Description
(発明の分野)
本発明は、薬剤および他の化合物の毒物学的なスクリーニングのための方法を提供する。本発明は、特に、ヒト胚性幹細胞(hESC)またはhESC由来系列特異的細胞(例えば、神経幹細胞、神経前駆細胞および神経細胞)である試薬、ならびにこれらの細胞を使用して薬学的化合物および他の化学物質の発生毒性作用または催奇形性作用を検出するための方法を提供する。より具体的には、本発明は、ヒトの発生の間のそれらの毒性を予測する、化合物の毒性を分析するためのインビトロ手段を提供する。毒性作用または催奇形性作用に対する候補予測バイオマーカーもまた、本明細書中で同定および提供される。
出生時欠損は、米国における乳児罹患率の主要な原因であり、生まれた乳児の33人のうちの1人が罹患する(非特許文献1;非特許文献2)か、または年間で約125,000人の新生児が罹患する。発生毒性は出生時欠損を引き起こし得、そして胚の死亡率、子宮内発育遅延(IUGR)、異常形態発生(例えば、骨格奇形)、および認識障害(例えば、自閉症)をもたらし得る機能的な毒性を生じ得ることが、理解される。化学物質の曝露がこれらの障害の発症において役割を果たし得るという役割についての、増大する関心が存在する。実際に、全ての出生時欠損のうちの5%〜10%は公知の催奇形性薬剤への胎内曝露によって引き起こされることが、推定される(非特許文献3)。

本発明は、未分化のヒト胚性幹細胞(hESC)またはhESC由来系列特異的細胞(例えば、神経幹細胞、神経前駆細胞および神経細胞)を使用する、薬学的化学物質および非薬学的化学物質の毒性および催奇形性のインビトロスクリーニングのための試薬および方法を提供する。本発明は、ヒト胚性幹細胞(hESC)またはhESC由来系列特異的細胞(例えば、神経幹細胞、神経前駆細胞および神経細胞)を使用する、薬剤および他の化合物の毒性(特に、発生毒性および催奇形性)を容易に決定するためのヒト特異的インビトロ方法を提供する。本明細書中に提供されるように、hESCまたはhESC由来系列特異的細胞(例えば、神経幹細胞、神経前駆細胞および神経細胞)は、化合物の毒性作用(特に、ヒトの発生に対するその毒性作用および催奇形性作用)を評価するために有用であり、したがって、種間の動物モデルに関連する制限を克服する。特に、本発明は、hES細胞またはhESC由来系列特異的細胞(例えば、神経幹細胞、神経前駆細胞および神経細胞)の代謝産物プロフィールがヒトの発生の公知である撹乱因子に対する応答において変化されることを示す。
本発明は、ヒト胚性幹細胞メタボロームを使用して発生毒性を評価するための試薬(ヒト胚性幹細胞(hESC)またはそれから産生されたhESC由来系列特異的細胞(例えば、神経幹細胞、神経前駆細胞および神経細胞)を含む)を提供する。ヒト胚性幹細胞は、インビトロでの器官形成を再現する移植前のヒト胚から直接単離される多能性であり、自己再生性の細胞である。系列特異的前駆細胞は、hES細胞に由来し、そして特定の細胞系列に入るが、依然としてその特定の系列内の細胞型に関して多分化能を保持する。例えば、神経前駆体は、神経分化を果たすが、依然としてその神経細胞型に関して限定されていないままである。また、高分化細胞型(例えば、ニューロン)は、本発明の方法の範囲内である。ヒトの発生および疾患の生物化学的経路は、hESCおよび/またはhESC由来系列特異的細胞において活性である。なぜならば、それらの細胞は、機能的な体細胞への分化を再現するからである。発生の間におけるこれらの経路の破壊は、神経管閉鎖不全(NTD)および認識障害などの障害に寄与する。環境因子(すなわち、化学物質または薬物)は、特定の獲得した先天性障害の個体発生に関与する。初期のヒトの発生の間どの経路が特に環境の作用に感受性であるのかという疑問は、未解決のままである。
(発生毒性のスクリーニング)
発生毒性試験のためのモデル系としてのhESCの効力を示すために、hESCを、公知の催奇形物質であるバルプロ酸塩(VPA)によって処理した。バルプロ酸塩は、癲癇および双極性障害の臨床適用を有する一般的な抗躁うつ病薬および抗痙攣薬であり(Williamsら、2001、Dev Med Child Neuro 43:202−6)、バルプロ酸塩は、発生の異常に関連する(Meadorら、2006、Neurology 67:407−412)。しかし、バルプロ酸塩が発生上の欠損をもたらす機構は、神経系の感受性の増大にかかわらず、完全に理解されていない(Bjerkedalら、1982、Lance 2:109:Wyszynskiら、2005、Neurology 64:961−5;Rasalamら、2005、Dev Med Child Neuro 47:551−555)。バルプロ酸塩に対する曝露は、一般集団の10〜20倍で二分脊椎および神経管閉鎖不全(NTD;Bjerkedalら、1982、Lancet 2:109)の明白な増大を引き起こし、そして認識障害(例えば、自閉症)(Adabら、2004、J Neurol Neurosurg Psychiatry 75:1575−83)を引き起こす。しかし、VPAは癲癇および双極性障害の臨床適用を有する抗痙攣薬である(Williamsら、2001、Dev Med Child Neurol 43:202−06)ので、処置は、一般に、妊娠全体にわたって持続されなければならない。
(遺伝子発現分析)
実施例1に示す分析の効力を、遺伝子発現研究によって確認し、遺伝子発現の変化が、hESCのVPA処理後に観察された。VPA処理は、hESCにとって有害でなく、そのhESCは、催奇形物質曝露後の複数回の継代にわたって生存可能なままであり、したがって遺伝子発現分析を行えるようにした。
(ヒト胚性幹細胞メタボローム)
催奇形物質曝露後の代謝産物プロフィール
催奇形物質であるバルプロ酸塩に対するhES細胞の曝露は、妊娠および発生の間の重要な経路を含む異なる代謝経路における有意な変化を誘導した。妊娠の間に活性化された代替的な代謝経路を、図5に示し、トリプトファンが、キヌレニンに変換される。本発明のこの局面を調査するために、hESCを、実施例1に記載したように培養し、そしてバルプロ酸塩処理の手順を、そこに記載したように行った。処理1(24時間)は、hES細胞を22μMバルプロ酸塩に24時間にわたって曝露し、次いで上清および細胞ペレットを回収した。第2の処理群(4日間)において、hESC細胞を、22μMのVPAに4日間にわたって曝露し、そして4日目に回収した。第3の処理群または延長した培養(EC、8日間)において、hESC細胞は、バルプロ酸塩を4日間にわたって受容し、次いで標準的なhES細胞培地においてさらに4日間にわたって培養を行った。細胞および上清を、8日目に回収した。各処理は、1つの実験あたり合計で6個の6ウェル培養皿による並行したコントロール群(1回の処理あたり2個の6ウェル培養皿)を有した。
変化倍率を、バルプロ酸塩処理hES細胞および未処理hES細胞の最小二乗平均の%の違いとして示す。p値を、ANOVAによって決定した。質量は、ESI−TOF−MSによって検出した平均中性質量であり、そしてRTは、分子が溶出した平均保持時間である。0.05未満のp値は太字である。
(キヌレニン:発生毒性およびCNS障害の診断および処置のためのバイオマーカー)
キヌレニンは、バルプロ酸塩処理hES細胞において検出されることが実施例3に示された。キヌレニン(グルタメートおよびピログルタミン酸と一緒に)は、バルプロ酸塩処理ヒト胚性幹細胞(hES) 対 コントロールにおいて示差的に産生された。キヌレニンは、化学物質の乳児における神経発達障害およびインビトロ発生毒性の同定のために有用な新規バイオマーカーである。この実施例は、神経発達障害についてのバイオマーカー(催奇形物質処理hESCにおいて示差的に産生される細胞産物を含む)の同定を記載する。
(キヌレニン経路の遺伝子発現分析)
実施例4における分析の効力を、遺伝子発現の変化をhESCのVPA処理後に観察した遺伝子発現研究によって確認した。ヒト胚性幹細胞のバルプロ酸塩処理は、低分子キヌレニン(トリプトファンの異化における中間体代謝産物)の顕著なアップレギュレーションを誘導した。トリプトファンは、神経伝達物質セロトニン(5HT)の前駆体である。したがって、キヌレニンへのトリプトファンの代謝およびその反対の経路(セロトニン合成)における酵素の発現がヒト胚性幹細胞において変化したか否かを、キヌレニン経路の機構的特性およびバルプロ酸塩に対するその応答を試験するために調べた。
(出生前のアルコール曝露についての発生毒性学スクリーニング)
アルコールに対する応答において示差的に分泌される代謝産物、および胎児アルコール症候群に関与する経路を同定するために、ヒト胚性幹細胞を、0%エタノール、0.1%エタノールおよび0.3%エタノールによって4日間にわたって処理し、次いで実施例1においてバルプロ酸塩について上で記載した一般的な方法に従ってLC/ESI−TOF質量分析を行った。細胞外培地を、回収し、そして上記処理の24時間後および4日後に処理し、そして49,481種の質量シグナルが、3回の技術的な複製後に検出された。49,481種の質量シグナルのうち、1,860種の化合物が、少なくとも1回の処理において有意(p<0.05)に異なり、そして有意な時間変化(<1または>1)を有した(表7)。ビニングした質量は、中性質量をいくつかの異なるデータベースにおいて検索することによってインシリコで注釈をつけられた。これらのデータベースは、Metlin、Biological Magnetic Resonance Data Bank(BMRB)、NIST Chemistry WebBook、およびHuman Metabolome Databaseを含んだ。質量を、その中性質量が上記に列挙したデータベースのうちの1つにおいて注釈をつけられた公知の化合物の10ppm以内であった場合、同定されたと見なした。
(神経前駆細胞の発生毒性学スクリーニング)
胚の発生に対する催奇形物質のメタボロミクス評価は、専らhESCに限定されない。本発明の方法はまた、他の先祖幹細胞(神経前駆細胞などの系列制限性幹細胞を含む)に関して有用である。系列特異的幹細胞に対する毒性学スクリーニングの効力を示すために、hESCに由来するニューロン前駆体を、実施例1に記載した方法に従って1mMバルプロ酸塩によって処理した。
Claims (51)
- 分泌または排出される細胞代謝産物の集団を分離することによって、試験化合物に接触されたヒト胚性幹細胞(hESC)またはhESC由来系列特異的細胞において示差的に産生される10ダルトン〜1500ダルトンの分子量を有する1種以上の細胞代謝産物を同定するための方法であって、該方法が、
a)試験化合物の存在下または非存在下でhESCまたはhESC由来系列特異的細胞を培養する工程;
b)hESCまたはhESC由来系列特異的細胞から分泌または排出される10ダルトン〜1500ダルトンの細胞代謝産物の集団を分離する工程;
c)hESCまたはhESC由来系列特異的細胞において10ダルトン〜1500ダルトンの分子量を有する1種以上の示差的に産生される細胞代謝産物を検出する工程;ならびに
d)該試験化合物の存在下または非存在下で培養された細胞において示差的に産生される10ダルトン〜1500ダルトンの分子量を有する少なくとも1種の代謝産物を選択する工程;

- 試験化合物を、該試験化合物と接触させたヒト胚性幹細胞(hESC)またはhESC由来系列特異的細胞に対する毒性または催奇形性についてアッセイするための方法であって、該方法が、
a)試験化合物の存在下または非存在下でhESCまたはhESC由来系列特異的細胞を培養する工程;
b)工程a)において培養されたhESCまたはhESC由来系列特異的細胞から分泌または排出される10ダルトン〜1500ダルトンの細胞代謝産物の集団を分離する工程;
c)工程b)からの10ダルトン〜1500ダルトンの細胞代謝産物の集団をアッセイして、毒性化合物または催奇形性化合物と接触させたhESCによって産生された10ダルトン〜1500ダルトンの分子量を有する1種以上の細胞代謝産物を同定する工程;ならびに
d)hESCまたはhESC由来系列特異的細胞が該試験化合物の存在下または非存在下で10ダルトン〜1500ダルトンの分子量を有する1種以上の細胞代謝産物を産生する試験化合物を選択する工程であって、該細胞代謝産物が、毒性化合物または催奇形性化合物と接触させたhESCまたはhESC由来系列特異的細胞によって産生される、工程;
e)該試験化合物の存在下または非存在下で該細胞において示差的に産生される10ダルトン〜1500ダルトンの分子量を有する少なくとも1種の細胞代謝産物と、毒性化合物と接触させたヒト胚性幹細胞(hESC)またはhESC由来系列特異的細胞において示差的に産生される10ダルトン〜1500ダルトンの分子量を有する1種以上の細胞代謝産物を含むバイオマーカープロフィールとを比較する工程
を包含する、方法。 - 前記細胞代謝産物の少なくとも1種は、前記試験化合物の存在下において、該試験化合物の非存在下よりも多い量で産生される、請求項31に記載の方法。
- 前記細胞代謝産物の少なくとも1種は、前記試験化合物の非存在下において、該試験化合物の存在下よりも多い量で産生される、請求項31に記載の方法。
- 前記細胞代謝産物は、前記hESCまたはhESC由来系列特異的細胞から分泌または排出される、請求項31に記載の方法。
- 前記細胞代謝産物は、100ダルトン〜1000ダルトンの分子量を有する低分子代謝産物である、請求項31に記載の方法。
- 前記試験化合物は、毒性化合物または催奇形性化合物である、請求項31に記載の方法。
- 細胞代謝産物は、物理的分離方法を使用してアッセイされる、請求項31に記載の方法。
- 前記物理的分離方法は、液体クロマトグラフィ/エレクトロスプレーイオン化飛行時間型質量分析である、請求項37に記載の方法。
- 前記細胞代謝産物は、テトラヒドロフォレート、ジヒドロフォレートまたは葉酸代謝経路における他の代謝産物、グルタチオン、または酸化型グルタチオンである、請求項31に記載の方法。
- 前記細胞代謝産物は、キヌレニン、8−メトキシキヌレネート、N’−ホルミルキヌレニン、7,8−ジヒドロ−7,8−ジヒドロキシキヌレネート、5−ヒドロキシトリプトファン、N−アセチル−D−トリプトファン、グルタメート、ピログルタミン酸またはトリプトファン代謝経路もしくはグルタミン酸代謝経路における他の代謝産物、ヒスタミン、ドパミン、3,4−ジヒドロキシ酪酸、セロトニン、γ−アミノ酪酸(GABA)あるいは他の酪酸種である、請求項31に記載の方法。
- 複数種の細胞代謝産物が、同定される、請求項31に記載の方法。
- 前記複数種の同定された細胞代謝産物が、バイオマーカープロフィールを構成する、請求項41に記載の方法。
- バイオマーカープロフィールを構成する前記細胞代謝産物のうちの1種が、キヌレニンである、請求項42に記載の方法。
- 前記試験化合物が、毒性化合物または催奇形性化合物である、請求項43に記載の方法。
- 前記複数種の同定された細胞代謝産物が、毒性化合物または催奇形性化合物に対するhESC応答の特徴を示すバイオマーカープロフィールを構成する、請求項31に記載の方法。
- 前記試験化合物の存在下または非存在下で前記細胞において示差的に産生される10ダルトン〜1500ダルトンの分子量を有する少なくとも2種の細胞代謝産物が、検出される、請求項1に記載の方法。
- 前記試験化合物の存在下または非存在下で前記細胞において示差的に産生される10ダルトン〜1500ダルトンの分子量を有する少なくとも2種の細胞代謝産物が、検出される、請求項16に記載の方法。
- 前記試験化合物の存在下または非存在下で前記細胞において示差的に産生される10ダルトン〜1500ダルトンの分子量を有する少なくとも2種の細胞代謝産物が、検出される、請求項31に記載の方法。
- 毒性化合物としての試験化合物を同定するための、請求項46〜48のいずれか1項に記載のバイオマーカープロフィールの使用法。
- 前記バイオマーカープロフィールが、テトラヒドロフォレート、ジヒドロフォレートまたは葉酸代謝経路における他の代謝産物、グルタチオン、または酸化型グルタチオンを含む、毒性化合物としての試験化合物を同定するための、請求項49に記載のバイオマーカープロフィールの使用法。
- 前記バイオマーカープロフィールが、キヌレニン、8−メトキシキヌレネート、N’−ホルミルキヌレニン、7,8−ジヒドロ−7,8−ジヒドロキシキヌレネート、5−ヒドロキシトリプトファン、N−アセチル−D−トリプトファン、グルタメート、ピログルタミン酸またはトリプトファン代謝経路もしくはグルタミン酸代謝経路における他の代謝産物、ヒスタミン、ドパミン、3,4−ジヒドロキシ酪酸、セロトニン、γ−アミノ酪酸(GABA)あるいは他の酪酸種を含む、毒性化合物としての試験化合物を同定するための、請求項49に記載のバイオマーカープロフィールの使用法。
