Description
本発明は、分泌物モニター物品、並びに専門家又は訓練されていない使用者の両方に、高精度で細菌性膣症に関連する分泌物のような分泌された生物学的流体をモニターさせる前記物品を用いるための方法を提供する。本発明は、pHポリマーマトリックス、並びにその混合物を支持体に結合させる方法をさらに教示する。好ましい態様において、本物品は、カバー又は親水性被覆物により、ポリマーマトリックス指示薬と膣組織とを分離させる必要なく、膣に直接用い得る含浸された綿棒又はストリップの形態である。
製造方法
1)ポリマーマトリックス指示薬
段階1:アセトン中に酢酸セルロースを24%乃至28%溶解する。
段階2:その溶液にフタル酸ジオクチルを4%乃至22%添加する。
段階3:その溶液に塩化トリドデシルメチルアンモニウムを1%乃至5%添加する。
段階4:その溶液に2−エトキシエタノールを24%乃至28%添加する。
段階5:その溶液に二度蒸留した水24%乃至28%中に溶解したニトラジンイエロー溶液を0.2%乃至0.5%添加する。
段階6:その溶液を綿棒又はストリップに含浸させる。
7つの異なるpH値を有する100mMの燐酸塩クエン酸塩緩衝液を調製した。各緩衝液を4つの異なる濃度、50mM、20mM、10mM及び5mMに希釈し、それらのpHを1MのNaOH又は1MのHClを用いて調整した。本発明のニトラジンイエローポリマーマトリックスを各緩衝液中に浸し、色の変化を書きとめ、0=黄色、1=淡緑色、2=緑、3=暗緑色のような数値により表した。表2及び図3にまとめられた結果により示されるように、溶液が50mM又は100mMのイオン濃度を有する場合、pHが5より高い場合に、ニトラジンイエローpH指示薬ポリマーマトリックスは、その色が黄色から緑/青に変わった。イオン濃度が20mMより低い場合、4.3乃至5.0のpH動的範囲において色の変化が生じる。
膣分泌物の緩衝能力を検査するためには、膣分泌物に対する異なる緩衝溶液を0.1NのNaOHで滴定した。
物品は、埋め込まれた指示薬ストリップを有するシール又はパンティーライナーであることができる。そのストリップは、水溶液中、6.6のpKaを有するpH指示薬ニトラジンイエローを含有する。
段階1:5mlのアセトンに、150mgの酢酸セルロース、107μlのジブチルフタレート、23μlのアリコート、150μlの2−エトキシエタノール、及び150μlのDDW中に溶解した2.4mgのニトラジンイエローを添加する。
段階2:その混合物を数分間攪拌し、完全に溶解させる。
段階3:ポリエステルモノフィラメント網織物にそのポリマー溶液を塗布し、所望の生成物を与える。
特定の患者における尿路感染症の再発では、その患者が新たな尿路感染症に罹っているか否かについて迅速にかつ容易に診断する必要がある。現在、患者が尿路感染症に罹っているかを決定するために、患者は、予約をし、医師を訪ねなくてはならない。さらに、その患者が尿路感染症の再発を起こしやすい場合、その感染症が再発していないことを確かめるために医院に定期的に行かなくてはならない。使用者が再び尿路感染症に罹っているかを決定することができる器具があるなら、医院を訪れることによるストレスと消費する時間を最少にし、その感染のより迅速な診断がなされ、患者が受ける苦痛を低減させ、感染症のより時宣を得た治療をさせる。
1.化学的環境
a.流体のpHレベル−pKaよりも高いpHレベルは安定な色変化を与える。
b.溶液の緩衝能力−
c.溶液中のアンモニウム塩含量
2.生物学的環境
a.尿中のたんぱく質の存在は、安定な色変化を与え、その反応は可逆性ではない。
Claims (27)
- (イ)ヒトから分泌された生物学的流体を吸収するための支持体、並びに
(ロ)少なくとも一つのpH決定試薬及びイオンバランス試薬を含む、ある色を有するpH指示薬部分、
を含む、ヒトから分泌された生物学的流体を特定するための物品であって、
前記pH指示薬部分が、尿とイオンバランス試薬とでは異なった反応をし、それによって、尿に起因する偽陽性を乾燥により区別する、物品。 - pH指示薬部分がストリップの形態であり、前記ストリップを支持体から取り外すことができ、乾燥することができる、請求項1に記載の物品。
- 羊水の漏出を検出するための請求項1又2に記載の物品。
- 羊水によるpH指示薬部分の色の変化は乾燥によっても安定であるが、尿による色の変化は不安定であり、乾燥により可逆性である請求項3の物品。
- (イ)ヒトから分泌された生物学的流体を吸収するための支持体、並びに
(ロ)少なくとも一つのpH決定試薬及び疎水性材料を含む、ある色を有するpH指示薬部分、
を含む、正常な緩衝能力を有する生物学的流体と、前記の正常な緩衝能力と比べて低い緩衝能力を有する生物学的流体とを識別するための物品であって、
前記pH指示薬部分が、低い緩衝能力を有する生物学的流体と、正常な緩衝能力を有する生物学的流体とでは異なった反応を示し、これによって、低い緩衝能力を有する生物学的流体と、正常な緩衝能力を有する生物学的流体とを識別できる物品。 - 膣分泌物の識別に適する、請求項5に記載の物品。
- 細菌性膣症、トリコモナス、寄生虫感染症、真菌感染症又は酵母菌感染症に関連する膣分泌物の識別に適する、請求項6に記載の物品。
- 細菌性膣症、トリコモナス、寄生虫感染症、真菌感染症又は酵母菌感染症に関連する低い緩衝能力を有する膣分泌物によってpH指示薬部分の色は変化し、正常な緩衝能力を有する膣分泌物によってはpH指示薬部分の色は変化しない、請求項7に記載の物品。
- pH指示薬部分がさらにイオンバランス試薬を含む、請求項5−8のいずれか1項に記載の物品。
- pH指示薬部分がストリップの形態であり、前記ストリップを支持体から取り外すことができ、乾燥することができる、請求項9に記載の物品。
- 低い緩衝能力を有する膣分泌物によるpH指示薬部分の色の変化は乾燥によっても安定であるが、尿による色の変化は不安定であり、これにより、尿に起因する偽陽性を乾燥により区別できる請求項9または10に記載の物品。
- pH指示薬部分が、セルロース、ニトロセルロース、酢酸セルロース、エチルセルロース及びこれらの組み合わせから成る群から選択される疎水性ポリマーを含む、請求項1−11のいずれか一項に記載の物品。
- pH決定試薬が陰電荷官能基を有する、請求項1−12のいずれか一項に記載の物品。
- pH決定試薬が、ニトラジンイエロー、チモールブルー、ブロモチモールブルー、キシレノールブルー、ブロモキシレノールブルー、フェノールレッド、m−クレゾールパープル、クロロフェノールレッド、ブロモクレゾールパープル、アリザリン、ニュートラルレッド及びクレゾールレッドから成る群から選択される少なくとも1つである、請求項13に記載の物品。
- pH決定試薬がニトラジンイエローである、請求項14に記載の物品。
- イオンバランス試薬が塩化トリドデシルメチルアンモニウム、塩化トリカプリルメチルアンモニウム及び塩化セチルトリメチルアンモニウムから成る群から選択される少なくとも1つである、請求項1−4及び9−15のいずれか一項に記載の物品。
- 物品が、綿棒、ガーゼ、パンティーシールド、衛生ナプキン又はオムツの形態である、請求項1−16のいずれか一項に記載の物品。
- pH指示薬部分が、セルロース、ニトロセルロース、酢酸セルロース及びエチルセルロースから選択される少なくとも1つの疎水性ポリマー、pH決定試薬、イオンバランス試薬、可塑剤及び湿潤剤及び溶媒を含む組成物を用いて、支持体とは独立して形成されたポリマーマトリックスである、請求項1−17のいずれか一項に記載の物品。
- pH指示薬部分が、セルロース、ニトロセルロース、酢酸セルロース及びエチルセルロースから選択される少なくとも1つの疎水性ポリマー、pH決定試薬、イオンバランス試薬、可塑剤及び湿潤剤及び溶媒を含む組成物を用いて、支持体と一体的に形成されたポリマーマトリックスである、請求項1及び5−9のいずれか一項に記載の物品。
- 溶媒がポリマーを溶解する量である、請求項18または19に記載の物品。
- 溶媒が、アセトン、酢酸エチル、ジエチルエーテル、イソプロピルエーテル、t−ブチルメチルエーテル及びテトラヒドロフランから選択される少なくとも1つである、請求項20に記載の物品。
- ポリマーマトリックスにおいて、ポリマーが10乃至40%の量であり、pH決定試薬が0.05乃至5%の量であり、イオンバランス試薬が0.1乃至10%の量であり、可塑剤が1乃至30%の量であり、湿潤剤が10乃至40%の量であり、パーセントが、この混合物の総重量に基づいており、その混合物の総重量が100%である、請求項18−21のいずれか一項に記載の物品。
- ポリマーが24乃至28%の量であり、pH決定試薬が0.2乃至0.5%の量であり、イオンバランス試薬が1乃至5%の量であり、可塑剤が4乃至22%の量であり、湿潤剤が24乃至28%の量である、請求項22に記載の物品。
- 可塑剤が、ビス−(2−エチルヘキシル)セバケート、フタル酸ジエチル、フタル酸ジオクチル、ビス−(2−ブトキシエチル)アジペート及びフタル酸ジブチルから成る群から選択される少なくとも1つである、請求項18−23のいずれか一項に記載の物品。
- 湿潤剤が、トリエチレングリコール、エチレングリコール及び2−エトキシエタノールから成る群から選択される少なくとも1つである、請求項18−24のいずれか一項に記載の物品。
- (イ)ヒトから分泌された羊水及び尿を吸収することができる支持体、並びに
(ロ)ニトラジンイエロー及び塩化トリカプリルメチルアンモニウムを含む黄色のpH指示薬部分、
を含む、羊水漏出を検出するための物品であって、前記pH指示薬部分は支持体から取り除いて乾燥することができ、
pH指示薬部分は羊水又は尿により変色し、但し、羊水による変色は安定であるが、尿による変色は不安定であるので、尿に起因する偽陽性をpH指示薬部分の乾燥により区別することができる、羊水漏出を検出するための物品。 - (イ)膣分泌物及び尿を吸収することができる支持体、並びに
(ロ)ニトラジンイエロー、酢酸セルロース及び塩化トリカプリルメチルアンモニウムを含む黄色のpH指示薬部分、
を含む、膣感染症の罹患を検出するためのモニター物品であって、前記pH指示薬部分は支持体から取り除いて乾燥することができ、
pH指示薬部分は正常な膣分泌物により変色せず、しかし、膣感染症に罹患した膣分泌物または尿により変色し、
ただし、膣感染症に罹患した膣分泌物による変色は安定であるが、尿による変色は不安定であるので、尿に起因する偽陽性をpH指示薬部分の乾燥により区別することができる、膣感染症の罹患を検出するためのモニター物品。
