本発明は、対象ペプチドの発現を誘導するために利用される、マンニトール、グルシトールおよびアラビトールまたはそれらの誘導体または類似体から成る群より選択される炭素源の代謝のために必要な酵素を欠損している細菌細胞であって、前記対象ペプチドをコードする核酸配列が、マンニトール、グルシトール、アラビトールおよびそれらの誘導体または類似体から成る群より選択される炭素源によって誘導されるプロモーターに作動可能に連結されている、前記細菌細胞を提供する。本発明は、組換えペプチドを発現することができるいかなる細菌細胞の利用も考慮する。そのような細胞は当分野において周知である。一部の実施形態では、宿主細胞は大腸菌細胞であり得る。もう1つの実施形態では、細胞は、シュードモナス属または密接に関連する細胞から選択される。他の実施形態では、細菌細胞はシュードモナス・フルオレセンスである。一部の実施形態では、細菌細胞は、マンニトール、グルシトールまたはアラビトールまたはそれらの誘導体または類似体から成る群より選択される炭素源の代謝のために必要な酵素に関して欠損性であり得る。

ベクターは、1またはそれ以上の表現型選択マーカーおよびベクターの維持を確実にするためおよび、所望する場合は、宿主内での増幅を与えるための複製起点を含む。あるいは、ベクターは、宿主細胞のゲノム内への組み込みを可能にする配列をさらに提供することができる。本開示に従った形質転換のための適切な宿主は、炭素源誘導性プロモーターによって駆動される組換えペプチドを発現することができるいかなる細菌細胞も含む。一部の実施形態では、細菌は、エシェリキア(Escherichia)およびシュードモナス属内の様々な種を含み得る。
IV.細菌宿主細胞における組換えポリペプチドの発現
a)本発明において述べるように、宿主細胞、好ましくは大腸菌またはシュードモナス・フルオレセンスを提供すること;
b)宿主細胞を、少なくとも1個のマンニトール、グルシトールまたはアラビトール誘導性プロモーターに作動可能に連結された少なくとも1個の対象組換えポリペプチドを含む少なくとも1個の核酸発現ベクターでトランスフェクトし、宿主細胞を十分な増殖培地で増殖させること;および
c)マンニトール、グルシトールまたはアラビトール、またはそれらの類似体または誘導体を、対象ペプチドの発現を誘導することができる量で増殖培地に添加すること(但し、宿主細胞は、添加されたマンニトール、グルシトールまたはアラビトール、またはそれらの類似体または誘導体を分解または代謝することができないようにされている);および
d)対象標的組換えポリペプチドを発現すること
を含む。

蛍光分析
マンニトール、グリセロールおよびグルコース濃度の分析
MB101が、RXF01195から始まる、DSM50106に述べられているものに類似したmtlオペロンを有することを示した(mtlE)。4個の異なるシュードモナス菌株、P.エルギノーサPA0−1、P.フルオレセンスDSM50106、P.フルオレセンスPf0−1およびP.フルオレセンスMB101におけるmtlオペロンの遺伝子アラインメントを図3に示す。2種類の遺伝子配置が存在し、1つは転写アクチベーター遺伝子、mtlRがmtlオペロンのすぐ上流に位置し、1つはmtlRがmtlオペロンのすぐ上流に存在しないものであって、これはMB101およびDSM50106の場合である。
Claims (11)
- (a)配列番号9または配列番号12;および
(b)配列番号9または配列番号12に示すヌクレオチド配列と少なくとも95%の配列同一性を有するヌクレオチド配列
から成る群より選択される核酸配列を含むマンニトール誘導性プロモーターに作動可能に連結された、組換えポリペプチドをコードする少なくとも1個の核酸構築物を含む、シュードモナス・フルオレセンス細胞であって、
前記シュードモナス・フルオレセンス細胞が、マンニトールの代謝または分解を阻害するように遺伝的に変化している、前記シュードモナス・フルオレセンス細胞。 - 前記核酸構築物が、mtlアクチベータータンパク質結合領域として働く核酸配列をさらに含む、請求項1に記載の細胞。
- 遺伝的変化が、マンニトールまたはその誘導体もしくは類似体の代謝において必要な遺伝子の突然変異または欠失である、請求項1に記載の細胞。
- 前記遺伝子がmtlオペロンから選択される、請求項3に記載の細胞。
- 前記遺伝子が、mtlD、mtlYおよびmtlZ、またはそれらの組合せから成る群より選択される、請求項4に記載の細胞。
- a)組換えポリペプチドを発現することができるシュードモナス・フルオレセンス宿主細胞を提供すること;
b)i)配列番号9または配列番号12;および
ii)配列番号9または配列番号12に示すヌクレオチド配列と少なくとも95%の配列同一性を有するヌクレオチド配列
から成る群より選択される核酸配列を含むマンニトール誘導性プロモーターに作動可能に連結された組換えポリペプチドをコードする少なくとも1個の核酸構築物で前記宿主細胞を形質転換すること;
c)前記宿主細胞を増殖培地で増殖させること;
d)前記マンニトール誘導性プロモーターから対象ペプチドの発現を誘導することができる量で炭素源を増殖培地に添加すること;および
e)前記マンニトール誘導性プロモーターを誘導することによって対象組換えポリペプチドを発現すること;および
f)発現された組換えポリペプチドを単離すること
を含み、前記宿主細胞が、前記マンニトール誘導性プロモーターを誘導することができるマンニトールの代謝または分解を阻害するように遺伝的に変化している、シュードモナス・フルオレセンス宿主細胞において組換えポリペプチドを発現するための方法。 - 前記プロモーターが、mtlアクチベータータンパク質結合領域として働く核酸配列をさらに含む、請求項6に記載の方法。
- 前記遺伝的変化が、マンニトールまたはその誘導体もしくは類似体の代謝において必要な遺伝子の突然変異または欠失である、請求項6に記載の方法。
- 前記遺伝子がmtlオペロンから選択される、請求項7に記載の方法。
- 前記遺伝子が、mtlD、mtlYおよびmtlZ、またはそれらの組合せから成る群より選択される、請求項9に記載の方法。
- 前記シュードモナス・フルオレセンス宿主細胞が、マンニトールの代謝または分解を阻害するように遺伝的に変化している、請求項6に記載の方法。
