Description
本発明は、組織工学用途のための生体適合性で、生体吸収性で、注入でき、その場(in situ)で硬化するスカフォード(scaffold)として有用な、ポリ(カプロラクトン)とポリ(プロピレンフマレート)とのブロックコポリマーの合成に関する。前記ブロックコポリマーは、追加の架橋剤を用いて、または用いずに、酸化還元または光開始により架橋させることができる。従って、前記コポリマーは、自己架橋性でもあり、光架橋性でもある。
骨再生に関する臨床的要求は多岐にわたっており、米国では、癒合を達成するために骨移植処置を必要とする骨格欠損を有する患者が、毎年、おおよそ1,000,000人いる。これらは、原発性および転移性腫瘍の切除、骨格傷害後の骨量減少、骨欠損に伴う初回および修正全関節形成術、脊髄関節固定術、ならびに骨粗しょう症性不全骨折後の骨梁の空隙から生じる申請を含む。骨移植材料の選択、作製および適用の際に行う現行の臨床上の決定は、しばしば多くの要素を含む。構造的展望から、外科的処置計画を決定する前に幾つかの決定に取り組む必要がある。

530、1250および2000g・mol-1の公称分子量を有するPCLジオール類[α,ω−ジヒドロキシポリ(ε−カプロラクトン)]をAldrich Co.(ウィスコンシン州、ミルウォーキー)から購入し、これらは、H−[O(CH2)5CO−]mOCH2CH2−O−CH2CH2O[−OC(CH2)5O]n−Hの化学構造を有した。共重合の前に、一定量のPCLジオールを真空オーブンの中で一晩50℃で乾燥させた。本研究における他の全ての試薬もAldrich Co.から購入した。第一段階において、ジエチルフマレート259グラムおよび1,2−プロピレングリコール342グラムを、2L三つ口丸底フラスコの中で、架橋阻害剤としてヒドロキノン0.33グラムおよび重合触媒として塩化亜鉛2.04グラムと混合する。反応はフマル酸ジエステルを得るために、先ず100℃で1時間行い、その後150℃で7時間行う。過剰の1,2−プロピレングリコールおよび副生成物エタノールを除去する。第二段階において、中間体をエステル交換して、両末端にヒドロキシル基を有する直鎖状ポリ(プロピレンフマレート)(PPF)を形成する。ポリ(プロピレンフマレート)の分子量は、重合時間を変えることにより調節することができる。一般には、重合は、真空下、先ず100℃で、その後130℃で1時間または3時間行う。ポリ(プロピレンフマレート)ブロックを作った後、真空条件および油浴を止めることにより反応を停止させる。
熱架橋プロセスでは、過酸化ベンゾイル(BPO)およびN−ジメチルトルイジン(DMT)を、各々、フリーラジカル開始剤および促進剤として使用した。開始剤溶液100マイクロリットル(1−ビニル−2−ピロリジノン(NVP)250マイクロリットル中、BOP 50mg)および促進剤溶液40マイクロリットル(塩化メチレン980マイクロリットル中、DMT 20マイクロリットル)を、塩化メチレン500マイクロリットル中の1.5グラムP(PF−co−CL)溶液に添加し、入念に混合した。重合スカフォードを様々なテフロン金型に移入し、それらの金型を対流式オーブンの中に一晩置いて、架橋を助長した。骨格にPCLブロックを導入した後の向上した鎖可撓性のため、架橋剤をさらに添加せずとも全てのコポリマーを自己架橋させることができる。架橋後、周囲温度に冷却した後、架橋ポリマーを金型からはずした。コポリマー類とポロゲン(様々なサイズ分布を有する塩)との混合物に対して同様の架橋プロセスを行って、異なる孔隙率(これは、ポロゲンの含有率により調節することができる)を有するスカフォード類を作ることができる。架橋後、それらのスカフォード類を3日間、蒸留水中に置くことにより、塩を溶脱させた。それらのスカフォード類を真空下で少なくとも12時間乾燥させた。
Claims (24)
- カプロラクトン単位;および

- 前記ブロックコポリマーが5000ダルトン以上の数平均分子量を有する、請求項1に記載のブロックコポリマー。
- (i)ポリカプロラクトンジオールと(ii)ポリ(プロピレンフマレート)とを反応させることにより調製されるコポリマー。
- 前記ポリカプロラクトンジオールが、500〜10,000ダルトンの範囲の重量平均分子量を有する、請求項3に記載のコポリマー。
- ポリ(プロピレンフマレート)とポリ(ε−カプロラクトン)とのブロックコポリマー、および、
フリーラジカル開始剤または光開始剤
を含む、架橋性生分解性材料であって、
前記ブロックコポリマーが次式で表される、架橋性生分解性材料。
- 前記材料がフリーラジカル開始剤を含み、かつ前記材料が自己架橋性である、請求項5に記載の材料。
- 前記材料が光開始剤を含み、かつ前記材料が光架橋性である、請求項5に記載の材料。
- 前記材料が注入可能である、請求項5に記載の材料。
