Description
ここで報告される本発明の対象に関して、幾つかの側面を考慮する必要があり、それらの中には、
1)細胞の情報交換、シグナル伝達経路、並びに組織及び各種臓器機能の一般的な恒常性において基本的な役割を果たす「イオンチャンネル」と呼ばれる生体膜の内在タンパク質の存在;
2)自己免疫疾患の発症に関連する現象において明確な役割を果たすことが知られている免疫系の細胞、主にT-リンパ球におけるそのチャンネルの発現レベルの違い;
3)各種化学物質、天然リガンド、並びに天然において発見されたリガンドを再生するか又は類似の誘導体(ペプチド模倣物)を調製することで合成的に調製される物質を加えることによって、そのチャンネル機能を制御する可能性と、それによって疾患を治療する可能性;
について生じる知識に関連した、生化学、分子生物学、免疫学及び電気生理学の分野の重要な進歩がある。
このセクションで、発明者は、高い親和性及び選択性でリンパ球の「エフェクターメモリT細胞」(TEM)のKv1.3イオンチャンネルを遮断することが可能なリガンド(ペプチド又は有機化合物)を単に適用することによって幾つかの免疫疾患を制御する最先端の知識を示す。
1)MS患者の末梢血からのミエリン反応性T細胞は、Kv1.3高である[Wulff et al., 2003]。
2)健常対照群の末梢血からのミエリン反応性T細胞は、Kv1.3低である[Wulff et al., 2003]。
3)インスリンペプチド、オボアルブミン等の不適切な抗原又は通常のマイトジェンを持つMS患者のT細胞の刺激は、Kv1.3高IKCa1低イオンチャンネル表現型を持つTEMの発生を誘発しない。
4)Kv1.3高TEM細胞が、死後MS脳の炎症性浸潤、及び脱髄したMS病変部の柔組織において見られた[Rus et al., 2005]。
5)間接リウマチ(RA)に苦しむヒト患者の滑液から単離したT細胞は、同じドナーであるが末梢血から単離したT細胞と比較して多量のKv1.3を発現する。このKv1.3高T細胞は、それがTEM細胞であることを示すCCR7−であった[Beeton et al., 2006]。
6)I型糖尿病(T1DM)ヒト患者の末梢血リンパ球から生成した、T1DMに関連した自己抗原インスリン及びGAD65に特異な短期抗原特異性CD4+T細胞株は、CCR7抗原(CCR7−)の不足及びKv1.3高チャンネル表現型を含むTEM細胞の特性を示す[Beeton et al., 2006]。
1)TEMの特性(CCR7−,Kv1.3高)を持ち、ミエリン抗原に特異的である慢性的に活性化したヒトT細胞株[Wulff et al., 2003]又はRA患者の滑液から単離したTEM細胞の生体外での増殖は、ShK[Wulff et al., 2003]、ShK(L5)等のKv1.3特異的遮断ペプチド、もしくは小分子Kv1.3遮断薬PAP−1[Beeton et al., 2006]によって、完全且つ永久に抑制される。
2)他の高親和性Kv1.3遮断ペプチドであるマルガトキシン毒を用いた生体内での実験は、Kv1.3の遮断がミニブタにおける遅延型過敏症反応の阻害を導くことを示す。この反応は、エフェクターメモリT細胞の活性の良好な尺度になる[Koo et al., 1997]。
3)(エフェクター段階中)受容動物中にペプチドを繰り返し適用すると共に、MBPによって生体外で刺激する間(感作段階)の、ShK又はShK−Dap22を用いたMBP特異性ラットT細胞の治療は、実験的自己免疫性脳脊髄炎(AT−EAE)のルイスラットへの養子移植を妨げた[Beeton et al., 2001]。ラットのEAE[Ben Nun and Cohen, 1982]は、類似の病変形成及び神経学的異常を特徴とするヒト疾患MSに対して最も良く特徴付けられたモデルであり、T細胞集団を引き起こす疾患は、Kv1.3高チャンネル表現型を有するミエリン特異性TEMである。また、Kv1.3及びIKCa1チャンネル遮断薬の併用は、その発現を受けて投与した場合にEAEの兆候を改善した[Beeton et al., 2001]。
4)ダーク・アグーチ・ラットにおけるプリスタン誘導のMHCクラスII拘束性慢性関節炎モデル(PIA)は、ヒト疾患の間接リウマチに対するラットのモデルである。ShK(L5)の1日1回の投与は、試用期間(最長34日)中に疾患にかかった関節の数を大きく低減した[Beeton et al., 2006]。
5)実験的自己免疫性糖尿病(EAD,ヒトのT1DMに対するラットモデル)を妨げるKv1.3阻害剤の有効性は、MHCクラスII拘束性DP−BB/Wラットにおいて研究された[Beeton et al., 2006]。Kv1.3の高親和性及び選択性小分子遮断薬であるPAP−1の連日反復投与は、賦形剤のみで治療した対照動物と比較して、EADの症状を示すラットの割合を約50%まで低減した(110日齢で評価した)。これは、膵島内T細胞及びマクロファージ浸潤の減少を伴い、賦形剤で治療した対照と比較してPAP−1で治療したグループにおけるβ細胞の破壊を低減した(35〜70日齢で評価した)[Beeton et al., 2006]。
別段の定義がない限り、ここで用いるすべての技術的及び科学的用語は、本発明が属する当業者によって共通に理解されるものと同じ意味を有する。ここで用いるように、以下の用語は、特記されない限り、それらに帰する意味を有する。ここで説明するものと類似又は同等の方法及び材料はいずれも本発明の実施又は試験において使用できるが、好ましい方法及び材料を説明する。本発明の目的ため、以下に用語を定義する。
本発明の主題は、二つの新規なペプチド(Vm23及びVm24)、そのアミノ酸配列(SEQ ID NO:1及びSEQ ID NO:2)、並びに機能的に同等な類似体の可能性及び特定の免疫抑制剤としてのその使用の可能性について言及する。Vm23及びVm24は、亜型Kv1.3、特にはヒトリンパ球(hKv1.3)のカリウムイオンチャンネルの高選択性遮断薬であり、生体内でラットにおける遅延型過敏症反応への炎症反応を減少させることが示される。従って、それら二つのぺプチド及びその機能的に同等な類似体は、異常T細胞応答に関連した一部の免疫疾患の治療に使用されるリード化合物である。その免疫抑制剤及びそのhKv1.3への効果の詳細に入る前に、この分野におけるいくつかの基礎的な知識を復習することが重要である。
固相法によるペプチドの合成は、限定されないがポリスチレン、ポリアクリルアミド、特定の繊維、他の安定な重合体等の固相樹脂の使用を含む。固相樹脂の誘導体化は、C末端の酸を機能的に形成する方法としてクロロトリチルクロリド、2−クロロトリチルクロリド、ヒドロキシメチルフェニル、Sasrin等の適切なハンドルが使用できたり、限定されないが4−(2',4'−ジメトキシフェニル−Fmoc−アミノメチル)−フェノキシメチル基等の樹脂リンカーを介したタンパク質安定化によって準備されてもよい。
シクロスポリン、FK506等の免疫抑制剤は、その治療的使用を制限する重度の副作用を示す。その二つの化合物で行った研究は、その薬剤の投与によって望ましくない副作用に関与する分子機構の少なくとも一部を同定することができた。シクロスポリンは、多くの異なる組織に存在するタンパク質シクロフィリンと相互作用し、一方、FK506は、FK結合タンパク質を標的とするため毒性を引き起こし、また多くの異なる組織においても見られた。従って、重篤な副作用のない新規な免疫抑制剤を同定するための大変な努力があった。主な目標の一つは、Kv1.3イオンチャンネル等のT-リンパ球において主に発現する新規なターゲットを同定することである。T-リンパ球にて発現するKv1.3カリウムチャンネルは、特定の細胞機能に対して非常に重要であるが、このタンパク質に対するRNAコーディングもまた他の細胞(B-リンパ球、ミクログリア、マクロファージ、破骨細胞、血小板、及び一部の脳細胞)において見られる。しかしながら、T-リンパ球の場合に限り、Kv1.3は、膜電位を支配し、遮断に重要な機能的結果を有する。Kv1.3遮断薬の作用の異なる機構、及びKv1.3チャンネルの比較的制限された組織分布のため、Kv1.3の特異的で且つ高親和性の遮断薬は、シクロスポリン及びFK506よりも低い毒性の副作用を示すことが期待され、従って、自己免疫疾患の治療及び移植医療に有用であることが証明できる。
異物又は自己抗原に対する効果的な免疫反応の開発には、所定の抗原に特異的なリンパ球の活性化及び増殖が求められる。これは、免疫系の異なる細胞成分間の十分に調整された相互作用を必要とする。この過程の第1ステップは、プロフェッショナル抗原提示細胞による処理された抗原のリンパ球に対する提示である[Janeway et al., 2001]。電位開口型Kチャンネル、Kv1.3に特異的な遮断薬によって、T-リンパ球媒介免疫反応(例えば、遅延型過敏症)を制御することは、本発明の目的の範囲内である。従って、我々は、T細胞に対するリンパ球の活性化においてKチャンネルが関与する同化作用を制限する。しかしながら、我々は、Bリンパ球における特定のサブセットの増殖もまたKv1.3チャンネルの活性次第であることについて言及すべきである[Wulff et al., 2004]。
幾つかのKv1.3の高親和性ペプチド遮断薬は、IKCa1よりKv1.3に対して選択的であり、Vm24に対しても同様である。これらには、サソリ毒、例えば、マルガトキシン(MgTx)、ノキシウストキシン(Ntx)、カリオトキシン、アニュロックトキシン及びイソギンチャクから単離されたShK毒等が含まれる。しかしながら、神経興奮性及び筋肉興奮性に重要なイオンチャンネルもまた、ナノモル濃度〜ピコモル濃度での親和性を持つ毒によって阻害され、例えば、Kv1.1は、ShK[Kalman et al., 1998]及びカリオトキシン[Grissmer et al., 1994]によって阻害され、一方、Kv1.2は、MgTx[Koch et al., 1997]、Ntx[Grissmer et al., 1994]及びアニュロックトキシン[Bagdany et al, 2005]によって遮断される。
本発明は、本発明のペプチドを備える医薬品組成物を提供する。前述したように、該ペプチドは、T細胞の特異的なサブセットが活性化する場合に哺乳動物における免疫反応を抑制するのに有効である。特に、本発明のペプチドを備える医薬品組成物は、免疫反応が異種器官拒絶反応の結果であるか(例えば、心臓、肺、肝臓、腎臓又は膵臓)又はリンパ球TEMに関連した自己免疫疾患の結果である病状を治療又は予防するのに有効である。
本発明は、哺乳動物における免疫反応の抑制が必要とされる病状の治療的又は予防的処置の方法を提供するものであり、特にはかかる処置を必要とする対象においてT細胞が活性化する場合であり、ここで、上記病状には、異種器官拒絶反応(例えば、心臓、肺、肝臓、腎臓又は膵臓)又は自己免疫疾患(例えば、多発性硬化症、関節リウマチ、I型糖尿病、自己免疫性乾癬、紅斑性狼瘡、潰瘍性大腸炎、交感性眼炎及び骨吸収歯周病)が含まれる。該対象には、ほんの一部を挙げれば、ヒトの他、イヌ、ネコ、ヤギ、ウシ、ウマ、ブタ、ヒツジ等の他の哺乳動物を挙げることができる。一の態様において、本発明は、治療的又は予防的処置を必要とする対象についてのT細胞の細胞膜におけるKv1.3カリウムチャンネルの阻害に対する応答又は感受性によって治療可能な病状の予防的又は治療的処置の方法を提供する。該方法は、ヒト等のそれを必要とする対象に対して、上述した本発明のペプチドを効果的な量投与する工程を備える。一般に、このペプチドは、SEQ ID NO:1のVm24、SEQ ID NO:2のVm23というペプチドもしくは配列SEQ ID NO:3を有するペプチド、もしくはそれらの機能的に同等な類似体、又はその薬学的に許容される塩を備えてもよい。
治療的適用において、本発明のペプチドは、疾患又は望ましくない病状(例えば、それぞれ自己免疫疾患又は異種臓器拒絶反応)に既に苦しむ対象に対して、該疾患及びその合併症の進行を治療し、回復し、部分的に停止し又は検出可能な程度に遅くするのに十分な量で投与できる。本発明のペプチドの治療的適用で用いるのに効果的な量は、病状の重症度、対象の一般的症状及び投与経路によって決まることになる。治療的適用におけるペプチドの効果的な量は、一般に、キログラム当たり約0.1マイクログラム〜キログラム当たり約10マイクログラムの投与量当たりペプチド(又はその薬剤的に許容される塩)の範囲内である。
本発明のペプチドの治療上の使用と別に、これらのペプチドは、動物組織又は安定な培養物から得られる幅広い細胞において、Kv1.3チャンネルの発現レベルを検出し及び特徴づけるために使用され得る。T細胞におけるKv1.3の発現を特徴付けることの重要性は、すでに本明細書において取り上げられており、従って、本発明はまた、Kv1.3チャンネル発現細胞を生理的に特徴付けるための分子プローブとしての本発明のペプチドの使用に関するものである。Kv1.3発現の検出及び特徴付けは、都合よく標識化された本発明のペプチドを用いる幾つかの検出技術によって行われることができ、限定されるものではないが、フローサイトメトリー、通常の共焦点蛍光顕微鏡、蛍光全発光、放射性結合及び置換技術、並びに免疫学的プルダウンアッセイが挙げられる。所定の細胞において発現したKv1.3チャンネルの定量は、定量検出技術によって行われることができ、限定されるものではないが、共焦点レーザー走査顕微鏡によるチャンネル計数、免疫金検出並びに放射性結合及び置換技術が挙げられる。更に、幾つかの非必須アミノ酸のポリペプチド鎖又は側鎖の化学的修飾は、Kv1.3チャンネルの検出及び定量化に使用できる標識化された機能的に同等な類似体を提供することができる。同様に、その機能的に等価な類似体は、特異的なリガンドを検索するための分子プローブとして使用でき、但し、その新規なリガンドは、Kv1.3上にVm23、Vm24及びそれらの機能的に同等な類似体と同一の結合部位を共有する。かかる化学的修飾は、Kv1.3に対して高い親和性及び特異性を持つリガンドを与えるVm23、Vm24及びそれらの機能的に同等な類似体の構造決定を未修飾のままにしておくべきである。ポリペプチド鎖の化学修飾は、有効な分子プローブを得るための常法であり、広く利用できる幾つかの方法によって達成でき、当業者に常用されている。修飾によって与えられる標識には、限定されないが、放射性同位体部分、蛍光部分、化学発光部分又は発色性部分、及びタグタンパク質(抗体、ビオチン、緑色蛍光タンパク質又はその誘導体)との架橋結合又は融合が含まれる。
使用した全ての溶媒及び化学物質は分析グレードで、二回蒸留水は、以前説明したような手順によって使用された[Batista et al., 2007]。
上述した各種天然リガンドの単離に用いる手順は、クロマトグラフ技術を用いる。毒液を水中で可溶化し、10000xgで5分間遠心分離した。浮遊物を回収し、高速液体クロマトグラフィー(HPLC)で分離した。可溶性毒液タンパク質を1.0ミリグラム含有する100マイクロリットルを、HPLCシステム(Millenium Millipore, Milford, MA)のVydac(Hisperia, CA, USA)から得た分析C18逆相カラム(大きさ10×250mm,カタログ番号238TP)に適用した。溶液A(水中0.12%トリフルオロ酢酸TFAの水)から60%溶液B(アセトニトリル中0.10%TFA)までの線形勾配を用い、60分間流して、成分を精製した。230nmの吸光度にて検出を監視し、1ml/分の流速で溶出した。手動で画分を収集し、Savant Speed−Vac乾燥機を用いて乾燥させた。図1に示すように、80を超える異なる画分をこのHPLC分離から収集した。20〜35分の保持時間で溶出する画分は、通常、研究された他のサソリ毒液のKチャンネルに最も特異的なサソリ毒の溶出時間に対応する[Batista et al., 2007]。このため、この時間に溶出する毒液成分に専念するよう特に注意しなければならない。特に、23分と24分に溶出する二つの画分を更に分析した。なぜなら、この溶出時間からのペプチドの質量分析決定は、既知の他のKチャンネル特異性毒に対して見られた値と密接な関係があるからであり、それらは、約4000ダルトンの分子量を有する。それらの成分は依然として均質でないため、第二のクロマトグラフ分離を同一のHPLCシステムを用いて行い、但し、異なる勾配で溶出した(溶液Aから40%溶液B、60分間、Vydag,Hisperia CAのカタログ番号218TP54のC18カラムの使用)。図1の差し込み図に示されるように、主成分が、冒頭の各画分から単離された(アスタリスクでラベルされる)。左側の差し込み図は、23分で溶出する画分に対応しており、右側の差し込み図は、24分で溶出する画分に対応する。質量分析決定及び自動エドマン分解による配列決定による分析を受けて、両方のペプチドが均質であることが分かった。最初に、我々の実験条件で24分にて溶出したものを分析した。それは純粋であり、3864原子量単位(a.m.u.)の分子量が示された。本明細書において、我々は、一つの分子量単位を指定するため、置き換え可能なようにa.m.u.又はダルトン(Daと省略)を使用することになる。このため、そのペプチドはVm24と名付けられ、我々の実験条件において24分で溶出するV.メキシカナスからの毒液のペプチドを意味する。左側の差し込み図(図1)に示すクロマトグラムは、Vm23と名付けたペプチドの分離に対応する。それは、23分で溶出するV.メキシカナスからの毒液のペプチドを意味する。この成分の実験的分子量は、3665Daであると測定された。
毒液及び純ペプチドの効果は、サソリ毒液が種特異的な毒を含有することが知られているため、少なくとも三つの生物学的モデル:哺乳動物、コオロギ、甲殻類を用い、研究室で通常行われる。哺乳動物又は異なる種類の節足動物に特異的な毒がある([Possani et al., 1999]において概説されている)。本発明の目的のため、我々は、動物モデルとしてマウスを用いた実験を行ったが、これは、その結果が、毒液又は精製された毒と接触したヒトに起こる信頼性の高い徴候となるからである。V.メキシカナスの可溶性毒液を様々な量(体重20グラムのマウス当たり50〜200マイクログラムのタンパク質)で注射されたマウスは、中毒症状を示さなかった。通常、毒液が上記の物量で人に対して毒性を有する場合、興奮性、唾液分泌、呼吸促迫(呼吸困難)、後肢の麻痺、下痢、痙攣、更には死等、明らかな中毒症状が見られた[Possani el al., 1985]。注射された動物に上述の症状のいずれかが現れるがペプチド投与後24時間以内に回復したらこのペプチドは「毒性」であると言え、一方、マウスが死んだら、それは「致死」とよばれる。非毒成分は、中毒症状を誘発しないものであり、pH7.2のPBS食塩水が注射されたマウスと同様の挙動を起こす[Possani et al., 1985]。最終的には、比較的低い投与量での全毒液は毒性がないが、同様な投与量での精製されたペプチドは中毒症状を引き起こすことができ、これは、精製中、サンプルが特定の成分中に濃縮されるからである。このため、V.メキシカナスの可溶性毒液が200マイクログラム/20グラムマウス体重で毒性がないことを考慮して、純ペプチドVm24を様々な濃度でマウスに注射した。使用した最高投与量は、200マイクログラム/20グラム、即ち10000ミリグラム/キログラムマウス体重であり、中毒症状は観察されなかった。
二つの技術を用いた。自動エドマン分解と質量分析(MS)である。純毒の直接アミノ酸配列決定は、Beckman LF 3000タンパク質シークエンサー(Palo Alto,CA,USA)を用いて、該会社から供給された化学物質及び手順によって行われた。純ペプチドの還元・アルキル化したサンプルを、他のサソリ成分に関して前に説明された手順と同様な手順を用いて[Valdez et al., 2004; Batista et al., 2007; Diego-Garcia et al., 2007]、Arg−Cエンドペプチダーゼ(Roche Diagnostics, Basel, Switzerland)で酵素的に切断した。対応するペプチドをHPLCで精製し、配列決定した。質量分析については、サンプルを、Surveyor MSシリンジポンプ伝達システムを用いて、Finnigan LCQDUOイオントラップ質量分析計(San Jose, CA)中で直接適用した。サンプルの5%をナノスプレー源(0.5マイクロリットル/分)に入れるため、10マイクロリットル/分の溶出液を分割した。スプレー電圧を1.7kVに設定し、毛細管温度を130℃に設定した。MS/MS実験については、断片化ソースを、25Vの衝突エネルギー、35〜45%(任意単位)の正規化衝突エネルギー及び活性化した広帯域スキャンで操作した。全てのスペクトルを正イオンモードで得た。データ収集及びデータ解析をWindows(登録商標) NT PCシステム上のXcaliburソフトウェアで実行した。酵素的に生成したペプチドからのMS/MSスペクトルを手動及びSequestソフトウェアによって分析した[Batista et al., 2007]。
1)今までに既知である他のサソリ毒の全てと比較した一次構造は、50%を超える相違を有する(以下、例6参照)。この事実によって、ここで番号α−KTx21(例えばα−KTx21.1及びα−KTx21.2)と提案された新しいサブファミリー(今日まで未知である)の存在を証明する。それは、最初の開示であり、両ペプチドがイソギンチャク、ハチ及びヘビの毒ペプチドを含む他のリガンドと構造的に異なることを明確に示す。
2)両配列の位置10のアミノ酸までのN末端セグメントは、同一であり、ジスルフィド架橋を維持するシステインの8の位置の内の7が一次構造の同一位置にある。Vm23の八番目のシステイン(C8)は、C末端側の最末端の位置35に配置されており、Vm24と比較して一つ早いアミノ酸である。このため、Vm24は36の残基を有するが、Vm23は35のアミノ酸残基を有する。Vm23の最終システインはアミド化されていないが、我々は、構造的折り畳みがVm24のものと同じであることを仮定した。Vm23及びVm24の両方の生理学的効果が同程度であるため、N末端領域のアミノ酸配列が活性に重要であると見込まれる。Vm23及びVm24の一次構造を比較すると、五つの相違が位置10、13、17、23及び29で見られ、最後のアミノ酸とその前のアミノ酸の間に一つの挿入欠失がある(Vm23のY34及びVm24のY35)。最も変わりやすい領域は、両ペプチドの中央部であり(10〜30の残基で、6の相違のうち5が位置する)、おそらく該残基がペプチドの機能に重要でないことを示唆する。位置17(R/K)、23(N/S)及び29(R/K)の置換は、アルギニン(R)及びリシン(K)が帯電した塩基性アミノ酸である一方で、アスパラギン(N)及びセリン(S)は、非帯電の親水性アミノ酸残基であるため、保存的な修飾であることについて言及する価値がある。Vm24と比較して、位置35のチロシンの欠如(Vm23のシステインと置換)は、唯一つのチロシンが両ペプチドの同一な折り畳み及び機能に十分な量であることを示唆する。
3)このように、最も変わりやすい領域は中央部分に位置しており、保守的な置換を可能にする。これは、当業者が容易に設計でき、少なくとも83%の対配列同一性を共有する整列位置において類似の生理学的特性を持つアミノ酸による修飾又は置換が(ここではVm23及びVm24の場合が示される)、類似の特性を持つ機能的に同等な類似体を生成すると期待され、従って、本発明の範囲に含まれるべきである。
4)しかしながら、最も重要な特徴は、以下の例7〜10において説明するように、他の亜型のカリウムチャンネルと比べて、Vm23及びVm24の双方が有するhKv1.3への高い親和性である。Vm23及びVm24は、チャリーブドトキシン、アニュロックトキシン、BgK、ShK等の既知の他の遮断薬と比べてhKv1.3に対し高い親和性を有する[Panyi et al., 2006]。上述の他の遮断薬は、大きく異なるアミノ酸配列及び/又はジスルフィド対形成を有するか、又は異なる作用特異性及びKv1.3チャンネルに対する結合親和性を示す。Vm23及びVm24の一次構造の単純な分析から、それらが他のサソリ又はイソギンチャク毒と同様な方法でKv1.3に影響を与えるかは明らかでない。このように、専有情報及び使用を請求するVm23及びVm24の配列は、Kv1.3チャンネルに影響を与える他のペプチドの知識から明らかではない。本発明は、完全に異なる新規のアミノ酸配列(SEQ ID NO:1及びSEQ ID NO:2参照)を報告する。決定した配列が正しいことを示す更なる証拠は、合成的に調製したVm24で得られた結果からもたらされる。天然Vm24及び合成的に調製したVm24の双方は、以下に示すように(例9)、全く同じ生理作用を有する。
5)最後に、二つのペプチドで見出した他の重要な事実は、それらが実験動物に対し相対的に高い濃度(20グラムマウス体重当たり最大10000マイクログラム)で注射した場合毒性がないことである。これは、以前述べたように、既知の他のサソリ毒液、例えば、Centruroides noxiusからのCn2と比較してかなり顕著である[Zamudio et al., 1992]。Vm24よりも約800倍低い投与量でマウスに注入したCn2は、50%の死亡率をもたらす。
サソリ毒液は、成分の非常に複雑な混合物であり、イオンチャンネルに関し活性がある短鎖ペプチド及び長鎖ペプチド([Possani and Rodriguez de Ia Vega, 2006]において概説されている)、遊離アミン、ヌクレオチド、炭水化物、脂質(Possani et al., 1999において概説されている)、ホスホリパーゼ[Zamudio et al., 1997: Valdez et al., 2004]、ヒアルロニダーゼ、リゾチーム[Batista et al., 2007]等の酵素、及び未知の機能を有するその他多くのタンパク質成分[Diego-Garcia et al., 2007]を備える。加えて、サソリは、地表上で3億年以上も進化した非常に古代の生物であり、その餌を狩ったり又は捕食動物から自身を守るための特殊な手段を選択する時間があった。このため、その毒液における生物学的活性な成分の存在を捜し求めることは魅力的で且つ賢明である。質量分析法及び装置に関する最近の進歩の結果、今では全毒液のマスフィンガープリント分析を得ることが可能である。このため、V.メキシカナスの可溶性毒液で行った最初の研究の一つは、全ての成分についての分子量の同定であり、Finnigan LCQDUO(San Jose, CA)イオントラップ質量分析計(EIS/MS)及びAmersham Bioscienses(Uppsala, Sweden)からのマトリクス支援レーザー脱離イオン化飛行時間(MALDI−TOF)型Ettan MALDI−TOF/Pro装置を用いて同定される。使用した戦略は、精製分離の純ペプチド又はC18逆カラム(図1)において混合物として溶出する関連成分のファミリーによって予め選択し、次いで、質量分析によってその分子量を分析することであった。表1において、HPLCシステムから収集した画分の保持時間は、各画分で見られた成分の分子量に従ってリストに記載されている。340を超えた異なる分子量の成分を決定した。一部の成分は、HPLC分離システムの隣接する二つのサブ画分に現れるが、この場合、一つしか数えないことにについて言及する価値がある。また、一部の画分を同定していない(NDとラベルされている)。
Vm23のC末端アミノ酸配列での最終残基に関して、結果は明確である。本明細書の例1において説明した配列分析は、その配列は図2において示されるが、最終残基がアミド化されず遊離カルボキシルシステイン残基を有する一次構造を終結させるということをほぼ明確にする。
大きさが小さいため、Vm24の毒は、構造−機能の関係を研究するための理想分子である。該ペプチドは、位置6、12、16、21、26、31、33及び36に位置する8のシステイン残基を含有し、四つの分子内ジスルフィド結合を形成する。この例は、タンパク質酵素で切断した後、RP−HPLCにより精製したペプチドのエドマン分解及び質量分析決定を用いた、Vm24のジスルフィド結合パターンの決定について説明する。
この例では、Vm24の化学合成を説明する前に、ペプチド化学のテーマにおける一部の基本概念と、存在する生成物の自然抽出よりむしろ化学合成によってリガンドを生成する論理的証拠について取り上げることが重要である。
SEQ ID NO:1及びSEQ ID NO:2の配列は、短いペプチド鎖、リッチな塩基性アミノ酸残基、類似のシステインパターン等、α−KTxファミリー内で分類される他の短鎖サソリ毒[Tytgat et al., 1999]との類似点をいくらか示す。このファミリーの全てのメンバーは、構造的に関連しており、カリウムチャンネル遮断薬として類似の機能を果たす。それにもかからわず、それらは、特定の種類及び亜型のカリウムチャンネルに対して多様な選択性を示す[Rodriguez de la Vega and Possani, 2004; Panyi et al., 2006]。分類を提案した研究者の国際発表による論理的根拠は、所定のサブファミリーがそのメンバーの中で高い割合の類似性及び他のサブファミリーのメンバーとの低い同一性によって識別できることであった[Tytgat et al., 1999]。その後、この識別がほとんどのサブファミリーの薬理学的スペクトルを反映することが実証された[Rodriguez de la Vega et al., 2003; Zhu et al., 2004)]。従って、相対的に制限されたファミリーの多様性を−そのサイズを詰めて−考慮すると、所定の機能スペクトルを与える分子特性及び構造特性を同定することは重要である。この種の分析は、通常、配列比較及び系統発生学的推測により行われる。該比較の基礎にある考えは、同一系列に属するタンパク質が一つ又は複数の先祖遺伝子の複製及び分岐に従って又は付随した種分化の現象に関係しているという仮説によるものであり、生物情報学分析によるその進化史の再構築を可能にし、所定の配列空間内での適応度地形を浄化するのに役立つ[Thornton and DeSaIIe, 2000; Orengo and Thornton, 2005]。
Vm24によるhKv1.3チャンネルの高親和性遮断
Vm24によるhKv1.3チャンネルの遮断は、ヒト末梢血T細胞で内因的に発現したチャンネルにおいて特徴付けられた[Peter et al., 2001; Bagdany et al]。電気生理学的実験用のT細胞を得るための手順の簡単な説明は以下の通りである。ヘパリン化ヒト末梢静脈血を健康なボランティアから得た。単核細胞をFicoll−Hypaque濃度勾配遠心分離によって分離した。収集した細胞を、25mMのHEPES緩衝液(pH7.4)を含有するCa2+及びMg2+遊離ハンク溶液で2回洗浄した。37℃の5%CO2恒温器中、10%のFCS/Hyclone(Logan,Utah,USA)、100マイクログラム/mlのペニシリン、100マイクログラム/mlのストレプトマイシン及び2mMのLグルタミンが補給されたRPMI−1640の24ウェル培養皿において、0.5×106/mlの密度で3〜4日間細胞を培養した。また、培地は、フィトヘマグルチニンA(PHA−P,Sigma-Aldrich Kft, Hungary)を6又は8マイクログラム/ml含有し、Kチャンネル発現を増加させた[Deutsch et al., 1986]。Tリンパ球は、Matteson及びDeutsch(Matteson et al., 1984)によって説明されるように、マウスの抗ヒトCD2(Becton-Dickinson, San Jose, CA, USA)とのインキュベーションと、その後のヤギ抗マウスIgG抗体(Biosource, Camarilo, CA, USA)でコーティングしたペトリ皿に対する選択的接着とによって電流を記録するために選択された。上記皿を、パッチクランプ試験のため、1mlの正常な細胞外バス培地(以下、参照)で5回穏やかに洗浄した。
例7の導入にて概要を述べたように、ペプチドが選択的免疫抑制剤であるために最も重要な要求の一つは、IKCa1を越えたKv1.3への選択性である。また、IKCa1チャンネルは、T細胞で内因的に発現する[Grissmer et al., 1993]。IKCa1チャンネルによって運ばれる電流は、1μMの遊離Ca2+濃度を有するピペット注入溶液を用いて測定でき、そのチャンネルを完全に活性化させるのに十分である[Grissmer et al., 1993]。しかしながら、同一細胞中でのKv1.3チャンネルの同時存在は、IKCa1チャンネルの薬理学的な特徴付けを困難にし、Kv1.3チャンネルを活性化しない場合の膜電位の研究を制限する[Grissmer et al., 1993; Bagdany et al., 2005]。このことと、刺激したT細胞中でさえ比較的少数のIKCa1チャンネルとが、我々に組換えチャンネルを用いたIKCa1薬理学を研究させる動機付けとなった。
本研究で用いた全てのチャンネル構築は、薬理学的アッセイ及び生理学的アッセイにおいて日常的に使用されており、その適用性はリストに挙げた参考文献において確認されている。
一過性形質転換:Cos−7細胞は、ラットKv2.1(rKv2.1、Dr. S. Korn, U. of Connectientから贈り物)[Immke et al., 1999];ヒトKv1.2(hKv1.2、Kv1.2の完全コード配列を含有するpcDNA3/Hygroベクター、Dr. S. Grissmer, U. of Ulm, Germenyからの贈り物)[Visan et al., 2004];ヒトKv1.4(hKv1.4ΔN:Kv1.4の不活性化球欠失変異体、D. Fedia, University of British Columbia, Vancouver, Canadaからの贈り物)[Kurata et al., 2004];及びヒトNav1.5(R. Horn, Thomas Jefferson University, Philadelphia, PA, USAからの贈り物)[O'Leary et al., 1995; Ahern et al., 2005]チャンネルを発現するのに使用された。tsA−201細胞は、hBKチャンネル(hSlo1遺伝子(U11058)、pCI−ネオプラスミド、Toshinori Hoshi, Uniersity of Pennsylvania, Philadelphia, PAからの贈り物)[Avdonin et al., 2003]を発現するのに使用された。その全てのチャンネルクローンは、製造者のプロトコールに従い、リポフェクタミン2000試薬を用いて、緑色蛍光タンパク質(GFP)をモル比1:5でコード化したプラスミドにより、一過性に同時形質導入され(Invitrogen, Carlsbad, CA, USA)、標準条件下で培養された。形質導入後、電流を2〜3日間記録した。GFP陽性細胞の形質導入をNikon TE2000U蛍光顕微鏡において同定し、電流の記録に用いた(同時形質導入では>70%の成功率)。
安定な細胞株:mKv1.1チャンネルを安定に発現するL929細胞及びhKv1.5チャンネルを安定に発現するMEL細胞は、前に説明され[Grissmer et al., 1994]、Dr.Heike Wulff(UC Davis, CA, USA)の贈り物であった。hERGチャンネルは、HEK−293細胞株において安定な方法で発現した。
例5で説明したように、理論上及び実施上の考慮すべき事項がVm24の化学合成に導いた。説明のとおり、合成した毒の構造及び純度を分析的HPLC、アミノ酸配列及び質量分析決定によって確認した。その全てのアプローチは、合成したVm24(sVm24)の一次配列が天然ペプチドのものと一致することを示した。更に、sVm24の生成に対するプロトコールは、折り畳みが天然ペプチドの場合と同じジスルフィド対形成に制限されるように設計されたチオール基のための異なる保護基を用いた。しかしながら、このデータは、ペプチドの生理学的活性が維持されることを保証しない。高親和性の遮断を媒介するチャンネル及びペプチドの相補的表面は、非常に複雑であり、天然ペプチドの構造からの最小限の逸脱は、Kv1.3を遮断する際のsVm24の効果を落とし得る[Giangiacomo et al., 2004]。
サソリ毒液は、大量の生物活性ペプチドを含む。所定の種の毒液は、V.メキシカナスのVm23及びVm24の場合のように、高度な配列同一性を有するペプチドを含む場合が多い(例1参照)。ペプチドの高度な配列同一性は、通常個々の分子に対して異なる生物活性を与える。これは、イオンチャンネルの外部前庭に位置する毒受容体とのペプチド毒の相互作用が、長距離静電相互作用、短距離静電相互作用及び密接相互作用を含めた複数の機構によって決定され、その要因の正味効果は、結合親和性及び選択性を決定する[Park and Miller, 1992; Peter et al., 2001; Giangiacomo et al., 2004]。以前示されたことには、同じサソリの二つのペプチド(P.imperatorのPi2及びPi3)は、単一のアミノ酸のみが異なり、その両方がサブナノモル濃度の濃度にてKv1.3を遮断する[Peter et al., 2001]。また、Miller及び共同研究者の先駆けの研究から[Goldstein et al., 1994]明らかであるのは、チャリーブドトキシンの幾つかの位置における非保存的なアミノ酸置換でさえ十分に許容され、毒は、Shakerカリウムチャンネルに対する親和性を維持する(例えば、T9K及びN22K)。また、Vm23とVm24間の差異がKv1.3に対する親和性及び特異性を変化させるのに重要でないことを検証するため、Vm23の薬理学的プロファイルを以下に示されるように詳細に試験した。
世界中の何億人もの人々は、限定されないが、特に、多発性硬化症、関節リウマチ、I型糖尿病、自己免疫性乾癬、紅斑性狼瘡、潰瘍性大腸炎、交感性眼炎、骨吸収歯周病、免疫性血小板減少性紫斑病、自己免疫性リンパ球増殖性症候群等の自己免疫疾患によって影響される。最近考えられていることには、これらの疾患の発症は、エフェクターメモリT細胞(TEM)に形質転換される休止状態の疾患特異的な自己反応性T細胞の活性化を伴う。自己反応性T細胞は、組織内への移動、炎症サイトカインの分泌及び即時性エフェクター効果の発揮による反復自己抗原刺激及び炎症損傷への寄与のため、未処理状態と連続活性化メモリT細胞を区別し得る[Sallusto et al., 1999]。遅延型過敏症(DTH)に関する機構は、TEM細胞に引き起こされる皮膚損傷の他の例である[Soler et al., 2003]。また、多くの自己免疫疾患の発症は、メモリB細胞、特にはクラス転換CD27+IgD−サブセットに属するものに起因することがある[Iglesias et al., 2001; O'Connor et al., 2001; Corcione et al., 2004]。これらの理由により、細胞の他のリンパ球サブセットの活性を損なわず、選択的にTEM及びクラス転換メモリB細胞を標的とできる治療薬を開発することが望ましく、この方法で急性免疫反応の障害を避ける。本発明の例7において述べたように、電圧開口型Kv1.3チャンネルは、TEM及びクラス転換メモリB細胞の免疫修飾のための新規な治療上の標的である。TEM細胞は活性化時にkV1.3を上方制御し、その抗原駆動の増殖は、Kv1.3チャンネルを遮断する既知の物質に対してかなり感受性がある[Wulff et al., 2003]。逆に、未処理細胞及びTEMは、Kv1.3遮断薬に対して感受性がほとんどなく、カルシウムで活性化したKチャンネルKCa3.1を上方制御することで、Kv1.3に対してすぐに抵抗性を持つようになる[Wulff et al., 2003; Chandy et al., 2004]。分化過程の間、B細胞及びT細胞は、KCa3.1からKv1.3にそのカリウムチャンネル依存性を変化させる[Wullf et al., 2004]。この事実により、Kv1.3チャンネル遮断薬は、未処理細胞及びCD27+IgD+メモリ細胞に影響を与えることなく、該細胞の増殖を阻害する。従って、Kv1.3チャンネルに特異的な遮断薬の使用は、TEM及びクラス転換メモリB細胞に優先的に影響を与え、免疫反応の大半を落とすことがないが、自己免疫疾患の結果として発生した健康状態を改善する。Kv1.3チャンネルの遮断は、動物モデルにおいて、実験的な自己免疫性脳脊髄炎(EAE)、歯周病骨吸収及びDTH反応を、明らかな副作用を引き起こすことなく、改善する[Koo et al., 1997; Beeton et al., 2001; Valverde et al., 2004]。ペプチドによるKv1.3チャンネルの遮断はすぐに逆にできるため、化学療法剤又はモノクローナル抗体を用いたときの場合ではなく、正常なレベルに戻るまで数ヶ月かかる治療の過程を制御することができる。明らかなことには、主な問題は、この治療上の処置に対して非常に選択的なペプチドを発見することである[Chandy et al., 2004]。
Claims (31)
- 配列番号1又は配列番号2から構成される単離精製されたペプチドであり、高い親和性及び特異性でカリウムチャンネルKv1.3を遮断することができることを特徴とする単離精製されたペプチド。
- 哺乳類細胞においてKv1.3カリウムチャンネル活性を遮断するのにペプチドと前記哺乳類細胞とを接触させることを備えた哺乳類細胞におけるKv1.3カリウムチャンネル活性阻害用の配列番号1もしくは配列番号2のアミノ酸配列、又はその薬学的に許容される塩を有するペプチド。
- 前記哺乳類細胞がヒトリンパ球細胞であることを特徴とする請求項2に記載のペプチド。
- Kv1.3カリウムチャンネル遮断活性を有するペプチドであって、
前記ペプチドが、T-リンパ球細胞をペプチドと接触させることを備えたT-リンパ球細胞におけるカルシウムシグナル経路減衰用の配列番号1又は配列番号2から構成されるアミノ酸配列を有することを特徴とするペプチド。 - 哺乳類細胞においてKv1.3カリウムチャンネル活性を阻害する生体外での方法であって、
前記哺乳類細胞において前記チャンネル活性を遮断するのに効果的な量のペプチドと、前記哺乳類細胞とを接触させる工程を含み、
前記ペプチドが、配列番号1もしくは配列番号2のアミノ酸配列、又はその薬学的に許容される塩を有することを特徴とする生体外での方法。 - 前記哺乳類細胞がヒトリンパ球細胞であることを特徴とする請求項5に記載の生体外での方法。
- T-リンパ球細胞においてカルシウムシグナル経路を減衰させる生体外での方法であって、
前記T-リンパ球細胞を、Kv1.3カリウムチャンネル遮断活性を有するペプチドの効果的な量と接触させる工程を含み、
前記ペプチドが、配列番号1又は配列番号2から構成されるアミノ酸配列を有することを特徴とする生体外での方法。 - Kv1.3カリウムチャンネル遮断活性を有するペプチドであって、
前記ペプチドが、T-細胞の集団をペプチドと接触させることを備えた哺乳動物の免疫系におけるT-細胞活性化過程抑制用の配列番号1もしくは配列番号2から構成されるアミノ酸配列、又はその薬学的に許容される塩を有することを特徴とするペプチド。 - 前記T-細胞の活性化が、前記哺乳動物の免疫反応によって引き起こされることを特徴とする請求項8に記載のペプチド。
- 前記免疫反応が、異種臓器拒絶反応の結果であるか又は自己免疫疾患の結果であることを特徴とする請求項8又は9に記載のペプチド。
- 前記哺乳動物がヒトであることを特徴とする請求項8〜10のいずれかに記載のペプチド。
- Kv1.3カリウムチャンネル遮断活性を有するペプチドを含む組成物であって、
前記ペプチドが、哺乳動物の免疫反応抑制用の配列番号1もしくは配列番号2から構成されるアミノ酸配列、又はその薬学的に許容される塩を有することを特徴とする組成物。 - 前記免疫反応が、異種臓器拒絶反応の結果であるか又は自己免疫疾患の結果であることを特徴とする請求項12に記載の組成物。
- Kv1.3カリウムチャンネル遮断活性を有するペプチドを含む組成物であって、
前記ペプチドが、異種臓器拒絶反応の予防的な処置又は治療上の処置を必要とする対象における異種臓器拒絶反応の予防的な処置又は治療上の処置用の配列番号1もしくは配列番号2から構成されるアミノ酸配列、又はその薬学的に許容される塩を有することを特徴とする組成物。 - 拒絶された臓器が、心臓、肺、肝臓、腎臓又は膵臓であることを特徴とする請求項14に記載の組成物。
- 前記処置を必要とする対象がヒトであることを特徴とする請求項14又は15に記載の組成物。
- Kv1.3カリウムチャンネル遮断活性を有するペプチドを含む組成物であって、
前記ペプチドが、リンパ球TEMに関連した自己免疫疾患の予防的な処置又は治療上の処置を必要とする対象におけるリンパ球TEMに関連した自己免疫疾患の予防的な処置又は治療上の処置用の配列番号1もしくは配列番号2から構成されるアミノ酸配列、又はその薬学的に許容される塩を有することを特徴とする組成物。 - 前記処置を必要とする対象がヒトであることを特徴とする請求項17に記載の組成物。
- 前記リンパ球TEMに関連した自己免疫疾患が、多発性硬化症、関節リウマチ、I型糖尿病、自己免疫性乾癬、紅斑性狼瘡、潰瘍性大腸炎、交感性眼炎、骨吸収歯周病、免疫性血小板減少性紫斑病及び自己免疫性リンパ球増殖性症候群からなる群から選択されることを特徴とする請求項17又は18に記載の組成物。
- 更に、前記対象に対して少なくとも一つの追加の免疫抑制剤を投与することを含むことを特徴とする請求項17、18又は19に記載の組成物。
- 前記追加の免疫抑制剤が、シクロスポリン、ラパマイシン、アザチオプリン、プレドニゾン、ShK毒素、ShK誘導体、デオキシスパガリン、及びそれらの塩からなる群から選択されることを特徴とする請求項20に記載の組成物。
- Kv1.3カリウムチャンネル遮断活性を有する少なくとも一つのペプチドと、薬学的に許容されるキャリアーと、を含む医薬品組成物であって、
前記ペプチドが、配列番号1もしくは配列番号2から構成されるアミノ酸配列、又はその薬学的に許容される塩を有することを特徴とする医薬品組成物。 - 少なくとも一つの追加の免疫抑制剤を更に含み、
前記追加の免疫抑制剤が、シクロスポリン、ラパマイシン、アザチオプリン、プレドニゾン、ShK毒素、ShK誘導体、デオキシスパガリン、及びそれらの塩からなる群から選択されることを特徴とする請求項22に記載の医薬品組成物。 - 局所的、全身的、静脈内、腹腔内、筋肉内、皮下、鼻腔内、経皮的もしくは経口的に投与されるか、又は皮内注射、気管支内点滴、胃腸内送達もしくは経粘膜的送達によって投与されることを特徴とする請求項14〜21のいずれかに記載の組成物。
- Kv1.3カリウムチャンネルに結合する高い親和性及び特異性を有する標識ペプチドであって、配列番号1又は配列番号2から構成されるアミノ酸配列を有することを特徴とする標識ペプチド。
- 標識付け部分が、放射性同位体、蛍光部分、化学発光部分、発色団、ビオチンリガンド及びタンパク質からなる群から選択されることを特徴とする請求項25に記載の標識ペプチド。
- 標識付けタンパク質が、抗体、又は緑色蛍光タンパク質、又は異なる発光スペクトルを有する前記緑色蛍光タンパク質由来のタンパク質であることを特徴とする請求項26に記載の標識ペプチド。
- Kv1.3チャンネルを発現する細胞を同定する生体外での方法であって、
a)Kv1.3カリウムチャンネルに結合する高い親和性及び特異性を有する標識ペプチドと標的細胞の集団を接触させる工程であって、前記標識ペプチドが、配列番号1又は配列番号2から構成されるアミノ酸配列を有し、標識化されている工程と、
b)検出技術によって、前記標的細胞の集団に存在するKv1.3カリウムチャンネルに結合した標識ペプチドを検出する工程と
を含むことを特徴とする生体外での方法。 - 前記細胞がヒトリンパ球であることを特徴とする請求項28に記載の生体外での方法。
- 所定の細胞に発現したKv1.3チャンネルの数を定量化する生体外での方法であって、
a)Kv1.3カリウムチャンネルに結合する高い親和性及び特異性を有する標識ペプチドと前記細胞を接触させる工程であって、前記標識ペプチドが、配列番号1又は配列番号2から構成されるアミノ酸配列を有し、標識化されている工程と、
b)定量的な検出技術によって、前記標的細胞の集団に存在するKv1.3カリウムチャンネルに結合した標識ペプチドを検出する工程と
を含むことを特徴とする生体外での方法。 - 前記細胞がヒトリンパ球であることを特徴とする請求項30に記載の生体外での方法。
