Description
Claims (10)
- 長手方向軸を有するカラムと、カラムの内圧を検知する圧力センサー及び/又はカラムから流出する液体の量を測定する流量測定装置と、可動アダプターとを備えるカラム充填装置であって、
上記可動アダプターが、制御ユニットで監視しながら移動させることができ、当該装置が、自動化ソフトウェア及びハードウェアを具備する圧縮手段であって、カラムの長手方向軸に沿って、カラム内のベッド媒体の集密層を圧縮して、所定量まで圧縮された圧縮充填ベッドを形成する圧縮手段を備えており、
上記制御ユニットが、上記圧力センサー及び/又は流量測定装置からの信号を監視・解析して、可動アダプターでの集密層の圧縮を開始するブレイクポイントを決定するソフトウェアを備えており、該ブレイクポイントが、カラム内の圧力の低下及び/又はカラムから流出する流体の流量の低下が起きた時点で生じたと決定される
カラム充填装置。 - 前記ソフトウェアが、前記ブレイクポイントから所定量のベッド圧縮を達成するために可動アダプターが移動しなければならない距離を計算し、該距離に相当する位置への可動アダプターの移動を制御できる、請求項1記載のカラム充填装置。
- 前記ソフトウェアが、ブレイクポイントの位置に対応するオペレータ読取り可能な信号を生成する、請求項1記載のカラム充填装置。
- 前記ソフトウェアが、前記ブレイクポイントから所定量のベッド圧縮を達成するために可動アダプターが移動しなければならない距離を計算し、該距離に相当するオペレータ読取り可能な信号を生成できるものである、請求項3記載のカラム充填装置。
- 前記カラムがクロマトグラフィーカラムであり、前記ベッドがクロマトグラフィー媒体を含む、請求項1記載のカラム充填装置。
- カラムの充填方法であって、
自動化ソフトウェア及びハードウェアを具備する自動制御ユニットが設けられたカラム充填装置を用意する段階、
カラムの内圧を検知して、上記自動制御ユニットで監視可能な圧力信号を生成する圧力センサーを上記カラム充填装置に設ける段階、及び/又は
カラムから流出する流体の量を測定して、上記自動制御ユニットで監視可能な流量信号を生成し、上記自動制御ユニットを用いて、ベッド媒体の集密層を収容したカラム内での可動アダプターの移動と、カラム内の圧力及びカラムから流出する流体の流量のいずれかとを監視する流量測定装置を上記カラム充填装置に設ける段階、及び
上記自動制御ユニットを用いて、上記圧力センサーからの圧力信号及び/又は流量測定装置からの流量信号を解析して、可動アダプターでの集密層の圧縮を開始するブレイクポイントを決定する段階であって、上記ブレイクポイントが、上記圧力信号及び/又は上記流量信号の低下が起きた時点で生じたと決定される、段階
を含んでなる方法。 - ブレイクポイントを決定した後、前記ソフトウェアを用いて、前記ブレイクポイントから所定量のベッド圧縮を達成するために可動アダプターが移動しなければならない距離を計算し、該距離に相当する位置まで前記可動アダプターを移動させる段階をさらに含む、請求項6記載の方法。
- ブレイクポイントの位置に対応するオペレータ読取り可能な信号を生成する段階をさらに含む、請求項6記載の方法。
- ブレイクポイントを決定した後、上記ソフトウェアを用いて、前記ブレイクポイントから所定量のベッド圧縮を達成するために可動アダプターが移動しなければならない距離を計算し、該距離に相当するオペレータ読取り可能な信号を生成する段階をさらに含む、請求項8記載の方法。
- 当該方法が、クロマトグラフィーカラムで実施され、前記ベッドがクロマトグラフィー媒体を含む、請求項6記載の方法。
