Description
(1)並行する複数の微小層流を形成することのできる培養基材上で細胞を培養する工程、
(2)微小層流のそれぞれに、異なる形質遺伝子を有する発現ベクター溶液を流す工程、
を含み、並行する微小層流のそれぞれに位置する培養細胞を異なる形質に転換することを特徴とする形質転換細胞集団の作製方法である。
並行する3本の微小流路(それぞれ幅100μm)を培養基材上に備えた微小流路チップを作成した。各微小流路の一端には溶液導入部を、また3本の微量流路から合流する溶液排出部を他端に形成した(図1参照)。
実施例2:微小流路内での細胞培養
NIH-3T3細胞を、10%FBSを添加したDMEM培地に懸濁し、この細胞懸濁液を微小流路チップの溶液導入部から3本の微小流路に導入した。液流が停止したことを確認した後、5%CO2、37℃のチャンバー内で静置した。細胞が十分に伸展した後に流路内の培養液を置換し、培養を継続した。
実施例3:微小流路内の培養細胞の形質転換
細胞がコンフルエント状態に達した後、微小流路の一つの溶液導入部から、GFP発現ベクターを含むリポフェクション試薬を導入し、残り二つの溶液導入部からは培養液を導入した。溶液排出部からリポフェクション試薬と培養液を吸引しながら、微小層流を30分間維持し、発現ベクターを細胞に接触させた。その後、全ての溶液導入部から培養液を導入してリポフェクション試薬を洗浄し、液流を止めた状態で培養を継続した。
Claims (2)
- 複数の異なる形質転換細胞集団を作製する方法であって、
(1)並行する複数の微小層流を形成することのできる培養基材上で細胞を培養する工程、
(2)微小層流のそれぞれに、異なる形質遺伝子を有する発現ベクター溶液を流す工程、
を含み、並行する微小層流のそれぞれに位置する培養細胞を異なる形質に転換することを特徴とする形質転換細胞集団の作製方法。 - 請求項1記載の方法で作製された形質転換細胞集団であって、培養基材上の並行する複数の微小層流のそれぞれに位置する培養細胞が異なる形質転換細胞であることを特徴とする形質転換細胞集団。
