Description
本発明は、CD1aに特異的に結合することのできるヒト結合分子の同定、これらの結合分子を含む免疫複合体及び前記結合分子を得る方法に関する。本発明は更に、医学における前記ヒト結合分子の利用方法、特に腫瘍性疾患及びランゲルハンス細胞組織球増加症の診断、予防及び/又は治療のための利用方法を含むものである。
CD1分子は樹状細胞、単球及びいくつかの胸腺細胞の表面に発現する分子のファミリーである。CD1分子は、抗原提示に関わるという点でMHCクラスI分子に類似している。ヒトでは5種類のCD1分子、CD1a、CD1b、CD1c、CD1d及びCD1e、がこれまで同定されている。これら5種類のCD1遺伝子産物のうち4種類が血清学的に定義されている。これらはCD1a、CD1b、CD1c及びCD1dと呼ばれ、固有の重鎖により区別され、それぞれの分子量は各々、約49kDa、約45kDa、約43kDa及び約48kDaである。
以下に、本発明で用いられる用語の定義を示す。
急性骨髄性白血病
本願明細書に用いられる用語「急性骨髄性白血病」とは、骨髄由来の前駆細胞の無制限な増殖により特徴づけられる。骨髄由来の前駆細胞は、骨髄性前駆細胞、骨髄単球性前駆細胞、未熟巨核芽細胞を含むが、これらに限定されるものではない。急性骨髄性白血病(AML)のサブタイプには、FAB分類によれば、FAB−M0、FAB−M1、FAB−M2、FAB−M3、FAB−M4、FAB−M5、FAB−M6及びFAB−M7がある。
本願明細書に用いられる用語「アミノ酸配列」とは、天然又は合成の分子、及びペプチド、オリゴペプチド、ポリペプチド又はタンパク質配列のことを示す。
本願明細書に用いられる用語「アポトーシス」とは、細胞分化の特定の段階及び特定の刺激に対する反応の中で起きる細胞的事象の複雑なカスケードにより起こる、秩序だった又は制御された全ての細胞死を意味する。アポトーシスは1種類以上の特徴的な細胞変化によって特徴づけられ、及び/又はこれに付随する。特徴的な細胞変化には、細胞質の凝縮、細胞膜微絨毛の消失、核の断片化、染色体DNAの分解又はミトコンドリア機能の喪失が含まれるが、これらに限定されるものではない。アポトーシスは、例えば、細胞生死判別試験、FACS分析又はDNA電気泳動法により決定し測定することができるが、これら全ては当該技術分野において既知のものである。
本願明細書に用いられる用語「結合分子」とは、キメラ化、ヒト化又はヒトモノクローナル抗体のような、無傷の免疫グロブリン、若しくは免疫グロブリンの結合パートナー、即ちCD1aに対する特異的な結合を無傷の免疫グロブリンと競合するような免疫グロブリン断片を有する抗原結合領域及び/又は可変領域を意味する。構造によらず、抗原結合断片は無傷の免疫グロブリンにより認識されるのと同じ抗原に結合する。用語「結合分子」が本願明細書に用いられる際には、無傷の抗体のクラス及びサブクラスの免疫グロブリンが含まれる。この中には、IgA、IgD、IgE、IgG、及びIgMが含まれ、これらの内いくつかは更にサブクラス(アイソタイプ)、即ち、IgA1、IgA2、IgG1、IgG2、IgG3及びIgG4に分類され、これはその抗原結合断片についても同様である。
本願明細書に用いられる用語「生物学的標本」とは、様々な標本の種類を含んでおり、血液や生物由来の他の液体標本、生検標本や組織培養物といった固形組織標本、又はここから採取した細胞若しくはその子孫を含む。この用語は、採取してから薬剤処理、可溶化、タンパク質やポリヌクレオチドといった、特定の成分を濃縮するなどのいかなる操作を加えた標本も含まれる。この用語はいかなる動物種から採取した様々な臨床検体を含んでおり、培養した細胞、細胞上清及び細胞溶解物もこれに含まれる。
本願明細書に用いられる用語「慢性骨髄性白血病」とは、骨髄及び骨髄以外の部位における骨髄造血細胞の無制限な増殖により特徴づけられ、悪性の骨髄芽球は分化して骨髄球、後骨髄球、杆状球及び顆粒球を作ることができる。
本願明細書に用いられる用語「相補性決定領域」とは、免疫グロブリンのような結合分子の可変領域内の配列を意味しており、抗原上で認識される抗原決定基と形状及び電荷の分布が相補的な抗原結合部位を作る。CDR領域は、タンパク質又はタンパク質断片の直線状抗原決定基、不連続抗原決定基又は立体構造抗原決定基に特異的であり、これは元々の立体構造のタンパク質上、又は、場合によっては、変性した、即ちSDSにより可溶化したタンパク質上に存在する。抗原決定基にはタンパク質の翻訳後修飾も含まれる。
本願明細書に用いられる用語「欠失」とは、アミノ酸又は核酸配列の変化であって、親分子、即ち通常の天然由来の分子と比べてそれぞれ1個以上のアミノ酸又は核酸が存在しないことを意味する。
本願明細書に用いられる用語「発現調節核酸配列」とは、特定のホスト生物体において機能を有して結合したコード配列の発現に必要となる及び/又は影響を与えるポリヌクレオチド配列を意味する。2つの核酸配列が機能を有して結合している場合には、同じ配向であり、同じリーディングフレームでもある。これらは通常、原則的には隣接しているが、このことは必須ではない。発現調節核酸配列、例えば、なかでも適切な転写開始、終止、プロモーター、エンハンサー配列;レプレッサー又は活性化配列;スプライシングやポリアデニル化シグナルといった効率的なRNA処理シグナル;細胞質mRNAを安定化する配列;翻訳効率を高める配列(即ち、リボソーム結合部位);タンパク質の安定性を高める配列;必要に応じて、タンパク質の分泌を高める配列は、選択されたホスト生物体において活性を示すいかなる核酸配列であっても良く、タンパク質をコードする遺伝子に由来する、ホスト生物体に相同又は非相同のものであっても良い。
本願明細書に用いられる用語「機能的変異体」とは、親結合分子の核酸及び/又はアミノ酸配列と比べ、核酸及び/又はアミノ酸配列が1箇所以上変化しているが、結合パートナー、即ちCD1aに親結合分子と競合して結合することがまだできる核酸及び/又はアミノ酸配列からなる結合分子を意味する。言い換えれば、親結合分子の核酸及び/又はアミノ酸配列の修飾は、その核酸配列によりコードされる又はそのアミノ酸配列を含む結合分子の結合特性に有意な影響を与えないか又は変化させることはなく、即ち、結合分子はまだその標的を認識し結合できる。機能的変異体は、核酸及びアミノ酸の置換、付加及び欠失を含め、保存配列修飾を有する。これらの修飾は、例えば部位特異的変異導入やランダムPCRによる変異導入のような、当該技術分野で知られる標準的な技術により導入することができる。
本願明細書に用いられる用語「ホスト」とは、クローニングベクターや発現ベクターといいたベクターを導入する生物体又は細胞を指し示すことを意図している。生物体又は細胞は、原核生物であっても、真核生物であってもよい。この用語が特定の対象となる生物体又は細胞だけでなく、そのような生物体や細胞の子孫も指し示すことを意図していることを理解されたい。ある修飾は変異又は環境の影響によりそれ以降の世代に生じることがあるので、そのような子孫は事実上、親生物体又は細胞と同質ではないが、このような子孫も本願明細書で用いられる用語「ホスト」の範囲内に含まれる。
用語「ヒト」とは、本願明細書で定義された結合分子について用いられ、ヒトに直接由来する分子、又はヒトの配列に基づく分子を意味する。結合分子がヒトの配列に由来する又は基づくもので、その後修飾を受けた場合でも、本願明細書を通じて用いられているようにヒトであると考える。言い換えれば、用語「ヒト」は、結合分子について用いる場合には、ヒト生殖細胞の免疫グロブリン配列に由来する可変領域及び定常領域であってヒト若しくはヒトリンパ球に又は修飾形に生じる又は生じていない可変領域又は定常領域に基づくものを含むことを意図している。従って、ヒト結合分子はヒト生殖細胞系列の免疫グロブリン配列にコードされていないアミノ酸残基を含むことができ、置換及び/又は欠失(即ち、試験管内における例えばランダム又は部位特異的変異導入によって、又は導入された変異生体内での体細胞突然によって導入された変異)を有することができる。本願明細書で用いられる「基づく」とはまた、核酸配列がテンプレートから正確にコピーされている、若しくは例えば変異性(error-prone)PCRなどにより軽微な変異が入っている、又は核酸配列がテンプレートに完全に合わせて、若しくは軽微な修飾を伴って合成で作られる状況も意味する。ヒト配列に基づく半合成分子もまた本願明細書で用いるヒトであると考えられる。
用語「免疫リポソーム」とは、本願明細書で定義された結合分子を有するリポソームであって、標的部分として作用してリポソームを結合分子の結合パートナーに特異的に結合させることのできるものを意味する。結合パートナーは、体液に存在しても良いし、細胞表面に結合していても良い。
用語「挿入」は、用語「付加」としても知られるが、アミノ酸又はヌクレオチド配列の変化であって、親分子、即ち通常は天然由来の分子と比べ、アミノ酸又はヌクレオチド残基が1以上追加されるものを意味する。
本願明細書に用いられる用語「内在化する(internalising)結合分子」とは、結合した標的細胞内に内在化する(internalised)ことができる本願明細書で定義された結合分子を意味する。言い換えれば、結合分子は取り込まれる、即ち標的細胞の外側(細胞表面)から内側(例えば、エンドソーム分画内、若しくは他の分画、又は細胞の細胞質内に)に、結合分子の結合パートナーに結合して直ちに標的細胞により輸送される。
本願明細書に用いられる用語「単離された」とは、本願明細書で定義された結合分子について用いる場合には、結合分子が他のタンパク質又はポリペプチドを実質的に含まないこと、特に異なった抗原特異性を有する他の結合分子を含まず、他の細胞の物質及び/又は化学物質を含まないことを意味する。例えば、結合分子を組換え技術で生成した場合には、好ましくはこれらの結合分子は培養液を含まず、結合分子が化学合成により生成された場合には、好ましくはこれらの結合分子は化学前駆物質又は他の化学物質を実質的に含まず、即ち、これらはタンパク質の合成に関わる化学前駆物質又は他の化学物質から分離されている。用語「単離された」は、本願明細書で定義された結合分子をコードする核酸分子について用いる場合には、本結合分子をコードするヌクレオチド配列が他のヌクレオチド配列、特にCD1a以外の結合パートナーに結合する結合分子をコードするヌクレオチド配列を含まない核酸分子を指し示すことを意図している。更に、用語「単離された」は、天然の核酸分子がその天然のホスト中に本来伴っている他の細胞成分、例えば、リボソーム、ポリメラーゼ、核酸分子が本来関連するゲノム配列から核酸分子が実質的に分離されていることを意味する。更に、用語「単離された」核酸分子は、cDNA分子のように、他の細胞物質を実質的に含まず、組換え技術により生成した場合には培地を実質的に含まず、化学的に合成された場合には化学前駆物質又は他の化学物質を実質的に含まない。
用語「ランゲルハンス細胞組織球増加症(LCH)」とは、ランゲルハンス細胞に類似した樹状細胞の局所性又は全身性の増殖により特徴づけられる疾患を意味する。これらの細胞は、骨髄に由来し、通常は主に皮膚に存在する。LCHでは、これらの細胞は増殖し、とりわけ骨、肝臓、肺及び脳のような他の多くの臓器に影響を与えることもあるし、与えないこともある。LCHは、組織球増加性疾患の中では最も頻度の多い疾患である。本疾患は、全年齢に発症するが、0〜4歳の子供に最も頻度が高い。臨床症状は、自然に治癒する可能性のある局所の皮膚の発赤又は単独の骨病変から、最終的には患者の死に至る広範囲の内臓障害及び機能不全まで、様々である。
本願明細書に用いられる用語「リポソーム」とは、様々な種類の脂質、好ましくは両親媒性脂質、リン脂質及び/又は界面活性剤から成る層により囲まれた小胞であり、この小胞はこれらの分子から、超音波処理又はリン脂質界面活性剤複合体からの界面活性剤の除去といった当該技術分野において知られている技術により人工的に作られる。この層は疎水性部分と親水性部分とを有する分子により形成された2本層であり、ここで疎水性部分は親水性培地中で会合して層の内部を形成し、親水性部分は培地と接したままとなる。この層は内部を囲んで封入し、この中には、全体的に又は部分的に、水相、固体、ゲル、気体相又は非水性の液体を含むことができる。リポソームは、核酸分子、結合分子、タンパク質、毒性物質及び他の物質又は化合物といった1種類以上の分子を、リポソームと細胞膜との融合により動物細胞のような細胞に輸送するのに有用であり、それはリポフェクションとも称される過程である。分子はリポソームの内部に含まれても、脂質層内に含まれても、脂質層の外表面に接着していても良い。
本願明細書に用いられる用語「モノクローナル抗体」とは、特定の抗原決定基に対して単一の結合特異性及び親和性を示すモノクローナル抗体を意味する。従って、用語「ヒトモノクローナル抗体」とはヒト生殖細胞系列の免疫グロブリン配列由来、又は完全に合成若しくは半合成配列由来の可変領域及び定常領域を有する、単一の結合特異性を示す抗体を意味する。
本願明細書に用いられる用語「骨髄異形成症候群」とは、骨髄の初期造血細胞に由来する密接に関連し合ったクローン性造血疾患の不均質なグループを含む。全ての疾患は、形態及び成熟が損なわれた細胞性骨髄(骨髄異形成性造血:dysmyelopoiesis)、および末梢血血球減少により特徴づけられ、無効な血液細胞が作られる結果となる。言い換えると、成熟過程にある血球細胞が十分に成熟して血液中に入る前に骨髄中で頻繁に細胞死を起こすため、血球濃度が低くなるのである。骨髄異形成症候群の患者では、白血病性芽球と呼ばれる、極めて未成熟な骨髄細胞の蓄積も見ることがある。
本願明細書に用いられる用語「天然由来」とは、物について用いるとき、その物が自然界に見つけられる、という事実を示す。例えば、自然から単離することのできる生物体に存在し、研究室でヒトに修飾を加えられていないポリペプチド又はポリヌクレオチド配列は天然由来である。
本願明細書に用いられる用語「腫瘍細胞」とは、明らかな生理学的機能を有しない不所望な新しい自律的増殖により生じた細胞を意味する。腫瘍細胞は更に、形質転換細胞及び血液の癌(良性及び悪性)を含む癌細胞を含む。
本願明細書に用いられる用語「核酸分子」とは、ヌクレオチドの高分子形態の物を意味し、RNA、cDNA、ゲノムDNA、並びにこれらの合成形態及び混合高分子のセンス鎖及びアンチセンス鎖の両方を含む。ヌクレオチドは、リボヌクレオチド、デオキシヌクレオチド、又はいずれかの種類のヌクレオチドの修飾した形態を指す。この用語は、単鎖又は2本鎖体のDNAも指す。加えて、ポリヌクレオチドは、天然由来及び/又は非天然由来の結合により互いに結合した、自然起源及び修飾ヌクレオチドの一方又は両方を含むことができる。核酸分子は、当業者に容易に理解されるように、化学的若しくは生物学的に修飾されても良いし、又は天然のものではない若しくは誘導化ヌクレオチド塩基を含んでも良い。そのような修飾には、例えば、標識化、メチル化、天然由来のヌクレオチドのうち1つ以上を類似物質で置換すること、ヌクレオチド間修飾があり、ヌクレオチド間修飾の例には、非荷電結合(例えば、メチルスルホン酸、ホスホトリエステル、ホスホアミド、カルバミン酸等)、荷電結合(例えば、ホスホロチオ酸、ホスホロジチオ酸等)、ペンダント部分(例えば、ポリペプチド)、挿入剤(例えば、アクリジン、プソラレン等)、キレート剤、アルキル化剤、及び修飾結合(例えば、アルファアノマー核酸等)などがある。上記の用語は、1本鎖、2本鎖、部分的2本鎖、3本鎖、ヘアピン状、環状及びパドロック体といった、任意の位相形態も含むことも意図している。水素結合又は他の化学的相互作用を介して指定配列に結合できるようにポリヌクレオチドを模倣した合成分子も含まれる。このような分子は当該技術分野で知られており、例えば、分子骨格でリン酸結合をペプチド結合で置換した分子がある。核酸配列を参照することにより、別に特定されている場合を除いてはその相補鎖も含むことになる。従って、特定の配列を有する核酸分子を参照することにより、相補配列とともに相補鎖も含むと理解されるであろう。相補鎖は、例えば、アンチセンス療法、ハイブリダイゼーションプローブ及びPCRプライマーにも有用である。
用語「機能を有して結合した」とは、物理的に結合した2つ以上の核酸配列構成要素で、互いに機能的な関連のあるもののことを指し示す。例えば、プロモーターがコード配列の転写又は発現を開始又は制御しうる場合には、プロモーターはコード配列と機能を有して結合しており、この場合には、コード配列はプロモーターの「制御下」にあると理解するべきである。一般に、2つの核酸配列が機能を有して結合している場合には、これらは同じ配向であり、通常同じリーディングフレームでもある。これらは通常、原則的には隣接しているが、このことは必須ではない。
「薬学的に許容される賦形剤」により、薬剤、医薬品又は結合分子といった活性分子と組み合わせて好ましい又は使いやすい投薬形態を調製する任意の不活性物質を意味する。「薬学的に許容される賦形剤」は、使用投与量及び濃度では服用者に無害であり、薬剤、医薬品又は結合分子を含有する製剤の他の構成成分に適合する賦形剤である。
本願明細書に用いられる用語「特異的に結合する」とは、例えば抗体のような結合分子と、例えば抗原のようなその結合バートナーの相互作用に関して、その相互作用が結合分子状の、例えば抗原決定基即ち抗原決定基のような特定の構造が存在することに依存していることを意味する。言い換えれば、抗体は、結合パートナーが他の分子が混合した中にある場合でさえ、結合パートナーに選択的に結合又は認識する。この結合は、共有相互作用若しくは非共有相互作用、又はその両者の組合せにより媒介される。
本願明細書で用いる「置換」とは、1つ以上のアミノ酸又はヌクレオチドを、各々異なったアミノ酸又はヌクレオチドで置き換えることを意味する。
用語「治療に有効な量」とは、CD1a分子が役割を果たす若しくは関連する疾患又は病気を予防及び/又は治療する、又はそのような病気又は疾患に関連した状態を改善する効果のある本願明細書で定義される結合分子の量を指し示す。
用語「治療」とは、治療上の処置だけでなく、予防又は阻止手段も指す。治療を必要とする人には、CD1a分子が役割を果たす又は関連する病気又は疾患に既に罹患している人だけでなく、そのような病気又は疾患を予防すべき人も含まれる。
用語「ベクター」とは、ホストに導入するために第2の核酸分子を挿入することができる核酸分子であって、それはホストで複製され、場合によっては発現する。言い換えれば、ベクターとは、ベクターが関連している第2の核酸分子を運ぶことのできるものである。クローニングベクター及び発現ベクターは本願明細書に用いる用語「ベクター」において想定されているものである。ベクターには、プラスミド、コスミド、細菌性人工染色体(BAC)及び酵母人工染色体(YAC)が含まれるが、これらに限定されるものではなく、バクテリオファージ又は植物又は動物(ヒトを含む)ウイルスに由来するベクターも含まれる。ベクターはここに提案されたホストにより認識される複製起点を有しており、発現ベクターの場合には、ホストにより認識されるプロモーター及び他の制御領域を有する。第2の核酸分子を有するベクターは形質転換、トランスフェクション又はウイルス侵入機構を利用することにより導入することができる。あるベクターは導入されたホストの中で自律的に複製することができる(例えば、細菌性の複製起点を有する細菌性ベクター)。他のベクターではホスト内に導入されるとホストのゲノムに統合され、それによりホストのゲノムとともに複製される。
本発明において、ヒトCD1aに結合することのできるいくつかのヒト結合分子が、ファージディスプレイ法を用いることにより同定され、得られた。更に、このようなヒト結合分子を生成する方法、並びにヒト結合分子を、とりわけ腫瘍性の疾患及び病気の診断、予防及び治療に使用する方法を記載した。
本発明は、ヒトCD1aに特異的に結合することのできるヒト結合分子を含む。本結合分子は、ヒトCD1a断片、本発明のヒト結合分子により認識されるCD1aの抗原決定基を少なくとも有するCD1a断片に結合することもでき、特に特異的に結合することができる。CD1aの自然起源の切断型又は分泌型、天然由来の変異体(例えば、オルタナティブスプライシングを受けたもの)及び天然由来の対立遺伝子変異体に特異的に結合できるヒト結合分子も本発明の一部である。本発明の結合分子は、修飾により結合分子のCD1a分子への結合が失われない限り、CD1aの天然由来ではない変異体又は類似体にも特異的に結合することができる。
とする光学的現象を用いて、測定することができる。
試験管内における樹状細胞の生成
培養樹状細胞(DC)は軟膜から単球を分離することによって得られるものであり、フィコール密度勾配円心を用いて顆粒球を除去し、ヒツジ赤血球ロゼット形成(SRBC−resetting)によりT細胞を除去し、次にプラスチックに2時間37℃で接着させて他の細胞を除去し、単球を得た。得られた単球をGM−CSF及びIL−4存在下で5日間培養してこれらの細胞を未成熟DC細胞に分化させた(主に、CD1a+/CD83−の表現形により特徴づけられる。)。更に、GM−CSF、IL−4、PGE2、IL−1β、IL−6及びTNF−アルファ存在下で2日間培養することにより、成熟DCが得られる(主に、CD1a+/CD83+の表現形により特徴づけられる。)(Jonuleit et al.,1997)(図1参照)
細胞分類による樹状細胞上の単鎖Fv断片を有するファージ抗体の選択
5日及び7日間培養したあとで、約1.5×107個のDC由来の未成熟及び成熟単球がそれぞれ組織培養フラスコから得られ、これを0.5mlの半合成ファージscFv抗体ディスプレイライブラリーと混合した。このヒト単鎖抗体断片ライブラリーはde Kruif et al.,(1995b)に記載されているとおりに構成された。短くいえば、49種類の生殖細胞系列VH遺伝子を有するライブラリーを〜108種類の合成重鎖CDR3領域及び7種類の軽鎖と融合した。CDR3領域の長さは6〜15アミノ酸であった。軽鎖はVκ1〜Vκ4及びVλ1〜Vλ3ファミリの一員によりコードされていた。最終的なライブラリには約4×108種類の個別クローンが含まれていた。
CD1aが選択したscFvファージ抗体により認識される抗原と同一であることの確認
全体で3回の選択を行い、2回目と3回目の後にはファージ抗体について、様々な細胞種に特異的に結合するかについて試験した。細胞は、例えば、培養DC、PBL、扁桃及び滑液細胞懸濁液、K562、U266、IM9、U937、THP、CEM、Fravel、HL60、Raji、RPMI8226、HepgII、HELA、HT29及びJurkattを含むいくつかの細胞株、CD1a分子で形質転換したC1R細胞株、並びにCD80、CD83及びCD86分子で形質転換したA431細胞株であった。細胞の染色のために、ファージ抗体100μlを、4%粉乳を含有するPBS50μlを添加することにより15分室温でブロッキングした。次に、1%BSAを含有するPBS50μl中の細胞5×105個をブロッキングしたファージ抗体に加え、得られた混合液を1時間氷上にて培養した。この後、得られた混合液を1%BSAを含有するPBS200μlにて3回洗浄した。細胞に結合したファージ抗体を検出するために、細胞を800倍希釈したヒツジ抗M13ポリクローナル抗体20μlにて20分氷上で培養した。その後、細胞を1%BSAを含有するPBSで洗浄し、700倍希釈したPE結合ロバ抗ヒツジポリクローナル抗体にて20分氷上で培養した。
ヒトCD1a特異的scFvの特性
ヒトCD1aを形質転換した細胞株にだけ結合した20種類のファージ抗体(scFv)クローンから、プラスミドDNAを取り出し、ヌクレオチド配列を標準的な技術に従い決定した。短く言えば、20種類のクローンすべてのVH及びVLのヌクレオチド配列を、プライマーM13REV(5‘−AACAGCTATGACCATG(配列番号23)及びfdSeq(5’−GAATTTTCTGTATGAGG(配列番号24))を用いて、Taqシークエンスキットにより以下のサイクリングプロトコール(25サイクル)に従ってシークエンス反応で決定した。サイクリング反応:94℃10分間(変性)、50℃5秒間(アニーリング反応)及び60℃4分間(伸長反応)。沈殿したDNAをサンプルバッファーに溶解し、ABIPRISM自動蛍光シーケンサーに流して解析した。得られたヌクレオチド配列を、抗体に関する通常の当業者に周知のVBASEデータベースと比較し、各々の鎖の遺伝子ファミリーを決定した。20種類のクローンには6種類の異なったscFvが含まれていることがわかった。これらはSC02−113、SC02−114、SC02−115、SC02−116、SC02−117及びSC02−118と呼ぶ(表1参照)。SC02−113、SC02−114、SC02−115、SC02−116、SC02−117及びSC02−118のヌクレオチド配列は各々、配列番号25、配列番号27、配列番号29、配列番号31、配列番号33及び配列番号35に示した(表1参照)。SC02−113、SC02−114、SC02−115、SC02−116、SC02−117及びSC02−118のアミノ酸配列は各々、配列番号26、配列番号28、配列番号30、配列番号32、配列番号34及び配列番号36に示した(表1参照)。ヒトCD1a特異的scFvのVH及びVL遺伝子特性並びに重鎖CDR3配列もまた表1に示した。
選択した抗CD1a単鎖Fvから完全ヒト免疫グロブリン分子(ヒトモノクローナル抗CD1a抗体)の構築
SC02−113、SC02−114、SC02−115、SC02−116、SC02−117及びSC02−118と呼ばれるscFvの重鎖及び軽鎖可変領域を、オリゴヌクレオチドを用いてPCRにて増幅し、制限酵素部位及び/又はIgG発現ベクターpSyn−C03−Hcγ1(配列番号37を見よ)、pSyn−C05−Cκ(配列番号38を見よ)及びpSyn−C04−Cλ(配列番号39を見よ)の中で発現させるための配列を付加した。SC02−113、SC02−114、SC02−115、SC02−116、SC02−117及びSC02−118と呼ばれるscFvの重鎖可変領域をベクターpSyn−C03−HCγ1にクローニングした。SC02−113、SC02−114、SC02−115、SC02−116、SC02−117及びSC02−118と呼ばれるscFvで共通している軽鎖可変領域をベクターpSyn−C05−Cκにクローニングした。SC02−115と呼ばれるscFv軽鎖可変領域をベクターpSyn−C04−Cλにクローニングした。scFvSC02−113、SC02−114、SC02−116、SC02−117及びSC02−118の間で共通しているVL遺伝子をまずオリゴヌクレオチド5K−I(配列番号40)及びsy3K−C(配列番号41)(下記参照)を用いて増幅し、PCR産物をベクターpSyn−C05−Cκにクローニングした。scFvSSC02−115のVL遺伝子をまずオリゴヌクレオチドsy5L−A(配列番号42)及び3L−B(配列番号43)(下記参照)を用いて増幅し、PCR産物をベクターpSyn−C04−Cλにクローニングした。すべてのコンストラクトのヌクレオチド配列を当業者に知られる標準的技術により確認した。

ヒトモノクローナル抗CD1a抗体による免疫組織化学
ヒトモノクローナル抗CD1a抗体が正常組織に対して結合する能力について、免疫組織化学により分析した。この目的で、副腎、膀胱、脳(小脳及び大脳)、血管(大動脈及び冠動脈)、卵管、食道、胃(前庭及び胃体)、十二指腸、回腸、直腸、心臓、腎臓、肝臓、肺、リンパ節、卵巣、膵臓、副甲状腺、末梢神経、下垂体、胎盤、前立腺、唾液腺、皮膚、脊髄、脾臓、横紋筋、精巣、扁桃、甲状腺、尿管及び子宮(頚部及び内膜)の正常組織の凍結切片を切り出し、スライドグラスにマウントして室温で乾燥させた。切片を過酸化水素0.03%を含有するアジ化ナトリウム50mMで内因性ペルオキシダーゼをブロッキングし、次に所定のプロトコールに従って内因性ビオチンをブロッキングした(X0590、Dako)。次に、ビオチン化した抗ヒトCD1aヒトIgGにより60分間室温で培養する前に、切片をBSA4%及び正常ヒト血清10%を含有するPBSでブロッキングした。結合したIgG分子を検出するために、切片をストレプトアビジン結合ホースラディッシュペルオキシダーゼ(Dako)で培養し、次にジアミノベンジジン(Sigma)で培養し、褐色結晶の局所沈着を得た。切片をヘマトキシリンにより対比染色し、切片中の有核細胞を可視化した。分析する前に、切片を脱水し、スライドをオイキット(BDH)により封止した。
新鮮腫瘍標本及び腫瘍細胞株におけるCD1a分子発現のヒトモノクローナル抗CD1a抗体によるフローサイトメトリー分析
抗ヒト抗CD1a IgG1抗体パネルを、新鮮腫瘍サンプルである急性骨髄性白血病(AML)、急性非リンパ性白血病(ANLL)、慢性骨髄性白血病の急性転化(CML−BC)、大型顆粒球性リンパ球性白血病(LGLL)、B細胞性慢性リンパ性白血病(B−CLL)及び細胞株SUP−T1でのCD1aの発現を評価するために用いた。この目的のために、2×105個の単核球をフィコールプラーク勾配分離法(新鮮標本について)又は提供者の説明書に従った培養(細胞株について)により調製し、本発明のヒトモノクローナル抗CD1a IgG1抗体により4℃で0〜50μg1/dlの範囲の濃度で染色した。02−113、02−114、02−115、02−116、02−117及び02−118と称する抗体の結合はビオチン化したヤギ抗ヒトIgG(Fc特異的、Caltag)、次にストレプトアビジンフィコエリトリン(Caltag)を用いて可視化した。染色した細胞はフローサイトメトリーにより分析した。抗CD1aマウスモノクローナル抗体で確立されている表現形に従って、中程度から高度の染色レベルが腫瘍細胞の亜集団で予想される。ヒト抗CD1aモノクローナル抗体が治療分子として有用であるためには中程度から高度の染色レベルが必要であることが予想される。
ヒト抗CD1aモノクローナル抗体の内在化(internalisation)研究
ヒトモノクローナル抗ヒトCD1a IgG1抗体の内在化能を測定するために、Jurkatt細胞のようなCD1a発現細胞株により内在化分析を行った。抗ヒトCD1a IgG1抗体を以下の染料により標識したオレゴングリーン−480−SEにより染色した。染料0.1mgをDMSO 10μlに溶解し、抗体0.4mgを加えてPBSで最終容量を0.4mlにして1時間室温で培養した。次に、この混合液をPBSで平衡化したセファデックスG25カラムに搭載した。標識した抗体をPBSで溶出し、着色した分画を集めた。5×105個のJurkatt細胞を飽和濃度の適当な抗体とともに氷上で30分間搭載した。結合しなかった抗体を氷冷したFBS10%含有RPMI 1640培地で3回洗浄して除いた。次に、細胞を培地50μl中に再懸濁し、4℃(内在化しない)又は抗体が内在化することのできる37℃で1時間培養した。氷冷したPBSで3回洗浄した後、細胞表面に結合した抗体を2.5μg/dlサブチリシンにより4℃1時間で剥がした。細胞を氷冷した1%BSA含有PBSで再び洗浄し、標本をフローサイトメトリーで分析した。4℃(内在化の生じない温度)で培養した細胞については、細胞に結合したIgGは細胞からはがされており、これは蛍光が失われたことにより示されている。他方で、細胞を抗体が内在化することのできる37℃で培養した場合には、サブチリシンを用いて細胞表面のCD1a分子に結合した抗体を除去した後も細胞には蛍光が残った。このことは抗CD1抗体が細胞内に内在化してプロテアーゼ処理に対して抵抗性となったことを示している。対照的に、ネガティブコントロール抗体である抗CD20抗体(これは内在化することはない。Ghetie et al.(2001)参照)は両方の温度で剥がれており、これは、この抗体が標的には結合するが細胞内には内在化しないことを示している。他方で、抗CD1a抗体の内在化により、抗CD1a抗体に結合したある毒性分子を有する免疫複合体について、その毒性分子が十分な機能/活性を示すために細胞膜を通過する必要がある場合にその効果が高められる。他方で、内在化することのできない抗CD1抗体は放射性複合体には有用でない可能性がある。このような複合体は細胞傷害性を発揮するのに細胞膜を通ることは必要ない。加えて、標的細胞の表面に結合する抗体はナチュラルキラー細胞(ADCC)のような細胞傷害性白血球を誘導するか、又は補体(CDC)の沈着を招く。抗体が結合することによる急速なCD1aの内在化はこれらの過程に干渉しうる。
腫瘍細胞におけるヒト抗CD1a抗体を用いた抗体依存性細胞傷害(ADCC)及び補体依存性細胞障害(CDC)の誘導
ヒトモノクローナルヒト抗CD1a抗体が抗体依存性細胞傷害(ADCC)及び補体依存性細胞障害(CDC)を誘導する能力を測定するために、通常の51Cr放出分析を行った。ADCCの実験にはHL60又はU937腫瘍標的細胞を100μCiの51Cr(英国バッキンガムシャー州にあるAmersham社)で37℃、1時間標識した。何回も洗浄した後、標的細胞をUボトムマイクロタイタープレートに5×103個/ウェルの濃度でプレートした。単離したヒト末梢血単核球を次に加え、いくつかのエフェクターと標的の比が80:1〜10:1になるようにした。細胞混合液を3連で37℃で培養し、その培養にあたり様々な濃度のヒトモノクローナル抗CD1a抗体を存在させ、最終容量を150μlとした。4時間培養した後、上清の一部を採取し、51Cr濃度を測定した。特異的溶解の割合を、数式:%特異的溶解=([実験的cpm−基礎cpm]/[最大cpm−基礎cpm])×100%を用いて計算した。最大溶解は標的細胞を1%トライトンX−100により標的細胞を溶解して測定し、基礎放出量は標的細胞を培地のみで培養して測定した。
CD1aで形質転換したマウス腫瘍モデルにおけるヒト抗CD1a抗体の生体内での抗腫瘍有効性
ヒト抗CD1a抗体の生体内での抗腫瘍有効性を調べるために、ヒトCD1aで形質転換したマウスB16腫瘍モデルを用いた動物実験を行った。この腫瘍は腫瘍学の文献(Fidler et al.(1976)を参照)では膨大な記載があり、例えば同系のC57B1/6では急速に成長する。抗CD1a治療の効果を試験するために、C57B1/6、C57B1/6ヌード、SCID又はNOD−SCIDマウスに第0日目に1×106個のB16腫瘍細胞を皮下に接種した。CD1aのcDNAを1つの隣接部位(flank)にセンス方向(即ち、正常に発現する)に形質導入したB16細胞を、各々のマウスに接種した。アンチセンス方向(即ち、発現しない)でCD1aのcDNAを形質導入したB16細胞を、逆の隣接部位(flank)でマウスに接種し、ネガティブコントロールとした。いくつかの実験では、腫瘍の成長をリアルタイムで検出するためにCD1a−B16細胞にルシフェラーゼを共に形質導入した。動物には第1日目にヒト抗CD1a抗体240μgを腹腔内に(i.p.)投与し、その後抗体80μgを更に3回、第3日目、第6日目及び第9日目に投与した。28日間にわたって動物の腫瘍の成長をモニターし、腫瘍の大きさが2cm3を超えた時点又は腫瘍の潰瘍ができた後に殺した。腫瘍の大きさは、蛍光像により(ルシフェラーゼを形質転換したB16細胞の場合)又は直接キャリパーによりモニターした。抗体治療を受けた動物における腫瘍の成長を、賦形剤である生理食塩水で治療した又はヒトIgG1コントロール抗体により治療したコントロールグループと比較することによりにより、抗体の抗腫瘍有効性を分析した。

BIACORE3000はリアルタイム表面プラズモン共鳴の技術に適用されるものであるが、上記のように調製した抗CD1a抗体及びマウス抗CD1aコントロール抗体についての親和性測定を実施するのに用いた。まず、リガンド、即ちCD1aタンパク質(Abnova社)を研究グレードCM54フローチャネル(Fc)センサーチップ上にアミノカップリングを用いてリガンドとして固定した。簡潔には、CM5チップ(BIACORE)のデキストラン基質をN−ヒドロキシサクシニミド(NHS)及びN−エチル−N‘−(ジメチルアミノプロピル)カルボジイミド(EDC)の混合物により活性化した。次に、CD1aタンパク質(好ましくは30μg/ml)を基質上の活性化カルボキシル基にカップリングさせた。カップリングした後、エタノールアミン−HCL(pH8.5)を加え、残っている活性化カルボキシル基を不活性化する。4つのチャネルのうち1つを活性化し、CD1aタンパク質のカップリングなしで不活性化してコントロールフローチャネルとした。この後、動態分析を行った。この目的のために、抗CD1a抗体の会合速度(Ka)、解離速度(Kd)及び親和性(KD)を調べた。0.01〜1000nMまでの一連の濃度に25℃を適用した(希釈因子2、希釈バッファーはHBS−EP)。4つのチャネルのうち1つを参照用フローセルとして用い、HBS−EPバッファーの注入をコントロール注入とした。各抗体50μlを20μl/分の一定の流速で注入した。注入の最後に、HBS−EPを750秒用いて、その後に製造者の取扱説明書に従い再生バッファー(例えばグリシン、pH2)のパルスによりCM5チップを再生する。この実験を3回繰り返し、分析のばらつきを測定する。次に、BIACORE評価ソフトを用いて一連の濃度における濃度の会合及び解離カーブを適合させる。これらの適合に基づいて、親和性を決定する。2価抗体の考え得る親和性効果を最小にするために、1)0.1−10×KDの濃度範囲、2)Ka及びKd率の低い標準誤差値、3)低い残余カイ2乗(Chi2)値、4)高い精度(好ましくは20%未満)、の4つの重要なパラメータを評価に含めた。抗CD1a抗体は0.1−50nMの範囲の高い親和性で優先的に結合する。

Claims (16)
- ヒトCD1aと特異的に結合可能なヒトモノクローナル抗体であって、配列番号8で表されるアミノ酸配列を備える重鎖可変領域および配列番号20で表されるアミノ酸配列を備える軽鎖可変領域を含むことを特徴とする、ヒトモノクローナル抗体。
- 抗体はIgG1であることを特徴とする請求項1に記載のヒトモノクローナル抗体。
- 抗体は、抗体依存性細胞障害、補体依存性細胞障害およびインターナリゼーション(内在化)からなる群より選ばれる少なくとも1つの活性を有することを特徴とする請求項1または2に記載のヒトモノクローナル抗体。
- 請求項1〜3のいずれか一項に記載のヒトモノクローナル抗体を含む免疫複合体であって、さらに少なくとも1種のタグを含むことを特徴とする免疫複合体。
- タグは、有毒物質、放射性物質、リポソーム、酵素およびその組合せからなる群より選ばれることを特徴とする請求項4に記載の免疫複合体。
- 請求項1〜3のいずれか一項に記載のヒトモノクローナル抗体をコード化することを特徴とする核酸分子。
- 請求項6に記載の少なくとも1種の核酸分子を含むことを特徴とするベクター。
- 請求項7に記載の少なくとも1種のベクターを含むことを特徴とする宿主細胞。
- 宿主細胞はヒト細胞であることを特徴とする請求項8に記載の宿主細胞。
- 請求項1〜3のいずれか一項に記載のヒトモノクローナル抗体を生産する方法であって、モノクローナル抗体の発現を促す条件下に請求項8または9に記載の宿主細胞を培養することを含むことを特徴とする方法。
- さらに、発現されたヒトモノクローナル抗体を回収することを含むことを特徴とする請求項10に記載の方法。
- 請求項1〜3のいずれか一項に記載のヒトモノクローナル抗体または請求項4または5に記載の免疫複合体を含むことを特徴とする組成物。
- 請求項1〜3のいずれか一項に記載のヒトモノクローナル抗体、請求項4または5に記載の免疫複合体または請求項11に記載の組成物を含む製薬上の組成物であって、さらに少なくとも1種の薬学的に許容可能な賦形剤を含むことを特徴とする製薬上の組成物。
- さらに、少なくとも1種の他の治療上の薬剤を含むことを特徴とする請求項13に記載の製薬上の組成物。
- 薬としての使用のための、請求項1〜3のいずれか一項に記載のヒトモノクローナル抗体、請求項4または5に記載の免疫複合体、請求項11に記載の組成物または請求項13または14に記載の製薬上の組成物。
- ヒトCD1aを検出する方法であって、以下のステップ
a.サンプルを請求項1〜3のいずれか一項に記載のヒトモノクローナル抗体、請求項4または5に記載の免疫複合体の診断上効果的な量で接触させること、および
b.ヒトモノクローナル抗体または免疫複合体がサンプルの化合物と特異的に結合するかどうか定めること
を含むことを特徴とする方法。
