Description
本願は、2003年4月7日に出願された英国特許出願No.0 307 999.3に優先権主張し、この出願の全内容が、引用することによって本明細書に組み入れられる。
微小流体系の使用は現在、分析化学、薬剤探求、診断法、コンビナトリアル合成及びバイオテクノロジーを含む種々の分野で十分に確立されている。そのような系はまた、コンビナトリアルライブラリーの合成又はスクリーニング、ポストゲノムの特性決定等における場合であり得るように、試料体積が低くあり得る重要な用途を有する。
導管構造における状態又は導管構造の状態に依存する、少なくとも1種の生成物の所定の特性を表すセンサー信号を生じるように適合されたセンサー、
導管構造における状態又は導管構造の状態を変えるように適合された手段、及び
センサー信号に依存して、手段に、導管構造における状態又は導管構造の状態を変えさせるように適合されたコンピュータ
を有する系が提供される。
(i) 共通流動導管11における反応条件の最適化、例えば収率の最適化もしくは特定の結果の生成、又は
(ii) 所定の特性がセンサー27により検知されるか、又は所定の値を有するように検知される、共通流動導管11における反応生成物
を生じるための操作又は生成するための試みをさせる。
この例では、系20は、反応の化学特性、例えば試薬A及びBの反応における特定の反応生成物の収率、時間分解反応もしくは生成物領域19の相対的な量又は特定の異性体の量を最適化するために使用される。
この例では、系20は、チップ1によって種々の異なる試薬の組合せをランし、反応生成物を1種以上のバイオセンサーに通して製薬用途、例えば薬剤発見における可能な使用のための試験をするために使用される。言い換えれば、系20は、反応生成物の高処理量スクリーニング(HTS)のために使用される。ここで、閉ループ制御は、製薬用途のさらなる研究のために、試薬ライブラリー23から反応生成物を見出すように作動する。
この例においては、系20のチップ1は、図3に概略的に示された導管構造103を有する。導管構造103は、試薬Aのための入口枝導管107及び試薬B(この場合には、入口枝導管107から外れている(下流である))のための入口枝導管109を有する。導管構造103はさらに、別の試薬Cのための1個以上の追加の入口枝導管125を有する。各入口枝導管107、109、125は、独立して動作可能なように、弁130を有する。再び、層流動流が、共通流動導管111中に生成される。
Claims (7)
- 流体が相互作用して少なくとも1種の生成物を生じることができる微小流体導管構造と、前記導管構造内の状態を自律的に制御するための自動化された閉ループ制御メカニズムとを有し、制御メカニズムが:
前記導管構造内の状態に依存する、少なくとも1種の生成物の所定の特性を表すセンサー信号を生じるように適合されたセンサーと;
分類された試薬のアレイから導管構造へ試薬を移動させるための移動メカニズムと;
前記センサー信号を受取り、前記センサー信号に依存して、前記移動メカニズムに、前記導管構造内における試薬の組み合わせを変化させるように適合されたコンピュータとを有し、
前記コンピュータは所定の特性に関連する所定の目標がプログラムされ、前記コンピュータはセンサー信号を前記所定の目標と比較し、当該比較に基づいて、前記所定の目標に到達するように、前記移動メカニズムに前記試薬の組み合わせを変化させるように適合され、
前記コンピュータは前記試薬のアレイの分類と、前記所定の目標に到達するため前記試薬の組み合わせのヒューリスティック変化をもたらす反復アルゴリズムとがプログラムされた系。 - 前記センサーが、少なくとも1種の生成物の所定の化学的特性を表すセンサー信号を生じるように適合される請求項1記載の系。
- 前記センサーが、少なくとも1種の生成物の所定の生物学的特性を表すセンサー信号を生じるように適合される請求項1記載の系。
- 前記所定の目標が同一性、純度、転化率、異性体比、収率、不純物プロファイル又は色に関する請求項2記載の系。
- 前記所定の目標が活性、効能、選択性又は持続時間に関連する請求項3記載の系。
- 統合された、コンピュータ制御された系である請求項1記載の系。
- コンピュータが、試薬の組合せが定の目標を満たす所定の生物学的特性を有する生成物を生じるまで、微小流体導管構造を通って異なる組合せの試薬を自律的に走らせるように閉ループ制御メカニズムをヒューリスティックに操作する工程とを含む、
請求項1に記載された系にて実現される化合物のスクリーニング方法。
