Claims (13)
- 標的物質と検出用物質との物質間の相互作用を検出するための表面であって、
該表面に末端部位が固定化された一本鎖DNAと該一本鎖DNAと相補的な塩基配列を備える一本鎖DNAによって形成され、前記標的物質と前記検出用物質との複合体が結合可能な所定塩基配列部位を有する二本鎖DNAと、
前記二本鎖DNAの相補的塩基対部位に結合する、前記標的物質と同一の物質とインターカレーターとからなる結合体と、
から構成された検出表面。 - 前記標的物質はホルモンであることを特徴とする請求項1記載の検出表面。
- 前記標的物質はホルモンであり、前記検出用物質はホルモンレセプターであり、前記所定配列部位はホルモン応答配列部位であることを特徴とする請求項2に記載の検出表面。
- 前記標的物質はホルモンであり、前記検出用物質はリガンド依存性のホルモンレセプターであり、前記所定配列部位はホルモン応答配列部位であることを特徴とする請求項2または3に記載の検出表面。
- 前記ホルモンは、ステロイドホルモンである請求項2から4のいずれか一項に記載の検出表面。
- 前記ステロイドホルモンは、グルココルチコイドである請求項5記載の検出表面。
- 請求項1から6のいずれか一項に記載の検出表面を少なくとも備えることを特徴とするセンサチップ。
- 請求項1から6のいずれか一項に記載の検出表面と、
該検出表面における前記二本鎖DNAから一本鎖DNAへの解離の発生を検出可能な検出手段と、
を少なくとも用いることを特徴とするセンサ装置。 - 二本鎖DNAを検出表面に固定化する手順と、
前記二本鎖DNAの相補塩基対部位に、検出対象であるホルモンと同一のホルモンとインターカレーターとからなる結合体を結合させる手順と、
検出対象であるホルモンとホルモンレセプターとの複合体と、所定の転写因子とが、前記二本鎖のホルモン応答配列部位に協同作用することにより、前記二本鎖DNAを一本鎖DNAに解離させる手順と、
前記解離の発生を検出する手順と、
を含むホルモンとホルモンレセプターとの結合を検出する方法。 - 前記二本鎖DNAが一本鎖DNAへ解離する際に、前記結合体が前記検出表面上の媒質中へ放出される手順と、
放出された前記結合体を構成する前記ホルモンが、前記ホルモンレセプターとの結合を介して、前記解離を促進する手順と、
を含むことを特徴とする請求項9記載の方法。 - 前記検出を表面プラズモン共鳴(Surface Plasmon Resonance)原理に基づいて行うことを特徴とする請求項9または10に記載の方法。
- 前記二本鎖DNAの固定化されている側のDNA鎖の末端部位に標識した誘電体と表面との間の距離が、前記二本鎖DNAが一本鎖DNAへ解離する際に変化することに起因する誘電率の変化を、前記表面プラズモン共鳴原理によって検出することを特徴とする請求項9から11のいずれか一項に記載の方法。
- 前記検出を、次の(1)、(2)のいずれかを光学的に測定する手順に基づいて行うことを特徴とする請求項9から12のいずれか一項に記載の方法。
(1)前記二本鎖DNAに標識された蛍光物質の蛍光強度の変化。
(2)前記二本鎖DNAに挿入された蛍光インターカレーターの発色の変化。
