- Acyclic And Carbocyclic Compounds In Medicinal Compositions (AREA)
Description
(1) フッ素含有物および酸を含み、う蝕性糖質を含まないことを特徴とする口腔用組成物。
(2) 歯の耐酸性を向上させる機能を有することを特徴とする、(1)に記載の口腔用組成物。
(3) 口腔内に適用したときに、唾液中のフッ化物イオン濃度が0.1〜100ppmとなる量のフッ素含有物を含むことを特徴とする、(1)又は(2)に記載の口腔用組成物。
(4) 口腔内に適用したときに、唾液のpHを6〜8に上昇させ、かつ、該pH下における唾液中のフッ化物イオン濃度が0.1〜10ppmとなることを特徴とする、(1)〜(3)のいずれかに記載の口腔用組成物。
(5) フッ素含有物が天然物である、(1)〜(4)のいずれかに記載の口腔用組成物。
(6) 天然物が茶抽出物である、(5)に記載の口腔用組成物。
(7) 酸が有機酸である、(1)〜(6)のいずれかに記載の口腔用組成物。
(8) 有機酸が、クエン酸、リンゴ酸、及び酒石酸から成る群より選ばれる1種または2種以上である、(7)に記載の口腔用組成物。
(9) 口腔用組成物が、食品または医薬品である、(1)〜(8)のいずれかに記載の口腔用組成物。
(10) 食品がチューインガムである、(9)に記載の口腔用組成物。
フッ素含有物として緑茶抽出物であるカメリアエキスMJ(太陽化学製)を使用し、表1に示した配合にて、1枚の重量が2.5gの板状のチューインガムを常法により調製した。なお、カメリアエキスMJのフッ化物イオン濃度は1500 ppm(mg/1000g)であることから、1枚のガム当りのフッ化物イオン濃度は1.5ppmである。
表2に示した配合にて、1枚の重量が2.5gの板状のチューインガムを常法により調製した。

表4に示した配合にて、1枚の重量が2.5gの板状のチューインガムを常法により調製した(1枚のガム当りのフッ化物イオン濃度は13.5 ppm)。

A.試験方法
(1)試料の調製
(1−1)標準試料
視診により健全と認められ、フッ化物使用歴のないヒト小臼歯由来の健全なエナメル質を試料ブロックとして用いた。試料ブロックは、エナメル質表面(3mmx4mm)以外をネイルバーニッシュで被覆した。この試料ブロックを、37℃に加温したpH 5.0の緩衝液(乳酸;0.1M, Ca2+;3.0mM,PO4 3-;1.8mM)の中に3日間浸漬して歯の表層を一部脱灰させた後、蒸留水で緩衝液を除去した。ここで得られた試料ブロックを標準試料とした。標準試料のブロックの一部は(2)のミネラル喪失量の測定に使用し、残りは以下の咀嚼後試料の調製に供した。
図1に示すようなガム咀嚼試験装置((A)単独、(B)装着した状態)に標準試料を2個はめ込み、エナメル質表面のみが露出するようにワックスで固定した。被験者は、前記試験装置を下顎に装着し、チューインガム1枚を20分間咀嚼した。咀嚼後、チューインガムを口腔内から取り除き、試験装置のみを更に、20分間口腔内に保持した。このチューインガム咀嚼試験を、1日に4回(朝食、昼食、夕食後と就寝前)、2週間、連続して実施した後の試料ブロックを咀嚼後試料とした。咀嚼後試料のブロックの一つは(2)のミネラル喪失量の測定に使用し、もう一つは以下の酸処理試料の調製に供した。
チューインガム咀嚼試験終了後、咀嚼後試料を試験装置より取り外し、これを37℃に加温したpH 5.0の緩衝液(乳酸;0.1M,Ca2+;3.0mM,PO4 3-;1.8mM)の中に3日間浸漬し、その後蒸留水で、緩衝液を除去した。ここで得られた試料ブロックを酸処理試料とした。酸処理試料は(2)のミネラル喪失量の測定に使用した。
(1)で得られた標準試料、咀嚼後試料および酸処理試料を、被験歯面の厚みが200μmになるように切片を作成した。この切片をアルミステップウェッジと共に、2kV、20mA、20分の条件で、マイクロラジオグラフィー(HB−50;HITEX社製)にて撮影し、ミネラル濃度分布を算出した。ミネラル濃度分布の算出にあたっては、1試料につき、深さ方向に2μmごと、25μmのライン幅でデンシトメータにて濃淡を測定してミネラル濃度を求め、下記式を用いてミネラル喪失量(△Z)を算出した。図2は、歯牙内部のミネラル濃度およびミネラル喪失量を模式的に示したグラフである。
実施例1、比較例1、および比較例2のチューインガムを用い、各群(実施例1、比較例1、および比較例2)10名からなる被験者によって、上記の試験方法に従って、咀嚼後試料および酸処理試料を調製した。なお、被験者の構成は、健常成人30名(男性15名、女性15名、平均年齢28.5才)であった。
5名の被験者の下顎に、pH電極を付けた図1の試験装置を装着した。また、同被験者の首筋には比較電極を装着した。この状態で被験者に実施例2、比較例3のチューインガム各1枚を咀嚼させ、口腔内唾液のpH変化を経時的に測定した。咀嚼前の口腔内唾液のpHは4.8〜7.0の範囲であった。表7に各被験者の咀嚼前、咀嚼後2分及び5分後のpH値を示す。
pH条件及びフッ化物イオン濃度が耐酸性にどのような影響を与えるかを調べるため、牛歯(10例)を使い、以下の耐酸性のモデル試験を行った。

Claims (3)
- 茶抽出物由来のフッ素含有物および0.1〜0.2重量%の有機酸を含み、う蝕性糖質を含まないこと、ならびに、口腔内に適用したときに、唾液のpHを7〜8に上昇させ、かつ、該pH下における唾液中のフッ化物イオン濃度が0.1〜10ppmとなることを特徴とする食品。
- 有機酸が、クエン酸、リンゴ酸、及び酒石酸から成る群より選ばれる1種または2種以上である、請求項1に記載の食品。
- 食品が、チューインガム、キャンデー、錠菓、またはフィルム菓子である、請求項1または2に記載の食品。
