Description
本出願は2008年6月13日に出願された米国仮特許出願第61/061,159号の利益を主張し、その内容は参照により本願明細書に引用されたものとする。
Claims (19)
- 細胞物質からRNAを抽出するための試薬であって、
(a)1pg/μL〜10pg/μLのヘパリン;
(b)細胞タンパク質のジスルフィド結合を還元するのに効果的な1mM〜10mMの還元剤;
(c)RNA二次及び三次構造を破壊するのに十分な量のキレート剤;
(d)トリス塩基、トリス-HCl、ビス-トリス塩基、ビス-トリス-HCl及びHEPESからなる群より選ばれる1mM〜100mMの緩衝剤;及び
(e)1mM〜100mMのアルカリ金属ハロゲン化物
を含む試薬。 - 前記ヘパリンが前記試薬の1pg/μL〜3pg/μLを構成する請求項1記載の試薬。
- 前記還元剤が前記試薬の1mM〜5mMを構成する請求項1記載の試薬。
- 前記還元剤がジチオスレイトール、β-メルカプトエタノール及びトリス(2-カルボキシ-エチル)ホスフィンからなる群より選ばれるメンバーである請求項1記載の試薬。
- 前記キレート剤がエチレンジアミン四酢酸;エチレングリコール四酢酸;クエン酸;サリチル酸;エチレンジアミン四酢酸、エチレングリコール四酢酸、クエン酸及びサリチル酸の塩;フタル酸;ヒスチジン;ヒスチジノール二塩酸塩;及び8-ヒドロキシキノリンからなる群より選ばれるメンバーである請求項1記載の試薬。
- 前記キレート剤がエチレンジアミン四酢酸である請求項1記載の試薬。
- 前記キレート剤が前記試薬の0.3mM〜3mMを構成する請求項1記載の試薬。
- 前記緩衝剤が、前記試薬のpHを7.5〜8.0に維持する前記試薬の量を構成する請求項1記載の試薬。
- 前記アルカリ金属ハロゲン化物が塩化ナトリウム、塩化カリウム及び塩化リチウムからなる群より選ばれる請求項1記載の試薬。
- 前記アルカリ金属ハロゲン化物がアルカリ金属塩化物であり、前記試薬の10mM〜50mMを構成する請求項1記載の試薬。
- 非イオン性洗剤を2.5%以下の容量で更に含む請求項1記載の試薬。
- 前記試薬が陽イオン洗剤及び陰イオン洗剤を欠く請求項1記載の試薬。
- 細胞物質からRNAを抽出する方法であって、前記細胞物質を請求項1の試薬に前記RNAを上清に十分抽出できる程度に接触させ、前記細胞物質の残りから前記上清を回収することを含む方法。
- 有機溶媒で前記上清を抽出することなく行われる請求項13記載の方法。
- 前記細胞物質からゲノムDNAを除去するための前記細胞物質の酵素処理を行なうことなく行われる、請求項13記載の方法。
- 前記細胞物質中のタンパク質を変性させるための前記細胞物質の熱処理を行なうことなく行われる、請求項13記載の方法。
- 前記ヘパリンが前記試薬の1pg/μL〜3pg/μLを構成し、前記還元剤が前記試薬の1mM〜5mMを構成し、前記キレート剤がエチレンジアミン四酢酸であり、前記試薬の0.3mM〜3mMを構成し、前記緩衝剤が前記の試薬のpHを7.5〜8.0に維持する前記試薬の量を構成し、前記アルカリ金属ハロゲン化物が前記試薬の10mM〜50mMを構成する請求項13記載の方法。
- 前記試薬が非イオン性洗剤を2.5%以下の容量で更に含み、イオン性洗剤を欠く請求項13記載の方法。
- 前記試薬が有機溶媒を欠く、請求項1記載の試薬。
