Description
1.酵素サンプルおよび試薬を含む液体をプレートのウエルに加える。
2.(酵素反応用に)インキュベートする。
3.沈殿物質(通常はトリクロロ酢酸[TCA]、普通は5%の最終濃度)を加えて反応を停止させ、タンパク質を沈殿させる。
4.4℃または氷上でインキュベートする。
5.未反応または遊離成分を洗浄除去する。(遊離物を定量化することが必要な場合は回収する。)
6.沈殿物(フィルターに結合したもの)および/または濾過物(遊離物)
を計数する。
Electroscan ESEM3(Philips Electron Opticsから入手可能)を使用して、膜サンプルの湿潤性を評価した。処理したサンプルからの膜の一部分を凍結割断し(以下でさらに記載するように)、次いで断面を分析するために、5℃に冷却したペルティエステージに載せた。断面サンプルは、疎水性側を画像フレームの上部に向けた。ESEMチャンバーを、ポンプを用いて大気圧から5トールの設定圧力に圧力を下げ、次いで水蒸気で洗浄し、浸水用対照物を使用して10トールにした。次いで浸水用対照物を放出させた。チャンバーの圧力が再度5トールに達したとき、チャンバーを再度水蒸気で洗浄し10トールにした。この手順を3回続けて、チャンバー中の空気を水蒸気に交換した。次いでチャンバーの圧力を7トールに設定した。チャンバーの圧力およびステージの温度に対するわずかな制御を行って、液体状の水をサンプルの突出部に保った。膜が濡れた後に、代表的な画像を得た。500倍の倍率で15KVのビーム加速電圧を使用して、画像を得た。画像は、Orion Image Acquisition Systemによって得た。
Claims (7)
- 第1の多孔質表面と、
第2の多孔質表面と、
膜内全体に前記第1の多孔質表面から前記第2の多孔質表面まで延在し、厚さを画定する、親水性材料でできた連続的な多孔構造と、
疎水性のフリーラジカル重合ポリマーもしくはフリーラジカル重合架橋ポリマーの被覆層と
を有する、多孔膜であって、
前記被覆層が、前記第1の多孔質表面の少なくとも一部を覆い、かつ前記第1の多孔質表面から前記多孔構造中へと前記多孔膜の厚さの8〜80%の深さまで延在する
ことを特徴とする、多孔膜。 - 前記親水性材料が親水性ポリマー材料を含む、請求項1に記載の膜。
- 前記親水性ポリマー材料が芳香族ポリスルホンである、請求項2に記載の膜。
- 前記親水性ポリマー材料がポリ二フッ化ビニリデンである、請求項2に記載の膜。
- 前記被覆層が、16mN/mより大きい表面張力を有する液体で濡れない、請求項1に記載の膜。
- 前記被覆層が、21mN/mより大きい表面張力を有する液体で濡れない、請求項1に記載の膜。
- 第1の多孔質表面と、
第2の多孔質表面と、
膜内全体に前記第1の多孔質表面から前記第2の多孔質表面まで延在する、親水性材料でできた連続的な多孔構造と、
空間的に制御された疎水性フリーラジカル重合ポリマー被覆層と
を有する、多孔膜の作製方法であって、前記被覆層は、前記第1の多孔質表面から前記多孔構造中へと前記多孔膜の厚さの8〜80%の深さまで延在し、かつ前記第1の多孔質表面の少なくとも一部を覆うものであり、前記方法が、
(a)少なくとも1つのフリーラジカル重合可能なモノマー、前記少なくとも1つのモノマー用の重合開始剤および架橋剤を成分として、これら3つの成分のための溶媒中に含む溶液を用意するステップ、
(b)多孔膜を前記溶液に接触させるステップ、
(c)前記連続的な多孔構造内の或る深さまで拡散する重合防止剤と、膜とを前記第1の多孔質表面から接触させ、ここで前記重合防止剤は、接触した前記第1の多孔質表面の少なくとも一部の上および前記重合防止剤が拡散した間隙の表面上にて重合が起こるのを防止できるステップ、および
(d)疎水性の架橋フリーラジカル重合ポリマー被覆を、前記溶液から、前記重合防止剤に接触していない前記膜の前記第2の多孔質表面上に形成させるステップ
を含む
ことを特徴とする、方法。
