Claims (5)
- 循環マクロファージの特徴を決定し、かつ/または分類する方法であって、
・採取した全血試料を、勾配遠心分離を行ってマクロファージを単離する工程、
・該マクロファージ細胞を穿孔処理する工程、
・該細胞を、前立腺特異抗原(PSA)、サイトケラチンおよび/または上皮膜抗原と、抗原抗体反応する少なくとも1つの選択された抗体で、細胞内染色する工程、および
・該前処理済み細胞のフローサイトメトリー分析を行い、続いて複数の細胞の統計学的評価を行う工程、を含む方法。 - フローサイトメトリーを実施した後にアイソタイプコントロールと染色のヒストグラム分析を行うことを特徴とする、請求項1の方法。
- 散在性前立腺腫瘍の貧食によって取り込まれた組織細胞の一部を人体外で検出するための請求項1または2に記載の方法。
- 貧食によって取り込まれた前記物質が前立腺関連であるかどうかを、前記マクロファージにおける前立腺特異抗原(PSA)の前記染色によって決定することを特徴とする、請求項3の方法。
- 請求項1に記載の循環マクロファージの特徴を決定し、かつ/または分類する方法を実施するための分析装置であって、
採取した血液をヘパリン処理するための手段、
マクロファージを単離するための勾配遠心分離器、
該マクロファージ細胞を穿孔処理する細胞穿孔手段、
前記前処理済み細胞を前立腺特異抗原(PSA)、サイトケラチンおよび/または上皮膜抗原と、抗原抗体反応する少なくとも1つの選択された抗体で、細胞内染色するための機器、および
腫瘍の初期診断を目的として単離および前処理済み細胞の細胞内構造を決定するためのコンピュータ支援評価ユニットを装備したフローサイトメーターを備える分析装置。
