遅れを示し、溶液が凍結した後に、細胞単位で凍結した。
3.培養液、保存液の種類により、細胞凍結の様式に明らかな相違が認められた。培養液中での細胞凍結は、第1回目の冷却に限り、細胞ごとに明瞭な発熱を発生させるが、2回目以降は、潜熱発生は認められなかった。一方、保存液中では、細胞ごとの凍結に伴う潜熱発生は、その発生数・強度ともに大幅に減少するが、冷却を繰り返しても、潜熱発生は引き続き観測可能であった。保存液中での、細胞単位の潜熱発生に伴う温度上昇は、培養液中の場合の温度上昇に比べて、約1/10であった。
4.細胞は、細胞内液の凍結による体積膨張で膜がダメージを受けることが知られている。このため保存液が用いられるが、DMSO、グリセリン、エチレングリコール、アルコール類、糖、など水と水素結合的な結びつけをして結晶化を妨げるのが有効とされている。本測定装置では細胞一つ一つについて凍結状況が把握でき、またコントロール(たとえば培養液だけの堀合など)の温度上昇と比較することで、結晶化の阻害の程度をスクリーニングすることができた。
5.昇温過程の融解解析を合わせ、昇温時の再結晶化によるダメージを推定できる事が示された。
厚さ1μmの分子量5000のポリビニルアルコール薄膜の一方の面を金電極、もう一方の赤外線カメラ側をネサガラスとしてはさみ、電界強度を100kV/cmから増大させた。なお、前提技術2を用いて撮影した。
図29は前提例3の結果を示す。
前提例4は、面状試料の一部を交流的(周波数fの三角波)に加熱し温度波を拡散させ、全体の温度変化を赤外線カメラで観測したものである。なお、前提技術2を用いて撮影した。
前提例5は、支持膜上の薄膜の熱拡散率測定を行ったものである。なお、前提技術2を用いて撮影した。
前提例6は、ほぼ平行とみなせる平板試料について、平板試料の裏面全体に電極を塗布し、熱拡散長さを勘案した周波数の温度波を与えるものである。なお、前提技術2を用いて撮影した。
以下に、本発明の実施形態を説明する。
試料温度測定装置の構成
図1は、本実施形態による試料温度測定装置の概念図を示す。この試料温度測定装置100は、大きく分けて、試料固定基板である試料固定容器6と電源7及び試料温度分析装置とにより構成している。前記試料温度分析装置は、赤外線放射温度計(赤外線カメラ)1と、赤外線カメラ2が装着される顕微鏡2と、赤外線カメラ1の撮影データを処理するデータ処理装置3と、試料固定容器6が保持されるステージ4から構成される。
試料固定容器6は、図2に示すように、第2の電極をなす上部電極12と第1の電極をなす下部電極11の間に、スペーサー(第1の電極と第2の電極間に、試料を含む液体等を収容する領域を形成する)13を配置し、複数の微細孔9をアレイ状に形成した絶縁体10をスペーサー13と下部電極11で挟んだ構造を有する。なお、後述するように、微細孔9は、下部電極11上に配置した絶縁膜に一般的なフォトリソグラフィーとエッチングにより形成することができる。
上部電極12は第2の基板であるパイレックス(登録商標)ガラス板5(図2および図6においては図示を省略)に、下部電極11は第1の基板であるサファイアのガラス板20(図2においては図示を省略)に、それぞれITOを成膜したものを用いることができる。
スペーサー13はシリコンシートの中央に試料領域8が開口形成されている。また、図2に示すように、スペーサー13には、試料領域8に試料を導入、排出するために、試料を導入する導入流路16及びそれに連通する導入口14と、試料を排出する排出流路17及びそれに連通する排出口15が設けられている。
複数の微細孔9を有する絶縁体10は、図4に示すフォトリソグラフィーとエッチングによる方法によりガラス板20上の下部電極11に一体形成により作製される。この一体成形は以下の方法により行える。
細胞接着試薬(例えば抗体としての細胞接着試薬、ポリ‐L‐リジン)により微細孔9の中に固定した細胞を微細孔9の底面に接着している。したがって、容器6内の溶媒の温度上昇による熱対流が生じても、あるいは後述するように、細胞を微細孔9から脱離させる方向に作用する負の誘電泳動力が細胞に作用しても、細胞が微細孔9から出る事を抑制できるようにしている。
上部電極12と下部電極11の間に電圧を印加する電源7は、上部電極12と下部電極11の電極間に周波電圧である交流電圧を印加するための交流電源として、信号発生器を用いた。なお周波電圧の波形に特に制限はなく、正弦波、矩形波、台形波等を選択可能であり、1つの微細孔に1つの細胞が誘導されて固定されるように誘電泳動力が作用すればどのような波形でも良い。
本実施形態では、細胞懸濁液をスペーサー13の導入口14より分注器を用いて注入する。その後、例えば約5分間静置し細胞を重力沈降させてから、電源7(交流電源)により、誘電泳動力発生用である第1の周波数で第1の電圧の交流電圧を上部電極12と下部電極11の間に第1の時間(t1)印加する。この誘電泳動力用の交流電圧の印加により、微細孔9への電解集中により正の誘電泳動力が作用し、アレイ状に形成した複数の微細孔9に対し、1つの微細孔9に略1個の細胞を固定することができる。なお、誘電泳動力用の交流電圧の第1の周波数としては、例えば100kHz以上の高い周波数の場合、細胞には正の誘電泳動力が作用し、細胞は電界集中が生じている微細孔に向かって動き固定される。
生体研究では、単一細胞レベルでの熱分析、例えば生死判定への応用が可能である。試料となる生体材料は、水を含む系で導電性があること、薄い(小さい)試料であることが好ましい。また、細胞間の個体差を熱的に観測する方法への展開も可能である。
上部電極12は、縦70mm×横40mm×厚さ1mmのパイレックス(登録商標)ガラス板5に、下部電極11には縦70mm×横40mm×厚さ0.42mmのサファイアガラス板20に、それぞれITOを成膜(膜厚150nm)したものを用いた。
スペーサー13は、縦40mm×横40mm×厚さ1.5mmのシリコンシートの中央を縦20mm×横20mmの開口を形成するようにくりぬいて試料領域8を形成した。
下部電極部24には、以下の処理により、細胞接着試薬であるポリ−L−リジンをコーティングした。すなわち、ポリ−L−リジン水溶液(0.01wt%)に下部電極部24を約5分間浸し、微細孔9の底面の下部電極11および絶縁体10にポリ−L−リジンを物理吸着させた後、純水で洗浄した。このようにする事で、微細孔9の中に固定した細胞が微細孔9の底面に接着し、容器6内の溶媒の温度上昇による熱対流が生じても、あるいは後述するように、細胞を微細孔9から脱離させる方向に作用する負の誘電泳動力が細胞に作用しても、細胞が微細孔9から出る事を抑制できる。
上部電極12と下部電極11の間に電圧を印加する電源7を構成する信号発生器として、エヌエフ回路設計ブロック製、WF1966を用いた。
試料温度分析装置は、赤外線カメラ1として、InSb、320×256FPA素子、波長域3−5μmを使用し、赤外線顕微鏡2として10倍光学系を用いた。
下部電極部24を備えた試料固定容器6、電源7及び試料温度分析装置を用いて、後述する実験を行った。まず、マウスミエローマ細胞を、300mMの濃度のマンニトール水溶液に懸濁させ、0.74×106個/mLの密度になるように細胞懸濁液を調整した。
第2の周波数として1Hzの交流電圧を印加した場合、微細孔9に固定された細胞は電圧印加直後から電圧印加前後の温度変化を測定したところ、赤外線顕微鏡では、図5に示すように、1Hzの交流電圧印加直後に振動を伴った急激な輻射強度の上昇が観察された。また印加した交流電圧とほぼ同周期で温度(輻射率)振動が観察され、1周期あたりの温度上昇は約1℃であった。さらに、数回の温度振動を繰り返しながら平均温度は上昇し、ある一定値に達すると変動しなくなった。
細胞を添加しない場合について、実施例1と同様に、試料固定容器6における微細孔9内の温度変化を測定した。まず、300mMの濃度のマンニトール水溶液を調整し、600μLをスペーサー13の導入口14より1mL容量の分注器を用いて注入した。その後、約5分間静置した後、交流電源により誘電泳動力発生用の第1の電圧20Vpp、第1の周波数3MHzの交流電圧を上部電極12と下部電極11の間に2分間印加した。
実施例1と同様に、まず、マウスミエローマ細胞を、300mMの濃度のマンニトール水溶液に懸濁させ、0.74×106個/mLの密度になるように細胞懸濁液を調整した。

(6)化学反応、潜熱等での発熱の周囲への拡散過程
(7)応力下での材料の変形または破壊に伴う吸発熱の観察
(8)物質表面からの水の蒸発過程の観察
(9)微粒子1個ごとの熱的特性の測定及び熱的解析

Claims (10)
- 液体中の微粒子からなる試料が入り込むサイズの試料固定領域を有する試料固定基板と、
前記試料を前記試料固定基板に固定する試料固定手段と、
前記試料固定基板に固定された試料に温度変化を与える温度変化手段と、
前記温度変化手段により温度変化が与えられている試料を赤外線像として拡大する赤外線像拡大手段と、
前記赤外線像拡大手段で拡大した赤外線像を測定する赤外線測定手段と、
前記赤外線測定手段で測定した赤外線像データを取り込み、赤外線像データに基づいて前記試料の熱的特性を測定するデータ処理手段と、
を備え、
前記試料固定基板は、
前記試料固定領域をなす多数の微細孔が厚み方向に貫通形成された絶縁体と、
上面に第1の電極が形成され、前記第1の電極に前記絶縁体の下面が当接する第1の基板と、
前記第1の電極と離隔対向して前記絶縁体を挟んで配置した第2の電極と、を有していて、前記第1の基板と前記第1の電極を赤外線に対して透明とし、
さらに、前記温度変化手段は、前記第1の電極と前記第2の電極間に電圧を印加することで、前記試料に温度変化を与える温度変化用の電圧印加手段であることを特徴とする試料温度測定装置。 - 前記赤外線測定手段は、赤外線カメラであることを特徴とする請求項1に記載の試料温度測定装置。
- 前記試料固定手段は、前記第1の電極と前記第2の電極間に電圧を印加することで、前記試料に誘電泳動力を付与する誘電泳動力発生用の周波電圧印加手段であることを特徴とする請求項1または2に記載の試料温度測定装置。
- 前記温度変化用の電圧印加手段は、試料に応じて予め決められた時間だけ電圧を前記試料に印加することを特徴とする請求項1から3のいずれかに記載の試料温度測定装置。
- 前記試料固定基板の試料固定領域を数μm径の試料が入り込む孔により形成し、前記データ処理手段は、前記赤外線測定手段としての赤外線カメラにより撮影した前記試料の赤外線像を可視化熱分析することを特徴とする請求項1から4のいずれかに記載の試料温度測定装置。
- 液体中の微粒子からなる試料に誘電泳動力を与えることにより、試料固定基板に設けた前記試料が入り込むサイズの試料固定領域に前記試料を誘導して固定し、さらに前記試料固定領域に固定された前記試料に対して温度変化を与えながら前記試料の赤外線像を赤外線像拡大手段を通して赤外線測定手段により測定する試料温度測定方法であって、
前記試料に対して温度変化を与える手段は、前記試料固定領域を挟んで配置した第1の電極と第2の電極と、前記第1の電極と前記第2の電極に電圧を印加する電圧印加手段であることを特徴とする試料温度測定方法。 - 前記赤外線測定手段により測定した赤外線像データに基づいて、前記試料の熱的特性を測定することを特徴とする請求項6に記載の試料温度測定方法。
- 前記試料が溶液中に存在する細胞であることを特徴とする請求項6または7に記載の試料温度測定方法。
- 前記温度変化用の電圧印加手段により前記第1の電極と前記第2の電極に予め決められた時間だけ電圧を印加することを特徴とする請求項6から8のいずれかに記載の試料温度測定方法。
- 前記試料固定基板の試料固定領域を数μm径の試料が入り込む孔により形成し、前記赤外線測定手段としての赤外線カメラにより撮影した前記試料の赤外線像を可視化熱分析することを特徴とする請求項6から9のいずれかに記載の試料温度測定方法。
