Description
本発明は、胃炎、胃潰瘍、膵臓炎、大腸炎、関節炎、糖尿病、動脈硬化、腎臓炎、肝炎、アルツハイマー性痴呆、パーキンソン病、ルーゲーリック病などの炎症性疾患で現われる炎症と細胞損傷の治療または予防に有用な薬学組成物、及びこのような薬学組成物を用いる治療または予防方法に関する。
炎症は、損傷を受けた細胞から由来する損傷因子と外来物質に対する血管反応及び細胞反応である。炎症反応を媒介する物質としては、1)アラキドン酸(arachidonic acid)の代謝物質であるプロスタグランジン(prostaglandin)、ロイコトリエン(leukotriene)及びリポキシン(lipoxins)、2)血小板活性化因子(platelet activation factor)、3)腫瘍壊死因子α(tumor necrosis factor−alpha)、インターロイキン‐1(IL‐1)などのサイトカイン(cytokines)とmonocyte chemoattractant protein(MCP‐1)、macrophage inflammatory protein‐1alpha(MCP‐1alpha)などのケモカイン(chemokines)、4)一酸化窒素(NO)、5)活性酸素、6)ヒスタミン(histamine)、セロトニンなどのような血管拡張因子などが知られている。炎症反応の主な目的は、外来物質及び損傷を受けた細胞と組織を除去することであるが、炎症反応はリューマチ性関節炎及び動脈硬化のような慢性疾患を誘発する原因になる。
したがって、本発明の技術的課題は、胃損傷、心血管障害などの副作用がなく安全であって炎症疾患の治療または予防に優れる薬学組成物、このような薬学組成物の炎症疾患治療または予防用途、及びこのような薬学組成物を投与することを含む炎症疾患治療または予防方法を提供することである。

また、本発明の望ましい一実施例である2‐ヒドロキシ‐5‐[2‐(4‐トリフルオロメチル‐フェニル)‐エチルアミノ]‐安息香酸(化合物2)は、以下のような反応式3によって製造することができるが、これに限定されることはない。
図1は、化合物2が胃の粘膜損傷を誘発せず安全であることを示す実験結果である。胃粘膜の損傷程度を確認するために、直接薬物を容量別に経口投与した。比較群としてアスピリンを使用した。

以下、本発明をより具体的に説明するために実施例などを挙げて説明する。しかし、本発明による実施例は多くの他の形態に変形することができ、本発明の範囲が後述する実施例に限定されると解釈されてはならない。本発明の実施例は、本発明の具体的理解を助けるために例示的に提供されるものである。
5‐アミノサリチル酸(0.51g、3.90mmol)を常温窒素気流下でN,N‐ジメチルホルムアミド(20.0ml)に入れて撹拌させた。トリエチルアミン(0.50ml)とヨウ化テトラブチルアンモニウム(10.1mg)を入れた後、30分間撹拌した。その後、1‐(2‐(ブロモエチル)‐4‐トリフルオロメチル)ベンゼンを添加し、常温で3時間撹拌した。反応を終結するために反応容器に氷を添加した。生成された結晶を濾過してアセトンとヘクサンを用いて撹拌させた後、再び濾過した。濾過された固体をエチルアセテートに溶かした後、0.5N塩酸と塩水で洗滌した。無水硫酸マグネシウムで乾燥した後、減圧蒸留して2‐ヒドロキシ‐5‐(4‐(トリフルオロメチル)ペニルエチルアミノ)安息香酸0.54g(21%収率)を得た。

前記合成例1で製造した2‐ヒドロキシ‐5‐[2‐(4‐トリフルオロメチル‐フェニル)‐エチルアミノ]‐安息香酸(10g、30.7mmol)を無水エタノール(100ml)に加えて50℃まで加温して完全に溶解させた。その後、溶液を0℃に冷却した。85%水酸化カリウム(2.03g、30.7mmol)と無水エタノール(20ml)溶液を使ってpHを6.8〜7.0に調節した後、室温で2時間撹拌した。沈殿した結晶を濾過及び乾燥して目的とする化合物である2‐ヒドロキシ‐5‐(4‐(トリフルオロメチル)ペニルエチルアミノ)安息香酸カリウム塩(10.4g)を93%の収率で得た。
<合成例3>2‐ヒドロキシ‐5‐[2‐(4‐ニトロペニル)エチルアミノ]‐安息香酸の製造
5‐アミノサリチル酸(500mg、3.26mmol)及び4‐ニトロペニルエチルブロマイド(900mg、3.92mmol)を使って合成例1と同様の方法によって、2‐ヒドロキシ‐5‐[2‐(4‐ニトロペニル)エチルアミノ]‐安息香酸890mg(50%収率)を淡い黄色固体相で得た。融点234〜236℃。
5‐アミノサリチル酸(500mg、3.26mmol)及び3‐(4‐ニトロペニル)プロピルブロマイド(950mg、3.92mmol)を使って合成例1と同様の方法により、2‐ヒドロキシ‐5‐[3‐(4‐ニトロペニル)‐n‐プロピルアミノ]‐安息香酸520mg(50%収率)を淡い黄色固体相で得た。融点229〜231℃。
C16H16N2O5の元素分析
他の化合物も前記合成例1と同様の方法で製造し、分析結果を下記表3にまとめて示した。
化合物18を下記反応式によって製造した。
化合物b:化合物aのジクロロメタン(200mL)溶液にN,N‐ジイソプロピルエチルアミン(80.3mL、461mmol)と塩化アセチル(21.9mL、307mmol)を0℃で添加した。反応混合物は室温まで加温し、5時間撹拌した。1N HCl(100mL)水溶液を反応混合物に添加した。反応物は層に分離され、水層はジクロロメタン(100mL)で抽出した。有機相を混合して塩水(100mL)で洗滌し、MgSO4で乾燥して濾過した後、濃縮した。粗生成物をジエチルエーテルで再結晶して白い固体相の化合物22(51.5g、71.7%)を生成した。
化合物18:化合物b(51.5g、110mmol)のジクロロメタン(200mL)溶液を0℃で4N HClの1,4‐ジオキサン溶液(200mL)で処理した。反応混合物は室温まで加温された。5時間撹拌した後、懸濁液は濃縮した。残渣はジエチルエーテル(500mL)で粉砕(triturate)した。ジクロロメタン(500mL)とジエチルエーテル(500mL)を用いて濾過及び洗滌し、白い固体相の化合物18(46.0g、82.5mmol、92.3%)を得た。
1−1.NO生成阻害効果
炎症反応に関与する炎症性因子であるNOに対する本発明の2‐ヒドロキシ安息香酸誘導体の阻害効果を評価した。BV2 ミクログリア細胞株にバクテリアトキシン炎症誘発物質であるリポ多糖体(LPS)と本発明の化合物をそれぞれ10uM、30uM、または100uMずつ同時に処理して24時間培養した。その後、培養液50ulを96ウェルプレートに採り、グリーズ試薬(Griess reagent)を50ulずつ加えて10分間室温で反応させた。その後、ELISAリーダーで540nmにおける吸光度を測定した。各化合物のBV2細胞株から出たIC50の結果は、下記表4に「NO in BV」の欄に示した。
BV2細胞株で2‐ヒドロキシ安息香酸誘導体の濃度別iNOS遺伝子の発現を観察した。BV2細胞株を1×106個になるように100mmディッシュに接種し、炎症誘導物質であるLPSと2‐ヒドロキシ安息香酸誘導体をそれぞれ1〜100uM濃度範囲で一定濃度間隔で同時に処理した。24時間培養した後、RNAを抽出した。抽出したRNAは逆転写酵素反応(reverse transcription)及び重合酵素反応(polymerase chain reaction)を行った後、TNF‐α、IL‐1β、及びiNOS遺伝子の発現様相を比べた。その結果を下記表4にまとめて示した。
BV2細胞株で2‐ヒドロキシ安息香酸誘導体のサイトカイン生成の抑制効果を観察した。BV2細胞株を1×106個になるように24ウェルプレートに接種し、炎症誘導物質であるLPSとそれぞれ100uMの2‐ヒドロキシ安息香酸誘導体または比較薬物とを同時に処理した。24時間培養した後、培養液を採った。培養液はELISA法により、サイトカインであるTNF‐αとIL‐6の量を測定した。その結果を下記表5にまとめて示した。
2‐ヒドロキシ安息香酸誘導体化合物の単回毒性を評価し、その結果を図32に示した。
従来のNSAID系消炎剤は胃粘膜損傷を起こす副作用があったため、抗炎症効果のある化合物2も胃粘膜損傷を誘発するか否かを実験した。比較群として30、100、及び300mg/kgのアスピリンを経口投与したときに発生する胃粘膜損傷を用いた。本発明による一実施例である化合物2は、かなり多い容量である1000mg/kgの経口投与群でも胃粘膜損傷を誘発せず、本発明の2‐ヒドロキシ安息香酸誘導体は非常に安全であることが分かった(図1)。
本発明による一実施例である2‐ヒドロキシ安息香酸誘導体の細胞保護効果を評価した。AGS細胞株(human gastric cancer cells)を96ウェルプレートに1×105個接種した。その後、それぞれにヘリコバクターを24時間処理(図3)、スリンダク(sulindoc)を16時間処理(図4)、H2O2を8時間処理(図5)、またはエタノールを24時間処理(図6)して胃粘膜細胞の死滅を誘導した。また、一部AGS細胞株には、本発明の一実施例である化合物2をそれぞれ10uM、30uM、または100uMずつ同時に処理して24時間培養した。MTT溶液をそれぞれのウェルに添加した後、4時間37℃で反応させた。その後、ELISAリーダーで540nmにおける吸光度を測定し、胃粘膜細胞の生存率を計算した。その結果、1uM及び10uMの化合物2では50%以上の細胞保護効果を見せ、100uMの化合物2は大半の細胞死滅モデルで70%以上の細胞保護効果を見せた。
5−1.アルコール及び塩酸誘導胃炎
本発明の2‐ヒドロキシ安息香酸誘導体がアルコール及び塩酸誘導胃炎に及ぶ効果を評価した。200〜250gの雄SDマウスを24時間絶食させた後、60% EtOH/150mM HClを経口投与して胃腸管出血を誘導した。胃腸管出血の30分前に容量別に化合物2を経口投与し、胃腸管出血の90分後に胃を摘出して胃損傷の程度を観察した。その結果、本発明による一実施例である化合物2は胃腸管出血を防御し、毒性もなかった。その結果を図8にまとめて示した。
本発明の2‐ヒドロキシ安息香酸誘導体がアルコール性胃炎に及ぼす効果を評価した。250gのマウスを24時間絶食させた後、アルコールを経口投与すれば胃腸管出血が起きるが(B)、化合物2を1時間前に前処理する場合(C)胃の出血が著しく減少した。その結果を図9のDに数値化して示した。このような結果から、本発明の化合物2はアルコール性胃炎に強力な保護作用を示すと言える。
本発明の2‐ヒドロキシ安息香酸誘導体が、NSAID誘導胃炎に及ぼす効果を評価した。NSAIDであるインドメタシンを6時間投与し胃損傷を弱く誘発させたモデル(図10のA)、及びインドメタシンを12時間投与してかなりの胃損傷を誘発させたモデル(図10のC)で、化合物2は卓越な胃保護効能を見せた(図10のB及びD)。
6−1.カラゲナン誘導炎症モデル:鎮痛効果の評価
本発明の2‐ヒドロキシ安息香酸誘導体の一例である化合物2が持つ関節炎モデルにおける鎮痛効能を評価した。2%カラゲナンを白いマウスの左側足部に注射した後、3〜300mg/kgの化合物2を経口投与してカラゲナンによって誘導される関節炎モデルにおける温熱性痛覚過敏症に及ぼす化合物2の影響を観察した。その結果を図10に示した。
ザイモサンを用いて本発明の2‐ヒドロキシ安息香酸誘導体の一例である化合物2が持つ関節炎モデルにおける抗炎症効能を評価した。1%ザイモサン0.5mlをエアパウチ内に注入し、ザイモサン投与の1時間前に3〜300mg/kgの化合物2を単回投与した。4時間後に関節炎の主な症状である浮腫と病因であるサイトカインの濃度を観察した。その結果、化合物2は30mg/kgから対照群に比べてTNF‐αの量を減少させる効果を示し、300mg/kgまで濃度依存的に減少させた。比較群として炎症疾患治療剤で知られているイブプロフェン(50mg/kg)を使った(図12)。IL‐1αも同様に濃度依存的に減少した(図13)。このような結果から本発明の2‐ヒドロキシ安息香酸誘導体は、炎症疾患、特に関節炎に効果があることを把握することができる。
本発明による一実施例である2‐ヒドロキシ安息香酸誘導体の有効性を評価するために、リュウマチ性関節炎動物モデルであるコラーゲン誘導関節炎モデルを使った。Bovine type IIコラーゲンをcomplete Freund’s adjuvantと混ぜて乳剤を製造した。8〜10週のDBA/1LacJマウスのしっぽの基始部に前記乳剤を皮内注射した。2週後に同様の方法で皮内注射した。2次コラーゲン皮内注射の1週間後、腹腔に化合物2、3、及び18を25mg/kg/day、比較群としてメトトレキサート1mg/kg/weekを注射し、対照群として10%ビヒクルを注射した(図15)。図14に、正常マウス(sham)、コラーゲンを投与したマウス(collagen)、及び化合物18を投与したマウス(化合物18、化合物18を25mg/kg/day注射)の足浮腫を撮影した写真を示した。

炎症性腸疾患モデルとして5%デキストラン硫酸ナトリウム(DSS)モデルを使用した。5%DSSを飲み水に混ぜて経口投与した。5%デキストラン硫酸ナトリウム(DSS)モデルは上皮細胞に損傷を誘発し、主に左側大腸の潰瘍と大腸長さの縮小などが観察される。このようなモデルは投与容量に応じて一定程度の炎症が誘発される比較的簡単且つ再現性の高いモデルである。したがって、新しい薬剤の効能評価に多く使用される。
急性膵臓炎モデルとして、セルレイン誘導膵臓炎モデルを使用した。セルレインはコレシストキニンの類似体であって、膵臓で膵臓消化酵素の分泌を誘発するホルモンである。腹腔注射でセルレイン(50μg/kg)を投与することで膵臓に消化酵素を過度に分泌させた。2時間が経過した後、LPSを投与することで浮腫を誘発させた。セルレイン投与時、脾臓を基準にして全体的に浮腫が過度に起きたことが観察された。また、25mg/kgの化合物18を投与したとき、その浮腫を減少させることを確認することができた(図21)。図21のDは、このような浮腫によって増加する膵臓の重さを測定した結果である。化合物18の投与群は、膵臓の重さを有意義に減少させることを確認することができる。
9−1.ELISA分析によるサイトカインの減少効果
3.5ヶ月から10.5ヶ月までのAPP/PS1痴呆マウスに、化合物2を25mg/kg/dayあるいはイブプロフェン62.5mg/kg/dayを飼料に混合して長期投与した。総7ヶ月間薬物を投与した後、ELISAを通じてTNF‐α、IL‐1β、及びIL‐6の量を定量的に分析した(mean±SEM、n=3−5)。図25において「*」は、Student−Newman−Keuls testによる一元変量分析(One−way ANOVA)時、一般飼料のみを供給したAPP/PS1痴呆マウスに比べてp<0.05の有意差を見せることを示す。
アルツハイマー病で脳血管損傷が発生することは広く知られている。脳血管障壁の損傷に対する化合物2と比較群であるイブプロフェンの効果を評価した。5ヶ月齢のAPP/PS1痴呆マウスに2% Evans blue dye 3ml/gを血管に注入した。薬物は、生後2ヶ月から5ヶ月まで化合物2(25mg/kg/day)または比較薬物であるイブプロフェン(62.5mg/kg/day)を飼料に混合して投与した。その結果、図26のBと図26のFのように、正常マウスに比べてcortex、hippocampus、及びthalamusでEvans blue dyeの透過度が増加することを確認することができた。また、図26に示されたように、化合物2の投与群(図26のC及びG)及びイブプロフェンの投与群(図26のD及びH)で脳血管障壁の保護効果を確認することができた。
チオフラビン色素染色法を用いて本発明の2‐ヒドロキシ安息香酸誘導体が痴呆に及ぼす影響を評価した。総7ヶ月間(3.5〜10.5ヶ月齢APP/PS1)化合物2を25mg/kg/dayずつ投与した。化合物2の投与群は一般飼料のみを供給したAPP/PS1痴呆マウスに比べてアミロイドプラークの量が約53%減少した(図27)。また、総4ヶ月間(8.5〜12.5ヶ月齢APP/PS1)化合物1を25mg/kg/day投与した群は、一般飼料のみを供給したAPP/PS1痴呆マウスに比べてアミロイドプラークの量が約49.3%減少した。
3.5ヶ月から10.5ヶ月までのAPP/PS1痴呆マウスに化合物2を25mg/kg/day飼料に混合して長期投与した。総7ヶ月間薬物を投与した後、行動改善効果を検証するために高架式十字迷路テストを行った。高架式十字迷路は2つのopen arms(30cm×6cm×0.5cm)と2つのclosed arms(30cm×6cm×15cm)で構成され、6cm×6cmのcenter platformがある。高架式十字迷路テストにおいて、中央地点にopen armsに向かって慎重にマウスをおいた後、5分間実験動物がopen armに留まる時間を測定して分析した(mean±SEM、n=3−5)。図28において「*」は、Student−Newman−Keuls testによる一元変量分析時、APP/PS1痴呆マウスに比べてp<0.05の有意差を見せることを示す。
10−1.ミクログリア活性化の減少効果
退行性脳疾患の1つであるALS(amyotrophic lateral sclerosis)における薬物効果を測定するために、実際患者と類似の病理的特徴を持つ形質組み替え動物モデルG93A(Glycine⇒Alanine)マウスを用いた。
一般飼料のみを提供した16週齢のG93Aマウス及び5mg/kg/dの化合物2投与群の脊髄部位を摘出した。RNAを分離した後、RT−PCRを通じて炎症性サイトカインであるTNF‐α及びIL‐1β mRNAの発現程度を確認した。その結果、化合物2の投与群は炎症性サイトカインを有効に減らすことが観察できた(図30)。
C57/BL6マウス(雄/8週齢)にMPTP(40mg/kg)のみを皮下注射するか、またはMPTPを注射する30分前から化合物2を50mg/kgずつ毎日腹腔注射した。2日後に脳組織を摘出した。脳組織をCD11b免疫染色し、反応させた後、DAB(diaminobenzidine)で発色させた。光学顕微鏡でミクログリア(脳疾患モデルにおける炎症の1つのマーカー)の活性化程度を観察した(図31)。
本発明の2‐ヒドロキシ安息香酸誘導体は、細胞培養実験によってアルコール誘導、ヘリコバクター・ピロリ誘導、NSAID誘導、及びH2O2誘導された細胞の死滅を効果的に防止した。本発明の化合物の効果は従来の胃炎などの細胞防御効果を持つ薬物であるムコスタまたはPPIに比べて卓越であった。また、本発明の化合物は動物モデルにおける実験結果から、アルコール誘導による胃炎などの治療に優れた効果を示し、消炎鎮痛剤として使われているNSAIDによる胃炎を緩和した。アスピリンと比較実験した結果、アスピリンよりさらに高容量を投与したマウスの胃で出血を誘導せず、安全性が確保された。したがって、本発明の2‐ヒドロキシ安息香酸誘導体は胃炎の治療及び予防に非常に有効である。
シクロオキシゲナーゼ(COX)の産物であるプロスタグランジンとリポオキシゲナーゼの産物であるロイコトリエン(LTB4)は、潰瘍性大腸炎のような炎症性疾患の炎症反応で重要な役割を占めると知られている。5‐リポオキシゲナーゼ(5‐LOX)の抑制剤であるジレウトン(zileuton)とNSAIDであるスルファサラジンは、腸損傷動物モデルにおける炎症の程度を現わす指標として使われるMPO(myeloperoxidase)の活性を減少させた(Singh VP et al.、Indian J Exp Biol.2004;42(7):667−73)。また、COX‐2選択的抑制剤であるニメスリド(nimesulide)は、2つの異なる(酢酸誘導及びLTB4誘導IBD)腸損傷動物モデルにおいて保護効果を見せた。ニメスリドは炎症反応においてMPO活性を著しく抑制した(Singh VP et al.、Prostaglandins Other Lipid Mediat.2003;71(3−4):163−75)。したがって、抗炎症効果のある本発明の化合物は大腸炎の治療に非常に有効に利用することができる。
本発明の2‐ヒドロキシ安息香酸誘導体は、コラーゲン誘導関節炎モデルにおいて従来関節炎治療剤として処方されているが副作用のあるメトトレキサートに比べ、ほぼ同じかまたは多少優れた効果を見せた。また、本発明の化合物はカラゲナン誘導痛覚過敏症に対する鎮痛緩和効果及びザイモサン誘導炎症性サイトカインの生成を抑制した。したがって、本発明の化合物は従来の炎症治療剤として使われる薬物と同等の効能を持ちながらも、安全性が確保されたリュウマチ性関節炎治療剤として使用することができる。
急性膵臓炎は、膵液の消化酵素または胆石症による胆汁が膵臓内に逆流して膵臓組織を自己消化して生じる炎症である。少しの浮腫からひどい出血まで多様な症状を現わし、これにより膵臓のさまざまな損傷をもたらす。膵臓炎は炎症と関連があるという報告が数多くあり、COX抑制剤による膵臓炎モデルの保護効果、及び炎症マーカーであるTNF‐α及びプロスタグランジン生成の抑制効果が報告された(Song AM et al.、Am J Physiol Gastrointest Liver Physiol.2002;283(5):G1166−74)。本発明の化合物は膵臓炎の治療に非常に有効に利用することができる。
第2型糖尿病で抗炎症及び鎮痛の役割が重要視されている。さらに、抗炎症効果を持っている多くの治療剤が、第2型糖尿病の兆候を減少させるか又は発病時点を遅らせるという証拠が報告されている(Deans KA et al.、Diabetes Technol Ther.2006;8(1):18−27)。抗炎症化合物であるリソフィリン(lisofylline)は、ストレプトゾトシン投与マウスにおいてIFN‐γとTNF‐αを抑制することで糖尿の症状を50%以上減少させた(Yang Z et al.、Pancreas.2003;26(4):e99−104)。したがって、抗炎症効果のある本発明の化合物は糖尿病性炎症及び痛症の治療及び/または予防に非常に有効に利用することができる。
アポリポタンパク質E‐欠乏(apoE(−/−))マウスの初期アテローム性動脈硬化症(atherosclerotic lesion)において、選択的なCOX‐2抑制剤(例えば、ロフェコキシブ及びNS‐398)と非選択的なCOX抑制剤(例えば、インドメタシン)はアテローム性動脈硬化症をそれぞれ約35〜38%及び約38〜51%減少させた(Burleigh ME J Mol Cell Cardiol.2005 Sep;39(3):443−52)。したがって、抗炎症効果のある本発明の化合物は、動脈硬化の治療に有効に利用することができる。
アルツハイマー性痴呆は痴呆の原因のうち最も一般的な形態である。病理組織学的には、神経繊維のもつれ、アミロイドプラーク、深刻な神経細胞の死滅などがアルツハイマー性痴呆の特徴である。
近年、アルツハイマー性痴呆が炎症と関連があるという論文が数多く報告されている。痴呆動物モデルでミクログリア及び炎症性サイトカインの生成が増加するという報告が多い(Minghetti L.Current Opinion in Neurology 2005、18:315−321)。また、炎症を抑制する薬物の投与はアルツハイマー性痴呆動物モデルで保護効果を奏し得ると知られている(Townsend KP and Pratico D.FASEB J.2005;19(12):1592−601)。
ルーゲーリック病は、筋萎縮性側索硬化症(ALS:amyotrophic lateral sclerosis)、運動(うんどう)ニューロン疾患(MND:motor neuron disease)などと呼ばれる病気であり、運動神経細胞が退行性変化によって漸次損傷することがこの疾患の特徴である。
また、ALSが炎症と関連があるという論文が数多く報告されているが、ALS動物モデルであるG93Aマウスでミクログリア及び炎症性サイトカインの生成が増加するという報告が多く(Weydt P and Moller T.Neuroreport.2005、25;16(6):527−31)、炎症を抑制する薬物の投与はALS動物モデルにおける保護効果を奏し得ることも知られている(West M et al.、J Neurochem.2004;91(1):133−43)。
したがって、本発明の化合物はルーゲーリック病の治療に有効に利用することができる。
パーキンソン病は、黒質に存在するドーパミン神経細胞の死滅が伴われ、振戦、筋肉硬直、非正常的姿勢、運動不能などの多様な症状を現わす退行性神経系疾患である。
パーキンソン病が炎症と関連があるという論文が数多く報告されている。パーキンソン病でミクログリア及び炎症性サイトカインの生成が増加するという報告が多く(Gao HM、Trends Pharmacol Sci.2003;24(8):395−401;Minghetti L.Curr Opin Neurol.2005;18(3):315−21)、炎症を抑制する薬物の投与はパーキンソン病動物モデルにおける保護効果を奏すると知られている(Gao HM、Trends Pharmacol Sci.2003;24(8):395−401)。
本発明は、炎症疾患の治療または予防に有用な前記化学式1で表れる2‐ヒドロキシ安息香酸誘導体又はこれの薬学的に許容可能な塩を含むことを特徴とする薬学組成物、及びこのような薬学組成物を用いる炎症疾患の治療または予防方法を提供する。本発明による薬学組成物は、胃炎、大腸炎、関節炎、糖尿病炎症、膵臓炎、動脈硬化、腎臓炎、肝炎、アルツハイマー性痴呆、ルーゲーリック病、パーキンソン病などのような炎症疾患の治療または予防に非常に有効であるだけでなく、安全性に優れる。
Claims (3)
- 前記2‐ヒドロキシ安息香酸誘導体は、2‐ヒドロキシ‐5‐フェネチルアミノ‐安息香酸、2‐ヒドロキシ‐5‐[2‐(4‐トリフルオロメチル‐フェニル)‐エチルアミノ]‐安息香酸、2‐ヒドロキシ‐5‐[2‐(3‐トリフルオロメチル‐フェニル)‐エチルアミノ]‐安息香酸、5‐[2‐(3,5‐ビス‐トリフルオロメチル‐フェニル)‐エチルアミノ]‐2‐ヒドロキシ‐安息香酸、2‐ヒドロキシ‐5‐[2‐(2‐ニトロ‐フェニル)‐エチルアミノ]‐安息香酸、5‐[2‐(4‐クロロ‐フェニル)‐エチルアミノ]‐2‐ヒドロキシ‐安息香酸、5‐[2‐(3,4‐ジフルオロ‐フェニル)‐エチルアミノ]‐2‐ヒドロキシ‐安息香酸、5‐[2‐(3,4‐ジクロロ‐フェニル)‐エチルアミノ]‐2‐ヒドロキシ‐安息香酸、5‐[2‐(4‐フルオロ‐2‐トリフルオロメチル‐フェニル)‐エチルアミノ]‐2‐ヒドロキシ‐安息香酸、5‐[2‐(2‐フルオロ‐4‐トリフルオロメチル‐フェニル)‐エチルアミノ]‐2‐ヒドロキシ‐安息香酸、2‐ヒドロキシ‐5‐[2‐(4‐メトキシ‐フェニル)‐エチルアミノ]‐安息香酸、2‐ヒドロキシ‐5‐(2‐o‐トリル‐エチルアミノ)‐安息香酸、2‐ヒドロキシ‐5‐(3‐フェニル‐プロピルアミノ)‐安息香酸、2‐ヒドロキシ‐5‐[3‐(4‐トリフルオロメチル‐フェニル)‐プロピルアミノ]‐安息香酸、5‐[3‐(4‐フルオロ‐フェニル)‐プロピルアミノ]‐2‐ヒドロキシ‐安息香酸、5‐[3‐(3,4‐ジクロロ‐フェニル)‐プロピルアミノ]‐2‐ヒドロキシ‐安息香酸、2‐ヒドロキシ‐5‐(3‐p‐トリル‐プロピルアミノ)‐安息香酸、2‐アセトキシ‐5‐[2‐(4‐トリフルオロメチル‐フェニル)‐エチルアミノ]‐安息香酸、及び5‐[2‐(2‐クロロ‐フェニル)‐エチルアミノ]‐2‐ヒドロキシ‐安息香酸からなる群より選択されたいずれか1つ以上であることを特徴とする請求項1に記載の炎症疾患の治療または予防用薬学組成物。
- 前記炎症疾患は、胃炎、大腸炎、膵臓炎、関節炎、糖尿病炎症、炎症性の痛症、腎臓炎、肝炎、大腸炎、胃潰瘍、及び動脈硬化、アルツハイマー性痴呆、パーキンソン病、もしくはルーゲーリック病において引き起こされる炎症からなる群より選択されたいずれか1つであることを特徴とする請求項1に記載の炎症疾患の治療または予防用薬学組成物。
