Description
[1] 以下の(a)又は(b)の組換えダニアレルゲン、
(a) 配列番号14で表わされるアミノ酸配列を含む組換えダニアレルゲン
(b) 配列番号14で表わされるアミノ酸配列において1若しくは数個のアミノ酸が欠失、置換若しくは付加されたアミノ酸配列を含み、かつダニアレルゲン活性を有する組換えダニアレルゲン
[2] 以下の(a)又は(b)のダニアレルゲンをコードする遺伝子、
(a) 配列番号14で表わされるアミノ酸配列を含むダニアレルゲン
(b) 配列番号14で表わされるアミノ酸配列において1若しくは数個のアミノ酸が欠失、置換若しくは付加されたアミノ酸配列を含み、かつダニアレルゲン活性を有するダニアレルゲン
[3] 以下の(c)又は(d)のDNAを含む遺伝子、
(c) 配列番号13で表わされる塩基配列を含むDNA
(d) 配列番号13の塩基配列を含むDNAと相補的な配列を含むDNAとストリンジェントな条件下でハイブリダイズし、ダニアレルゲン活性を有するタンパク質をコードするDNA
[4] [1]のダニアレルゲンの断片ペプチド、
[5] 配列番号5から7で表わされるアミノ酸配列を少なくとも1つ含むアミノ酸配列からなる[4]の断片ペプチド、
[6] [4]又は[5]の断片ペプチドをコードするダニアレルゲンの断片遺伝子、
[7] [2]若しくは[3]の遺伝子又は[6]に記載の断片遺伝子を含有する組換えベクター、
[8] [1]のダニアレルゲンと他のタンパク質との融合タンパク質、
[9] [7]の発現ベクターで形質転換された細菌、酵母、昆虫細胞又は動物細胞、
[10] [9]の細菌、酵母、昆虫細胞又は動物細胞を該遺伝子の発現可能な条件下で培養して、組換えダニアレルゲンを産生させ、次いで該組換えダニアレルゲンを回収することを特徴とする組換えダニアレルゲンの製造方法、
[11] [9]の細菌、酵母、昆虫細胞又は動物細胞を該遺伝子の発現可能な条件下で培養して、融合組換えダニアレルゲンを産生させ、該融合組換えダニアレルゲンを回収し、次いで、融合している他のタンパク質を脱離せしめることを特徴とする組換えダニアレルゲンの製造方法、
[12] [1]の組換えダニアレルゲン、[4]の断片ペプチド又は[8]記載の融合タンパク質を有効成分とするダニアレルギー疾患治療剤、
[13] [1]の組換えダニアレルゲン、[4]の断片ペプチド又は[8]記載の融合タンパク質を有効成分とするダニアレルギー疾患診断薬、
[14] [1]に記載のダニアレルゲンに対する抗体、
[15] モノクローナル抗体である[14]のダニアレルゲンに対する抗体、
[16] [15]のモノクローナル抗体を産生するハイブリドーマ、
[17] [14]又は[15]の抗体を用いる屋内塵中のダニアレルゲンの免疫学的測定方法、ならびに
[18] ELISAである[17]の屋内塵中のダニアレルゲンの免疫学的測定方法。
[19] [12]のダニアレルギー疾患治療剤を非ヒト動物に投与し、減感作により非ヒト動物のアレルギー疾患を治療する方法。
[20] 非ヒト動物がイヌである[19]の方法。
(1) ダニアレルゲンSpot39/40タンパク質の単離及び部分配列決定
ダニ抽出物を2次元電気泳動にかけ、臨床的にダニアレルギーと診断された動物から得られたダニアレルゲン特異的IgEを用いて2次元電気泳動により分離されたダニアレルギーを特定する。
抽出したダニアレルゲンを用いて、公知の方法で部分配列決定を行うことができる。部分配列を決定する公知の方法として、MS/MS解析によるde novo sequencingやペプチドマッピングがある。
ダニよりmRNAを抽出し、該mRNAを鋳型にしてダニアレルゲンcDNAを合成し、cDNAライブラリーを作製し、スクリーニングすることにより、本発明のダニアレルゲンをコードするDNAを得ることができる。
本発明は、ダニアレルゲンであるSpot39/40タンパク質の断片であって、配列番号5から7に示されるアミノ酸配列の少なくとも1つを含むアミノ酸配列からなるダニアレルゲンを含む。
コナヒョウヒダニ(Dermatophagoides farinae)はマウス・ラット・ハムスター用粉末飼料(オリエンタル酵母工業株式会社、Tokyo,Japan)と襖の混合培地(7:3)中で湿度75%、25℃において30日間培養したものを用いた。ダニ培養物を飽和食塩水に懸濁して遠心分離した。その上清を含む浮遊物を濾過した。濾紙上(Tokyo Roshi Kaisha,Ltd.110 mm)に回収されたダニ虫体を、使用するまで−80℃で保存した。
ダニ虫体抽出物(Dfb)をTCA沈殿処理して、等電点3〜10、分子量1万〜20万までの成分を分離する2次元電気泳動を行って銀染色した結果、約500個のスポットが認められた(図1)。
Claims (13)
- 以下の(a)〜(c)のいずれかの組換えダニアレルゲン。
(a) 配列番号14で表わされるアミノ酸配列を含む組換えダニアレルゲン
(b) 配列番号14で表わされるアミノ酸配列において1若しくは数個のアミノ酸が欠失、置換若しくは付加されたアミノ酸配列を含み、かつダニアレルゲン活性を有する組換えダニアレルゲン
(c) 配列番号14で表わされるアミノ酸配列と90%以上の同一性を有するアミノ酸配列を含み、かつダニアレルゲン活性を有する組換えダニアレルゲン - 以下の(a)〜(c)のいずれかのダニアレルゲンをコードする遺伝子。
(a) 配列番号14で表わされるアミノ酸配列を含むダニアレルゲン
(b) 配列番号14で表わされるアミノ酸配列において1若しくは数個のアミノ酸が欠失、置換若しくは付加されたアミノ酸配列を含み、かつダニアレルゲン活性を有するダニアレルゲン
(c) 配列番号14で表わされるアミノ酸配列と90%以上の同一性を有するアミノ酸配列を含み、かつダニアレルゲン活性を有する組換えダニアレルゲン - 以下の(d)又は(e)のDNAを含む遺伝子。
(d) 配列番号13で表わされる塩基配列を含むDNA
(e) 配列番号13の塩基配列を含むDNAと相補的な配列を含むDNAとストリンジェントな条件下でハイブリダイズし、ダニアレルゲン活性を有するタンパク質をコードするDNA - 請求項2又は3に記載の遺伝子を含有する組換えベクター。
- 請求項1記載のダニアレルゲンと他のタンパク質との融合タンパク質。
- 請求項4記載の発現ベクターで形質転換された細菌、酵母、昆虫細胞又は動物細胞。
- 請求項6記載の細菌、酵母、昆虫細胞又は動物細胞を該遺伝子の発現可能な条件下で培養して、組換えダニアレルゲンを産生させ、次いで該組換えダニアレルゲンを回収することを特徴とする組換えダニアレルゲンの製造方法。
- 請求項6記載の細菌、酵母、昆虫細胞又は動物細胞を該遺伝子の発現可能な条件下で培養して、融合組換えダニアレルゲンを産生させ、該融合組換えダニアレルゲンを回収し、次いで、融合している他のタンパク質を脱離せしめることを特徴とする組換えダニアレルゲンの製造方法。
- 請求項1に記載のダニアレルゲンに対する抗体。
- モノクローナル抗体である請求項9記載のダニアレルゲンに対する抗体。
- 請求項10記載のモノクローナル抗体を産生するハイブリドーマ。
- 請求項9又は10に記載の抗体を用いる屋内塵中のダニアレルゲンの免疫学的測定方法。
- ELISAである請求項12記載の屋内塵中のダニアレルゲンの免疫学的測定方法。
