Description
A−Y−L (I)
で示されるトランスフェクション用化合物またはその塩、異性体もしくは混合物、とを含む組成物に関する。一般式(I)において、
・Aは4,5−二置換2−デオキシストレプタミン環の種類のアミノグリコシドを意味し、
・Yはスペーサーを意味し、そして
・Lは下記のいずれかの基を意味する:
−(R1)R2基(式中、R1およびR2は互いに独立して、水素原子もしくは脂肪性脂肪族鎖を意味するか、またはR1もしくはR2の一方は存在せず、ただしR1とR2の少なくとも一方は脂肪性脂肪族鎖を意味する)、または
−N−R3もしくは−O−R3基(式中、R3はステロイド誘導体を意味する)。
・2−デオキシストレプタミン核が4位と5位において置換されている、4,5−二置換2−デオキシストレプタミン型アミノグリコシド、および
・2−デオキシストレプタミン核が4位と6位において置換されている、4,6−二置換2−デオキシストレプタミン型アミノグリコシド。
−C(O)−;
−NH−C(O)−CH2−CH2−;
−W−(CH2−)k−W'−;および
−(CH2−)i−W−(CH2−)j−。
本発明の目的にとって、用語「ステロイド誘導体」とは、コレスタン型の多環式化合物を意味するものである。これらの化合物は天然でもそうでなくてもよく、好ましくはコレステロール、コレスタノール、3−α−シクロ−5−α−コレスタン−6−β−オール、コール酸、ギ酸コレステリル、ギ酸コレスタニル、3−α−5−シクロ−5−α−コレスタン−6−β−イル・フォルミエート(ギ酸エステル)、コレステリルアミン、6−(1,5−ジメチルヘキシル)−3a,5a−ジメチルヘキサデカヒドロシクロペンタ[a]シクロプロパ[2,3]シクロペンタ[1,2−f]ナフタレン−10−イルアミン、およびコレスタニルアミンから選ばれる。
より正確には、本発明の1目的は、疾患、特に遺伝子産物の過剰発現から生ずる疾患の治療を意図した医薬品の製造のために本発明に係る組成物を使用することである。この医薬品中に含まれているsiRNAは該遺伝子産物のmRNAを特異的に標的とし、それにより該疾患をin vivoまたはex vivoで治療する。
(1)請求項1に係る組成物を用意し、そして
(2)該細胞を該組成物と接触させる。
1.1:材料および方法
1.1.1:核酸、カチオン性脂質および複合体の調製
3'−ローダミン標識抗GFP siRNAおよび5'−ローダミン標識対照siRNAはQiagen社(米国カリフォルニア州チャツワース)により供給された。ストック。非標識の抗GFP siRNAはEurogentec社(ベルギー、スラン)により供給された。抗GFP siRNAはセンス鎖について下記配列を有していた:GCAAGCUGACCCUGAAGUUCAU(配列番号1)。GAPDH mRNAを標的とするサイレンサーTMGAPDH siRNA(ヒト、マウス、ラット)はAmbion社(英国、ケンブリッジ州)により供給された。ヒト抗ラミンA/C siRNAは、Santa Cruz Biotechnology社(米国カリフォルニア州サンタクルーズ)により供給された。抗GFP siRNAヘアピンをコードするプラスミドpTERは、pCDNA4TO(Invtrogen Life Technologies、米国カリフォルニア州カールスバッド)のBglIIとHindIIとの間に修飾Hlプロモーターを挿入することにより得たものであり、A. Polesskaya氏(フランス、ビルジュイフ)の好意により供給された。プラスミド類は、EndoFreeプラスミド精製カラム(Qiagen、米国カリフォルニア州チャツワース)を用いて組換え大腸菌(Escherichia coli)から精製した。脂質アミノグリコシド誘導体およびBGTCは既に述べたようにして(非特許文献1〜3)合成した。
d2GFP(ヒト肺がん)細胞を、フラスコ内で1%ストレプトマイシン/ペニシリン、ゲネチシン0.8mg/ml(GIBCO、Invitrogen Life Technologies社、米国カリフォルニア州カールスバッド)および10%ウシ胎仔血清(Eurobio、フランス、クールブフ)を加えた、グルコース、l−グルタミンおよびピルビン酸(ピルベート)を含有するD−MEM中で、5%CO2/加湿雰囲気において37℃で培養した。トランスフェクションの1日前に、細胞を65000の細胞数で24ウェルの培養プレートに移した。それにより24時間後に約70〜80%コンフルエンス(集密)の状態になった。各ウェルにおいて、500μlの血清およびゲネシチン不含有D−MEM中の複合体5050μlを添加することによりトランスフェクションを行った。2時間後、トランスフェクション培地を、10%ウシ胎仔血清および1%ストレプトマイシン/ペニシリンを含有する1mlのD−MEMで置換した。トランスフェクション実験を3回実施し、残留GFP発現をトランスフェクションの24時間後に定量した。経時的実験のために、トランスフェクションを下記の変更点を除いて上記と同様に実施した。トランスフェクションした細胞を3日目ごとに、GFP定量測定の日のコンフルエンスが90%となるように適当な細胞数で新たな24ウェル培養プレートに移した。トランスフェクション実験は3回実施し、残留GFP発現をトランスフェクションから1、2、3、5、6および9日後に測定した。
24時間後、トランスフェクションした細胞を500μlのPBSで2回洗浄し、プロテアーゼ阻害剤カクテル(Roche Diagnostics、ドイツ、マンハイム)を加えたレポーター溶解緩衝液(Promega、ウイスコンシン州マディソン)で溶解させた。1回の凍結−解凍(−80℃/20℃)により完全な溶解を確保した。その後、サンプルを10000gで5分間遠心した。GFP蛍光測定を、180μl量の上清でのアッセイにより、それぞれ485nmおよび535nmの励起波長および発光波長のVictor2(Perkin Elmer、フランス、レジュリ)を用いて実施した。蛍光は、BCAアッセイキット(Pierce、米国イリノイ州ロックフォード)を用いて得られた、サンプルの全タンパク質濃度に対して正規化された。残留GFP発光は、対照サンプルの蛍光の%として表された。
トランスフェクションされた細胞の蛍光顕微鏡検査は、倒立型蛍光顕微鏡Axiovert 200M (CarlZeiss、ドイツ、ゲッティンゲン)を用いて実施した。
臭化エチジウム蛍光測定を励起波長および発光波長がそれぞれ260nmおよび590nmのKontron SFM25蛍光分光光度計で行った。蛍光は10μg/mlの核酸(siRNAまたはDNA)で調製されたサンプルに臭化エチジウム(最終濃度5μM)を添加した直後から監視した。
動的光散乱測定は、Zetasizer 300HSA(英国、ウースター州モルバン)を用いて20℃で行った。さまざまな量のカチオン性脂質を含有する核酸10μg/mlで調製されたサンプルについて測定値を監視した。
10μgのsiRNA/mlと適当量のカチオン性脂質とを用いてsiRNA/カチオン性脂質複合体を調製した。5μl量のサンプルを、多孔質炭素被覆銅グリッド上に付着させ、過剰分を濾紙で吸い取った後、グリッドを液体窒素で冷却された液体エタン浴(Leica EM CPC)中に投入した。この標本をクライオホルダー(Gatan)を用いて−170℃の温度に保持し、200kVおよび低線量条件下では50000Xの標準倍率で動作するFEI Tecnai F20電子顕微鏡で観察した。画像は2K×2K Gatant低速スキャンCCDカメラで記録した。
TRIzol試薬(Invitrogen、フランス、セルジ・ポントワ)とRnesyTMミニキット (Qiagen、米国カリフォルニア州チャツワース)とを用いて全RNAを抽出した。mRNAの精製を、OligotexTM mRNAミディキット (Qiagen、米国カリフォルニア州チャツワース) を用いて行った。SuperScriptTM III (Invitrogen Life Technologies社、米国カリフォルニア州カールスバッド) を用いて、逆転写を供給業者の記載通りに実施した。ABI PrismTM7900HT配列検出システム (Applied Biosystems) を用いてリアルタイム定量PCRを実施し、得られたデータを、Applied Biosystems SDS 2.1ソフトウェアによる機器スペクトル補償後に取得した。GFPリバースプライマーの配列は、CGGGCATGGCGGACTT(配列番号2)、FAMプローブの配列はCAGCACGACTTCTTC(配列番号3)、そしてGFPフォワードプライマーの配列はGCTACCCCGACCACATGAAG(配列番号4)であった。各実験に対して対照の無DNAサンプルについても実施した。サイクリング条件は、95℃で10分間の高温開始後に、95℃で15秒と60℃で1分の40サイクルを含んでいた。ゲル電気泳動を用いて、適当なサイズの単一アンプリコンを検出した。データの正規化のためにHPRT遺伝子を用いた。値は、対照siRNAで得られた量に対する、抗GFPもしくはGAPDHもしくはラミンA/C mRNAがトランスフェクションされた細胞内で得られた正規化されたGFPもしくはGAPDHもしくはラミンA/C mRNA量の%に対応していた。

まず、我々は、細胞質GFPを発現する形質転換細胞株(d2GFP)内での抗GFP siRNA分子の導入を通してGFP発現を阻害するために、プラスミドDNAトランスフェクションに現在使用されているカチオン性ベクターの能力を評価した。抗GFP siRNA分子は、BGTC/DOPEまたはDC−Chol/DOPEまたはPEIとの複合体であった。トランスフェクションから1日後に、GFP発現の阻害を蛍光GFP測定により監視した。図1は、正電荷モル数/負電荷モル数として表されるカチオン性ベクター/siRNA電荷比が増大するにつれて、残留GFP発現が低下したことを示している。BGTC、DC−CholおよびPEIのカチオン性リポソームと複合体化されたsiRNAで得られた残留GFP発現は、それぞれ約32%、52%および55%であった。これらの結果は、siRNAにより媒介される遺伝子発現の阻害効率が向上した新規ベクター開発の必要性を浮かび上がらせるものであった。その後の実験における遺伝子発現阻害の比較のための標準的カチオン性ベクターとしてBGTC−DOPEリポソームを選択した。
脂質性アミノグリコシド誘導体は、アミノグリコシド先端基に自己凝集性の炭化水素系後端部(テイル部)が結合してなる両親媒性物質である。図2は、本試験で使用された種々の脂質性アミノグリコシド誘導体を示す。それらは、カチオン性先端基としてのトブラマイシン、カナマイシン、パロモマイシンまたはネオマイシンが、スクシニル・スペーサーを介して、ジオレイル鎖に結合したものからなる。トブラマイシンおよびカナマイシンは4,6−二置換2−デオキシストレプタミン環の種類に属し、パロモマイシンおよびネオマイシンは4,5−二置換2−デオキシストレプタミン環の種類に属する。
我々は、DNAまたはsiRNAとBGTCのカチオンリポソームまたは脂質性アミノグリコシド誘導体との会合から生ずる複合体の物理化学的性質を、カチオン性脂質/核酸電荷比との関係として検討した(図3)。電荷比を算出するために、1μgのsiRNAは3ナノモルの荷電リン酸分であり、BGTC、DOST、DOSK、DOSPおよびDOSNはそれぞれ2、4、3、4および6個の正電荷をもつと仮定した。
BGTC/DOPE小胞標本を、30〜70nmの範囲内からなる直径の単層膜ラメラリポソームから作製した(図4A)。BGTC−DOPEリポソームをsiRNA溶液と混合すると、サイズが200〜500nmの範囲内の小さな密集した丸みを帯びた構造物の生成を生じた(図4B)。これらの構造物は恐らくsiRNA分子に対応する電子密度の高い電子緻密層により離間された数層の脂質層のスタック(積み重ね)からなるものであった。7.0nmの測定距離は、脂質二重層(二分子層)の厚さおよび二本鎖siRNA分子の直径に対応していた。よく配向した集中構造では、間隔が3.0nmの微細なスジ模様が見えた。これは恐らく2層の脂質膜間の規則的配置を示していよう(図4C)。
図5Aは、BGTC/siRNA電荷比が増大するにつれて、BGTC−DOPE/siRNA複合体によるd2−GFPトランスフェクション細胞のGFP蛍光レベルが次第に低下したことを示している。これに対し、DOST−DOPE/siRNA複合体は、DOST/siRNA電荷比が4の場合でGFP蛍光の11%への急激な低下を生じ、より高い電荷比でもこの最小値にとどまった。意外にも、DOSK−DOPE/siRNA複合体ではGFP蛍光は漸減を示し、DOSK/siRNA電荷比12で41%の最小蛍光レベルに到達した。4,5−二置換型のDOSP−DOPEおよびDOSN−DOPEから得られたアミノグリコシド誘導体は両方とも、カチオン性脂質/siRNA電荷比の増加に伴ってGFP発現の漸減を示し、カチオン性脂質/siRNA電荷比8で10%の最小残留蛍光レベルに到達した。各種カチオン性脂質による同じ条件での対照siRNAのトランスフェクションは、非トランスフェクション細胞(データを示さず)に比べて、GFP発現に影響しなかった。その後の実験に関して、カチオン性脂質/siRNA電荷比を、BGTCおよびDOSTにより複合体化したsiRNAでは4、DOSPによるものでは10、そしてDOSNおよびDOSKによるものでは12に設定した。
GFP発現性ヒト肺がんH1299細胞を対照siRNAまたは3'−ローダミン標識抗GFP siRNAでトランスフェクションした。siRNA分子はDOSPの不存在下(「裸(naked)」siRNA)または存在下で処方したが、いずれの場合もDOPEは使用しなかった。トランスフェクションしたH1299細胞を、GFP蛍光を視覚化するためにFITCフィルターを用いて、またはsiRNA内部移行(内在化)を視覚化するためにローダミンフィルターを用いて観察した。
図7は、DOSP−DOPEが、X-tremeGeneおよびリポフェクタミン(Lipofectamine)TM2000で観察されたものに比べて、d2GFPでトランスフェクションされた細胞中のGFP蛍光レベルのより大きな減少を生じたことを示している。結果は、DOSP−DOPE/siRNA複合体で得られた残留GFP発現により正規化された。DOSP−DOPEとRNAiFectTM試薬との間には有意差は認められなかった。
次に、カチオン性リポソーム/siRNA複合体のGFP発現に及ぼす影響を時間との関係として検討した(図8)。DOSN−DOPEまたはDOSP−DOPEと複合体化されたsiRNAでトランスフェクションされた細胞については、残留GFP発現は5日間は約10%であり、次いで次第に増大して9日目に100%に戻った。DOST−DOPE/siRNA複合体でトランスフェクションされた細胞は同様のGFP発現の経時変化を示したが、残留GFP発現は約20%であった。図8はまた、DOSK−DOPEリポソームは2日目でGFP蛍光減少の最大値に至ったことを示している。
我々は、アミノグリコシド誘導体が、d2GFP細胞中に、siRNAだけでなく、GFPに抗して小ヘアピンRNAをコードするプラスミドDNAもまた導入する可能性についても評価した(図9)。結果は、カチオン性脂質/DNA電荷比が増大するにつれてGFP発現が減少し、カチオン性脂質に依存した最小蛍光値に到達することを示した。例外はDOSN−DOPE/DNA複合体で、カチオン性脂質/DNA電荷比の増大によるGFP発現の増大を生じた。これらの結果は、ルシフェラーゼをコードするプラスミドのトランスフェクションで得られたものとよく一致しており、GFP発現阻害がトランスフェクション効率と相関していることを示した。
脂質性アミノグリコシド誘導体/siRNA複合体のコロイド(としての)安定性は、電荷比に依存し、それぞれ負荷電、中性または正荷電の複合体に対応する、主要なゾーンA、BおよびCを決定した。このコロイド安定性挙動は、DNA分子とのリポポリアミンミセル(非特許文献4)、ビスグアニジニウムリポソーム(非特許文献6および本研究)またはDOTAPリポソームの会合から得られた複合体で既に認められていたものであった。しかし、ゾーンBは、DNAを新たに合成された脂質性アミノグリコシド誘導体と複合体化させたものと似ていたが、siRNAとの複合体化では縮小していた。
Claims (19)
- siRNAと、一般式(I) :
A−Y−L (I)
で示されるトランスフェクション用化合物またはその塩、異性体もしくはそれらの混合物、とを含む組成物。
上記式中、
・Aは4, 5−二置換2−デオキシストレプタミン環の種類のアミノグリコシドを意味し、
・Yはスペーサーを意味し、そして
・Lは下記のいずれかの基を意味する:
−(R1) R2基(式中、R1およびR2は互いに独立して、水素原子もしくは脂肪性脂肪族鎖を意味するか、またはR1もしくはR2の一方は存在せず、ただしR1とR2の少なくとも一方は脂肪性脂肪族鎖を意味する)、または
−N−R3もしくは−O−R3基(式中、R3はステロイド誘導体を意味する)。 - 前記4, 5−二置換2−デオキシストレプタミン環の種類のアミノグリコシドAが、パラモマイシン、ネオマイシン、およびリボスタマイシンから選ばれる、請求項1に記載の組成物。
- スペーサーYが、炭素数1〜6のアルキル官能基、ケトン官能基、エステル官能基、エーテル官能基、アミノ官能基、アミド官能基、アミジン官能基、カルバメート官能基、チオカルバメート官能基、グリセロール、尿素、チオ尿素、および芳香環から選ばれた少なくとも1種の化学官能基から構成される、請求項1または2に記載の組成物。
- スペーサーYが次式で示される基から選ばれる、請求項3に記載の組成物:
−C(O)−;
−NH−C(O)−CH2−CH2−;
−W−(CH2 −) k−W'−;および
−(CH2−) i −W−(CH2−) j −。
上記式中、i,jおよびkは1〜6(両端を含む)から選ばれた整数であり、WおよびW' は互いに同一でも異なっていてもよく、それぞれケトン官能基、エステル官能基、エーテル官能基、アミノ官能基、アミド官能基、アミジン官能基、カルバメート官能基、チオカルバメート官能基、グリセロール、尿素、チオ尿素、および芳香環から選ばれた基である。 - Lが−(R1) R2基を意味し、少なくとも1種の脂肪性脂肪族鎖が炭素数10〜22の飽和または不飽和アルキル基から選ばれる、請求項1〜4のいずれかに記載の組成物。
- Lがジミリストイル、ジオレイルおよびジステアリルから選ばれる、請求項5に記載の組成物。
- 少なくとも1種のアジュバントをさらに含有する、請求項1〜7のいずれかに記載の組成物。
- 前記少なくとも1種のアジュバントが、脂質、ペプチド、タンパク質およびポリマーの少なくとも1種である、請求項8に記載の組成物。
- 前記少なくとも1種のアジュバントが中性脂質から選ばれる請求項9に記載の組成物。
- 前記中性脂質が、両性イオン性または生理条件下でイオン性電荷を失う天然および合成脂質から選ばれる、請求項10に記載の組成物。
- 前記中性脂質がジオレイルホスファチジルエタノールアミン、オレイルパルミトイルホスファチジルエタノールアミン、ジステアロイル−、ジパルミトイル−およびジミリストイル−ホスファチジルエタノールアミン類、ならびに1〜3回N−メチル化されたそれらの誘導体、さらにはホスファチジルグリセロール類、ジアシルグリセロール類、グリコシルジアシルグリセロール類、セレブロシド類、スフィンゴ脂質類、アシアロガングリオシド類、ジオレイルホスファチジルコリン、ならびにコレステロールから選ばれる、請求項11に記載の組成物。
- 細胞外または細胞内標的化エレメントをさらに含む、請求項1〜12のいずれかに記載の組成物。
- 前記標的化エレメントが、糖、ペプチド、タンパク質、オリゴヌクレオチド、脂質、神経メディエータ、ホルモン、ビタミン、およびそれらの誘導体から選ばれる、請求項13に記載の組成物。
- 前記標的化エレメントが、請求項1に記載のトランスフェクション用化合物またはsiRNAのいずれかに共有結合している、請求項13に記載の組成物。
- 注射用処方組成物に対して薬学的に許容される賦形剤をさらに含有する請求項1〜15のいずれかに記載の組成物。
- 皮膚および/または粘膜への投与に対して薬学的に許容される賦形剤をさらに含有する請求項1〜15のいずれかに記載の組成物。
- 被験者における標的遺伝子発現の阻害用の医薬品の製造のための、請求項1〜17のいずれかに記載の組成物の使用であり、ここで、該組成物中に含まれるsiRNAは該標的遺伝子に特異的に向けられる、前記使用。
- 下記工程を含む、siRNAをin vitroで細胞内に移入する方法:
(1) 請求項1〜17のいずれかに記載の組成物を用意し、そして
(2) 該細胞を前記組成物と接触させる。
