本発明は、ヒトの不快な体臭の発生を抑えるデオドラント剤に関する。

【従来の技術及び発明が解決しようとする課題】
体臭は汗の臭いを始め、口臭、頭皮臭、足臭等の体全体に及ぶ。その中でも特に汗の臭いについては、「わきが」に代表される腋臭と体全体に及ぶ酸臭が混合して構成されている。特に腋臭は嫌悪感を催す臭いの代表として、近年ますますその防御が望まれている。
腋臭のもととなる汗が分泌されるアポクリン腺は、エックリン汗腺とは異なり腋窩や乳輪,陰部等に多く存在するが、それらの部位に局在するのものではなく、体幹部に広く存在しており(Pinkus H; Mehregan AH; Adnexal Nevi and Benign Adnexoid Tumors, in A Guide to Dermatohistopathlogy;2nd ed, pp528, pp29, by Appleton-Century-Crofts, New York、1976)、清潔志向が進む近年においては、斯かる腋臭を持続的に除去することが課題となっている。

アポクリン腺から分泌された汗はそれ自体では強い臭気を発することはないが、皮膚表面に存在する皮膚常在菌によって臭気物質に変化することが知られており(Leyden JJ, et al, J Invest Dermatol, 77: 413-416, 1981)、最近では、におい分子の一つである3−メチル−2−ヘキセン酸(3M2H)がキャリア蛋白質であるアポリポプロテインD(Apolipoprotein D)と結合した形で皮膚表面へ分泌されることも報告されている(Zeng C, et al, Proc Natl Acad Sci U S A, 93 : 6626-6630, 1996)。
また、におい分子の種類も、脂肪酸(Zeng XN, et al; Analysis of characteristic odors from human male axillae., J Chem Ecol, 17: 1469-1492, 1991)や男性ホルモンの誘導体(Labows JN. et al., Steroids, 34 :249-258, 1979、Gower DB, et al., J Steroid Biochem Mol Biol, 48:409-418, 1994)他、多岐わたることが報告されている。

一方、これまで体臭を防止するための主たる方法としては、体表の微生物を抑える技術(殺菌剤、抗菌剤等)、汗の分泌を収斂作用で抑える技術(金属酸化物などの収斂剤等)、発生した不快な体臭を消臭する技術及び香りによるマスキング技術等が報告されている。

しかしながら、これらの技術はいずれも腋臭に代表される不快な体臭を根本から絶つものではなく、その効果も充分ではない。また、抗菌技術によれば、不快臭の発生原因菌以外に皮膚常在菌をも殺菌することで皮膚の一次バリアー機能を低下させるおそれも示唆されている。

本発明は、安全性が高く、汗の臭いを始めとする、不快な体臭、特に腋臭の発生を根本的に抑制できるデオドラント剤を提供することを目的とする。

【課題を解決するための手段】
本発明者らは、アポクリン汗腺におけるキャリアタンパク質の一つとして知られているアポリポプロテインDの合成・分泌を抑制すれば、腋臭の原因となるにおい分子の皮膚表面への放出を抑制できると考え、当該タンパク質の合成・分泌を抑制する物質について検討したところ、特定の植物抽出物にアポリポプロテインDの分泌抑制作用があり、デオドラント剤として有用であることを見出した。

すなわち本発明は、オウバク、ゴバイシ、コメ、ログウッド、アルニカ、ウイキョウ、エンメイソウ、オウレン、サイシン、サンショウ、レンゲソウ、セイヨウキヅタ、ケイヒ、メリッサ、マロニエ、アセンヤク、コウホネ、ツボクサ及びワレモコウから選ばれる植物又はその抽出物からなるデオドラント剤及びアポリポプロテインD分泌抑制剤を提供するものである。

【発明の実施の形態】
本発明のデオドラント剤及びアポリポプロテインD分泌抑制剤として用いられる植物は、オウバク(黄柏 PHELLODENDRI CORTEX;キハダ Phellodendron amurens Ruprecht)、ゴバイシ(五倍子 GALLAE RHOIS;ヌルデ Rhus javanica L, Rhus chinensis Miller)、コメ(イネ Oryza sativa L.の種子)、ログウッド(魯格烏特;Haematoxylon campechianum L.)、アルニカ(アルニカArnica montana L.)、ウイキョウ(茴香 フェンネル FOENICULI FRUCTUS;ウイキョウ Foeniculum vulgare Miller)、エンメイソウ(延命草ISODONIS HERBA;ヒキオコシ Isodon japonicus Hara)、オウレン(黄連 COPTIDIS RHIZOMA;オウレン Coptis japonica Makino)、サイシン(細辛 ASIASARI RADIX;ウスバサイシン Asiasarum sieboldii F. Maekawa又はケイリンサイシンAsiasarum heterotropoides F. Maekawa var. mandshuricum F. 当該植物又はその抽出物は、本発明のデオドラント剤及びアポリポプロテインD分泌抑制剤としてそのまま用いることもできるが、当該抽出物を希釈調製して又は濃縮若しくは凍結乾燥した後、粉末又はペースト状に調製して用いることもできる。
斯かる植物又はその抽出物は、後記実施例に示すように、におい分子のキャリア蛋白質であるアポリポプロテインDの分泌量を減少させ、腋臭抑制効果を発揮する。従って、これらはにおいの原因物質の発生を根本的に抑制できるデオドランド剤として有用である。

本発明のデオドランド剤及びアポリポプロテインD分泌抑制剤は、化粧料、外用医薬品又は医薬部外品等の製剤、例えばクリーム、乳液、ローション、パウダー、スプレー、スティック、シート又は湿布等の貼付剤として用いることができ、またいくつかの使用方法を併用することも可能である。

化粧料、外用医薬品又は医薬部外品として用いる場合のデオドランド剤又はアポリポプロテインD分泌抑制剤の含有量は、一般的に0.1〜20重量%とするのが好ましく、特に0.5〜10重量%とするのが好ましい。また、植物抽出物としての含有量は、一般的に固形分換算で0.00001〜10重量%とするのが好ましく、特に0.0005〜5重量%とするのが好ましい。

本発明のデオドランド剤及びアポリポプロテインD分泌抑制剤は、足、腋、頭部、陰部等不快臭の発生しやすい箇所に適用することにより、体臭の発生を制御することができる。斯かる場合の製剤の使用量は、有効成分の含有量により異なるが例えば液状製剤の場合皮膚面1cm2当たり1〜20mg、固形状の製剤の場合、同じく1〜50mgとするのが好ましい。

【実施例】
製造例1 オウバク抽出物の調製
オウバク(キハダ Phellodendron amurense Ruprechtのコルク層を除いた樹皮)40gに50v/v%エタノール水溶液400mLを加え、室温で7日間抽出後、濾過して抽出液を得た(収量:350mL、蒸発残分:0.87w/v%)。

実施例1 アポリポプロテインD分泌抑制作用
(1)細胞培養およびサンプル添加
細胞培養は乳がん由来の細胞株T−47D(ATCC No.HTB-133)を用い、10%FCS添加DMEM培地(LIFE TECHNOLOGIES社 Cat.11995-065)にて、37℃、5%−CO2、加湿で継体培養を行った。
6wellプレートに40×104cell/wellとなるように接種し、10%FCS(チャコール処理)添加DMEM培地で2日培養を行った。培地を除去し、PBS(LIFE TECHNOLOGIES社 Cat.14190-144)で二回洗浄の後、レチノイン酸10-6Mと各種エキス0.1%を添加したDMEM培地をいれる。二日後に、培地交換しさらに二日後培地を回収し、ウェスタンブロッティングを行った。

(2)ウェスタンブロッティング
前処理:回収した培地から遠心により浮遊する細胞を除去し、400μLをマイクロコン10で限外ろ過濃縮しその後、電気泳動のサンプルバッファー(2mL 10%SDS, 2mL 2-mercapoethanol, 2.5mL 1M Tris pH 6.8, 3.5mL 70%sucrose)10μLを添加し、95℃で5分間加熱処理後、2000rpm,3minの遠心で回収した。
SDS−PAGE:Tris/Glycine/SDSバッファー(BIO-RAD社製、Cat.161-0772)、10%SDS−ポリアクリルアミドゲル(BIO-RAD社製、Cat.161-J331)を用いて、前処理を施した培地濃縮液を1ウェルにつき、マイクロコン10(MILLIPORE社製、Cat.42407)1本分(培地400μL相当)ずつアプライし、10〜20mAの定電流で電気泳動を行った。
セミドライブロッティング:ブロッティングバッファーは0.01%SDS・20%メタノール含有Tris/Glycineバッファー(BIO-RAD社製、Cat.161-0734)。ゲル1枚につきブロッティング用濾紙(BIO-RAD社製、Cat.170-3932)2枚をブロッティングバッファーに浸し、メタノール化したPVDFメンブレンと電気泳動したゲルもブロッティングバッファーも浸した。Trans-Blot SD(BIO-RAD社製、Cat.170-3957)に下から、濾紙2枚、表を下にしたPVDFメンブレン、表を下にしたゲル、濾紙2枚の順にセットし、20V、60分通電した。
ブロッキング:ブロックエース(雪印社製、UK-B25)に、通電後PBSで洗浄したメンブレンを浸漬した。(約20時間冷蔵保存)
1次抗体:anti-HUMAN Apolipoprotein D(RDI社製、Cat.RDI-APODabm)をブロックエース中に300倍希釈になるよう加え、室温で1時間振盪した。
2次抗体:メンブレンを1×TBST(DAKO社製、Cat.S3306を希釈)で2回リンスし、更に15分1回、5分2回振盪しながら洗浄した。Anti-mouse IgG F(ab')2(アマシャム ファルマシア社製, NA931)をブロックエースで30000倍に希釈して用い、メンブレンを浸漬し1時間振盪した。
検出:メンブレンを1×TBSTで2回リンスし、更に15分1回、5分2回振盪しながら洗浄した。SuperSignal West Femto Maximum Sensitivity Substrate(Pierce社製, Cat.34095)中に5分浸漬した。その後オートラジオグラフィーフィルムに露光してアポリポプロテインDを検出した。フィルムをスキャナー(EPSON社製、GT-9500WIN)でコンピューターに読み込んで、Lane&Spot Analyzer(ATTO社製)でバンドの濃さを数値化し、エキス無添加サンプルに対するアポリポプロテインD量の比を算出した。結果を表2に示す。

実施例2 体臭抑制試験
体臭が強く、かつ腋臭を有している被験者20名の片方の腋にプラセボサンプル(表3)を、もう片方の腋にオウバク抽出物サンプル(表3)を4週間、1日2回塗布した。体臭の評価は専門のパネラー2名が、使用前と使用後の腋窩部分の体臭を、下記の5段階で評価した。結果を表4に示す。
